The Effect of HRE-Regulated VEGF Expression and Transfection on Neural Stem Cells in Rats.

In this study, we analyzed neural stem cells transfected with the HRE-VEGF gene in groups experiencing different periods of hypoxia. The results of RT-PCR showed that the expression of vascular endothelial growth factor (VEGF) mRNA gradually increased with the prolonged period of hypoxia (p < 0.05). The results from the western-blot test showed that expression of the VEGF protein increased with as the period of hypoxia increased (p < 0.05). The results of MTT combined with Elisa reagent showed that with the prolonged period of hypoxia, the secretion of VEGF protein increased, and that the prol

Deep learning-based classification of multi-categorical Alzheimer's disease data.

It is urgent to find the appropriate technology for the early detection of Alzheimer's disease (AD) due to the unknown AD etiopathologies that bring about serious social problems. Early detection of mild cognitive impairment (MCI) has pivotal importance in delaying or preventing the AD onset. Herein, we utilize deep learning (DL) techniques for the purpose of multiclass classification between normal control, MCI, and AD subjects. We used multi-categorical data from the Alzheimer's Disease Neuroimaging Initiative (ADNI) including brain imaging measurements, cognitive test results, cerebrospinal

A historical, systematic review of handedness origins.

Handedness ontogenesis is still under debate in science. This systematic review analyzed articles regarding the theories and basis of handedness formation, highlighting the historical knowledge path that this literature underwent. Cochrane Library, LILACS, Web of Sciences, Science Direct and PubMed databases were searched. This review included review studies with handedness as the main topic. Only papers written in English with analyses exclusively in neurotypical humans (any age range) were included. Different approaches (genetic, neural, social, and behavioural) were reviewed in light of gro

Distinct effects of prefrontal and parietal cortex inactivations on an accumulation of evidence task in the rat.

Numerous brain regions have been shown to have neural correlates of gradually accumulating evidence for decision-making, but the causal roles of these regions in decisions driven by accumulation of evidence have yet to be determined. Here, in rats performing an auditory evidence accumulation task, we inactivated the frontal orienting fields (FOF) and posterior parietal cortex (PPC), two rat cortical regions that have neural correlates of accumulating evidence and that have been proposed as central to decision-making. We used a detailed model of the decision process to analyze the effect of ina

Nerve growth factor β polypeptide (NGFB) genetic variability: association with the methadone dose required for effective maintenance treatment.

Opioid addiction is a chronic disease with high genetic contribution and a large inter-individual variability in therapeutic response. The goal of this study was to identify pharmacodynamic factors that modulate methadone dose requirement. The neurotrophin family is involved in neural plasticity, learning, memory and behavior and deregulated neural plasticity may underlie the pathophysiology of drug addiction. Brain-derived neurotrophic factor (BDNF) was shown to affect the response to methadone maintenance treatment. This study explores the effects of polymorphisms in the nerve growth factor

Application of functional near-infrared spectroscopy in older adults with balance impairment: A review.

As the population ages, the issue of falls among the elderly is becoming increasingly prominent, and balance dysfunction is one of the main risk factors for falls. Accurately assessing balance function in older adults and elucidating its underlying neural mechanisms holds significant clinical importance for predicting fall risk and developing personalized interventions. In recent years, functional near-infrared spectroscopy (fNIRS), as a non-invasive brain imaging technology, has demonstrated unique advantages in revealing the neural mechanisms underlying balance control, owing to its good por

Neuromodulation for restoring and amplifying brain function.

Neuromodulation aims to improve brain function by altering neural activity. While many rehabilitation strategies seek to restore normal, healthy-like dynamics, some adopt a complementary strategy, using neuromodulation to strengthen repurposed processes that support function. Here we formalize these approaches as 'restorative normalization' (RN) and 'compensatory amplification' (CA), respectively. Drawing on cognitive neurophysiology, we evaluate the following four factors that enable CA to be effective: multiple realizability, multiscale neuroplasticity, precision readiness and activity selec

Closing the loop in DBS: A data-driven approach.

Deep brain stimulation (DBS) has transformed the treatment of movement disorders like Parkinson's Disease (PD). Innovations in DBS technology and experimentation have fostered adaptive DBS (aDBS), which employs a closed-loop system that senses physiological biomarkers to inform precise neuromodulation and personalized therapy. This review analyzes several promising advances in aDBS, including biomarker detection, control policies, mechanisms of efficacy, and a data-driven approach using artificial intelligence to decode motor states from neural signals. Investigations into data-driven approach

In vitro induction of patterned branchial arch-like aggregate from human pluripotent stem cells.

Early patterning of neural crest cells (NCCs) in the craniofacial primordium is important for subsequent development of proper craniofacial structures. However, because of the complexity of the environment of developing tissues, surveying the early specification and patterning of NCCs is difficult. In this study, we develop a simplified in vitro 3D model using human pluripotent stem cells to analyze the early stages of facial development. In this model, cranial NCC-like cells spontaneously differentiate from neural plate border-like cells into maxillary arch-like mesenchyme after a long-term c

The Neural and Computational Architecture of Feedback Dynamics in Mouse Cortex during Stimulus Report.

