[Sirenomelia in one of the twins].

A case of rarely occurring malformation of the embryonal period of development, a diploid variant of sirenomelia in one of the twins, is described. Roentgenology revealed a paired set of bones of the extremities. There was aplasia of the external sex organs with formed gonads, atresia of the urethra and anus, aplasia of the renal arteries, hypoplasia of the right and aplasia of the left kidney, colonic-urinary bladder fistula. Operative intervention (epicystostomy, colostomy) was applied. The baby died on the 8th day with renal insufficiency and disorders of the cerebral circulation.

Nuclear donor cell lines considerably influence cloning efficiency and the incidence of large offspring syndrome in bovine somatic cell nuclear transfer.

Total five ear skin fibroblast lines (named F1, F2, F3, F4 and F5) from different newborn Holstein cows have been used as nuclear donor cells for producing cloned cows by somatic cell nuclear transfer (SCNT). The effects of these cell lines on both in vitro and in vivo developmental rates of cloned embryos, post-natal survivability and incidence of large offspring syndrome (LOS) were examined in this study. We found that the different cell lines possessed the same capacity to support pre-implantation development of cloned embryos, the cleavage and blastocyst formation rates ranged from 80.2 ±

Cloned cattle fetuses with the same nuclear genetics are more variable than contemporary half-siblings resulting from artificial insemination and exhibit fetal and placental growth deregulation even in the first trimester.

The cloning of cattle by somatic cell nuclear transfer (NT) is associated with a high incidence of abnormal placentation, excessive fluid accumulation in the fetal sacs (hydrops syndrome), and fetal overgrowth. Fetal and placental development was investigated at Day 50, during placentome formation; at Day 100, when placentation was completed; and at Day 150, when the hydrops syndrome frequently develops. The NT fetuses were compared with contemporary half-siblings generated from in vitro-produced embryos or by artificial insemination (AI). Fetal cotyledon formation and vascularization of the c

The development and maintenance of the mononuclear phagocyte system of the chick is controlled by signals from the macrophage colony-stimulating factor receptor.

BACKGROUND: Macrophages have many functions in development and homeostasis as well as innate immunity. Recent studies in mammals suggest that cells arising in the yolk sac give rise to self-renewing macrophage populations that persist in adult tissues. Macrophage proliferation and differentiation is controlled by macrophage colony-stimulating factor (CSF1) and interleukin 34 (IL34), both agonists of the CSF1 receptor (CSF1R). In the current manuscript we describe the origin, function and regulation of macrophages, and the role of CSF1R signaling during embryonic development, using the chick as

Consistent left-right asymmetry cannot be established by late organizers in Xenopus unless the late organizer is a conjoined twin.

How embryos consistently orient asymmetries of the left-right (LR) axis is an intriguing question, as no macroscopic environmental cues reliably distinguish left from right. Especially unclear are the events coordinating LR patterning with the establishment of the dorsoventral (DV) axes and midline determination in early embryos. In frog embryos, consistent physiological and molecular asymmetries manifest by the second cell cleavage; however, models based on extracellular fluid flow at the node predict correct de novo asymmetry orientation during neurulation. We addressed these issues in Xenop

Effects of Thyroid Hormone on Tissue Hypoxia: Relevance to Sepsis Therapy.

Tissue hypoxia occurs in various conditions such as myocardial or brain ischemia and infarction, sepsis, and trauma, and induces cellular damage and tissue remodeling with recapitulation of fetal-like reprogramming, which eventually results in organ failure. Analogies seem to exist between the damaged hypoxic and developing organs, indicating that a regulatory network which drives embryonic organ development may control aspects of heart (or tissue) repair. In this context, thyroid hormone (TH), which is a critical regulator of organ maturation, physiologic angiogenesis, and mitochondrial bioge

Developmental and transcriptomic alterations in bovine blastocysts derived from oocytes of donors exposed to toxic tall fescue.

