MR Imaging-Based Evaluations of Olfactory Bulb Atrophy in Patients with Olfactory Dysfunction.

BACKGROUND AND PURPOSE: Although the olfactory bulb volume as assessed with MR imaging is known to reflect olfactory function, it is not always measured during olfactory pathway assessments in clinical settings. We aimed to evaluate the utility of visual olfactory bulb atrophy and neuropathy analyses using MR imaging in patients with olfactory dysfunction. MATERIALS AND METHODS: Thirty-four patients who presented with subjective olfactory loss between March 2016 and February 2017 were included. Patients underwent a nasal endoscopic examination, olfactory testing with the Korean Version of the

The Olfactory Bulb: An Immunosensory Effector Organ during Neurotropic Viral Infections.

In 1935, the olfactory route was hypothesized to be a portal for virus entry into the central nervous system (CNS). This hypothesis was based on experiments in which nasophayngeal infection with poliovirus in monkeys was prevented from spreading to their CNS via transection of olfactory tracts between the olfactory neuroepithelium (ONE) of the nasal cavity and the olfactory bulb (OB). Since then, numerous neurotropic viruses have been observed to enter the CNS via retrograde transport along axons of olfactory sensory neurons whose cell bodies reside in the ONE. Importantly, this route of infec

Compensatory rapid switching of binasal inputs in the olfactory cortex.

Odors are inhaled through the nostrils into two segregated nasal passages and detected by sensory neurons in the bilateral olfactory epithelia. Airflow through the two nasal passages is usually asymmetrical because of alternating changes in nasal mucosal congestion. Here we show that neurons in the anterior olfactory nucleus (AON) of the adult rat olfactory cortex are ordinarily dominated by ipsi-nasal inputs and that binasal neurons in the AON respond to ipsilateral and contralateral nasal inputs with nearly equivalent odorant category selectivity. Deprivation of ipsilateral nasal inputs by u

Evidence for glutamate as the olfactory receptor cell neurotransmitter.

1. Synaptic transmission between olfactory receptor neurons and mitral/tufted cells was examined using a whole-cell recording technique in a hemisected preparation of the turtle olfactory bulb. To determine the olfactory receptor neuron transmitter, we isolated components of the synaptic response of mitral/tufted cells to olfactory nerve stimulation using postsynaptic receptor antagonists. 2. Low-intensity stimulation of the olfactory nerve evoked monosynaptic excitatory postsynaptic potentials in mitral/tufted cells that consisted of a rapid and prolonged depolarization with little contributi

Role of polysialic acid on outgrowth of rat olfactory receptor neurons.

Towards elucidating the role of polysialic acid (PSA) in developing olfactory neuron of the rat, we injected neuraminidase (endo-N) into the olfactory nerve pathway under whole embryo culture, then employed immunohistochemistry to (i) detect expression of highly sialylated neural cell adhesion molecules (NCAM-H) and (ii) identify olfactory neurons via anti-microtubule-associated protein 1B (MAP1B) antibody. Olfactory axonal outgrowth from basal lamina occurred at the 31-somite stage and reached the olfactory bulb primordium at the 42-somite stage, being coincident with the timing and expressio

Severity of olfactory deficits is reflected in functional brain networks-An fMRI study.

Even though deficits in olfactory function affect a considerable part of the population, the neuronal basis of olfactory deficits remains scarcely investigated. To achieve a better understanding of how smell loss affects neural activation patterns and functional networks, we set out to investigate patients with olfactory dysfunction using functional magnetic resonance imaging (fMRI) and olfactory stimulation. We used patients' scores on a standardized olfactory test as continuous measure of olfactory function. 48 patients (mean olfactory threshold discrimination identification (TDI) score = 16

Amygdalar Gating of Early Sensory Processing through Interactions with Locus Coeruleus.

Fear- and stress-induced activity in the amygdala has been hypothesized to influence sensory brain regions through the influence of the amygdala on neuromodulatory centers. To directly examine this relationship, we used optical imaging to observe odor-evoked activity in populations of olfactory bulb inhibitory interneurons and of synaptic terminals of olfactory sensory neurons (the primary sensory neurons of the olfactory system, which provide the initial olfactory input to the brain) during pharmacological inactivation of amygdala and locus coeruleus (LC) in mice. Although the amygdala does n

A macroglomerulus in the antennal lobe of leaf-cutting ant workers and its possible functional significance.

Ants have a well-developed olfactory system, and pheromone communication is essential for regulating social life within their colonies. We compared the organization of primary olfactory centers (antennal lobes, ALs) in the brain of two closely related species of leaf-cutting ants (Atta vollenweideri, Atta sexdens). Both species express a striking size polymorphism associated with polyethism. We discovered that the ALs of large workers contain a substantially enlarged glomerulus (macroglomerulus, MG) at the entrance of the antennal nerve. This is the first description of an MG in non-sexual ind

Children who can't smell the coffee: isolated congenital anosmia.

