Residual structure and dynamics in Parkinson's disease-associated mutants of alpha-synuclein.

alpha-Synuclein (alpha S) is a pre-synaptic protein that has been implicated as a possible causative agent in the pathogenesis of Parkinson's disease (PD). Two autosomal dominant missense mutations in the alpha S gene are associated with early onset PD. Because alpha S is found in an aggregated fibrillar form in the Lewy body deposits characteristic of Parkinson's patients, aggregation of the protein is believed to be related to its involvement in the disease process. The wild type (WT) and early onset mutants A30P and A53T display diverse in vitro aggregation kinetics even though the gross ph

Expression of mutant alpha-synuclein causes increased susceptibility to dopamine toxicity.

Mutations of the alpha-synuclein gene have been identified in autosomal dominant Parkinson's disease (PD). Transgenic mice overexpressing wild-type human alpha-synuclein develop motor impairments, intraneuronal inclusions and loss of dopaminergic terminals in the striatum. To study the mechanism of action through which mutant alpha-synuclein toxicity is mediated, we have generated stable, inducible cell models expressing wild-type or PD-associated mutant (G209A) alpha-synuclein in human-derived HEK293 cells. Increased expression of either wild-type or mutant alpha-synuclein resulted in the for

Both familial Parkinson's disease mutations accelerate alpha-synuclein aggregation.

Parkinson's disease (PD) is a neurodegenerative disorder that is pathologically characterized by the presence of intracytoplasmic Lewy bodies, the major component of which are filaments consisting of alpha-synuclein. Two recently identified point mutations in alpha-synuclein are the only known genetic causes of PD, but their pathogenic mechanism is not understood. Here we show that both wild type and mutant alpha-synuclein form insoluble fibrillar aggregates with antiparallel beta-sheet structure upon incubation at physiological temperature in vitro. Importantly, aggregate formation is acceler

Genetic polymorphism in the persyn (gamma-synuclein) gene and the risk of Alzheimer's disease.

Alzheimer's disease (AD) is a complex disease with the possible involvement of several genes. Genetic studies on sporadic late-onset AD have determined APOE*4 to be the major risk factor. Members of the synuclein gene family are potential candidates for the risk of AD. The persyn gene (gamma-synuclein) has recently been characterized and a common polymorphism (Glu110Val) has been identified. In this study we investigated the association of this polymorphism with sporadic late-onset AD patients. We screened DNA samples of 313 late-onset cases and 352 controls. No significant association was obs

The NACP/synuclein gene: chromosomal assignment and screening for alterations in Alzheimer disease.

The major component of the vascular and plaque amyloid deposits in Alzheimer disease is the amyloid beta peptide (A beta). A second intrinsic component of amyloid, the NAC (non-A beta component of amyloid) peptide, has recently been identified, and its precursor protein was named NACP. A computer homology search allowed us to establish that the human NACP gene was homologous to the rat synuclein gene. We mapped the NACP/synuclein gene to chromosome 4 and cloned three alternatively spliced transcripts in lymphocytes derived from a normal subject. We analyzed by RT-PCR and direct sequencing the

Mechanistic insights into the inhibition of α-synuclein aggregation by sugar-based nanoparticles.

The pathological accumulation of α-synuclein (α-syn) into amyloid fibrils is a key hallmark of Parkinson's disease and related synucleinopathies. Inhibiting the early stages of α-syn aggregation remains a major therapeutic challenge. In this work, we report the fabrication and investigation of five sugar-based nanoparticles (NPs), including glucose, fructose, maltose, sucrose, and trehalose, and their ability to inhibit α-synuclein aggregation. Using a combination of biophysical strategies, comprising thioflavin-T fluorescence, dynamic light scattering, circular dichroism, and confocal microsc

Target engagement and efficacy in disease-modifying trials of synuclein-related disorders.

Disease-modifying trials in Parkinson's disease (PD), multiple system atrophy (MSA), and dementia with Lewy bodies (DLB) increasingly incorporate fluid and imaging biomarkers to demonstrate target engagement, de-risk subsequent phases of drug development and provide mechanistic evidence of biological activity. Whether biomarker modulation predicts clinical benefit remains unclear. We conducted a PRISMA-compliant review of phase II and III disease-modifying trials (2020-2025) that included at least one biomarker intended to assess target engagement. Twenty-six trials met inclusion criteria (PD=

Membrane Phase, Charge, and Curvature Regulate α-Synuclein Binding Dynamics.

