Single-strand conformation polymorphism-based analysis reveals genetic variation within Spirometra erinacei (Cestoda: Pseudophyllidea) from Australia.

This study examined genetic variability within Spirometra erinacei (Cestoda: Pseudophyllidea) from different host species and geographical origins in Australia using a polymerase chain reaction (PCR)-based mutation detection approach, followed by DNA sequencing. Part of the cytochrome c oxidase subunit 1 gene (p cox 1) was amplified by PCR, scanned for sequence variation by single-strand conformation polymorphism (SSCP), and representative samples from different host species were selected for DNA sequencing. While no variation in SSCP profiles was detected among S. erinacei samples from dog, f

Apolipoprotein E polymorphism: A new genetic marker of hip fracture risk--The Study of Osteoporotic Fractures.

Hip fractures are common and devastating events. The apolipoprotein E*4 (APOE) allele, associated with Alzheimer's disease, has also been associated with osteoporosis in hemodialysis patients. We prospectively studied 1750 women, age >/=65 years, who underwent measurements of hip and calcaneal bone mineral density (BMD), were typed for APOE and followed for approximately 7.0 years for the occurrence of fractures and falls. Women with at least one APOE*4 allele had an increased risk of hip fracture, relative hazard (RH) (95% confidence interval) = 1.90 (1.05-3.41) and wrist fracture, RH = 1.67

Genetic risk for renal artery stenosis: association with deletion polymorphism in angiotensin 1-converting enzyme gene.

Atherosclerotic renal artery disease is an important secondary cause of hypertension. Currently, there is great interest in possible genetic determinants of cardiovascular disease. The ACE-D allele has been reported to be associated with increased risk of myocardial infarction as well as coronary re-stenosis after angioplasty. We therefore assessed whether this allele is also linked to renovascular disease by studying 56 Caucasian subjects with atherosclerotic renal artery stenosis and 74 age, sex and race matched control subjects. Genetic analysis for the ACE I/D polymorphism was performed on

Genetic identification of pregnancies responsible for choriocarcinomas after multiple pregnancies by restriction fragment length polymorphism analysis.

The pregnancies responsible for two cases of choriocarcinomas were determined by the restriction fragment length polymorphism of human leukocyte antigen-DQ genes in the tumors and leukocytes from the patients, their husbands, and their children. The first case was preceded by three normal pregnancies, and the second case was preceded by two normal pregnancies and a complete hydatidiform mole. Comparison of the restriction fragment length polymorphism patterns revealed that the first case of choriocarcinoma carried the parental genome and originated from the third normal pregnancy, whereas the

Impact of genetic polymorphisms of the renin-angiotensin system and of non-genetic factors on kidney transplant function--a single-center experience.

Renin-angiotensin-aldosterone system (RAAS) polymorphisms such as the angiotensinogen-gene-M235T-, the angiotensin-conversion enzyme (ACE)-gene I/D- and the angiotensin-II-type 1-receptor-(AT1R)-A1166C-polymorphism have been implicated in renal insufficiency and hypertension. We studied the association of these RAAS genotypes and non-genetic factors with transplant function and hypertension after renal graft transplantation (NTX). A total of 229 renal graft recipients, transplanted at a single center, were monitored up to 54 months and genotyped using polymerase chain reaction. The prevalence

Genetic analysis of chromosome 20-related posterior polymorphous corneal dystrophy: genetic heterogeneity and exclusion of three candidate genes.

PURPOSE: Posterior polymorphous corneal dystrophy (PPCD) is a genetically heterogeneous autosomal dominant condition which maps to the pericentromeric region of chromosome 20. Mutations in the VSX1 transcription factor have been reported in patients affected with PPCD, keratoconus, or a combination of both phenotypes. However, no mutation was identified in the coding region of VSX1 in the family used for the original mapping. To clarify the genetic basis of PPCD1, a thorough analysis was performed on the original PPCD1 family and two other PPCD1-linked families. As part of the analysis, the ex

Genetic polymorphisms and hypertension in Indian populations: a systematic review and integrative bioinformatics approach.

BACKGROUND: Hypertension continues to be a major public health issue in India, with increasing incidence associated with both lifestyle factors and genetic susceptibility. Despite genome-wide association studies identifying several loci related with blood pressure worldwide, there is a paucity of integration of these findings within the genetically varied Indian setting. OBJECTIVE: This systematic review aims to compile current information on single-nucleotide polymorphisms linked to hypertension in Indian communities and to elucidate their biological significance by bioinformatics analysis.

Influence of genetics on clinical trajectories in children with MASLD: role of the PNPLA3 I148M polymorphism.

