Activity-dependent neuroprotective protein (ADNP) derived peptide NAP prevents musculoskeletal abnormalities in ovariectomized mice.

Estrogen is an essential hormone that critically impacts bodily and brain functions, supporting learning, memory, and motor activities. A decrease in estrogen levels is associated with cognitive decline and motor dysfunction, such as muscle weakness. While conventional hormone replacement treatments (HRT) exist, those have limitations and potentially severe side effects. NAP (davunetide) is the smallest neuroprotective peptide site of activity-dependent neuroprotective protein (ADNP), a master regulator of cognition, essential for brain formation. It is known that NAP restores ADNP activity in

A multimodality intervention to improve musculoskeletal health, function, metabolism, and well-being in spinal cord injury: study protocol for the FIT-SCI randomized controlled trial.

BACKGROUND: A spinal cord injury (SCI) is a devastating, life-changing event that has profoundly deleterious effects on an individual's health and well-being. Dysregulation of neuromuscular, cardiometabolic, and endocrine organ systems following an SCI contribute to excess morbidity, mortality and a poor quality of life. As no effective treatments currently exist for SCI, the development of novel strategies to improve the functional and health status of individuals living with SCI are much needed. To address this knowledge gap, the current study will determine whether a Home-Based Multimodalit

Low-level, high-frequency mechanical signals enhance musculoskeletal development of young women with low BMD.

UNLABELLED: The potential for brief periods of low-magnitude, high-frequency mechanical signals to enhance the musculoskeletal system was evaluated in young women with low BMD. Twelve months of this noninvasive signal, induced as whole body vibration for at least 2 minutes each day, increased bone and muscle mass in the axial skeleton and lower extremities compared with controls. INTRODUCTION: The incidence of osteoporosis, a disease that manifests in the elderly, may be reduced by increasing peak bone mass in the young. Preliminary data indicate that extremely low-level mechanical signals ar…

Transitioning to expert-in-the-loop AI explainability in orthopaedic practice: Opportunities, challenges, and outlook.

Artificial intelligence (AI) is demonstrating very successful applications in orthopaedics, ranging from medical imaging interpretation and risk prediction to surgical planning and post operative monitoring. Yet, many AI systems remain difficult to trust in real-world clinical workflows because they are often optimized for standalone accuracy rather than clinically useable transparency, human AI teamwork, and safe escalation when the AI models are uncertain. Experts-in-the-loop explainability re frames explainable AI(XAI) as a workflow capability; orthopaedic surgeons, musculoskeletal radiolog

Low-Field MRI Reduces Cost and Environmental Impact While Maintaining Diagnostic Performance in Resource-Limited Settings: A Narrative Review With Cardiovascular Focus.

High-field MRI (1.5-3 T) delivers excellent images but requires costly, energy-intensive infrastructure. Emerging low-field systems (<1 T) cut equipment and operating costs and consume far less power, making them attractive for low- and middle-income countries (LMICs). We conducted a narrative review of human studies from LMICs that reported clinical, economic or environmental outcomes of low-field MRI, paying special attention to cardiovascular applications and implementation issues in Pakistan. Across included papers, low-field scanners lowered capital expenditure, energy consumption and sit

Glucosamine as a regulator of O-GlcNAc signaling: linking metabolism to disease pathogenesis.

BACKGROUND: Glucosamine, a naturally occurring amino sugar abundant in cartilage, has long been utilized as a dietary supplement to alleviate osteoarthritis (OA) and joint pain. Beyond its structural role in maintaining joint integrity, glucosamine is metabolized through the hexosamine biosynthetic pathway (HBP) to generate UDP-N-acetylglucosamine (UDP-GlcNAc), the indispensable donor substrate for protein O-linked N-acetylglucosamine (O-GlcNAc) modification. FINDINGS: Accumulating evidence indicates that glucosamine-driven modulation of HBP flux and the consequent alteration of O-GlcNAcylati

A 3D Bone Culture Platform Using Human Osteocytes and Decellularized Extracellular Matrix for Modeling Musculoskeletal Diseases.

Modeling musculoskeletal diseases such as osteoporosis requires in vitro platforms that accurately reproduce key features of human bone biology. Conventional 3D culture systems provide valuable insight into cell-cell and cell-matrix interactions, but their dependence on costly materials, bioprinters, or specialized infrastructure limits their broader adoption. Here, we describe a robust and cost-effective protocol for generating 3D bone constructs using human-derived osteocytes embedded in GelMA hydrogels, supplemented with decellularized extracellular matrix (dECM) obtained from donated femor

3D bioprinting of tissues and organs for systemic diseases and localized injuries.

Three-dimensional (3D) bioprinting integrates engineering, materials science, and biology to fabricate living tissues with precise spatial control. By enabling the layer-by-layer deposition of cells and biomaterials, it overcomes many limitations of traditional scaffold-based tissue engineering and offers new opportunities for regenerative and personalized medicine. This review presents a comprehensive overview of recent advances in 3D bioprinting. It introduces a systematic, ASTM-aligned classification of key bioprinting modalities, extrusion, jetting, and vat photopolymerization, along with

Neuropeptide Y signaling in osteoarthritis pain: neuroimmune mechanisms, joint remodeling, and translational opportunities.

Osteoarthritis is a leading cause of chronic musculoskeletal pain, but pain severity is not fully explained by cartilage loss or radiographic damage. Neuropeptide Y is a multifunctional neuropeptide with receptor- and compartment-dependent effects on nociceptive signaling, sympathetic activity, immune regulation, and skeletal remodeling. Direct osteoarthritis-specific evidence links synovial-fluid neuropeptide Y to pain in some human knee osteoarthritis cohorts, NPY2R signaling to chondrocyte hypertrophy and matrix catabolism, and osteocyte-derived neuropeptide Y to experimental osteoarthritis
