On the shape of the radiation survival curve in tumor spheroids: The role of oxygen heterogeneity.

BACKGROUND: The shape of cell survival curves at the tissue level remains an open question in radiobiology. While homogeneous cell populations (so-called single cells) typically exhibit an exponential decrease in survival with increasing dose, it is unclear whether this behavior persists in multicellular systems, where oxygen and nutrient gradients introduce additional complexity. PURPOSE: This study investigates how oxygen diffusion and consumption in tumor spheroids modify the aggregate radiation response and whether the resulting survival curves follow a purely exponential or a linear-quad

Cell-type-specific radiosensitivity in the human parotid gland and its implications for radiotherapy-induced dysfunction.

BACKGROUND: The human parotid gland, vital for oral health and digestion, comprises mainly acinar and ductal cells, each with unique responses to environmental stimuli such as radiation. Understanding the differential effects of radiation on these cells is crucial for therapeutic advancements in treating radiation-induced salivary gland damage. METHODS: Acinar and ductal cells were isolated from human parotid glands and cultured in both 2D and 3D environments. Morphology, proliferation capacity, and transepithelial electrical resistance (TER) were assessed to characterize each cell type. To e

Beyond DNA damage: 3D tumor models and the integrin mechanobiology of radioresistance.

Despite major advances in radiation delivery, clinical outcomes remain constrained by tumor biology rather than technology. Conventional radiobiology has relied on reductionist two-dimensional (2D) systems that fail to capture the spatial, mechanical, and multicellular organization of tumors. Three-dimensional (3D) tumor models now resolve radiation response as a mechanobiological process coordinated across the extracellular matrix (ECM), adhesion signaling, cytoskeleton, and nucleus. This review focuses on a specific and we argue, underappreciated intersection: how 3D models expose integrin-m

The mechanism of breast cancer stem cells and tumor microenvironment promoting radioresistance in breast cancer and its intervention strategies.

BACKGROUND: Breast cancer (BC) is the most common malignant tumor among women worldwide. Radiotherapy (RT) is a primary treatment modality; however, local recurrence driven by radioresistance frequently undermines its efficacy. MECHANISMS: Growing evidence indicates that breast cancer stem cells (BCSCs) and the tumor microenvironment (TME) cooperatively regulate multiple signaling pathways, thereby reducing RT efficacy. BCSCs exhibit intrinsic radioresistance through enhanced DNA repair capacity (Section 2.2.1), maintenance of redox homeostasis via the NRF2-KEAP1 and HIF-1α axes (Section 2.2.

Potential radiosensitive germline biomarkers in the testes of wild mice after the Fukushima accident.

We investigated potential germline-specific radiosensitive biomarkers in the testes of large Japanese field mice (Apodemus speciosus) exposed to low-dose-rate (LDR) radiation after the Fukushima accident. Fukushima wild mice testes were analysed via RNA-sequencing to identify genes differentially expressed in the breeding and non-breeding seasons when compared to controls. Results revealed significant changes during the breeding season, with Lsp1 showing a considerable upregulation, while Ptprk and Tspear exhibited significant reductions. Conversely, in the non-breeding season, Fmo2 and Fmo2 (

Effect of Autophagy Inhibitors on Radiosensitivity in DNA Repair-Proficient and -Deficient Glioma Cells.

Background and Objectives: The development of radioresistance is a fundamental barrier to successful glioblastoma therapy. Autophagy is thought to play a role in facilitating the DNA repair of DNA damage foci in radiation-exposed tumor cells, thus, potentially contributing to their restoration of proliferative capacity and development of resistance in vitro. However, the effect of autophagy inhibitors on DNA damage repair is not fully clear and requires further investigation. Materials and Methods: In this work, we utilized M059K (DNA-PKcs proficient) and M059J (DNA-PKcs deficient) glioma cell

Radiosensitivity in patients affected by ARPC1B deficiency: a new disease trait?

