Nuclear factor-kappaB p50 subunits in chronic periodontitis and Porphyromonas gingivalis lipopolysaccharide-pulsed dendritic cells.

The pathogenesis of a number of chronic inflammatory diseases has been linked to dysregulated functioning of nuclear transcription factor (NF)-kappaB. In the present study, we examine NF-kappaB activation in human oral mucosal tissues of chronic periodontitis (CP) patients. Electrophoretic mobility shift assay and DNA-binding enzyme-linked immunosorbent assays indicate elevated levels of transcriptionally repressive p50 subunits and an increased p50/p65 ratio in CP tissues compared to healthy controls. Because Porphyromonas gingivalis has been recognized to be a causal factor in CP, we used P.…

Porphyromonas gingivalis fimbriae-dependent interleukin-6 autocrine regulation by increase of gp130 in endothelial cells.

BACKGROUND AND OBJECTIVE: Local persistent infection by Porphyromonas gingivalis leads to inflammatory systemic diseases, such as atherosclerosis. We have reported previously that avirulent P. gingivalis fimbriae-dependent invasion into endothelial cells might be involved in progression of atherosclerosis. Although interleukin-6 (IL-6) regulates progression of atherosclerosis, little is known about the relationship of P. gingivalis fimbriae-dependent invasion to IL-6 regulation in endothelial cells. MATERIAL AND METHODS: We examined the secretion of IL-6 and the expression of the IL-6 signal …

Porphyromonas gingivalis infection accelerates intimal thickening in iliac arteries in a balloon-injured rabbit model.

BACKGROUND: Current epidemiologic data suggest that a localized infection (periodontitis) can disseminate into the distant tissues, and subgingival bacteria can migrate in the bloodstream, thereby contributing to independent systemic disease processes. To test this hypothesis, we investigated the effect of repeated systemic inoculations with Porphyromonas gingivalis (Pg) on intimal hyperplasia in iliac arteries in a rabbit model of balloon injury. METHODS: One week after single balloon injury to the iliac artery, 30 male New Zealand rabbits were randomly assigned to intravenous inoculation wi…

Engagement of specific innate immune signaling pathways during Porphyromonas gingivalis induced chronic inflammation and atherosclerosis.

BACKGROUND AND OBJECTIVE: Although mannitol has been widely used in hospitals to treat patients with high intracranial pressure (ICP) secondary to intracerebral haemorrhage (ICH), no universal agreement has been reached regarding the optimal dosage of this agent for achieving appropriate intracranial decompression. The aim of this study was to investigate the effects of different mannitol dosages on ICP and the effects of other factors, such as sex, age, haemorrhage location and haematoma volume, on the ICP-lowering effect of mannitol. The data obtained were then used to construct a formula fo

Genotypic characterization of Porphyromonas gingivalis isolated from Swedish patients with periodontitis and from periodontal abscesses.

INTRODUCTION: A significant genetic polymorphism has been shown for Porphyromonas gingivalis isolates from different geographical areas. It is, however, possible that genetic similarities can be found among isolates obtained from a more specific population. The aim of the present study was to evaluate genetic heterogeneity among P. gingivalis isolates obtained from Swedish subjects with chronic periodontitis and from periodontal abscess lesions. METHODS: A total of 78 P. gingivalis strains, including 55 fresh clinical isolates obtained from 52 Swedish periodontitis subjects, eight isolates fr…

Roles of the host oxidative immune response and bacterial antioxidant rubrerythrin during Porphyromonas gingivalis infection.

The efficient clearance of microbes by neutrophils requires the concerted action of reactive oxygen species and microbicidal components within leukocyte secretory granules. Rubrerythrin (Rbr) is a nonheme iron protein that protects many air-sensitive bacteria against oxidative stress. Using oxidative burst-knockout (NADPH oxidase-null) mice and an rbr gene knockout bacterial strain, we investigated the interplay between the phagocytic oxidative burst of the host and the oxidative stress response of the anaerobic periodontal pathogen Porphyromonas gingivalis. Rbr ensured the proliferation of P.…

Inhibition of trypsin-like cysteine proteinases (gingipains) from Porphyromonas gingivalis by tetracycline and its analogues.

Extracellular cysteine proteinases, referred to as gingipains, are considered important virulence factors for Porphyromonas gingivalis, a bacterium recognized as a major etiologic agent of chronic periodontitis. We investigated the effect of tetracycline and its analogues, doxycycline and minocycline, on the enzymatic activities of gingipains. Tetracyclines at 100 microM totally inhibited the amidolytic activity of arginine-specific gingipains (HRgpA and RgpB). In contrast, inhibition of Kgp was less efficient and required a somewhat higher concentration of the antibiotic to achieve the same e…

Tetracyclines inhibit Porphyromonas gingivalis-induced alveolar bone loss in rats by a non-antimicrobial mechanism.

