We encountered a child with an intraosseous small round cell tumor that was negative for LCA, CD20 (L26), and CD3 and positive for vimentin, CD99 (MIC-2), and periodic acid-Schiff. The tumor exhibited rosette-like formations. This case was initially interpreted as Ewing's sarcoma (ES); however, additional studies revealed positivity for CD79a, CD43, and TdT expression, and an immunoglobulin heavy chain gene rearrangement (IgH-R) by polymerase chain reaction (PCR) established this to be a precursor B-lymphoblastic lymphoma. Because the differential diagnosis of ES and lymphoblastic lymphoma can

In a 9 year old boy, a destructive lesion in the diaphysis of the right femur was wrongly diagnosed as a Ewing's sarcoma on the basis of the radiologic findings and fine needle aspiration cytology report. The clinical and radiologic picture was suggestive of Ewing's sarcoma, but an open biopsy of the lesion revealed a histopathological pathological picture of eosinophilic granuloma. A brief review of the literature is given, together with discussion on the differential diagnosis of a mid shaft femoral lesion in young patients.

The results of a pilot study using adjuvant chemotherapy and sequential half-body irradiation (HBI) for nonmetastatic Ewing's sarcoma are presented. Seventeen patients received Cyclophosphamide, Vincristine, and Adriamycin (8 cycles), followed by sequential radiation treatment of the upper (500 cGy) and lower (600 cGy) half body. Survival at 3 years was 49%. These results are contrasted with those for 18 concurrently treated patients who received standard adjuvant therapy. Overall 5-year survival and relapse-free survival for these 35 consecutive patients was 61 and 53%. The pilot protocol was

A rare case of a primary Ewing's sarcoma arising from the intracranial portion of the right orbital roof is presented. Clinical presentation was that of a rapidly growing tender exostosis, associated with headaches. History indicated that the tumor may have been present for more than 1 year. Skull x-rays showed marked diastasis of the coronal suture and a "mottled" appearance on the right frontal bone suggestive of a sarcoma. Bone scan further confirmed the presence of a malignant bone tumor. Computerized tomography scan showed the intracranial extension of the tumor and a follow-up scan allow

BACKGROUND: Ewing sarcoma (ES) is one of the most frequent sarcomas in children and young adults. Dose-intensified induction with vincristine, doxorubicin, cyclophosphamide, ifosfamide and etoposide (VDC/IE) proved to be more efficient than vincristine, ifosfamide, doxorubicin and etoposide (VIDE) and has emerged as a new standard. Here, we analyzed peripheral blood (pB) and stem-cell yields of adult patients after the induction with VDC/IE and compared them with historic control after VIDE induction.
METHODS: Stem-cell collections of 6 adult ES patients (all male) after VDC/IE were analyzed.

UNLABELLED: Genes encoding ETS family transcription factors are altered by chromosomal rearrangement in 60% to 70% of prostate cancers and nearly all Ewing sarcomas. Ewing sarcoma rearrangements result in chimeric fusion of ETS proteins to the RNA-binding protein EWSR1. Prostate cancer rearrangements result in aberrant expression of ETS proteins such as ETV1, ETV4, ETV5, or ERG that can interact with wild-type EWSR1, suggesting common mechanisms between these diseases. In this study, we find that ETV1, ETV4, and ETV5 can phenocopy EWSR1::FLI1 in Ewing sarcoma cell lines. However, rescue of EWS

Clinical features, aside from disease stage, have proven insufficient to identify patients at extremely high or low risk. There are few established biological markers for EwS. The discovery of new prognostic and/or predictive biomarkers could improve our understanding of tumor heterogeneity, facilitate individual risk stratification, and aid targeted therapies. We initiated a pan-European study using formalin-fixed, prospectively collected, primary tumors prior to chemotherapy from 335 patients in two clinical trial registries, EURO-Ewing 99 and Ewing 2008, to analyze correlation and survival

OBJECTIVE: To evaluate the prognostic and predictive value of metabolic and volumetric parameters obtained from baseline (PET1) and post-chemotherapy (PET2) 18 F-FDG PET/CT in pediatric Ewing sarcoma (EWS) patients, focusing on their associations with histopathologic response, metastatic presentation, overall survival (OS), and progression-free survival (PFS).
METHODS: This retrospective single tertiary academic center study included 55 pediatric EWS patients who underwent PET1. Twenty-four patients also had PET2. SUVmax, SUVpeak, metabolic tumor volume (MTV), total lesion glycolysis (TLG) an

Ewing sarcoma (EwS) shows a limited clinical response to poly (ADP-ribose) polymerase (PARP) inhibitors (PARPi), despite promising preclinical data. In this study, we compared five PARPi with different PARP-trapping capacities in PDX-derived cell lines and mouse models. Talazoparib, the strongest PARP-trapping agent, showed markedly greater efficacy than olaparib or veliparib. It triggered extensive DNA damage, micronuclei formation, and activation of the cyclic GMP-AMP synthase (cGAS)/stimulator of interferon genes (STING) pathway, leading to robust type I interferon and pro-inflammatory cyto