Actin-related protein 2/3 complex subunit 1B (ARPC1B) deficiency is a recently described inborn error of immunity (IEI) presenting with combined immunodeficiency and characterized by recurrent infections and thrombocytopenia. Manifestations of immune dysregulation, including colitis, vasculitis, and severe dermatitis, associated with eosinophilia, hyper-IgA, and hyper-IgE are also described in ARPC1B-deficient patients. To date, hematopoietic stem cell transplantation seems to be the only curative option for patients. ARPC1B is part of the actin-related protein 2/3 complex (Arp2/3) and coopera

Targeting Glucose Metabolism of Cancer Cells with Dichloroacetate to Radiosensitize High-Grade Gliomas.

As the cornerstone of high-grade glioma (HGG) treatment, radiotherapy temporarily controls tumor cells via inducing oxidative stress and subsequent DNA breaks. However, almost all HGGs recur within months. Therefore, it is important to understand the underlying mechanisms of radioresistance, so that novel strategies can be developed to improve the effectiveness of radiotherapy. While currently poorly understood, radioresistance appears to be predominantly driven by altered metabolism and hypoxia. Glucose is a central macronutrient, and its metabolism is rewired in HGG cells, increasing glycoly

Zymolytic Grain Extract (ZGE) Significantly Extends the Lifespan and Enhances the Environmental Stress Resistance of Caenorhabditis elegans.

Many reports have shown that grains play an important role in our daily lives and can provide energy and nutrients to protect us from various diseases, and they are considered to be indispensable parts of our lives. It has been reported that some constituents in grains could exert functional effects against HIV infections and multiple cancers. Zymolytic grain can produce some new useful molecules and thus support the cell nutrients in the human body. In this study, the effects of zymolytic grain extract (ZGE) supernatants on the changes of nematode indicators were investigated, including lifes

Laser therapy and photosensitive medication: a review of the evidence.

In the 2009 guidelines from the BMLA, the use of non-essential aesthetic lasers was contraindicated in patients receiving medication that causes whole-body photosensitisation as well as those causing local light sensitisation. Following this and anecdotal advice, many laser centres refuse to treat patients who are on known photosensitive medication. Therefore, specific patient cohorts that would benefit from laser therapy are being denied because of medications, such as long-term antibiotics for chronic facial acne. This article reviews the published literature on lasers and photosensitive med

DdrB stimulates single-stranded DNA annealing and facilitates RecA-independent DNA repair in Deinococcus radiodurans.

The bacterium Deinococcus radiodurans can survive extremely high exposure to ionizing radiation. The repair mechanisms involved in this extraordinary ability are still being investigated. ddrB is one gene that is highly up-regulated after irradiation, and it has been proposed to be involved in RecA-independent repair in D. radiodurans. Here we cloned, expressed and characterized ddrB in order to define its roles in the radioresistance of D. radiodurans. DdrB preferentially binds to single-stranded DNA. Moreover, it interacts directly with single-stranded binding protein of D. radiodurans DrSSB

Reinvestigation of the classification of five cell strains of xeroderma pigmentosum group E with reclassification of three of them.

Xeroderma pigmentosum is a photosensitive syndrome caused by a defect in nucleotide excision repair or postreplication repair. Individuals of xeroderma pigmentosum group E (xeroderma pigmentosum E) have a mild clinical form of the disease and their cells exhibit a high level of nucleotide excision repair as measured by unscheduled DNA synthesis, as well as biochemical heterogeneity. Cell strains from one group of xeroderma pigmentosum E patients have normal damage-specific DNA binding activity (Ddb+), whereas others do not (Ddb-). Using a refinement of a previously reported cell fusion complem

[The relation of circadian variations of heuristic behavior and CNS radioresistance in animals].

There has been studied the influence of g-radiation (60Co, 62.5 Gy, craniocaudal) on circadian dynamics of heuristic behaviour (the elements of rational-discriminative activity) of male white rats. There has been found equivocal nature of radiation action: mostly manifestations of some symptoms of neurologic disturbances observed in definite daily periods make difficult realizing behavioural act, but in the other cases such event is not observed (acrophases of both processes coincide). After disappearance of observed neurologic manifestations of central nervous system damage (symptoms of early

Establishment of two permanent human bone marrow stromal cell lines with long-term post irradiation feeder capacity.

