Orally available designed miniproteins inhibit enterotoxigenic Bacteroides fragilis pathology by blocking toxin receptor binding.

Toxigenic bacterial infections in the gut are a significant contributor to the global burden of disease. Advanced tools for protein and live biotherapeutic engineering offer potentially transformative strategies for treating such diseases, while avoiding the collateral effects of traditional antibacterials. Here we used de novo protein design to identify inhibitors of the metzincin family protease Bacteroides fragilis toxin (BFT). These inhibitors, which bind distal to the active site, interfere with toxin-mediated E-cadherin cleavage and downstream proinflammatory signaling by blocking claudi

A 5-transcript signature for discriminating viral and bacterial etiology in pediatric pneumonia.

Pneumonia stands as the primary cause of death among children under five, yet current diagnosis methods often result in inadequate or unnecessary treatments. Our research seeks to address this gap by identifying host transcriptomic biomarkers in the blood of children with definitive viral and bacterial pneumonia. We performed RNA sequencing on 192 prospectively collected whole blood samples, including 38 controls and 154 pneumonia cases, uncovering a 5-transcript signature (genes FAM20A, BAG3, TDRD9, MXRA7, and KLF14) that effectively distinguishes bacterial from viral pneumonia (area under th

Phenotypic Tracking of Antibiotic Resistance Spread via Transformation from Environment to Clinic by Reverse D2O Single-Cell Raman Probing.

The rapid spread of antibiotic resistance threatens our fight against bacterial infections. Environments are an abundant reservoir of potentially transferable resistance to pathogens. However, the trajectory of antibiotic resistance genes (ARGs) spreading from environment to clinic and the associated risk remain poorly understood. Here, single-cell Raman spectroscopy combined with reverse D2O labeling (Raman-rD2O) was developed as a sensitive and rapid phenotypic tool to track the spread of plasmid-borne ARGs from soil to clinical bacteria via transformation. Based on the activity of bacteria

A Half-Century Perspective on the Microbial Bioremediation of PCBs.

Microbial biotransformation of PCBs was first observed in the field and in laboratory studies nearly five decades ago. Following years of investigation (on both aerobic biodegradation or mineralization and anaerobic dechlorination), bioremediation technologies are emerging for PCB-contaminated sediments. This perspective examines the arc of scientific and engineering developments that eventually brought us to today's approach of combining anaerobic and aerobic metabolism in an in situ bioaugmentation application. Although an understanding of aerobic biodegradation of lesser chlorinated bipheny

Optimization of nitrogen removal and microbial communities in biofloc systems via carbon-to-nitrogen ratio regulation and Candida utilis.

Biofloc-based systems provide a sustainable strategy for nitrogen management in intensive aquaculture, but their efficiency depends strongly on carbon availability and microbial community regulation. In this study, we evaluated the combined effects of carbon-to-nitrogen (C: N) ratio regulation and Candida utilis supplementation on nitrogen removal and microbial community dynamics in a biofloc-based Chinese soft-shelled turtle culture system. Three C: N ratios (10, 15, and 20) were established using glucose as the carbon source, with or without C. utilis addition. Water quality parameters, feed

In Search of a Mechanistic Link between Chlamydia trachomatis-Induced Cellular Pathophysiology and Oncogenesis.

Centrosome duplication and cell cycle progression are essential cellular processes that must be tightly controlled to ensure cellular integrity. Despite their complex regulatory mechanisms, microbial pathogens have evolved sophisticated strategies to co-opt these processes to promote infection. While misregulation of these processes can greatly benefit the pathogen, the consequences to the host cell can be devastating. During infection, the obligate intracellular pathogen Chlamydia trachomatis induces gross cellular abnormalities, including supernumerary centrosomes, multipolar spindles, and d

Biotreatment potential of co-contaminants hexavalent chromium and polychlorinated biphenyls in industrial wastewater: Individual and simultaneous prospects.

Co-existence of polychlorinated biphenyls (PCBs) and hexavalent chromium (Cr(VI)) in the environment due to effluent from industries has aggravated the pollution problem. Both contaminants can alter chemical interactions, processes and impair enzymatic activities in the ecosystem that results in negative impacts on aquatic and terrestrial life. Previously, research has been performed for the fate and transfer of these contaminants individually, but simultaneous removal approaches have not received much attention. Cr(VI) exists in a highly toxic form in the environment once released, whereas lo

Optimizing bacterial expression of catalytically active human cytochromes P450: comparison of CYP2C8 and CYP2C9.

1. Methods for the co-expression in Escherichia coli of human cytochrome P450 (CYP) 2C8 and CYP2C9 with NADPH-cytochrome P450 reductase (OxR) to produce a catalytically active system were compared. 2. Approaches assessed were expression of a CYP:OxR fusion construct, bicistronic plasmids, simultaneous transformation with CYP and OxR plasmids, and separate expression of CYP and OxR with reconstitution of activity by mixing the bacterial membranes. Two N-terminal modifications (Delta3-20 and 17alpha-leader) of the individual P450s were additionally investigated. 3. Each approach gave efficient e

Integrating acoustic microfluidics with spectroscopic analysis for efficient bacterial lysis and molecular characterisation.

