Electrogenerated chemiluminescence of solutions, films, and nanoparticles of dithienylbenzothiadiazole-based donor-acceptor-donor red fluorophore. Fluorescence quenching study of organic nanoparticles.

We report here the electrochemistry, spectroscopy, and electrogenerated chemiluminescence (ECL) from a solution, film, and nanoparticles (NPs) of a red-emitting dithienylbenzothiadiazole molecular fluorophore [4,7-bis(4-(n-hexyl)-5-(3,5-di(1-naphthyl)phenyl)thiophen-2-yl)-2,1,3-benzothiadiazole, 1a], which has a donor-acceptor-donor configuration. In addition, the quenching of the fluorescence of the organic NPs by KI was investigated. The 1a film and NPs exhibit two absorbance peaks at 350 and ~504 nm that are red-shifted compared to those of 1a dissolved in solution (340 and 486 nm). Fluores…

Nanoparticle contrast-enhanced computed tomography of sporadic aortic aneurysm and dissection: Effect of nanoparticle size and contrast agent dose.

Background: Aortopathies, such as aortic aneurysm and dissection (AAD), are associated with enhanced aortic wall permeability and endothelial dysfunction. We previously demonstrated that nanoparticle contrast-enhanced computed tomography (nCECT), which detects enhanced aortic wall permeability, could enable non-invasive detection of early AAD before its progresses to fatal aortic rupture. This study investigated the effect of nanoparticle contrast agent (NPCA) size and dose on detection of aortopathy by nCECT. Methods: In vivo studies were performed in a mouse model of sporadic AAD induced by

Lipid Nanoparticles and PEG: Time Frame of Immune Checkpoint Blockade Can Be Controlled by Adjusting the Rate of Cellular Uptake of Nanoparticles.

The engineerability of lipid nanoparticles (LNPs) and their ability to deliver nucleic acids make LNPs attractive tools for cancer immunotherapy. LNP-based gene delivery can be employed for various approaches in cancer immunotherapy, including encoding tumor-associated antigens and silencing of negative immune checkpoint proteins. For example, LNPs carrying small interfering RNAs can offer several advantages, including sustained and durable inhibition of an immune checkpoint protein. Due to their tunable design, modifying the lipid composition of LNPs can regulate the rate of their uptake by i…

PLGA/TPGS nanoparticles for docetaxel delivery: The pegylation effect on nanoparticle physicochemical properties and uptake and cytotoxicity in prostate cancer cells.

Prostate cancer is the most common malignancy in men worldwide and docetaxel (DTX) is the treatment of choice. However, both the drug and formulation excipients for drug solubilization can cause side effects. In this context, the development of polymeric nanoparticles offers advantages to improve drug delivery and reduce toxicity. In the present work, factorial design was used to evaluate the effect of the amount of poly(L-lactide-co-glycolide) (PLGA) or poly(L-lactide-co-glycolide acid-polyethylene glycol) (PLGA-PEG), D-Alpha-Tocopheryl Polyethylene Glycol Succinate (TPGS) and ratio between a…

Nanoparticle-induced inflammation and fibrosis in ex vivo murine precision-cut liver slices and effects of nanoparticle exposure conditions.

Chronic exposure and accumulation of persistent nanomaterials by cells have led to safety concerns on potential long-term effects induced by nanoparticles, including chronic inflammation and fibrosis. With this in mind, we used murine precision-cut liver tissue slices to test potential induction of inflammation and onset of fibrosis upon 72 h exposure to different nanomaterials (0-200 µg/ml). Tissue slices were chosen as an advanced ex vivo 3D model to better resemble the complexity of the in vivo tissue environment, with a focus on the liver where most nanomaterials accumulate. Effects on the…

Differences in Engineered Nanoparticle Surface Physicochemistry Revealed by Investigation of Changes in Copper Bioavailability During Sorption to Nanoparticles in the Aqueous Phase.

Sorption of chemical substances to nanoparticles (NPs) in the aqueous phase strongly influences NP physicochemisty, and investigations of these complex interactions can provide important insights into the environmental fate of NPs. The objective of the present study was to use differences in copper (Cu) bioavailability to investigate aqueous-phase sorption with NPs that had different physicochemical characteristics (silicon [Si], perovskite, and titanium dioxide NPs [TiO2 NPs]). Sorption of Cu with NPs was assessed by the presence of adsorbent in water and onto the NP surface after ultracentri…

Unravelling the Mechanistic Understanding of Metal Nanoparticle-Induced Reactive Oxygen Species Formation: Insights from a Cu Nanoparticle Study.

