Early development of the pineal photoreceptors prior to the retinal differentiation in the embryonic rainbow trout, Oncorhynchus mykiss (Teleostei).

The development of the pineal and retinal photoreceptors in the embryonic rainbow trout, Oncorhynchus mykiss was investigated by means of light and electron microscopy. The pineal photoreceptors endowed with photoreceptive outer segments and signal-transmitting synapses appeared first 15 days after fertilization, and well developed by 21 days. In contrast, the retinal photoreceptors appeared as late as 27 days after fertilization (1 day before hatching). This temporal disparity in photoreceptor development has been compared with the photoperiodic formation of otolith increments, which is initi…

The effects and physiological consequences of raised levels of cortisol on rainbow trout (Oncorhynchus mykiss) erythrocyte beta-adrenoreceptors.

We have investigated the influence of cortisol on the beta-adrenoreceptor population of rainbow trout [Oncorhynchus mykiss (Walbaum)] erythrocytes and determined what impact it has on the adrenergic responsiveness of erythrocytes in vitro to exogenous catecholamines. To do so, the erythrocyte beta-adrenoreceptors were characterized in fish with chronically elevated plasma cortisol levels (118 +/- 5.9 ng ml-1, greater than 10 days) and compared with shams, using radioreceptor assay techniques. The number of 'internalized', low-affinity receptors was increased when cortisol levels were raised, b…

Intracellular and extracellular acid-base status and H+ exchange with the environment after exhaustive exercise in the rainbow trout.

Exhaustive exercise induced a severe short-lived (0-1 h) respiratory, and longer-lived (0-4 h) metabolic, acidosis in the extracellular fluid of the rainbow trout. Blood 'lactate' load exceeded blood 'metabolic acid' load from 1-12 h after exercise. Over-compensation occurred, so that by 8-12 h, metabolic alkalosis prevailed, but by 24 h, resting acid-base status had been restored. Acid-base changes were similar, and lactate levels identical, in arterial and venous blood. However, at rest venous RBC pHi was significantly higher than arterial (7.42 versus 7.31). After exercise, arterial RBC pHi…

A study of the effects of hypoxia on acid-base status of rainbow trout blood using an extracorporeal blood circulation.

(1) The use of an extracorporeal blood circulation method has made it possible to obtain information on the time course of possible modifications in acid-base status of trout blood during hypoxia. (2) Two series of experiments are described: (a) Continuous measurement of arterial pH, PCO2 and PO2 during 24 h of moderate hypoxia (60 Torr). (b) Continuous measurement of acid-base status during 24 h of deep hypoxia (40 Torr). (3) The results of the first series show that hypoxia provokes a stimulation of ventilatory frequency and amplitude which is associated with an increased output of CO2 from …

Relative biopotency of dietary ergocalciferol and cholecalciferol and the role of and requirement for vitamin D in rainbow trout (Salmo gairdneri).

A growth assay was conducted for six consecutive 28-day periods by using triplicate groups of 110 rainbow trout with an average initial body weight of 3.0 g. Ergocalciferol (vitamin D2 or D2) and cholecalciferol (vitamin D3 or D3) were included to provide levels of 200, 400 and 800 iu/kg in a semipurified casein, gelatin diet. Further treatments with 0 vitamin D and 1600 IU/kg of D3 were also included. The resulting growth curves were significant for parallelism. Statistical analysis showed that D3 was 3.27 times as potent as D2 (limits 2.33 to 4.58). The dietary requirement for D3 was found t…

Morphological study on the regeneration of the retina in the rainbow trout after ouabain-induced damage: evidence for dedifferentiation of photoreceptors.

The retina of the rainbow trout is capable of marginal regeneration after ouabain-induced degeneration (intraocular injection of 5 microliters 10(-4) M ouabain). In the central area where the pigment epithelium proliferates to a multicellular layer, the neural retina does not regenerate up to 182 days after injection of ouabain. The regeneration process begins in the marginal growth zone with an increase in the mitotic rate; the growth zone itself is not damaged after ouabain administration. The proliferate differentiates with time into a newly layered retina; this portion of the retina is cal…

Virus-susceptibility of juvenile rainbow trout (Oncorhynchus mykiss) exposed to sodium fluoride: immune and thyroid hormone system investigations.

Endocrine disruptors (EDs) are widely distributed in terrestrial and aquatic ecosystemsand are increasingly recognized for their potential impacts on organism health. Exposure to EDs is known to affect endocrine systems, such as the thyroid hormone system (THS), but can also manipulate the immune system (IS) of organisms. This study aimed to investigate the potential impact of the ED sodium fluoride (NaF) on the THS and pathogen resistance in rainbow trout exposed for seven months to 1.5 or 5 mg/L of fluoride (F-). Viral resistance was assessed by subjecting the fish to an infectious challenge…

Blanching pretreatment enhances puffing quality of rainbow trout (Oncorhynchus mykiss) skin by modifying protein structure and water distribution.

The high production of farmed rainbow trout (Oncorhynchus mykiss) generates large amounts of underutilized by-products, particularly skin. Its dense structure restricts water mobility, resulting in a hard texture and poor crispness in puffed products. To address this issue, blanching was employed as an efficient and economical pretreatment to modify protein structure and regulate water distribution. By combining pre-drying with puffing, intact puffed fish skin was obtained. Blanching at 60 °C yielded the highest expansion ratio (202.56 ± 3.51%) and optimal crispness (0.26 ± 0.01 mm). Criticall…

Xenobiotic Metabolizing Enzyme Response of Rainbow Trout (Oncorhynchus mykiss) to Benzophenone-3 and N,N-diethyl-m-toluamide.

Benzophenone-3 (BP-3) and N,N-diethyl-m-toluamide (DEET) are ingredients of personal care products. Despite the frequent occurrence of both compounds in aquatic environments and the fact that fish are exposed to these chemicals together, their combined effects on xenobiotic metabolizing enzymes and antioxidant enzyme activities have not been studied in aquatic organisms. The aim of this study was to determine the effects of the combination of BP-3 and DEET on cytochrome P450 and antioxidant system in rainbow trout (Oncorhynchus mykiss). For this purpose, juvenile male rainbow trout samples wer…

Anti-inflammatory effects of taurocholic acid and tauroursodeoxycholic acid from rainbow trout spleen extract via NF-κB suppression.

Inflammation is an essential innate immune response, yet excessive or persistent inflammation contributes to chronic inflammatory diseases. Our previous studies demonstrated that rainbow trout spleen extracts possess potent anti-inflammatory activity, primarily concentrated in the butanol fraction (OSB), through suppression of NF-κB signaling and macrophage M1 polarization. However, the complex composition of OSB and the requirement for high effective concentrations limited its translational potential. In this study, we try to identify the key bioactive components responsible for OSB's anti-in…

Comparison of anticoagulant vacutainer blood collection tubes on rainbow trout (Oncorhynchus mykiss) leukocyte viability during long-term storage.

Peripheral blood leukocytes (PBL) are critical for understanding systemic immune responses and assessing the organism's health. In immunological studies, particularly with aquaculture-relevant species like Rainbow trout (Oncorhynchus mykiss), ensuring PBL viability during sample storage is important, especially when samples must be transported over long distances. Anticoagulants are essential for preventing blood clotting and preserving cellular integrity; however, their effects on leukocyte populations in salmonids remain poorly studied. This study aimed to evaluate the impact of four anticoa…
