Involvement of the hypothalamus-raphe magnus-spinal defecation center axis in stress-induced defecation in rats.

Acute psychological stress stimulates colonic motility; however, the specific descending neural pathways linking the brain to the distal colon remain incompletely characterized. Here, we provide evidence that the hypothalamus‒raphe magnus‒spinal cord‒pelvic nerve pathway mediates stress-induced defecation in male Sprague-Dawley rats. Using urethane-anesthetized rats, we used viral tracing, pharmacological, and chemogenetic approaches to show that hypothalamic neurons projecting to the raphe magnus nucleus activate descending serotonergic neurons probably via oxytocinergic signaling, thereby st…

Electrical devices for visual restoration.

Given the rising number of patients with blindness from macular, optic nerve, and visual pathway disease, there is considerable interest in the potential of electrical stimulation devices to restore vision. Electrical devices for restoration of visual function can be grouped into three categories: (1) visual prostheses whose goal is to bypass damaged areas and directly activate downstream intact portions of the visual pathway; (2) electric field stimulation whose goal is to activate endogenous transcriptional and molecular signaling pathways to promote neuroprotection and neuro-regeneration; a

Extracellular fluid pathway inside the facial nerve fascicles.

The extracellular fluid pathway in the facial nerve and the diffusion of a tracer from the facial nerve to other cranial nerves was examined in the rabbit. Sodium fluorescein solution was injected into either the facial nerve fascicles or the epineurial connective tissue as a tracer at the stylomastoid foramen and then localized by fluorescence microscopy. In the facial nerve, fluorescence was observed in the endoneurium and external nerve sheath (epineurium and perineurium) through the geniculate ganglion following injection into the nerve fascicles. The vestibular, trigeminal, and glossophar

Selective magnocellular damage in melanoma-associated retinopathy: comparison with congenital stationary nightblindness.

Psychophysical methods for isolating and evaluating the function of specific neural pathways are used to characterize the visual losses in patients with melanoma-associated retinopathy (MAR). These are compared with those of congenital stationary nightblindness (CSNB), a condition which displays a similar grossly abnormal ERG and loss of rod function. In MAR patients achromatic contrast sensitivity was greatly reduced in the low spatial frequency range. Stimuli chosen to isolate the magnocellular pathway were seen badly, whereas stimuli signalled primarily by the midget of the parvocellular pa…

A Robust Single Primate Neuroepithelial Cell Clonal Expansion System for Neural Tube Development and Disease Studies.

Developing a model of primate neural tube (NT) development is important to promote many NT disorder studies in model organisms. Here, we report a robust and stable system to allow for clonal expansion of single monkey neuroepithelial stem cells (NESCs) to develop into miniature NT-like structures. Single NESCs can produce functional neurons in vitro, survive, and extensively regenerate neuron axons in monkey brain. NT formation and NESC maintenance depend on high metabolism activity and Wnt signaling. NESCs are regionally restricted to a telencephalic fate. Moreover, single NESCs can turn into

Cocaine-induced stereotypy is linked to an imbalance between the medial prefrontal and sensorimotor circuits of the basal ganglia.

The dysfunction of basal ganglia circuits related to stereotyped motor activity was analysed using the well-established model of cocaine-induced stereotypy in the rat. We examined and compared the neurochemical and electrophysiological effects occurring in medial prefrontal and sensorimotor basal ganglia circuits of the dorsal striatum after cocaine injection in sensitized and non-sensitized rats. Acute injections of cocaine (25 mg/kg), not inducing stereotyped behaviour, affected both medial prefrontal and sensorimotor circuits in a similar way: (i) a mild and delayed increase and decrease of

Blockade of GABA, type A, receptors in the rat pontine reticular formation induces rapid eye movement sleep that is dependent upon the cholinergic system.

The brainstem reticular formation is an area important to the control of rapid eye movement (REM) sleep. The antagonist of GABA-type A (GABA(A)) receptors, bicuculline methiodide (BMI), injected into the rat nucleus pontis oralis (PnO) of the reticular formation resulted in a long-lasting increase in REM sleep. Thus, one factor controlling REM sleep appears to be the number of functional GABA(A) receptors in the PnO. The long-lasting effect produced by BMI may result from secondary influences on other neurotransmitter systems known to have long-lasting effects. To study this question, rats wer

Transgenic mice over-expressing GABA(B)R1a receptors acquire an atypical absence epilepsy-like phenotype.

In this study, we tested whether over-expressing the GABA(B) receptor R1a subtype in transgenic mouse forebrain neurons would be sufficient to induce spontaneous absence seizures. As hypothesized, these transgenic mice develop spontaneous, recurrent, bilaterally synchronous, 3-6 Hz slow spike and wave discharges between 2 and 4 months of age. These discharges are blocked by ethosuximide and exacerbated by baclofen confirming their absence nature. The discharges occur coincident with absence-like behaviors such as staring, facial myoclonus, and whisker twitching. However, in contrast to typical

Improving antisaccade performance in adolescents with attention-deficit/hyperactivity disorder (ADHD).

The goal of the study was to examine the effects of task manipulations on antisaccade accuracy and response times (RTs) of adolescents with attention-deficit/hyperactivity disorder (ADHD), age-matched controls, 10-year-olds and young adults. Order effects were tested by administering the task at the beginning and end of the session. Other manipulations involved a visual landmark to reduce demands on working memory and internal generation of saccades; spatially specific and non-specific cues at three intervals; and central engagement of attention through perceptual and cognitive means at three

Prefrontal cortical projections to the rat dorsal raphe nucleus: ultrastructural features and associations with serotonin and gamma-aminobutyric acid neurons.