Humans can be exposed to engineered and nonintentionally formed metal and metal oxide nanoparticles (Me NPs) in occupational settings, in public transportation areas, or by means of contact with different consumer products. A critical factor in the toxic potency of Me NPs is their ability to induce oxidative stress. It is thus essential to assess the potential reactive oxygen species (ROS) formation properties of Me NPs. A common way to assess the relative extent of ROS formation in vitro is to use fluorescence spectroscopy with the DCFH-DA (2',7'-dichlorofluorescein diacetate) probe, with and…

Peptide ligands on the PEGylated nanoparticle surface and human serum composition are key factors for the interaction between immune cells and nanoparticles.

The architecture of a nanoparticles' surface formed due to a modification with a ligand and protein corona formation in biofluids is critical for interactions with cells in vivo. Here we studied interactions of immune cells with magnetic nanoparticles (MNPs) covalently modified with polyethylene glycol (PEG) and their counterparts conjugated with peptides: a pH (low) insertion peptide (pHLIP) and cycloRGD as a targeting ligand in human serum. The conjugation of MNPs-PEG with pHLIP, but not with cycloRGD, enhanced the association of these particles with mononuclear phagocytic cells in vitro and…

Fabrication of Glass-Insulated Ultramicrometer to Submicrometer Carbon Fiber Electrodes to Support a Single Nanoparticle and Nanoparticle Ensembles in Electrocatalytic Investigations.

We report a procedure to fabricate glass-insulated carbon fiber ultramicroelectrodes (CF UMEs) with sizes of 10 and 1 μm in diameter. CF UMEs of theses sizes are commonly insulated with polymer or epoxy resins, which typically have pinholes and are less stable and difficult to polish. Through judicious choice of polishing materials, the fabrication procedure reported here leads to CF UMEs with a surface geometry that, within experimental error, is close to that of an inlaid disk. We demonstrate how the hardness factor of commonly used polishing materials may alter the geometry of CF UMEs, in w

Nanoparticle strategies for cancer therapeutics: Nucleic acids, polyamines, bovine serum amine oxidase and iron oxide nanoparticles (Review).

Nanotechnology for cancer gene therapy is an emerging field. Nucleic acids, polyamine analogues and cytotoxic products of polyamine oxidation, generated in situ by an enzyme-catalyzed reaction, can be developed for nanotechnology-based cancer therapeutics with reduced systemic toxicity and improved therapeutic efficacy. Nucleic acid-based gene therapy approaches depend on the compaction of DNA/RNA to nanoparticles and polyamine analogues are excellent agents for the condensation of nucleic acids to nanoparticles. Polyamines and amine oxidases are found in higher levels in tumours compared to t…

The Development of Silver Nanoparticles-Based Colorimetric Uric Acid Detection: An Extension Study of Silver Nanoparticles Extraction from Water Hyacinth (Eichhornia crassipes).

This work is an extension of our previous work on UV-assisted green synthesis of silver nanoparticles (AgNPs) from water hyacinth leaf extract, which is focused on using their unique biogenic capping layer to mitigate matrix interference in clinical diagnostics. The synthesized AgNPs were evaluated for uric acid (UA) detection and used to fabricate a plasmonic colorimetric biosensing platform. The results show that the biosynthesized AgNPs effectively act as optical signal transducers in an uricase-based enzymatic system, in which the natural capping shield provides excellent electrosteric pro…

Quantitative Detection of Biological Nanoparticles Using Twilight Off-Axis Holographic Microscopy: Insights on Complex Formation between PEGylated Gold Nanoparticles and Lipid Vesicles.

The detection of biological nanoparticles (NPs), such as viruses and extracellular vesicles (EVs), plays a critical role in medical diagnostics. However, these particles are optically faint, making microscopic detection in complex solutions challenging. Recent advancements have demonstrated that distinguishing between metallic and dielectric signals with twilight off-axis holographic microscopy makes it possible to differentiate between metal and biological NPs and to quantify complexes formed from metal and biological NPs binding together. Here, this method is employed to investigate complex …