Fescue toxicosis, caused by the ingestion of ergot alkaloids present in endophyte-infected tall fescue (E + ), is associated with impaired reproductive performance in beef cattle. Because early embryonic development depends on maternally derived RNAs and proteins accumulated during oocyte maturation, disruption of oocyte contents by ergot alkaloids may affect embryonic development. This study evaluated the effects of maternal exposure to E + on reproductive performance and blastocyst gene expression in beef heifers. Twenty-four commercial Angus heifers were assigned to either E + (n = 12) or e

Suspect List-Guided High-Throughput Screening (SLIG-HTS) Identifies Environmental Substances Associated with Advanced Embryonic Developmental Timing.

Retardation of embryonic development by environmental contaminants has been extensively documented, whereas coordinated advancement in developmental timing has received far less attention despite its risks to organisms as well. To address this gap, we developed a Suspect List-Guided High-Throughput Screening strategy that integrates literature-informed suspect list construction with developmental trend-guided, stage-resolved end points evaluation to identify environmental substances associated with advanced embryonic developmental timing across a broad chemical space. By retrospectively screen

The basic helix-loop-helix transcription factor TCF4 recruits the Mediator Complex to activate gonadal genes and drive ovarian development.

The bipotential gonad is the precursor organ to both the ovary and testis and develops as part of the embryonic urogenital system. In mice, gonadogenesis initiates around embryonic day 9.5 (E9.5), when coelomic epithelial (CE) cells overlaying the mesonephric ducts proliferate and acquire competence to differentiate into the somatic cell types of the embryonic gonad, the pre-supporting cell (Sertoli cells in the testis and granulosa cells in the ovary) and the interstitial cell lineages. While some transcription factors that drive gonadal cell fate are known, we found that basic helix-loop-hel

Integrative single-cell RNA-seq and ATAC-seq analysis of myogenic differentiation in pig.

BACKGROUND: Skeletal muscle development is a multistep process whose understanding is central in a broad range of fields and applications, from the potential medical value to human society, to its economic value associated with improvement of agricultural animals. Skeletal muscle initiates in the somites, with muscle precursor cells generated in the dermomyotome and dermomyotome-derived myotome before muscle differentiation ensues, a developmentally regulated process that is well characterized in model organisms. However, the regulation of skeletal muscle ontogeny during embryonic development

Expressed proteins and activated pathways in conditioned embryo culture media from IVF patients are diverse according to infertility factors.

OBJECTIVE: Given that the embryo culture medium secretome reflects the embryo development, we hypothesize that protein profiles are affected according to infertility factors, which can be responsible for detrimental embryonic developmental competence. The aim of this study was to screen the protein profile of conditioned embryo culture media in patients presenting deep infiltrating endometriosis (ENDO) and polycystic ovarian syndrome (PCOS) undergoing IVF, by proteomics approaches. The control group was constituted by tubal factor patients. METHODS: Patients underwent in vitro fertilization (

Case Report: Case report: A rare case of middle-ear Rhabdomyosarcoma in a 4-year-old boy.

We present a rare case of a four-year-old boy with a botyroid embryonal rhabdomyosarcoma of the right middle ear. Rhabdomyosarcoma is a soft tissue malignancy which is thought to originate from embryonic mesenchymal cells of striated skeletal muscle.  It is a disease primarily of children and is exceptionally rare in parameningeal regions.  The diagnosis is often delayed and easily misdiagnosed as aural polyp. Therefore, advanced disease is common at the time of diagnosis.  A four-year-old boy presented with a four-month history of recurrent left ear blood and pus discharge, otalgia and fevers

Susceptibility of thymocytes for infection by chicken anemia virus is related to pre- and posthatching development.

To investigate the age-dependent mechanism of susceptibility for chicken anemia virus (CAV) infection, we inoculated embryos and chickens of ages between day 9 of embryonic development and day 28 after hatching with CAV. Chicken embryos inoculated at days 9 and 11 of development showed no CAV-infected cells in the thymus, nor in other lymphoid organs. Many CAV-infected cells were detected in the thymic cortex of all chicken embryos inoculated at days 13 and 16 of development and of all chickens inoculated 1, 3, and 7 days after hatching. All embryos and chickens that contained CAV-infected cel

Transcriptomic changes in embryonic blood of subsequent generations following in ovo stimulation with nutriepigenetic factors.

