LCT-EF258 with S17I Mutation in DprA Exhibits Horizontal Gene Transfer Deficiency After Spaceflight.

BACKGROUND: Space is a special environment in which microgravity and cosmic rays are the primary factors that induce gene mutations of microorganisms. In our previous studies, a single point mutation in the gene dprA was found in an Enterococcus faecium strain of LCT-EF258 after spaceflight. DNA processing protein A (DprA) plays a prominent role in the horizontal transfer of genes among bacteria (such as Streptococcus pneumoniae, Helicobacter pylori, Bacillus subtilis, and Rhodobacter capsulatus). However, the function of DprA in E. faecium remains unknown. Furthermore, E. faecium could acquir

Space flight-associated neuro-ocular syndrome (SANS).

Interesting novel and somewhat perplexing physiologic and pathologic neuro-ocular findings have been documented in astronauts during and after long duration space flight (LDSF). These findings collectively have been termed the "space flight-associated neuro-ocular syndrome" (SANS). The National Aeronautics and Space Administration (NASA) in the United States has meticulously and prospectively documented the clinical, ultrasound, optical coherence tomography imaging, and radiographic findings of SANS including unilateral and bilateral optic disc edema, globe flattening, choroidal and retinal fo

[Neuromuscular status of children of different gestational age on the stage of transition from intrauterine immersion to the earth's gravity].

The work was aimed at describing the neuromuscular status of premature baby in the context of the ontogenetic and zero gravity model using the results of superficial interference electromyography (IEMG). Throughout six postnatal weeks, IEMG of premature babies is similar to EMG of full-term child on the first days of extrauterine life; IEMG is characterized by a "simplified" temporal structure, low amplitude and frequency, IEMG dynamics of fullterm child is slow in contrast to premature baby; the reason seems to be maximum long intrauterine life during which the motor system gets better prepar

Role of afferent input in muscle atrophy.

It is well known that soleus muscle of rat atrophies following spaceflight or hindlimb suspension (Ohira et al., 1992). It is, further, reported that the electromyogram (EMG) of soleus muscle disappears immediately in response to unloading by exposure to actual micro-g environment (Kawano et al., 2002; Leterme and Falempin, 1998) and by hindlimb suspension of rats (Alford et al., 1987; Ohira et al., 2000). However, the EMG level is increased gradually to the control level following 7-10 days of continuous hindlimb suspension (Alford et al., 1987; Ohira, 2000), while muscle atrophy is progressi

The venous tone is not altered after three-week tail suspension in rats.

Cardiovascular deconditioning observed in humans during spaceflight has been suggested to be related in part to changes in venous compliance, mechanisms including skeletal muscle deconditioning. However, increased venous compliance was observed during very short term simulations (24 to 48 hours), and during an over 28-day simulation the hyperdistensibility tended to decrease whereas the muscular changes were still present (2). In the first case, muscular changes can not explain the venous alterations because of the short delay. In the second case, the relationship between muscular and venous a

Altered cell function in microgravity.

Physiological changes in humans during spaceflight upon return to earth have been attributed to systemic adaptation, response to stress, and lack of normal exercise. Studies from the Skylab, SL-3, and D-1 missions have demonstrated that significant physiological alterations are seen in single cell prokaryotes and eukaryotes, as well as in animal tissues. Basic cellular functions such as electrolyte concentration, cell growth rate, glucose utilization, bone formation, response to growth stimulation, and exocytosis are modified in microgravity. Many of the physiological changes seen in humans, v

Exploring Radiomics Features Based on H&E Images as Potential Biomarkers for Evaluating Muscle Atrophy: A Preliminary Study.

Radiomics features have been widely used as novel biomarkers in the diagnosis of various diseases, but whether radiomics features derived from hematoxylin and eosin (H&E) images can evaluate muscle atrophy has not been studied. Therefore, this study aims to establish a new biomarker based on H&E images using radiomics methods to quantitatively analyze H&E images, which is crucial for improving the accuracy of muscle atrophy assessment. Firstly, a weightless muscle atrophy model was established by laying macaques in bed, and H&E images of the shank muscle fibers of the control and bed rest (BR)

Integrated physiological characteristics and proteomes of Bacillus licheniformis response to 6-month spaceflight.

Bacillus licheniformis (BL) is used for clinical treatment of acute and chronic enteritis and diarrhea. The present study explored the physiological characteristics and proteomes of BL response to 6-month spaceflight aboard the China Space Station. 6-month spaceflight delayed BL lag phase, reduced cell wall thickness, and impaired BL biofilm formation and adhesion capacity to Caco-2 cells. After 6-month spaceflight, activity of β-amylase, alkaline protease and lipase in BL was increased by 21.6%,67.3% and 42.5%. The alkaline resistance of BL at pH 11.0 was increased by 29.6%. ibSLS as a biobank for democratizing access to multi-omics data and biospecimens from spaceflight research.

During spaceflight, astronauts are exposed to extreme conditions such as microgravity, cosmic radiation, and confinement, which can cause a wide range of health problems. To elucidate the molecular mechanisms underlying these issues and to develop intervention strategies for maintaining physiological homeostasis during space missions, space life science research using mouse models is actively conducted on the International Space Station (ISS). However, because of the high cost and technical complexity of spaceflight experiments, it is essential to maximize the scientific value of each mission

A computational model for retinal haemodynamics under gravitational and postural variations.