PURPOSE: Metabolic Dysfunction-Associated Steatotic Liver Disease (MASLD) is increasingly prevalent in pediatric populations, especially among children with obesity. Genetic risk variants for MASLD- particularly the 148M allele of the patatin-like phospholipase domain-containing 3 (PNPLA3) gene - have been linked to various clinical outcomes and have recently led to the identification of a “liver-specific” MASLD subtype, distinct from the classical cardiometabolic MASLD phenotype. While adult evidence shows divergent effects of this gene especially on extrahepatic outcomes, similar pediatric d

DNA Repair Gene Polymorphisms as Genetic Factors Associated with Presence of Seborrheic Keratoses.

INTRODUCTION: Seborrheic keratoses (SK) are common benign epidermal tumors. Their pathogenesis is unknown, and no pathological significance is ascribed to them, although they could be part of paraneoplastic syndromes, in addition to presenting a variety of somatic mutations. SKs are associated with increased age, family history, and sun-exposure. METHODS: This study aimed to analyze if SK was related to DNA repair genes polymorphisms and analyze if any epidemiological, clinical or environmental characteristics could modify their prevalence. It was conducted an epidemiological, analytical, obs

Genetic polymorphisms in the SLC27A5 gene are associated with biochemical characteristics of HBV patients in Yunnan.

BACKGROUND: Lipid droplets could influence hepatitis B virus (HBV) replication and release, as well as disease progression in patients with chronic HBV. The solute carrier family 27 member 5 (SLC27A5) gene is highly expressed in the liver and possessed bile acid-CoA ligase activity. However, no previous study has investigated the role of the SLC27A5 gene in HBV infection or disease progression. METHODS: A total of 400 chronic HBV patients and 300 controls were collected in Yunnan Province to genotype three single nucleotide polymorphisms (SNPs) of the SLC27A5 gene and investigate potential as

Genetic polymorphisms of GSTM1 and GSTT1 genes: effects on susceptibility to formaldehyde-induced hematotoxicity.

Formaldehyde (FA) is a known human carcinogen for the upper respiratory tract. However, its hematotoxicity remains unclear. This study was performed to assess probable effects of FA on blood parameters and to determine the common polymorphisms in the detoxification enzymes GSTM1 and GSTT1 as biomarkers of susceptibility. Sixty-four workers with high occupational FA exposure and 57 non-exposed controls were studied. Complete blood count was performed. Also, GSTM1/GSTT1 genotypes were determined. After adjusting for potential confounding variables, FA exposure was only associated with the levels

Genetic Polymorphisms as Key Modulators of Cardiovascular Risk from Endocrine-Disrupting Chemicals.

Environmental exposure to persistent and non-persistent endocrine-disrupting chemicals (EDCs), including per- and polyfluoroalkyl substances (PFAS), polychlorinated biphenyls (PCBs), polybrominated diphenyl ethers (PBDEs), polycyclic aromatic hydrocarbons (PAHs), dioxins, phthalates, and bisphenols, has been increasingly associated with elevated cardiovascular disease (CVD) risk. Emerging evidence suggests the importance of gene-environment interactions in modulating individual susceptibility to EDC-related cardiovascular effects. This review summarizes current knowledge by synthesizing the ma

Relationship Between Central Sensitization and Genetic Polymorphisms-A Case-Control Study in Fibromyalgia.

BACKGROUND: Fibromyalgia is a syndrome characterised by widespread chronic pain, which is believed to be mediated by a state of central sensitisation (CS). Based on the hypothesis that CS itself could have genetic determinants, our aim was to further explore this pathway. METHODS: This study included 201 women with fibromyalgia and 40 healthy women matched on age and menopausal status. The nociceptive flexion reflex (NFR) threshold was used as a surrogate marker of CS. Genotyping of 60 candidate-genes likely to influence the pathogenesis of fibromyalgia or chronic pain was explored in all sub

Host genetic factors modulating dengue virus: a systematic review of TLR polymorphisms.

BACKGROUND: Dengue virus (DENV) infection is a substantial global public health burden, affecting ~ 390 million people every year, with a mortality rate of ~ 2.5% of those requiring hospitalization. Despite implementing vaccination and outbreak controls, DENV infection incidence continues to rise, particularly in endemic tropical and subtropical regions. Toll-like receptors (TLRs), as key components of the innate immune system, play a crucial role in recognizing DENV and initiating immune responses. Genetic variations, such as single-nucleotide polymorphisms (SNPs), in TLR genes may alter thei

Genetic Association of PCSK5 and MUC2 Gene Polymorphisms with Recurrent Pregnancy Loss (RPL).

Recurrent pregnancy loss (RPL) is defined as the occurrence of two or more consecutive pregnancy losses before 20 weeks of gestation, encompassing both embryonic and fetal losses. Although previous studies have provided substantial insights into RPL, the causes in many cases remain unexplained. This lack of information has prompted continued investigation into various risk factors, including those identified through next-generation sequencing (NGS). In the present study, whole-exome sequencing (WES) was used to identify genes potentially associated with RPL and infertility, which may serve as
