The orchid Gastrodia-derived extracellular vesicles deliver miR535 to suppress a fungal effector and facilitate symbiosis with Armillaria.

Plant-derived extracellular vesicles (PDEVs) are nanoscale lipid bilayer structures that facilitate cross-kingdom communication by transporting bioactive molecules, including proteins and small RNAs. While their roles in plant immunity against fungal pathogens are well-established, their functions in beneficial or symbiotic interactions, particularly with mycorrhizal fungi, remain largely unexplored. Here, we investigated the function of PDEVs in the symbiosis between the fully mycoheterotrophic orchid Gastrodia elata and its fungal partner Armillaria gallica. We isolated and characterized G. …

WRKY transcription factors and plant defense responses: latest discoveries and future prospects.

WRKY transcription factors are among the largest families of transcriptional regulators. In this review, their pivotal role in modulating various signal transduction pathways during biotic and abiotic stresses is discussed. Transcription factors (TFs) are important constituents of plant signaling pathways that define plant responses against biotic and abiotic stimuli besides playing a role in response to internal signals which coordinate different interacting partners during developmental processes. WRKY TFs, deriving their nomenclature from their signature DNA-binding sequence, represent one

The MicroRNA156a-SPL module regulates kiwifruit resistance to Pseudomonas syringae pv. actinidiae.

MicroRNAs (miRNAs) are key regulators of plant growth, development, and immunity. However, their role in kiwifruit and other perennial fruit trees remains poorly characterized. Here, we demonstrate that Ac-miR156a functions as a negative regulator of kiwifruit resistance to Pseudomonas syringae pv. actinidiae (Psa). In the resistant cultivar "Jinkui," Ac-miR156a expression was significantly suppressed upon Psa infection, whereas it was markedly induced in the susceptible cultivar "Hongyang." Overexpression of miR156a increased kiwifruit susceptibility to Psa, while silencing miR156a via short-

Spatial organization of plant defense at the infection front.

Plants lack specialized immune cells and instead rely on coordinated cellular responses to restrict pathogen invasion while preserving tissue integrity. How plant immunity spatially organizes these responses remains unclear. Using live-cell reporters in Arabidopsis infected with Pseudomonas syringae, we show that effector-triggered immunity, superimposed on pattern-triggered immunity, establishes a sustained yet spatially confined immune architecture at the infection front. Defense activation persists for days but remains restricted to a narrow ring of cells surrounding viable bacterial microc…

The pathogenicity factor of DNA virus interacts with Arabidopsis deubiquitinases UBP12/13 to promote viral infection.

Plant DNA viruses, such as Cauliflower mosaic virus (CaMV) and Cabbage leaf curl virus (CaLCuV), are devastating pathogens that cause substantial yield losses in crops worldwide. These viruses assemble their genomes into minichromosomes with host histones. Histone post-translational modifications (PTMs) are important for viral replication and host resistance. In this study, we demonstrate that the CaMV pathogenicity factor P6 interacts with Arabidopsis deubiquitinases UBP12 and UBP13, which are typically negative regulators of plant immunity. Using affinity purification-mass spectrometry (AP-M…

The Arabidopsis Iron-Sulfur (Fe-S) Cluster Gene MFDX1 Plays a Role in Host and Nonhost Disease Resistance by Accumulation of Defense-Related Metabolites.

Until recently, genes from the iron-sulfur (Fe-S) cluster pathway were not known to have a role in plant disease resistance. The Nitrogen Fixation S (NIFS)-like 1 (NFS1) and Mitochondrial Ferredoxin-1 (MFDX1) genes are part of a set of 27 Fe-S cluster genes induced after infection with host and nonhost pathogens in Arabidopsis. A role for AtNFS1 in plant immunity was recently demonstrated. In this work, we showed that MFDX1 is also involved in plant defense. More specifically, Arabidopsis mfdx1 mutants were compromised for nonhost resistance against Pseudomonas syringae pv. tabaci, and showed …

The U-Box/ARM E3 ligase PUB13 regulates cell death, defense, and flowering time in Arabidopsis.

The components in plant signal transduction pathways are intertwined and affect each other to coordinate plant growth, development, and defenses to stresses. The role of ubiquitination in connecting these pathways, particularly plant innate immunity and flowering, is largely unknown. Here, we report the dual roles for the Arabidopsis (Arabidopsis thaliana) Plant U-box protein13 (PUB13) in defense and flowering time control. In vitro ubiquitination assays indicated that PUB13 is an active E3 ubiquitin ligase and that the intact U-box domain is required for the E3 ligase activity. Disruption of

Generation and immunogenicity of Japanese encephalitis virus envelope protein expressed in transgenic rice.

Transgenic plants have become attractive as bioreactors to produce heterologous proteins that can be developed as edible vaccines. In the present study, transgenic rice expressing the envelope protein (E) of Japanese encephalitis virus (JEV), under the control of a dual cauliflower mosaic virus (CaMV 35S) promoter, was generated by Agrobacterium-mediated transformation. Southern blot, Northern blot, Western blot and ELISA analyses confirmed that the E gene was integrated into transgenic rice and was expressed in the leaves at levels of 1.1-1.9 microg/mg of total soluble protein. After intraper

AtALMT9 Modulates Leaf Development and Disease Resistance in Arabidopsis thaliana.