Spaceflight-associated neuro-ocular syndrome presents a significant risk during long-duration spaceflight, yet the retinal mechanisms underlying these visual changes remain poorly characterized. We developed a physiologically detailed lumped-parameter model coupling a five-compartment retinal vascular network with a dynamic intraocular pressure module responsive to hydrostatic shifts. Head-down tilt experiments were reproduced to emulate cephalad fluid shifts in microgravity. Model outputs are closely aligned with experimental benchmarks. Intraocular pressure increased from 16.3 mmHg (upright)

Simulated Microgravity Inhibited the Proliferation of Bovine Muscle Satellite Cells.

This study aimed to assess the effects of simulated microgravity (SMG) on bovine muscle satellite cells (bSCs). The results demonstrated that bSCs maintained a fibroblast-like morphology throughout SMG exposure from day 3 to day 7. However, cells in the SMG group exhibited reduced proliferation compared to the control group, as indicated by lower cell density at both time points. In contrast, cell viability was higher in the SMG group than in the control group. Cell cycle analysis revealed a higher proportion of SMG-treated bSCs in the G0/G1 phase and a lower proportion in the S phase at day 3

Searching for Innovative Functional Foods: Correlation Between Chemopreventive Potential and Bioactive Compounds Accumulation in Brassica Sprouts Grown Under Altered Gravity Conditions.

The main aim of this study was to evaluate the effect of space-like environment on the chemopreventive activity of Brassica sprouts against thyroid cancer cells in vitro. For this purpose the sprouts of broccoli, kale, kohlrabi, and Brussels sprouts were cultivated in darkness and in microgravity for 5-7 days. Then, the sprouts' extracts were examined for cytotoxic and antiproliferative activity against thyroid cancer and normal cells. The tested microgravity environment stimulated the cytotoxic activity of kohlrabi sprouts, causing approximately 50% reduction in thyroid cancer cells' viabilit

Spaceflight-induced contractile and mitochondrial dysfunction in an automated heart-on-a-chip platform.

With current plans for manned missions to Mars and beyond, the need to better understand, prevent, and counteract the harmful effects of long-duration spaceflight on the body is becoming increasingly important. In this study, an automated heart-on-a-chip platform was flown to the International Space Station on a 1-mo mission during which contractile cardiac function was monitored in real-time. Upon return to Earth, engineered human heart tissues (EHTs) were further analyzed with ultrastructural imaging and RNA sequencing to investigate the impact of prolonged microgravity on cardiomyocyte func

Simulated microgravity reduces intracellular-free calcium concentration by inhibiting calcium channels in primary mouse osteoblasts.

Calcium homeostasis in osteoblasts plays fundamental roles in the physiology and pathology of bone tissue. Various types of mechanical stimuli promote osteogenesis and increase bone formation elicit increases in intracellular-free calcium concentration in osteoblasts. However, whether microgravity, a condition of mechanical unloading, exerts an influence on intracellular-free calcium concentration in osteoblasts or what mechanisms may underlie such an effect are unclear. Herein, we show that simulated microgravity reduces intracellular-free calcium concentration in primary mouse osteoblasts. I

Angular displacement perception modulated by force background.

We had recumbent subjects (n = 7) indicate the amplitude of imposed, passive yaw-axis body rotations in the 0, 1, and 1.8 g background force levels generated during parabolic flight maneuvers. The blindfolded subject, restrained in a cradle, aligned a gravity-neutral pointer with the subjective vertical while in an initial position and then tried to keep it aligned with the same external direction during a body rotation, lasting less than 1.5 s about the z-axis 30 degrees, 60 degrees, or 120 degrees in amplitude. All the rotations were above semicircular threshold levels for eliciting percepti

Intrathecal ligaments and nerve root tension: possible sources of lumbar pain during spaceflight.

UNLABELLED: Lumbar intrathecal ligaments have recently been demonstrated to randomly bind dorsal nerve roots to the dura within the lumbar vertebral column. Lengthening of the vertebral column and associated lumbar back pain experienced by astronauts is common in microgravity. This study was designed to investigate the relationship of lumbar intrathecal ligaments in spinal lengthening as a possible mechanism for back pain. METHODS: A two-part study was designed using 36 vertebral columns from embalmed cadavers. There were 12 vertebral columns studied in mid-sagittal section to demonstrate the

The biomedical challenges of space flight.

Space medicine has evolved considerably through past U.S. missions. It has been proven that humans can live and work in space for long durations and that humans are integral to mission success. The space medicine program of the National Aeronautics and Space Administration (NASA) looks toward future long-duration missions. Its goal is to overcome the biomedical challenges associated with maintaining the safety, health, and optimum performance of astronauts and cosmonauts. This program investigates the health effects of adaptation to microgravity: the nature of their pathologies, the effects of

Frog experiment onboard space station Mir.

Japanese tree frogs (Hyla japonica) showed unique postures and behavior during an 8-day flight to the Russian space station Mir. When floating in the air, the animals arched their back and extended their four limbs. This posture resembles that observed during jumping or parachuting of the animals on the ground. Frog sitting on a surface bent their neck backward sharply, did not fold their hind limbs completely, and pressed their abdomen against the substrate. They walked backwards in this posture. The typical posture resembles that adopted during the emetic behavior process on the ground, alth

The Frog in Space (FRIS) experiment onboard Space Station Mir: final report and follow-on studies.

The "Frog in Space" (FRIS) experiment marked a major step for Japanese space life science, on the occasion of the first space flight of a Japanese cosmonaut. At the core of FRIS were six Japanese tree frogs, Hyla japonica, flown on Space Station Mir for 8 days in 1990. The behavior of these frogs was observed and recorded under microgravity. The frogs took up a "parachuting" posture when drifting in a free volume on Mir. When perched on surfaces, they typically sat with their heads bent backward. Such a peculiar posture, after long exposure to microgravity, is discussed in light of motion sick
