BACKGROUND: Aquaporin-1 (AQ-1) is a transmembrane water channel protein reportedly expressed in continuous capillary endothelium and intestinal lacteals. We investigated endothelial AQ-1 expression in rat intestine and mesentery, and also in lymph nodes.
METHODS AND RESULTS: Rat intestine, mesentery, and lymph nodes were immunolabeled for AQ-1, revealing membrane expression in endothelial cells of vascular continuous capillaries and venules, and of initial and conducting lymphatics. Blood vessel profiles were identified with RECA-1 and circulating FITC-albumin. In nodes, capillaries and high …

P-selectin antibody has been shown to prevent microvascular damage after ischemia reperfusion (I/R). We investigated whether the treatment w … the protective effects of anti-P-selectin treatment with marked leukocyte adhesion on postcapillary venules and platelet-endothelial cell interactions in arterioles and venules and decreased capillary perfusion at reperfusion, thus suggesting that the mechanism of I/R injury is not critically dependent on P-selectin.

L-selectin regulates lymphocyte migration by mediating lymphocyte attachment to high endothelial venules of peripheral lymph nodes (PLN). L-selectin-deficient mice display a 70 to 90% reduction in the number of lymphocytes within PLNs and a 30 to 55% increase in spleen cellularity. The altered distribution of lymphocyte subpopulations in L-selectin-deficient mice resulted in significantly elevated serum IgM and IgG1 levels and augmented humoral immune responses to T cell-independent and T cell-dependent Ags following i.p. immunization. By contrast, s.c. immunization of L-selectin-deficient mic…

The ability of lymphocytes to recognize and bind to high endothelial venules (HEVs) is essential for lymphocyte migration from the blood into lymphoid tissues and into sites of inflammation. Endothelial cell binding capacity also critically determines the clinical usefulness of T-cell lines and clones in immunotherapy. In the present study, interleukin-2-dependent T-cell lines were derived from the blood, lamina propria of the gut, inflamed synovium, synovial fluid, and peripheral lymph nodes. After 3-8 weeks of culture, the expression of homing-associated molecules and binding to mucosal, syn…

To determine the effect of lung inflation and left atrial pressure on the hydrostatic pressure gradient for fluid flux across 20- to 60-microns-diam venules, we isolated and perfused the lungs from newborn rabbits, 7-14 days old. We used the micropuncture technique to measure venular pressures in some lungs and perivenular interstitial pressures in other lungs. For all lungs, we first measured venular or interstitial pressures at a constant airway pressure of 5 or 15 cmH2O with left atrial pressure greater than airway pressure (zone 3). For most lungs, we continued to measure venular or inters…

There is growing evidence that microvascular dysfunction is a pathology accompanying various injuries and conditions that produce chronic pain and may represent a significant contributing factor. Dysfunction that occurs within each component of the microvasculature, including arterioles, capillaries and venules impacts the health of surrounding tissue and produces pathology that can both initiate pain and influence pain sensitivity. This mini review will discuss evidence for a critical role of microvascular dysfunction or injury in pathologies that contribute to chronic pain conditions such as…

Leukocytoclastic vasculitis, also known as hypersensitivity angiitis, is a cutaneous, small vessel vasculitis of the dermal capillaries and venules. The predominant clinical presentation is palpable purpura. Multiple medications can cause leukocytoclastic vasculitis, as well as autoimmune diseases, infections, and malignancy. The disease process may be limited to only the skin or a manifestation of a systemic vasculitis or process. Treatment is centered on symptom management. Our patient is a 60-year-old female who presented with bilateral dry and wet tender ulcerations. She was previously tre…

Erythema induratum (EI), or nodular vasculitis (NV), is a type of panniculitis that is often associated with vasculitis affecting various-sized veins, venules, and arteries in reaction to various causative factors. Historically, EI was highly linked to tuberculosis, but in 1946, Montgomery first proposed the term NV to describe cases of EI not associated with tuberculosis. Only 2 reports of NV associated with inflammatory bowel disease have been reported in the literature. The authors report a 60-year-old woman with Crohn's disease presenting with exacerbation of NV in the setting of vedolizum…

Budd Chiari syndrome is an uncommon heterogeneous group of disorders which occur due to obstruction at any level from the hepatic venules to the junction of inferior vena cava and right atrium of heart which has significant morbidity and mortality. An early diagnosis of the disease is required for appropriate treatment. Due to the diffuse nature of the disease, normal biopsy findings do not exclude the disease. Proper clinical history and imaging are essential for definitive diagnosis. In this pictorial essay, we discuss the imaging spectrum of Budd Chiari syndrome.

