Maturity of judgement in decision making for predictive testing for nontreatable adult-onset neurogenetic conditions: a case against predictive testing of minors.

International guidelines developed to minimize harm from predictive testing for adult-onset, nontreatable neurogenetic conditions such as Hu … ity found that the 3 factors involved in maturity of judgement in decision making - responsibility, temperance and perspective - continue to develop into late adolescence. There is also evidence that the prefrontal areas of the brain, which are involved in executive functions such as decision making, are not fully developed until early adulthood. Combined wi …

[Alcohol-induced hypertriglyceridemia in subjects with decreased lipoprotein lipase--improvement after cessation of alcohol intake].

We systematically investigated the effect of lipoprotein lipase (LPL) and alcohol intake on the development of primary type IV hyperlipoproteinemia which is manifested as a moderate increase in serum triglycerides as a consequence of an elevated very low density lipoprotein concentration and a normal level of serum cholesterol. Twenty six type IV hyperlipoproteinemic and 28 normolipidemic males underwent measurement of the immunoreactive LPL mass in postheparin plasma (30 unit of heparin/kg of body weight) and were interviewed as to their alcohol consumption. They were divided into 4 groups, w

Improved method for acetaldehyde in plasma and hemoglobin-associated acetaldehyde: results in teetotalers and alcoholics reporting for treatment.

A fluorigenic high performance liquid chromatographic (HPLC) assay for the determination of acetaldehyde in plasma and hemoglobin-associated acetaldehyde is described. The assay is based on the reaction of acetaldehyde with two molecules of 1,3-cyclohexanedione in the presence of ammonium ion to form a fluorescent species followed by reverse phase HPLC separation of specific aldehyde derived compounds. The assay is specific; and has sensitivity in the picomole range, with intraassay precision of less that 3.5% and interassay precision of less than 15%. The total run time is less than 6 minutes

Clinical features of multiple sclerosis in a genetically homogeneous tropical population.

Individuals affected with multiple sclerosis (MS) from a genetically homogeneous Caucasian population in Antioquia, a tropical region of Colombia, were evaluated in order to observe the clinical behavior of the disease. The frequency of clinical manifestations in 65 patients with definite MS from Antioquia was compared with those reported from temperate regions. The most common manifestations were optic neuritis and motor symptoms with absence of cerebellar symptoms. This presentation is significantly different from the frequency distribution at onset in series from temperate regions. These di

Fatal human melioidosis in south-eastern Queensland.

Two simultaneous human cases of fatal melioidosis in temperate south-eastern Queensland involved patients who had had pre-existing multisystem illnesses, had sustained cutaneous lesions before illness onset, and died from overwhelming sepsis. Onset of disease was preceded by unseasonably heavy rainfall. These and other features of these cases suggest that the source of infection was local, in which case the endemicity of Burkholderia pseudomallei in temperate regional Australia may be broader than is currently recognised, and melioidosis may need to be considered in at-risk patients in these a

Pf Bacteriophage and Their Impact on Pseudomonas Virulence, Mammalian Immunity, and Chronic Infections.

Pf bacteriophage are temperate phages that infect the bacterium Pseudomonas aeruginosa, a major cause of chronic lung infections in cystic fibrosis (CF) and other settings. Pf and other temperate phages have evolved complex, mutualistic relationships with their bacterial hosts that impact both bacterial phenotypes and chronic infection. We and others have reported that Pf phages are a virulence factor that promote the pathogenesis of P. aeruginosa infections in animal models and are associated with worse skin and lung infections in humans. Here we review the biology of Pf phage and what is kno

Genotype patterns and evolutionary rates: uncovering Japanese encephalitis virus spread across Asia's climate regions.

Japanese Encephalitis Virus (JEV) is a highly endemic zoonotic virus, consistently found in Asia and parts of the Western Pacific, and it's a major cause of human encephalitis. JEV belongs to a family of antigenically related viruses such as West Nile Virus (WNV), Murray Valley encephalitis virus (MVEV), and Aichi Lake Fever Virus (ALFV) and is transmitted by mosquitoes. Persistent outbreaks of the disease necessitate detailed studies to understand their transmission dynamics and develop effective prevention strategies. This study explores the evolutionary dynamics and spatial transmission of

Burn size and environmental conditions modify thermoregulatory responses to exercise in burn survivors.

