--Paneth cell niche function is critical for intestinal stem cells in radiation injury but dispensable in GVHD.

Paneth cells (PCs) produce WNT3 and EGF in the small intestine and are thought to contribute to the intestinal stem cell (ISC) niche, which is a target of graft vs. host disease (GVHD). However, PC deletion models have failed to demonstrate substantial consequences for ISCs or other epithelial pathology. The physiologic role of PC niche function has thus remained opaque, and the significance of this function after bone marrow transplantation (BMT) is poorly understood. Using Lysozyme1-DTR mice to ablate PCs, we found stable WNT3 expression in vivo, and crypts from PC-ablated mice retained orga

Effect of curcumin intervention on radiation-induced injury: a GRADE-based systematic review and meta-analysis.

BACKGROUND: Radiation-induced injury (RII) is a common condition among patients receiving radiation therapy which affects the quality of life and clinical outcomes in cancer patients. Curcumin has shown promising therapeutic potential in the ameliorating RII through its radioprotective, anti-inflammatory and antioxidant properties. However, the findings across studies remain inconsistent. METHODS: A comprehensive systematic search of PubMed, Web of Science, Embase, Cochrane Library, CNKI, Wanfang, and VIP was conducted from inception to 2 July 2025. Risk of bias was assessed using the Cochrane

Application Mechanisms and Clinical Prospects of Brain-Computer Interface Technology in Radiation-Induced Brain Injury.

Radiation-induced brain injury (RIBI) is a common complication of radiotherapy that leads to neurological symptoms, significantly impairing patients' daily functioning and long-term rehabilitation. Consequently, the development of effective therapeutic strategies has received considerable attention. As an emerging approach in the management of neurological disorders, brain-computer interface (BCI) technology shows substantial potential for both the assessment and treatment of RIBI. This review synthesizes evidence retrieved from electronic databases and examines RIBI from the perspectives of i

Drosophila melanogaster as a Model for Gastrointestinal Radiation Injury: Conserved Mechanisms, Experimental Approaches, and Countermeasure Discovery.

The gastrointestinal tract is a critical target of acute radiation exposure. Severe radiation exposure can deplete epithelial stem and progenitor cells, compromise barrier integrity, alter host-microbe interactions, and drive fluid loss, inflammation, and systemic decline. Mammalian models remain essential for clinical translation, but their cost and complexity constrain sample sizes, statistical power, large-scale mechanistic discovery, and countermeasure screening. The adult Drosophila melanogaster midgut provides a complementary in vivo platform containing intestinal stem cells, absorptive

TO-207 Mitigates Radiation-Induced Hematopoietic Injury in a Murine Model of Lethal Total-Body Irradiation.

INTRODUCTION: Ionizing radiation exposure can cause acute radiation syndrome (ARS) with life-threatening multi-organ damage. Although several hematopoietic growth factors are FDA-approved as radiation countermeasures, alternative strategies that target inflammatory mechanisms remain urgently needed. TO-207, formerly known as JTE-607, is a selective multi-cytokine inhibitor that suppresses pro-inflammatory cytokine secretion in human monocytes and macrophages. TO-207 represents a mechanistically distinct radiation mitigator; however, whether it suppresses the acute post-irradiation cytokine sur

Chronic liver diseases aggravate radiation-induced intestinal injury through taurocholic acid dysregulation.

BACKGROUND: Chronic liver diseases (CLDs) are pervasive health issue worldwide, whereas radiation-induced intestinal injury is a frequent and consequential complication of abdominopelvic radiotherapy. Although these two pathological states often coexist clinically, the effects of CLDs on the development and severity of enteric radiation damage remain poorly understood. Therefore, the aim of this study was to investigate the effects of CLDs on radiation-induced intestinal injury and to elucidate the underlying mechanisms involved. METHODS: Mouse models of CCl4-induced liver injury or alcoholic

The role and therapeutic prospects of intercellular communication and RNA m6A modification in radiation-induced liver injury.

Radiation-induced liver injury (RILI) is a common and severe complication of radiotherapy for abdominal and thoracic malignancies. Ionizing radiation triggers widespread alterations of the N6-methyladenosine (m6A) modification landscape of hepatic RNA. Dysregulated m6A methylation disrupts hepatocyte homeostasis, immune cell activation and intercellular communication, and thus may contribute to the progression of RILI from acute inflammation to chronic hepatic fibrosis. Although intercellular crosstalk has been reported in RILI-related studies, its crosstalk with m6A modification remains poorl

Hemp seed oil mediates injury mitigation and anti-inflammation in radiated splenic T cells.

BACKGROUND: Exposure to acute radiation results in hematopoietic damage, immune defects, and organ injury, ultimately leading to severe lethality. However, few drugs or compounds have been reported to effectively mitigate the injuries induced by high-dose ionizing radiation. METHODS: To elucidate the radioprotective mechanisms of hemp seed oil against acute radiation, lethal radiation was applied to evaluate the radio-protective function. The survival rates of mice were recorded, and the immune populations, particularly T cells, in the spleen were analyzed using flow cytometry. The expression

Peripheral Blood Transcriptomic Profile Associated with Cutaneous Radiation Injury in Cancer Patients Undergoing Radiotherapy.