Wegener granulomatosis is a rare systemic idiopathic disease characterized by involvement of small vessels - medium and small arteries, venules, arterioles and ocasionally large arteries. This disease has predilection for the upper and lower respiratory tract and the kidney, with granulomatous inflamation and necrosis. Clinical manifestations and organ involvement of the disease vary widely. Early diagnosis and treatment may lead to a full recovery. Without treatment, Wegener's granulomatosis can be fatal. The authors present a case of a 33 year-old female, with severe disease, but with good o…

Telangiectasia macularis eruptiva perstans (TMEP) is an uncommon form of cutaneous mastocytosis that occurs exclusively in adults. Histologically, TMEP presents with scattered mast cells lined up around the dilated capillaries and venules of the superficial vascular plexus. In some cases, the number of mast cells falls within the range observed in normal skin and therefore cannot be detected by routine histologic examination. We used immunohistochemical staining for c-kit (CD 117) for the definitive diagnosis in two patients with TMEP. One of them was successfully treated with topical applicat…

Extensive and severe hepatic centrilobular hemorrhagic necrosis is a common finding in hepatic vein obstruction and Budd-Chiari syndrome. Some drugs, including allopurinol, can also cause this histopathologic appearance but to our knowledge in this setting the lesions are not so massive. Here we report a case of a 41-year-old female who developed fever, pruritic skin rash, jaundice, eosinophilia, abnormal liver function tests, and acute renal failure 3 weeks after the beginning of allopurinol treatment, complicated with severe hepatocyte necrosis around most terminal hepatic venules suggesting…

A 50-year-old man with cancer-associated retinopathy was investigated using light and electron microscopy, immunofluorescence studies, and western blotting. He had visual disturbance, ring-like scotoma, and night blindness bilaterally. There were narrowed retinal arterioles and dilated retinal venules. Oral corticosteroid therapy had positive effects. Immunostaining using the patient's serum revealed a positive reaction in the ganglion cell layer of normal retina. Western blotting showed that the patient's serum antibody reacted with normal retinal proteins of 24 and 48 kDa. Multiple metastase…

The permeability of the microcirculation to native and glycosylated albumin was tested in 25 non-diabetic Syrian hamsters. The microvasculature of the cheek pouch was studied by a fluorescent video-microscopy technique after the animals had been injected with fluorescent native or glycosylated albumin or with both. Native albumin remained in the cheek pouch microvasculature, whereas glycosylated albumin leaked out of the microvascular bed along the postcapillary and collecting venules. The extravascular leakage of glycosylated albumin, probably due to its electrical or conformational change, m…

This report describes two patients with epithelioid hemangioendothelioma presenting with multiple tumour nodules in the liver, but without evidence of tumour elsewhere. One patient died in liver failure twelve months after presentation. The other patient has been well for over two years. Histologically the tumours were remarkably similar, being composed of a proliferation of primitive endothelial cells embedded in a myxohyaline stroma, infiltrating central veins, hepatic sinusoids and, to a lesser extent, terminal portal venules. In each case an initial mistaken histological diagnosis of veno-…

Capillary malformation (CM), or port wine birthmark, is a congenital low-flow vascular malformation of dilated capillaries and postcapillary venules. It often affects the face and may darken, thicken, or develop nodules over time. Most CMs are isolated but can be associated with syndromes such as Sturge-Weber, characterized by a facial CM, leptomeningeal vascular malformation, and ocular findings such as glaucoma. CMs also occur in other syndromes, emphasizing the need for thorough clinical evaluation. Management is multidisciplinary, with pulsed-dye laser as the gold standard treatment; excis…

Microscopic polyangiitis (MPA) is a systemic disorder that affects small vessels, such as arterioles, venules or capillaries. Cerebrovascular disease has been rarely reported in connection with MPA; this complication is usually associated with fatal hemorrhage or hemorrhagic conversion. We report a case of MPA with multiple bilateral non-hemorrhagic cerebral infarctions in a 66-year-old woman who was undergoing steroid pulse therapy. The diagnosis of MPA was based on the presence of painful mononeuritis multiplex, pulmonary fibrosis, and increased myeloperoxidase activity and on the biopsy of …

The histological morphology of AILD-type T-cell lymphoma shows proliferation of small, arborizing high endothelial venules. Patients typically have generalized lymphadenopathy, fever, weight loss, skin rash, polyclonal hypergammaglobulinemia and autoimmune phenomena, and are susceptible to infections. The clinical course is usually aggressive. About 30% of patients have an initial remission on steroids alone, but most require some form of cytotoxic chemotherapy. Median survival ranges from 15 to 24 months, and it appears that the disease cannot be cured. We have used anti-angiogenetic therapy …