BACKGROUND: Epigenetic modifications regulate gene expression and are influenced by environmental factors, shaping phenotypic and clinical outcomes. These changes have the potential to persist across generations, although their stability may vary between tissues. This study aims to observe, using an in ovo model, the effects of single (F1) and repeated (F1-F3) prenatal stimulation with potential epigenetic factors on the transcriptome of embryonic blood in subsequent generations. METHOD: Using an in ovo model across three generations of Green-legged Partridgelike chickens (with additional F4

Innovation in basic science: stem cells and their role in the treatment of paediatric cardiac failure--opportunities and challenges.

Heart failure is a leading cause of death worldwide. Current therapies only delay progression of the cardiac disease or replace the diseased heart with cardiac transplantation. Stem cells represent a recently discovered novel approach to the treatment of cardiac failure that may facilitate the replacement of diseased cardiac tissue and subsequently lead to improved cardiac function and cardiac regeneration. A stem cell is defined as a cell with the properties of being clonogenic, self-renewing, and multipotent. In response to intercellular signalling or environmental stimuli, stem cells differ

Paternal contributions to the mammalian zygote: fertilization after sperm-egg fusion.

Mammalian fertilization has traditionally been regarded as a simple blending of two gametes, during which the haploid genome of the fertilizing spermatozoon constitutes the primary paternal contribution to the resulting embryo. In contrast to this view, new research provides evidence of important cytoplasmic contributions made by the fertilizing spermatozoon to the zygotic makeup, to the organization of preimplantation development, and even reproductive success of new forms of assisted fertilization. The central role of the sperm-contributed centriole in the reconstitution of zygotic centrosom

Comparative ultrastructure and opsin immunocytochemistry of the retina and pineal organ in fish.

The pineal organ and retina were compared in developing charr and cisco, further in adult cisco, eel, creek chub, dace, zebrafish and black moli by opsin immunocytochemistry. In prehatching charr embryos, retinal rods and cones and pinealocytes displayed well-developed outer segments and formed synapses. Differentiation of the retina started centrally but was more advanced in the dorso-caudal retina than rostroventrally. The pineal organ differentiated earlier distally than proximally. In the cisco, the pineal organ and retina differentiated around hatching. In charr embryos, further in the la

The origins of the circumventricular organs.

The circumventricular organs (CVOs) are specialised neuroepithelial structures found in the midline of the brain, grouped around the third and fourth ventricles. They mediate the communication between the brain and the periphery by performing sensory and secretory roles, facilitated by increased vascularisation and the absence of a blood-brain barrier. Surprisingly little is known about the origins of the CVOs (both developmental and evolutionary), but their functional and organisational similarities raise the question of the extent of their relationship. Here, I review our current knowledge o

CDK5RAP2 Is Required to Maintain the Germ Cell Pool during Embryonic Development.

Gene products linked to microcephaly have been studied foremost for their role in brain development, while their function in the development of other organs has been largely neglected. Here, we report the critical role of Cdk5rap2 in maintaining the germ cell pool during embryonic development. We highlight that infertility in Cdk5rap2 mutant mice is secondary to a lack of spermatogenic cells in adult mice as a result of an early developmental defect in the germ cells through mitotic delay, prolonged cell cycle, and apoptosis.

Partial dorsal agenesis accompanied with circumportal pancreas in a donor for islet transplantation.

As embryonic development of the pancreas progresses, a wide range of congenital anomalies and variations can occur. We report on the incidental observation of partial agenesis of the dorsal pancreas coexistent with cricumportal pancreas in a donor for islet transplantation. We successfully recovered a high yield of islets from such a pancreas, and the islet recipient achieved insulin independence after transplant. This case report discusses the suitability of the anomalous pancreas as a donor organ for islet transplant and the possible embryologic association between the two entities.