The peripheral blood transcriptomic profile associated with clinically observed cutaneous radiation injury (CRI) during fractionated radiotherapy remains incompletely characterized. We analyzed blood transcriptomes from 22 adults receiving radiotherapy: 10 with CRI and 12 without radiation-related skin injury at blood collection. RNA was assayed using two-color Agilent microarrays. Variance filtering retained 12,251 probes that were evaluated using four complementary analyses: preranked Gene-Set Enrichment Analysis (GSEA) for ranked gene-set enrichment, Gene-Set Variation Analysis (GSVA) for s

Gintonin enhances epithelial barrier function by activating NRF2 pathway in radiation-induced intestinal injury.

BACKGROUND: Because the intestine is a radio-sensitive organ in the body, and radiation-induced intestinal injury is a major clinical problem associated with radiotherapy or radiological accidents. Dysfunction of the epithelial barrier leads to bacterial translocation to other organs, resulting in severe inflammation. Recent findings suggest that gintonin (GT) suppresses oxidative stress and inflammation in neuroinflammatory diseases. PURPOSE: This study objected to elucidate the mitigating effects of GT on radiation-induced intestinal injury. METHODS: The therapeutic effects of GT were asse

Blockade of calcium-activated chloride channel ANO1 ameliorates ionizing radiation-induced intestinal injury.

INTRODUCTION: Radiation enteritis is one of the most frequent clinical complications of radiotherapy (RT), yet few effective strategies currently exist to protect against that. Anoctamin 1 (ANO1) functions both as a chloride channel and a signal transduction protein, influencing numerous pathophysiological processes. OBJECTIVES: This study aimed to investigate whether targeting ANO1 could mitigate radiation-induced enteritis while enhancing tumor radiosensitivity. METHODS: Quantitative PCR (qPCR) and Western blot (WB) were used to assess ANO1 expression and its changes after irradiation. Sur

Development of a noninvasive redox imaging method that can stably detect radiation-induced intestinal injury.

The intestine is an important organ in radiotherapy of the abdominal region, and radiation-induced intestinal injury (RIII) is an undesirable biological response to radiotherapy. Radiotherapy is known to induce oxidative stress associated with the generation of reactive oxygen species (ROS), which in turn plays an important role in RIII. However, these effects cannot be detected or predicted early using conventional imaging techniques such as computed tomography and magnetic resonance imaging (MRI). In this study, an intestinal redox imaging method using dynamic nuclear polarization (DNP) MRI

Discovery of a radiation countermeasure therapeutic for intestinal injury enabled by human organ chips combined with AI.

There is a need for better therapies for acute radiation injury (ARI) of the human intestine as current treatments offer limited efficacy. As the ileum is most sensitive to radiation in patients receiving cancer radiation therapy, we created human Organ Chip microfluidic culture models lined by primary patient-derived ileal epithelial cells interfaced with intestinal microvascular endothelium and exposed them to clinically relevant doses of γ-radiation. These Ileum Chips recapitulated key features of ARI, including cell loss, barrier dysfunction, and inflammation, as well as a therapeutic resp

Pathological Mechanisms of Radiation-Induced Lung Injury and Novel Nano-Drug Delivery Therapeutic Strategies.

Radiation-induced lung injury (RILI), encompassing early radiation pneumonitis (RP) and late radiation-induced lung fibrosis (RILF), remains a major dose-limiting complication of thoracic radiotherapy. The pathological processes involve oxidative stress, DNA damage, inflammatory cascades (notably IL-1, IL-6, TNF-α, and TGF-β), and fibrotic remodeling, yet current therapies-such as corticosteroids and antifibrotic agents-provide only partial relief and are often accompanied by significant side effects. Recent advances in nano-drug delivery systems (NDDS) offer new opportunities to overcome thes

Tmprss6 modulates radiation-induced liver injury through the hepcidin axis and PI3K/AKT pathway.

BACKGROUND: Radiation-induced liver injury (RILI) poses a significant challenge in abdomino-pelvic tumor radiotherapy, adversely impacting normal liver tissues. This study explores the role of Tmprss6, a gene encoding Matriptase2, in acute RILI development and its impact on hepatocyte apoptosis. METHODS: A RILI model was established using 30 Gy total liver irradiation in C57BL/6J mice. Expression of Tmprss6 and hepcidin post radiation was detected by immunohistochemistry staining, Western blot and quantitative real-time PCR. Tmprss6 knockout mice were established to demonstrate the contributi

DNAzyme hydrogels specifically inhibit the NLRP3 pathway to prevent radiation-induced skin injury in mice.

Radiation-induced skin injury (RISI) is a frequent complication of radiotherapy, yet current preventive strategies exhibit suboptimal efficacy. Our previous publications have consistently demonstrated the effectiveness of biomaterials and hydrogels in preventing RISI. Based on comprehensive literature reviews, we speculate that NLRP3 overexpression plays a central role in the development of RISI. Therefore, designing DNAzyme (DZ)-hydrogels with targeted inhibition of NLRP3 overexpression is crucial for preventing RISI.To achieve this, we designed and screened the optimal NLRP3-DZ using bioinfo

RNA-Seq Reveals Th17 Cell Differentiation Pathway as a Mechanism of Radiation-Induced Brain Injury.

Radiotherapy is a prevalent therapeutic modality for head and neck malignancies; however, it invariably results in radiation-induced damage to normal cerebral tissue, culminating in radiation-induced brain injury (RBI). Despite extensive research on radiation-induced neuroinflammation, the association between RBI and the Th17 cell differentiation pathway remains inadequately understood. C57BL/6 mice underwent a single administration of 30 Gy cranial irradiation to develop the RBI model. Cognitive function was evaluated through the Morris water maze (MWM), open field test, novel object recognit
