The choroid plexus (CP) epithelial cells establish an important blood-brain interface, the blood-cerebrospinal fluid barrier (BCSFB), which constitutes a complementary gateway to the blood-brain-barrier for the entrance of several molecules into the central nervous system (CNS). However, the mechanisms that operate at the BCSFB to regulate the molecular traffic are still poorly understood. The taste signalling machinery, present in many extra-oral tissues, is involved in the chemical sensing of the composition of body fluids. We have identified this pathway in rat CP and hypothesised that it c…

One of the most striking reported symptoms in CoViD-19 is loss of smell and taste. The frequency of these impairments and their specificity as a potential central nervous system function biomarker are of great interest as a diagnostic clue for CoViD-19 infection as opposed to other similar symptomatologic diseases and because of their implication in viral pathogenesis. Here severe CoViD-19 was investigated by comparing self-report vs. testing of smell and taste, thus the objective severity of olfactory impairment and their possible correlation with other symptoms. Because a significant discrep…

Pigments often inflict tissue-damaging and proaging toxicity on light illumination by generating free radicals and reactive oxygen species ( … e those photochemical parameters. Collectively, nucleophile sensitivity of TRPA1(A) allows flies to taste potential phototoxins for feeding deterrence, preventing postingestive photo-injury. Conversely, pigments need to bear high nucleophilicity (electron-donating propensity) to act as type 1 phototoxins, which is consistent with the fact that transferr …

Bitter taste receptors (T2Rs) are expressed in several tissues of the body and are involved in a variety of roles apart from bitter taste perception. Advanced glycation end-products (AGEs) are produced by glycation of amino acids in proteins. There are varying sources of AGEs, including dietary food products, as well as endogenous reactions within our body. Whether these AGEs are T2R ligands remains to be characterized. In this study, we selected two AGEs, namely, glyoxal-derived lysine dimer (GOLD) and carboxymethyllysine (CML), based on their predicted interaction with the well-studied T2R4,…

Over recent years, the presence of the sweet taste receptor TIR3 in rodent and human insulin-producing pancreatic islet β-cells was documented. The activation of this receptor by sweet-tasting sucralose mimics several biochemical and functional effects of D-glucose in the β-cells. The present study extends this analogy to the bioelectrical response of β-cells. In this respect, sucralose was inefficient in the absence of D-glucose, but induced on occasion electrical activity in mouse β-cells exposed to low non-stimulatory concentrations of the hexose and potentiated, in a concentration-related …

Bitter taste receptors (T2Rs) in the human airway detect harmful compounds, including secreted bacterial products. Here, using human primary sinonasal air-liquid interface cultures and tissue explants, we determined that activation of a subset of airway T2Rs expressed in nasal solitary chemosensory cells activates a calcium wave that propagates through gap junctions to the surrounding respiratory epithelial cells. The T2R-dependent calcium wave stimulated robust secretion of antimicrobial peptides into the mucus that was capable of killing a variety of respiratory pathogens. Furthermore, sweet…

BACKGROUND: Taste disorders are frequently observed in cancer patients undergoing chemotherapy and are serious adverse events which impair the quality of life (QoL) of the cancer patient. Nevertheless, taste disorder mechanisms in cancer patients undergoing chemotherapy have not yet been fully elucidated. The aim of this study was to reveal taste disorder-related peripheral mechanisms using the two-bottle preference test (TBPT) and histological examination of tongues by hematoxylin-eosin staining and immunohistochemistry with protein-gene product 9.5.
METHODS: In the TBPT, one bottle was fill…

The adaptive nature of taste discrimination has been questioned on the basis that marked differences in taste thresholds for soluble sugars across mammals may not correspond to distinct dietary tendencies and further imply inaccurate prediction of the energetic value of food. Appropriate models of the evolution of taste, however, require not only consideration of adaptations to diet but also recognition of phylogenetic and allometric effects. Here, we analyse these factors in primates exhibiting various diets and covering a wide range of body weights. Taste thresholds for sucrose and fructose …