This project tested the hypothesis that burn survivors can perform mild/moderate-intensity exercise in temperate and hot environments without excessive elevations in core body temperature. Burn survivors with low (23 ± 5%TBSA; N = 11), moderate (40 ± 5%TBSA; N = 9), and high (60 ± 8%TBSA; N = 9) burn injuries performed 60 minutes of cycle ergometry exercise (72 ± 15 watts) in a 25°C and 23% relative humidity environment (ie, temperate) and in a 40°C and 21% relative humidity environment (ie, hot). Absolute gastrointestinal temperatures (TGI) and changes in TGI (ΔTGI) were obtained. Participant

A comparative analysis of Aedes albopictus and Aedes aegypti subjected to diapause-inducing conditions reveals conserved and divergent aspects associated with diapause, as well as novel genes associated with its onset.

Aedes albopictus and Aedes aegypti are mosquito species that are distributed worldwide and transmit diverse arboviruses of medical importance, such as those causing yellow fever, dengue, chikungunya and Zika. A. albopictus embryos may remain viable for long periods in the environment due to their ability to become dormant through quiescence or diapause, a feature that contributes to their dispersion and hinders control actions. Diapause incidence can vary among natural populations of A. albopictus, but metabolic and genetic parameters associated with its induction still need to be better defin

Phage Milagro: a platform for engineering a broad host range virulent phage for Burkholderia.

Burkholderia infections are a significant concern in people with CF and other immunocompromising disorders, and are difficult to treat with conventional antibiotics due to their inherent drug resistance. Bacteriophages, or bacterial viruses, are now seen as a potential alternative therapy for these infections, but most of the naturally occurring phages are temperate and have narrow host ranges, which limit their utility as therapeutics. Here we describe the temperate Burkholderia phage Milagro and our efforts to engineer this phage into a potential therapeutic by expanding the phage host range

Non-migratory lineages experience higher diversification rates: global evidence from salmonid fishes.

Biologists have long sought to understand the forces driving speciation. Contrasts in dispersal ability (e.g., migration) are hypothesized to influence rates of lineage diversification. However, past inferences of migration-diversification relationships have been contradictory, suggesting the need for further investigation and more nuanced explanations. Here, we test for a relationship between diadromy (marine-freshwater migration) and diversification in salmonids, a dominant group of fishes native across the temperate Northern Hemisphere. We reconstruct a molecular phylogeny comprising 140 sp

The effect of harvest time across different climatic regions on the accumulation of phenolic compounds in Marselan grapes and wines.

Harvest time is a critical determinant of grape and wine quality. This study investigated the influence of harvest time on the phenolic compounds and physicochemical properties of Marselan grapes and wines across two climatic regions during the 2022 and 2023 vintages. Five sequential harvests (S1-S5) were conducted in Shandong (warm temperate monsoon climate), whereas three (N1-N3) were implemented in Ningxia (moderate temperate continental climate). In Shandong, harvest at S4 optimized the accumulation of total phenolic content (TPC), total flavan-3-ols (TFOC), total flavonoid content (TFC) a

Genetic Engineering of Lysogenic-Lytic Switch Genes Improves Burkholderia Phage Killing Efficacy.

Burkholderia pseudomallei, the causative agent of melioidosis, presents significant challenges in both treatment and environmental decontamination. Bacteriophages, or phages, are increasingly being explored as potential diagnostic, therapeutic, and biocontrol agents against this bacterial pathogen. Our recent investigation has shown that most B. pseudomallei genomes contained prophage(s) associated with specific tRNA gene loci, prompting us to explore these detectable prophages as sources of temperate phages for further applications. Transcriptomic profiling of B. pseudomallei Bp82, a model st

Global gram-negative diabetic foot infection prevalence and its associations with climate: a systematic review and meta-analysis.

BACKGROUND: The International Working Group on the Diabetic Foot guidelines state that Pseudomonas aeruginosa and overall gram-negative diabetic foot infections (psaDFI and gnDFI) are more common in tropical and subtropical zones vs. temperate zones, and in North Africa/Asia vs. Western Europe/North America based on non-systematic comparisons of cohort studies. METHODS: We performed a systematic review and meta-analysis of studies of any design and language published between January 1, 2010, and December 15, 2020, that reported the proportion of participants with DFIs and microbiological test

Management of alcoholic hepatitis.

About 5,000 people die each year from chronic liver disease in England and Wales alone. In many patients, the liver injury is due to chronic excessive alcohol consumption and manifests as alcoholic hepatitis (an acute inflammation of the liver), cirrhosis, or alcoholic hepatitis superimposed on a background of cirrhosis. Mild alcoholic hepatitis may be asymptomatic and reversible, but where the hepatitis is more severe, up to 65% of those affected die from it. The incidence of alcoholic hepatitis in the UK seems set to increase with the rise in heavy drinking, particularly among women. Here we