Two children with isolated congenital anosmia, a rare syndrome of deficient restricted neuronal migration, are presented with early diagnosis confirmed by standardized smell testing and detailed neuroimaging studies. Recognition of this disorder and its spectrum of presentations provides important insights into the molecular mechanisms underlying the development of the olfactory system.

Plasticity in the olfactory cortex: age-dependent effects of deafferentation.

In order to assess the role of input-target interactions in the development of olfactory cortex, the primary afferent fibers from the olfactory bulb to the superficial part of layer I of the cortex (layer Ia) were removed in developing and mature rats. After survival periods that vary from a few days to 2-6 months, changes were assessed in (1) the radial thickness of layer I, (2) the laminar distribution of intracortical associational fibers, which normally terminate in a deep part of layer I (layer Ib), and (3) the distribution of glia in layer I. The findings indicate that the lamination of

Transplants of olfactory mucosa in the rat brain I. A light microscopic study of transplant organization.

Olfactory mucosa from neonatal rats has been transplanted into the fourth cerebral ventricle or into the parietal cortex of neonatal and adult rats. In these ectopic locations, olfactory neurons continue to differentiate from the neurogenetic matrix (basal cells) of the neuroepithelium. Sensory axon bundles from the newly formed olfactory neurons penetrate the host brain where they branch without forming the characteristic olfactory glomeruli. From the base of the neuroepithelium neural elements migrate into the host cerebral tissue losing their epithelial organization.

LHRH systems in brain of platyfish.

The luteinizing hormone-releasing hormone (LHRH) system of the platyfish Xiphophorus has been studied using immunohistochemistry and retrograde transport of horseradish peroxidase (HRP). Three different populations of LHRH-positive cell bodies are present in the brain, one in the ventral telencephalon at the border to the olfactory bulb (nucleus of olfactoretinalis), one lateral to the n. preopticus (nucleus preopticus basalis lateralis) and one in the midbrain. LHRH neurons from the nucleus olfactoretinalis project via the medial olfactory tract to the olfactory bulb and to olfactory nerves.

The neuroplastic effect of olfactory training to the recovery of olfactory system in mouse model.

BACKGROUND: Several studies have reported the benefits of olfactory training (OT) in the olfactory nervous system of mouse models. Therefore, in this study we performed next-generation sequencing to evaluate the effects of OT on mRNA sequencing in the olfactory area. METHODS: Mice in each group were administered 300 mg of 3-methylindole per kilogram of mouse weight. The olfactory function was evaluated by a food-finding test once a week. The olfactory neuroepithelium was harvested for histologic examination and protein analysis. Subsequently, data analysis, gene ontology and pathway analysis,

Substrate-bound carbohydrates stimulate signal transduction and neurite outgrowth in an olfactory neuron cell line.

Subpopulations of olfactory receptor neurons, which are dispersed throughout the olfactory neuroepithelium, express specific cell surface carbohydrates and project to discrete regions of the olfactory bulb. Cell surface carbohydrates such as N-acetyl-lactosamine have been postulated to mediate sorting and selective fasciculation of discrete axon subpopulations during development of the olfactory pathway. Substrate-bound N-acetyl-lactosamine promotes neurite outgrowth by both clonal olfactory receptor neuron cell lines and olfactory receptor neurons in vitro, indicating that cell surface carboh

Impaired olfactory function in substance use disorder.

RATIONALE: The sense of smell plays a key role in guiding motivated behavior, and olfactory function is impaired in clinical populations with dysfunctional approach-avoidance behavior, including major depressive and alcohol use disorder (AUD). However, whether olfactory impairments are also observed in individuals with substance use disorders (SUDs) other than AUD is currently unknown. OBJECTIVES: This study aimed to evaluate the relationship between olfactory function and SUDs. METHODS: We tested olfaction in 40 individuals with substance use disorders (SUDs) other than AUD using the Sniffi

Tackling the Biological Meaning of the Human Olfactory Bulb Dyshomeostatic Proteome across Neurological Disorders: An Integrative Bioinformatic Approach.

Olfactory dysfunction is considered an early prodromal marker of many neurodegenerative diseases. Neuropathological changes and aberrant protein aggregates occur in the olfactory bulb (OB), triggering a tangled cascade of molecular events that is not completely understood across neurological disorders. This study aims to analyze commonalities and differences in the olfactory protein homeostasis across neurological backgrounds with different spectrums of smell dysfunction. For that, an integrative analysis was performed using OB proteomics datasets derived from subjects with Alzheimer's disease

An updated antennal lobe atlas for the yellow fever mosquito Aedes aegypti.

The yellow fever mosquito Aedes aegypti is a prolific vector of arboviral and filarial diseases that largely relies on its sense of smell to find humans. To facilitate in-depth analysis of the neural circuitry underlying Ae. aegypti olfactory-driven behaviors, we generated an updated in vitro atlas for the antennal lobe olfactory brain region of this disease vector using two independent neuronal staining methods. We performed morphological reconstructions with replicate fixed, dissected and stained brain samples from adult male and female Ae. aegypti of the LVPib12 genome reference strain and