Tetracyclines have been widely used as adjuncts in periodontal therapy due to the antimicrobial efficacy of these drugs. Recently, their ability to inhibit host-derived matrix metalloproteinases (collagenase and gelatinase) and bone resorption in organ culture has also been invoked as a therapeutic rationale. The current study was undertaken to determine whether tetracyclines can inhibit alveolar bone loss in vivo due to a non-antimicrobial action of these drugs. Experimental periodontitis was induced by inoculating adult, male Sprague-Dawley rats with P. gingivalis (strain 381) following kana

Early modulatory effects of Porphyromonas gingivalis and Lactobacillus salivarius on oncogenic processes in dysplastic oral keratinocytes.

BACKGROUND: Disruption of oral homeostasis can contribute to diseases such as oral cancer. This study investigates the early modulatory effects of the probiotic Lactobacillus salivarius and the pathogen Porphyromonas gingivalis on dysplastic oral keratinocytes (DOK). METHODS AND RESULTS: DOK cells were co-cultured with each bacterium at a multiplicity of infection (MOI) of 10 for 2 and 6 h. Following co-culture, RNA was extracted to assess the relative expression of key carcinogenesis-related genes (DUSP16, PIK3CA, AKT, mTOR, ALOX5, VEGF, COX2, HEY1, JAGGED1) via qPCR. Complementary assays inc…

Porphyromonas gingivalis produces a functional HemH ferrochelatase important for its survival in a heme-limited environment.

Porphyromonas gingivalis, the keystone pathogen responsible for dysbiosis in the oral microbiome, the development of periodontal diseases, and the contribution to systemic comorbidities, is a heme auxotroph. It encodes only four enzymes in the heme biosynthesis pathway (HemD, HemN, HemG, and HemH). Comparative genomic analyses demonstrated that, while many Bacteroidota members have lost most of the canonical heme biosynthetic enzymes, Porphyromonas species uniquely retained a ferrochelatase (HemH) homolog. This study aimed to functionally characterize the P. gingivalis HemH protein to prove it…

Diversity and assembly mechanisms of Porphyromonas gingivalis fimbriae: Conservation and structural heterogeneity of type V fimbriae.

BACKGROUND: Porphyromonas gingivalis is a periodontal pathogen that primarily mediates its interactions with host tissues and other bacteria through fimbrial structures. The diversity and assembly mechanisms of its fimbriae are fundamental to understanding its pathogenicity, and its roles in periodontal and systemic diseases. This bacterium mainly expresses two types of fimbriae, FimA and Mfa1. Recent structural and genetic studies have demonstrated that these fimbriae conform to the conserved assembly principles of Type V fimbriae shared by Bacteroidia-class bacteria. HIGHLIGHTS: This review…

Role of β-adrenergic signaling in the development of Porphyromonas gingivalis lipopolysaccharide-induced cardiac dysfunction in mice.

The aims of the present study were to investigate the changes of cardiac dysfunction induced by Porphyromonas gingivalis lipopolysaccharide (PG-LPS, 0.8 mg/kg/day for a week) in mice, and to elucidate the underlying mechanisms, as well as to evaluate the effect of the non-selective β-blocker propranolol (PPL) on the PG-LPS-induced changes. Since little is known about the involvement of the sympathetic nervous system in the development or exacerbation of periodontitis-induced cardiovascular disease (CVD), we first examined the effects of PG-LPS on heart variability. The normalized low frequency…

The Effect of Probiotic Lactobacillus Strains on Porphyromonas gingivalis, the Etiological Factor of Periodontal Diseases-A Systematic Review.

Periodontal diseases are multifactorial, involving dysbiotic microbial communities and impaired immune responses. Porphyromonas gingivalis is considered the critical bacterium in their pathogenesis. Periodontal diseases are related to biofilm formation; therefore, their antimicrobial treatment is difficult. The review's aim was to summarize the effect of probiotic Lactobacillus strains on P. gingivalis. A structured research of bibliographic databases was conducted according to PRISMA 2020 guidelines. We included all eligible records from 2014 to 2024. Seventy-two studies and seventeen Lactoba…

PorA of the Type IX Secretion Is a Ligand of the PorXY Two-Component Regulatory System in Porphyromonas gingivalis.

Porphyromonas gingivalis is an important bacterium associated with chronic periodontitis. The type IX secretion system (T9SS) in P. gingivalis secretes conserved C-terminal domain (CTD) containing proteins, which are also called T9SS cargo proteins, including gingipain proteinases, to the cell surface and extracellular milieu. We have shown that gene expression of some T9SS component proteins is regulated by a two-component regulatory system, PorX-PorY, an ECF sigma factor, SigP, and a T9SS cargo protein, PorA. As PorA has its own CTD, PorA is mainly localized as an A-LPS-bound form and PorV-b…

Гипоксия стимулирует развитие множественного склероза, улучшая воспаление активации в макрофаге с инфекцией Porphyromonas gingivalis.