Extra-skeletal Ewing sarcoma (EES) is an extremely rare condition, with only 10 cases reported worldwide. The challenge in this case remains the diagnosis, which relies on immunohistochemical and molecular testing: small round blue cells, CD99-positive expression and the presence of an EWSR1/FLI1 gene fusion. We herein report a 43-year-old male with a primary penile Ewing sarcoma, without apparent symptoms, and whose tumor type was difficult to identify on biopsy, with no definite histological diagnosis established before final surgery. A partial penectomy and adjuvant chemotherapy with vincri

Neuroblastoma and Ewing sarcoma are two round cell malignancies that can exhibit light microscopic overlap and potentially arise in the same anatomic location, leading to diagnostic uncertainty. Neoadjuvant chemotherapy-induced cytodifferentiation/maturation is a common feature in pediatric malignancies, whereas Ewing sarcomas often exhibit variable degrees of necrosis, inflammatory infiltrates, and stromal fibrosis after neoadjuvant treatment. Posttreatment neuroblastoma-like morphologic changes have been rarely reported in pediatric Ewing sarcoma. Herein, we report a 33-year-old man with a r

Primary Ewing sarcoma of the kidney (ESK) is an exceptionally rare and aggressive malignancy, representing less than 1% of renal tumors. It typically affects young adults and often presents at an advanced stage, leading to a poor prognosis. Diagnosis requires a multidisciplinary approach integrating histopathology, immunohistochemistry, and molecular analysis. A 29-year-old male patient presented with a one-and-a-half-year history of right flank pain and one year of painless, intermittent hematuria. Initial management at a local hospital for a suspected inflammatory lesion was unsuccessful. A

Ewing sarcoma, the second most common primary bone cancer in children, requires intensive treatment that may lead to significant long-term sequelae, particularly in infants. We retrospectively analyzed data from 1621 French patients treated between 1988 and 2015 within the EW88/93/97 or EE99 trials, focusing on 17 infants diagnosed before 24 months of age (1% of the cohort). Their outcomes were compared to the 1245 older pediatric patients (aged 2-18 years). Infant Ewing sarcomas were more frequently extraosseous (29% vs. 8%, p = 0.0037). Acute chemotherapy tolerance was comparable between inf

Ewing sarcoma (EWS) is a rare bone cancer that is most usually detected in children. Neutrophil extracellular traps (NETs) are closely related to the prognosis of cancer, but the significance of NET-related features in EWS remains uncertain. We constructed a NET signature utilizing four crucial NET-related genes in EWS to forecast prognosis and investigate the potential immunological value of this signature in EWS. EWS data were collected from the International Cancer Genome Consortium and Gene Expression Omnibus databases. We identified the subtypes mediated by NET-related genes in EWS and an

This letter aims to provide a forward-looking analysis of the recent preclinical study by Ou et al. (Angiogenesis, 2025) on the efficacy of the multi-kinase inhibitor KC1036 in Ewing sarcoma (ES).We conducted a critical appraisal of the reported data, focusing on the dual anti-angiogenic and direct anti-tumor mechanisms of KC1036. The analysis is contextualized within the current understanding of ES pathogenesis and treatment resistance. The original study compellingly demonstrates that KC1036, by concurrently inhibiting VEGFR and FGFR signaling, effectively suppresses ES growth. While these fi

INTRODUCTION AND IMPORTANCE: Multiple primary malignancies (MPMs) involve two or more distinct primary cancers in one individual, either simultaneously or at different times. The incidence of MPMs is rising due to advancements in cancer detection, improved survival rates, and long-term treatment effects. This case report, likely the first of its kind, highlights a rare instance of a 30-year-old female developing a carcinoid tumor 5 years after Ewing sarcoma, emphasizing the need for vigilant monitoring of cancer survivors.

INTRODUCTION: Sarcoma as a cause of laryngeal cancer is rare and is even rarer to have an Ewing sarcoma out of the bone presents as laryngeal cancer. In this report, we present this extremely rare case.
CASE PRESENTATION: A 41-year-old man was seen at the ENT clinic complaining of chronic hoarseness and a lump in his neck. Flexible laryngoscopy showed a large mass occupying the left side of the larynx and then a computerized tomography scan proved it. For further evaluation, the laryngoscopy was performed and the frozen section revealed a malignancy. Consequently, the surgical decision was ta

Intracranial myxoid mesenchymal tumor, FET::CREB fusion positive is a rare, recently described central nervous system neoplasm. It is characterized by EWSR1::CREB family transcription factor fusion, typically arises in children and adolescents, and is locally aggressive even after gross total resection. Currently, there are little data available to guide management and gauge long-term prognosis. Furthermore, there have been no reports of these lesions occurring simultaneously with other intracranial neoplasms or in patients with a history of malignancy. Here we describe the first case of a ver