α-Synuclein (αSyn) is an intrinsically disordered protein whose interactions with lipid membranes are central to both its physiological function and its role in synucleopathies. While membrane charge, phase, and curvature are each known to influence αSyn binding, these properties are typically examined independently, leaving their combined effects on both equilibrium and dynamic membrane association unresolved. Here, we systematically investigate how membrane phase and charge jointly regulate αSyn binding, curvature sensitivity, and exchange dynamics using fluorescence microscopy, circular dic

Aggregation and membrane activity of mutant A30P alpha-synuclein on mitochondrial membranes.

The A30P point mutation in the α-synuclein (α-syn) protein is linked to a familial form of Parkinson's disease (PD), the most common motor neurodegenerative disorder. The association of α-syn with lipids in cellular membranes plays a key role in modulating the aggregation behavior of the protein, as well as its toxicity. Here, we studied the impact of the A30P mutation on the interaction of α-syn with mitochondrial membranes, since the interplay between α-syn aggregation and mitochondrial dysfunction is central for the pathogenic process in PD, but still not completely understood. Experiments

AlphaB-crystallin modified by methylglyoxal prevents fibrillization of α-synuclein A53T.

AlphaB-crystallin is a small heat shock protein that is found in the eye lens, retina, heart, brain, skin, and skeletal muscle. Mutations of αB-crystallin lead to the development of cataracts, human muscle and heart disorders. The role of non-enzymatic glycation of αB-crystallin in the development of pathological processes has been little studied. The aim of this study was to evaluate the influence of enhanced methylglyoxal (MGO) level on the functions of αB-crystallin. We modified recombinant αB-crystallin (aB-Cr) by MGO and investigated the effect of this modification on different functions

Leveraging Supramolecular Polymers to Induce the Targeted Protein Degradation of α-Synuclein.

Halting the progression of neurodegenerative diseases remains one of the foremost challenges in medicinal chemistry due to the complex biology that drives disease progression. For example, a hallmark of synucleinopathies, such as Parkinson's disease, is the misfolding and aggregation of the protein α-Synuclein (α-Syn), driving the formation of toxic oligomers and fibrils that avoid natural intracellular clearance mechanisms, participate in unusual protein-protein interactions, and ultimately contribute to the death of dopaminergic neurons. The field of targeted protein degradation (TPD) has em

p53 regulates mitochondrial function and alpha-synuclein aggregation in Parkinson's disease.

Parkinson's disease (PD) is the second most common neurodegenerative disease, in which mitochondrial dysfunction and abnormal aggregation of alpha-synuclein (α-syn) play key roles in the pathology of PD. As a classic tumor suppressor, p53 has also been found to be involved in the pathological process of PD in recent years. However, the specific mechanism by which p53 regulates mitochondrial function and abnormal aggregation of α-syn is still unclear. Here, we observed that the expression of α-syn and p53 was increased and mitochondria were impaired in the MPTP-induced PD mouse model, leading u

Early α-Synuclein Heterodimerization Kinetics Predict Parkinson's Disease Onset.

Identifying physicochemical predictors that connect protein aggregation to clinical phenotypes remains challenging. Here, we examine whether simulated aggregation descriptors of the intrinsically disordered protein α-synuclein (αS) relate to the clinical age of onset in Parkinson's disease. Using a multiscale simulation framework, we combine coarse-grained (CG) models to quantify the kinetics of early dimerization with atomistic simulations to estimate dimer interaction energies, as well as CG thermodynamic protofilament binding free energies complemented by atomistic calculations. These analy

Divergent effects of pathological α-synuclein truncations and mutations on phase separation.

Phase separated condensates of α-synuclein (α-Syn) accelerate amyloid fibril formation, a process implicated in Parkinson's disease pathogenesis. Yet, the precise effects of pathologically relevant α-Syn sequence modifications on this process remain unclear. Here, we show that sequence truncations exert the strongest influence on condensate thermodynamics, material properties, and amyloid aggregation, whereas familial point mutations impart minimal effects. Among the tested familial variants (A30P, H50Q, G51D and A53T), only G51D forms condensates that show a reduced propensity for amyloid fib

Alpha-synuclein quantitative seed amplification assay predicts conversion to dementia.

INTRODUCTION: The alpha-synuclein seed amplification assay (SAA) has shown excellent performance in the detection of Lewy body pathology in cerebrospinal fluid (CSF). Lewy body pathology is prognostically relevant in patients at risk for dementia. Current assays only provide binary results, so there is a need to quantify the extent of pathology in living patients. METHODS: In addition to the "standard" SAA, we developed a quantitative SAA (qnSAA) and measured 432 CSF samples (216 baseline-follow-up pairs). RESULTS: qnSAA results correlated with cognitive performance. Seventy-five percent of