Human neonates are occasionally treated with diuretics, and we investigated whether this causes a long-term enhancement of salt preference. Salt preference was examined in children aged 4-11 years. Twenty one of the children had received furosemide therapy as preterm neonates, and 24 were preterm neonates from the same ward that had no furosemide therapy. No differences were found between the two groups in preferred concentration of NaCl in soup, in consumption of salty items, and in blood and urine sodium and creatinine. However, in a tested subsample, fractional excretion of sodium (FENa) wa

BACKGROUND: Sporadic observations suggest a possible taste impairment in AL amyloidosis, but the frequency and intensity of this sensory anomaly are not known.
MATERIALS AND METHODS: We submitted 21 AL amyloidotic patients, drawn from subjects referred to the Amyloidosis Study Center of the Department of Internal Medicine and Medical Therapy, University of Pavia, to suprathreshold scaling analysis.
RESULTS: Taste acuity was reduced in most of them. Every taste showed independent behavior, and 90% of these patients were hypogeusic. True ageusia for one or two tastes was observed in 35% of pat…

A prognostic test in cases of Bell's palsy must be accurate and applicable early in the course of the disease. The case history is examined in 239 patients with Bell's palsy. The prognostic value of the case history, taste impairment and sialometry is investigated in a series of untreated patients who were followed up for 12 months. Sialometry is advocated as a reliable progostic indicator in the individual patient if a strong stimulant is used. None of the other examined parameters discrimates between good and poor outcome with reasonable accuracy. A follow-up time of 12 months is advocated i…

Food intake was measured in 22 obese patients before and after jejunioleostomy for obesity. Most of the weight loss could be accounted for by the observed reduction of caloric intake. Malabsorption was also present as indicated by increased loss of fat in the stools, and decreased absorption of D-xylose and vitamin B12. A dislike for sweet tastes developed after surgery in most patients. Preferences for concentrated solutions of sucrose and glucose were reduced after patients showed a depression of food intake by a 440-calorie preload which had not been detected before surgery. These studies s…

Organoleptic properties are important factors in the development of pharmaceutical formulations, as they directly impact patient adherence and, consequently, treatment efficacy. Cannabinoid-based oils often possess unpleasant taste and texture, which can compromise treatment compliance. This preliminary proof-of-concept study aimed to develop oil-in-water (O/W) emulsions utilizing cannabidiol (CBD) and cannabigerol (CBG) rich extracts, evaluate their physicochemical stability under accelerated conditions, and perform an instrumental taste-related characterization. The formulations exhibited pH…

This study investigated the effect of Penicillium (Penicillium aethiopicum & Penicillium chrysogenum, PP), yeast (Candida parapsilosis & Rhodotorula mucilaginosa, CM) and mixture (MI) inoculation on the taste development of Jinhua ham. The results showed that Penicillium and yeast significantly increased acidic protease activity and reduced TVB-N values compared to the control. Three kinds of inoculation promoted the degradation of both myofibrillar and sarcoplasmic proteins, leading to a accumulation of peptides and free amino acids. It was strongly detected by the electronic tongue sensors P…

Sweet taste is a crucial chemosensory modality for detecting natural sugar compounds, which are primarily derived from angiosperms. In vertebrates, excluding birds, sweet taste is typically mediated by the Tas1r2-Tas1r3 heterodimer, and the receptor function often reflects dietary adaptations to sugar-rich diets. To gain insight into early vertebrate dietary transitions, we identified Tas1r genes in 58 vertebrate species and one outgroup and conducted functional assays in 10 representative species spanning six major clades, including one coelacanth, two amphibians, one squamate, two turtles, t…

Taste receptor family 1 (T1R) and taste receptor family 2 (T2R) are G protein-coupled receptors (GPCRs) responsible for sweet, umami, and bitter taste perception. Their widespread ectopic expression of T1Rs and T2Rs in extraoral tissues suggest important physiological functions beyond taste. However, their roles in immune regulation and mucosal immunity have not been comprehensively summarized. We aimed to review the current evidence on the roles of T1Rs and T2Rs in immune cell function, respiratory and intestinal mucosal immunity, and disease susceptibility. Relevant studies investigating the…