We describe the establishment of two permanent Simian virus 40-transformed human stromal cell lines, designated L87/4 and L88/5, derived from the bone marrow of a hematologically normal male patient. Both cell lines show a fibroblastoid morphology and do not express hematopoietic cell markers. L87/4 but not L88/5 expresses the macrophage marker CD68. The most remarkable feature of these new stromal cell lines is their ability to persist as growth-arrested adherent feeder cells after ionizing-irradiation at doses up to, and exceeding 20 Gy (L87/4). This renders them particularly useful for stud

Changes in normal and abnormal colony formation of thyroid cells in vivo, when transplanted at 1 day or 6 weeks after X-irradiation in situ.

Thyroid cells were given 5.5 Gy X-rays in situ and transplanted after 1 day or after 6 weeks to form colonies in fat pads. The colony-forming efficiency unexpectedly decreased by a factor of about 2 with this increase in delay time before transplantation. In addition, there was a concomitant marked increase above the control levels in the proportion of new structures of thyroid origin containing abnormal follicles. These quantitative and qualitative increases in injury may be related to the gradual expression of thyroid abnormalities following irradiation in situ, and they may have implication

Cellular radiosensitivity in ataxia-telangiectasia.

Hypersensitivity to both the cell-killing and chromosome-damaging effects of ionizing radiations, and other agents causing DNA breakage, is a consistent feature of cells from individuals with the cancer-prone disorder ataxia-telangiectasia (A-T). Evidence for a defect in DNA strand break rejoining is slight, but a higher-than-normal level of chromosomal breaks persists in irradiated A-T cells. There is also evidence for elevated frequencies of DNA recombination and deletion mutation in A-T cells; these responses may be linked through a loss of fidelity in rejoining DNA breaks through recombina

Inhibition and recovery of DNA synthesis in human tumor cell lines following radiation exposure.

Eight human tumor cell lines with radiosensitivities (D0) ranging from 1 to 3 Gy were analyzed for their response to radiation-induced inhibition of DNA synthesis. These cell lines differ in their sensitivity to induction of DNA double-strand breaks and in the rate at which they rejoin DNA double-strand breaks. Fifty-gray doses of gamma rays induced between 35 and 75% inhibition in rates of DNA synthesis. The magnitude of the inhibition was not related to cellular radiosensitivity, frequency of initial DNA double-strand breaks, or the rate of rejoining of DNA double-strand breaks. All the cell

Management of iatrogenic flat-back syndrome.

Sacral tumors represent a small subset of spinal lesions and typically include chordomas, metastases, other primary bone tumors, and benign schwannomas. Resection is the standard treatment for many sacral tumors, but many types of sacral lesions have the potential for recurrence after excision. In these cases, adjuvant radiotherapy is often beneficial. Although conventional radiotherapy plays an important role in the management of spinal lesions, the radiation doses required for adequate local control of many sacral lesions generally exceed the tolerance doses of normal tissues, thus limiting

Clinical outcomes among patients treated with Lu-PSMA-617 and metastasis-directed EBRT for metastatic prostate cancer.

Metastasis-directed external beam radiation therapy (EBRT) delivered for limited-volume disease, integrated with systemic radioligand therapy, may improve long-term disease control across the spectrum of metastatic prostate cancer. We examined 88 patients with metastatic prostate cancer who received both lutetium-177 PSMA-617 (LuPSMA) radiopharmaceutical therapy and metastasis-directed external beam radiation therapy (EBRT) between June 2022 and August 2025. EBRT was delivered to a median of three metastatic sites, most commonly bone (69.3 %) and lymph nodes (64.8 %). The PSA50 response rate t

Neck dissection prior to radiation therapy for squamous cell carcinoma of tongue base.

BACKGROUND: Squamous cell carcinoma of the tongue base can be treated with comparable control and survival rates when neck dissection (ND) is performed before radiation therapy (RT). METHODS: Fourteen patients were treated between 1990 and 2000. Tumor stage on presentation was: T2, 6; T3, 4; T4, 4. Average radiation dose was 7,268 cGy. Follow-up ranged from 2 to 11 years (median, 50 months). Ten patients (71.4%) who presented with cervical metastases underwent neck dissection. All neck dissections were performed prior to RT. RESULTS: The initial local control rate was 85.7%. The 10-year surv