Conscious reportability of visual input is associated with a bimodal neural response in the primary visual cortex (V1): an early-latency response coupled to stimulus features and a late-latency response coupled to stimulus report or detection. This late wave of activity, central to major theories of consciousness, is thought to be driven by the prefrontal cortex (PFC), responsible for "igniting" it. Here we analyzed two electrophysiological studies in mice performing different stimulus detection tasks and characterized neural activity profiles in three key cortical regions: V1, posterior parie

Music can be reconstructed from human auditory cortex activity using nonlinear decoding models.

Music is core to human experience, yet the precise neural dynamics underlying music perception remain unknown. We analyzed a unique intracranial electroencephalography (iEEG) dataset of 29 patients who listened to a Pink Floyd song and applied a stimulus reconstruction approach previously used in the speech domain. We successfully reconstructed a recognizable song from direct neural recordings and quantified the impact of different factors on decoding accuracy. Combining encoding and decoding analyses, we found a right-hemisphere dominance for music perception with a primary role of the superi

The Neural Mechanism of Number Line Bisection: A fMRI study.

The neural networks involved in number line bisection are poorly understood. We used functional magnetic resonance imaging (fMRI) to investigate these. fMRI was analyzed from 28 healthy volunteers who performed number and physical line bisection tasks (and their respective baselines). Whole brain analyses showed that these two bisection tasks shared common neural correlates in bilateral parietal-frontal networks; furthermore, bilateral parietal-frontal areas, right cerebellum, left insula and supplementary motor area (SMA) showed higher activity when contrasting the number line with a physical

A new mode of pancreatic islet innervation revealed by live imaging in zebrafish.

Pancreatic islets are innervated by autonomic and sensory nerves that influence their function. Analyzing the innervation process should provide insight into the nerve-endocrine interactions and their roles in development and disease. Here, using in vivo time-lapse imaging and genetic analyses in zebrafish, we determined the events leading to islet innervation. Comparable neural density in the absence of vasculature indicates that it is dispensable for early pancreatic innervation. Neural crest cells are in close contact with endocrine cells early in development. We find these cells give rise

Neural mobilization promotes nerve regeneration by nerve growth factor and myelin protein zero increased after sciatic nerve injury.

Neurotrophins are crucial in relation to axonal regrowth and remyelination following injury; and neural mobilization (NM) is a noninvasive therapy that clinically is effective in neuropathic pain treatment, but its mechanisms remains unclear. We examined the effects of NM on the regeneration of sciatic nerve after chronic constriction injury (CCI) in rats. The CCI was performed on adult male rats, submitted to 10 sessions of NM, starting 14 days after CCI. Then, the nerves were analyzed using transmission electron microscopy and western blot for neural growth factor (NGF) and myelin protein ze

DMPK protein isoforms are differentially expressed in myogenic and neural cell lineages.

Myotonic dystrophy type 1 (DM1) is a neuromuscular disorder caused by an unstable (CTG . CAG)n segment in the 3' untranslated region of the myotonic dystrophy protein kinase (DMPK) gene. It is commonly accepted that DMPK mRNA-based toxicity is the main contributor to DM1 manifestations; however, not much is known about the significance of the DMPK protein. To appreciate its normal and possible pathobiological role, we analyzed the patterns of DMPK splice isoform expression in mouse tissues. Long membrane-anchored DMPK dominated in heart, diaphragm, and skeletal muscle, whereas short cytosolic

[Neurophysiologic intraoperative monitoring to preserve cranial nerve function in base of skull surgery].

BACKGROUND: Neurophysiologic intraoperative monitoring (NIM) has gone through a renaissance since the advent of computer technology. Currently, both motor and sensory cranial nerves, including the IInd and VIIIth cranial nerves, can be intraoperatively controlled by means of small and mobile systems. PATIENTS AND METHODS: In order to estimate the value of NIM using the new generation of computer systems, we analyzed the records of 379 patients who underwent skull base surgery since 1996. These comprised NIM of the IInd, VIIth, VIIIth cranial nerves in most cases and of the IXth, Xth, XIth, an

The impact of transcranial ultrasound on auditory brainstem responses and neuronal activation in the cochlear nucleus.

OBJECTIVES: Transcranial ultrasound stimulation (TUS) is a non-invasive technique with therapeutic potential for neurodegenerative diseases. Given its increasing therapeutic relevance, it is essential to identify any unintended consequences, especially on vulnerable systems such as the auditory pathway. Specifically, the precise impact of TUS on auditory responses and neural activation patterns within the central auditory system remains unclear. We hypothesized that TUS could indirectly modulate central auditory processing, even at frequencies outside the direct hearing range, leading to obser

[Evaluation of the possibility of using neural networks for automatic diagnostics of obstructive urination].

INTRODUCTION: Obstructive type of urination requires accurate and timely diagnosis to prevent complications and improve the quality of life of patients. Traditional diagnostic methods such as uroflowmetry, although they remain the standard, have their limitations. In this context, videography of urine stream followed by image analysis is a more cost-effective and promising approach that allows for a more detailed picture of urination, accessible not only to urologists, but also to patients. AIM: To establish the possibility of recognizing and classifying graphs of the urination using neural n