Delaying plant aging and death and enhancing immune defenses of plant have beneficial effects on crop production and the continued development of the agricultural economy. In this study, we focused on AtALMT9, which is located in the vacuolar membrane. Specifically, we investigated its functions in Arabidopsis thaliana leaf development and disease resistance. Our physiological analysis revealed that AtALMT9 negatively regulates leaf development and the immune response to pathogenic microorganisms. Moreover, RNA-seq and qRT-PCR analyses indicated leaf aging- and immunity-related genes were sign

A conserved phospho-switch controls receptor kinase SDS2 activation and homeostasis in rice immunity.

Plant surface immune receptors are tightly controlled to maintain homeostasis and prevent overactivation. However, the mechanisms coordinating the balance remain largely unknown. The monocot-specific receptor-like kinase SDS2 regulates cell death and immunity in rice. Here, we identify Thr671, a conserved phospho-switch in RD-type Ser/Thr kinases across kingdoms, as the central node that orchestrates both immune activation and turnover of SDS2. Autophosphorylation of Thr671 initiates signaling but also primes SDS2 for ubiquitination and degradation by the E3 ligase SPL11. This degradation is a

Metabolic regulation of systemic acquired resistance.

Plants achieve an optimal balance between growth and defense by a fine-tuned biosynthesis and metabolic inactivation of immune-stimulating small molecules. Recent research illustrates that three common hubs are involved in the cooperative regulation of systemic acquired resistance (SAR) by the defense hormones N-hydroxypipecolic acid (NHP) and salicylic acid (SA). First, a common set of regulatory proteins is involved in their biosynthesis. Second, NHP and SA are glucosylated by the same glycosyltransferase, UGT76B1, and thereby inactivated in concert. And third, NHP confers immunity via the S

Xanthomonas phaseoli pv. Manihotis effector XopAG impairs cassava immunity by hijacking MeHSP26 to suppress flavonoid biosynthesis.

Cassava bacterial blight (CBB), caused by Xanthomonas phaseoli pv. manihotis (Xpm), is a major disease constraining cassava production. Although overexpression of the Xpm effector XopAG enhances Arabidopsis susceptibility to Pseudomonas syringae pv. tomato DC3000, its virulence mechanism in cassava remains unclear. The study showed that deletion of XopAG did not affect morphology or extracellular enzyme production of Xpm but significantly attenuated its virulence on cassava. Quantitative proteomic analysis revealed that 267 differentially accumulated proteins (DAPs) at 6 h post-inoculation (hp

iTRAQ-Based Proteomics Analysis of Response to Solanum tuberosum Leaves Treated with the Plant Phytotoxin Thaxtomin A.

Thaxtomin A (TA) is a phytotoxin secreted by Streptomyces scabies that causes common scab in potatoes. However, the mechanism of potato proteomic changes in response to TA is barely known. In this study, the proteomic changes in potato leaves treated with TA were determined using the Isobaric Tags for Relative and Absolute Quantitation (iTRAQ) technique. A total of 693 proteins were considered as differentially expressed proteins (DEPs) following a comparison of leaves treated with TA and sterile water (as a control). Among the identified DEPs, 460 and 233 were upregulated and downregulated, r

Oxicam-type non-steroidal anti-inflammatory drugs inhibit NPR1-mediated salicylic acid pathway.

Nonsteroidal anti-inflammatory drugs (NSAIDs), including salicylic acid (SA), target mammalian cyclooxygenases. In plants, SA is a defense hormone that regulates NON-EXPRESSOR OF PATHOGENESIS RELATED GENES 1 (NPR1), the master transcriptional regulator of immunity-related genes. We identify that the oxicam-type NSAIDs tenoxicam (TNX), meloxicam, and piroxicam, but not other types of NSAIDs, exhibit an inhibitory effect on immunity to bacteria and SA-dependent plant immune response. TNX treatment decreases NPR1 levels, independently from the proposed SA receptors NPR3 and NPR4. Instead, TNX ind

Tracking Oxidative Bursts in Tomato-Pathogen Interactions.

Reactive oxygen species (ROS), such as hydrogen peroxide (H₂O₂), hydroxyl radicals (•OH), and superoxide anions (O₂-), are central to plant stress signaling. Among these, the role of H₂O₂ is prominent due to its relatively stable nature and its ability to diffuse across membranes, making it an ideal candidate for intra- and intercellular signaling. In tomato (Solanum lycopersicum L.), foliar pathogens elicit a rapid oxidative burst characterized by elevated H₂O₂ levels. In addition to suppressing pathogens, the oxidative burst also initiates broader immune responses in the plant. Thus, assessi

Surface immune signaling unlocks NLR activation through mRNA alternative splicing.

Plants activate pattern-triggered immunity (PTI) and effector-triggered immunity (ETI) to combat pathogens. However, how these systems coordinate immune activation while preventing autoimmunity remains poorly understood. In this study, we uncovered a regulatory mechanism in which surface immune signaling unlocks nucleotide-binding leucine-rich repeat (NLR) immune receptor activation through mRNA splicing. We identified an N-terminal prodomain in the potato late blight resistance protein Rpi-vnt1.1 that inhibits resistosome formation, preventing potential autoactivation of this NLR. Upon pathog

A phosphorelay circuit drives extracellular alkalinization in receptor kinase-mediated immune and cell-wall damage signaling.

Extracellular alkalinization has long been recognized as a hallmark of plant cell-surface receptor activation, including during pattern-triggered immunity (PTI), yet the mechanisms driving elicitor-induced alkalinization and its role in plant signaling remain unclear. Here, we demonstrate that inhibition of autoinhibited H+-ATPases (AHAs) is required for elicitor-induced extracellular alkalinization. This alkalinization is essential for immune and cell-wall damage signaling mediated by diverse plasma membrane-localized receptor kinases (RKs), likely through modulation of ligand-receptor intera