Rapid and efficient bacterial lysis is crucial for point-of-care diagnostics, environmental monitoring, and integrated analytical platforms. However, conventional lysis methods often involve trade-offs between lysis efficiency, biomolecule integrity, and workflow complexity. Here, we present a reagent-free, non-thermal acoustofluidic platform that uses bulk acoustic waves (BAWs) to rapidly lyse both Gram-negative and Gram-positive bacteria via high energy acoustic streaming and localised shear forces. The method achieved lysis efficiencies of ∼82 % for Escherichia coli and ∼50 % for Enterococc

Primary involvement of pharynx and peyer's patch in inhalational and intestinal anthrax.

Bacillus anthracis causes three forms of anthrax: inhalational, gastrointestinal, and cutaneous. Anthrax is characterized by both toxemia, which is caused by secretion of immunomodulating toxins (lethal toxin and edema toxin), and septicemia, which is associated with bacterial encapsulation. Here we report that, contrary to the current view of B. anthracis pathogenesis, B. anthracis spores germinate and establish infections at the initial site of inoculation in both inhalational and cutaneous infections without needing to be transported to draining lymph nodes, and that inhaled spores establis

The multifunctional LigB adhesin binds homeostatic proteins with potential roles in cutaneous infection by pathogenic Leptospira interrogans.

Leptospirosis is a potentially fatal zoonotic disease in humans and animals caused by pathogenic spirochetes, such as Leptospira interrogans. The mode of transmission is commonly limited to the exposure of mucous membrane or damaged skin to water contaminated by leptospires shed in the urine of carriers, such as rats. Infection occurs during seasonal flooding of impoverished tropical urban habitats with large rat populations, but also during recreational activity in open water, suggesting it is very efficient. LigA and LigB are surface localized proteins in pathogenic Leptospira strains with p

Processes and their explanatory factors governing distribution of organic phosphorous pools in lake sediments.

The amount of organic phosphorus (OP) and its distribution among different pools in lake sediments depends on biotic and abiotic processes driving the OP fractionation. Key environmental factors governing these transformations processes between OP fractionations in sediments were studied on the basis of geochemical characteristics of OP pools in relation to environmental factors in the sediments. The results illustrate that the factors influencing the accumulation or depletion of different OP pools were intrinsically dependent on the composition of the deposited organic matter (OM). During the

Transfer of plasmid-borne beta-lactamase in Neisseria gonorrhoeae.

Neisseria gonorrhoeae strain GC82 contains a plasmid specifying a beta-lactamase (beta-Lam(+)). Mixed incubation of strain GC82 with a penicillin-susceptible (beta-Lam(-)), streptomycin-resistant mutant of strain GC9 results in the expression of beta-lactamase activity and streptomycin resistance in the transcipients. The frequency of transfer of the plasmid-specified resistance to penicillin seems to be proportional to the initial input ratio of the mating mixture of donor to recipient and to correlate positively with bacterial density. Cell-to-cell transmission of the deoxyribonucleic acid (

Lymphoma and the control of B cell growth and differentiation.

It is now widely accepted that lymphomagenesis is a multistep transformation process. A number of genetic changes and environmental and infectious factors contributing to the development and malignant progression of B-cell lymphoproliferative disorders are well documented. Reciprocal chromosomal translocations involving the immunoglobulin loci are a hallmark of most mature B cell lymphomas and lead to dysregulated expression of proto-oncogenes (c-myc) important for cell proliferation or genes involved in cell cycle progression (cyclin D1), differentiation block (bcl-6, PAX5) and cell survival

Selenium stable isotope ratios in California agricultural drainage water management systems.

Selenium stable isotope ratios are known to shift in predictable ways during various microbial, chemical, and biological processes, and can be used to better understand Se cycling in contaminated environments. In this study we used Se stable isotopes to discern the mechanisms controlling the transformation of oxidized, aqueous forms of Se to reduced, insoluble forms in sediments of Se-affected environments. We measured 80Se/76Se in surface waters, shallow ground waters, evaporites, digested plants and sediments, and sequential extracts from several sites where agricultural drainage water is pr

Hermansky-Pudlak syndrome: an immunologic assessment of 15 cases.

PURPOSE: The Hermansky-Pudlak syndrome is an autosomal recessive disorder consisting of the triad of oculocutaneous albinism, a storage pool platelet defect, and multisystem tissue deposition of ceroid pigment. Although the underlying metabolic defect has not been identified, secondary or associated effects on the immune system are suggested by reports of an association with disorders such as pulmonary fibrosis, granulomatous colitis, lupus, and frequent bacterial infections. We evaluated a large group of patients with the Hermansky-Pudlak syndrome to assess immune competence in this condition

Enhancement of antibacterial properties of α-tricalcium phosphate/honey biocement using glucose oxidase.

The antibacterial properties of a simple cement composite based on powdered α-tricalcium phosphate phase and liquid component containing honey and glucose oxidase were studied on Escherichia coli and Staphylococcus aureus. The content of active glucose substrate and glucose oxidase in the composites was around 2 wt% and up to 10 ppm, respectively. The applied amounts were sufficient to produce hydrogen peroxide at effective concentration levels (up to 800 μM) against the studied bacterial strains with low cytotoxicity to osteoblastic cells. The results showed strong adsorption of glucose oxida