Studies of human brain indicate that both the ventromedial prefrontal cortex (PFC) and the dorsal raphe nucleus (DRN) may be dysfunctional in major depressive illness, making it important to understand the functional interactions between these brain regions. Anatomical studies have shown that the PFC projects to the DRN, although the synaptic targets of this excitatory pathway have not yet been identified. Electrophysiological investigations in the rat DRN report that most serotonin neurons are inhibited by electrical stimulation of the PFC, suggesting that this pathway is more likely to synap

MicroRNA profiling reveals novel biomarkers for cardiovascular and psychological health in plateau psycho CVD.

This study aimed to explore the expression characteristics of miRNAs in cardiovascular diseases (CVD) and depression within a plateau environment, to better understand their potential role in Plateau Psycho-CVD. A prospective study design was employed to analyze circulating small RNAs from 20 subjects using high-throughput sequencing technology. Participants were divided into four groups (C, PPC, PP, PC) for comparative analysis. Differentially expressed miRNAs were selected for further functional enrichment analysis. The findings revealed that hsa-miR-1976 and hsa-miR-4685-3p were significant

Multifaceted role of oxidative stress in neurological disorders.

The intricate relationship between oxidative stress and neurological disorders stems from multiple factors inherent to central nervous system function. Neural tissue’s high oxygen consumption, elevated iron content, and hydrogen peroxide generation create conditions favorable for oxidative imbalance. The susceptibility of neuronal membranes to oxidative damage is particularly noteworthy due to their high polyunsaturated fatty acid composition. This vulnerability, coupled with disrupted redox homeostasis marked by excessive reactive oxygen species (ROS) and compromised antioxidant mechanisms, c

Distinct striatum pathways connected to salience network predict symptoms improvement and resilient functioning in schizophrenia following risperidone monotherapy.

Abnormal interactions between the striatum and salience network (SN) are considered as etiological and treatment-sensitive marker in schizophrenia. However, whether alterations in the intrinsic dynamics as reflected by resting-state functional connectivity (RSFC) between the striatum and salience network may predict treatment response to the widely used antipsychotic treatment strategies (risperidone, monotherapy) has not been examined systematically. To this end, treatment-naive first-episode schizophrenia patients (n = 41) underwent task-free resting-state fMRI assessment before (baseline) a

Protein Arginine Methyltransferase PRMT1 Is Essential for Palatogenesis.

Cleft palate is among the most common birth defects. Currently, only 30% of cases have identified genetic causes, whereas the etiology of the majority remains to be discovered. We identified a new regulator of palate development, protein arginine methyltransferase 1 (PRMT1), and demonstrated that disruption of PRMT1 function in neural crest cells caused complete cleft palate and craniofacial malformations. PRMT1 is the most highly expressed of the protein arginine methyltransferases, enzymes responsible for methylation of arginine motifs on histone and nonhistone proteins. PRMT1 regulates sign

Trafficking to the primary cilium membrane.

The primary cilium has been found to be associated with a number of cellular signaling pathways, such as vertebrate hedgehog signaling, and implicated in the pathogenesis of diseases affecting multiple organs, including the neural tube, kidney, and brain. The primary cilium is the site where a subset of the cell's membrane proteins is enriched. However, pathways that target and concentrate membrane proteins in cilia are not well understood. Processes determining the level of proteins in the ciliary membrane include entry into the compartment, removal, and retention by diffusion barriers such a

A brain centred view of psychiatric comorbidity in tinnitus: from otology to hodology.

Introduction. Comorbid psychiatric disorders are frequent among patients affected by tinnitus. There are mutual clinical influences between tinnitus and psychiatric disorders, as well as neurobiological relations based on partially overlapping hodological and neuroplastic phenomena. The aim of the present paper is to review the evidence of alterations in brain networks underlying tinnitus physiopathology and to discuss them in light of the current knowledge of the neurobiology of psychiatric disorders. Methods. Relevant literature was identified through a search on Medline and PubMed; search t

Low dose Hsp90 inhibitor 17AAG protects neural progenitor cells from ischemia induced death.

Stress adaptation effect provides cell protection against ischemia induced apoptosis. Whether this mechanism prevents other types of cell death in stroke is not well studied. This is an important question for regenerative medicine to treat stroke since other types of cell death such as necrosis are also prominent in the stroke brain apart from apoptosis. We report here that treatment with 17-N-Allylamino-17-demethoxygeldanamycin (17AAG), an Hsp90 inhibitor, protected neural progenitor cells (NPCs) against oxygen glucose deprivation (OGD) induced cell death in a dose dependent fashion. BMP signaling orchestrates photoreceptor specification in the zebrafish pineal gland in collaboration with Notch.

A variety of signaling pathways have been shown to regulate specification of neuronal subtype identity. However, the mechanisms by which future neurons simultaneously process information from multiple pathways to establish their identity remain poorly understood. The zebrafish pineal gland offers a simple system with which to address questions concerning the integration of signaling pathways during neural specification as it contains only two types of neurons - photoreceptors and projection neurons. We have previously shown that Notch signaling inhibits the projection neuron fate. Here, we sho

Folate deprivation, the methionine cycle, and Alzheimer's disease.

Folate deficiency is associated with increase in homocysteine levels. Abnormal plasma levels of that neurotoxic nonproteinogenic amino acid is implicated in many pathological conditions including cardiovascular diseases, neural tube defects, and is now recognized as a risk factor in Alzheimer's disease (AD) dementia. Homocysteine elimination is regulated by two metabolic pathways, namely, the transmethylation and the transsulfuration pathways. Its elimination via these two metabolic pathways is modulated by folate, a member of the B-vitamin family. Folate provides, via its metabolic end produc

Improving prognosis in type 2 diabetes. Diabetic neuropathy is in trouble.

Painful sensory syndromes and the anesthetic foot result in much clinical morbidity and patient unhappiness in diabetes. As yet, a satisfactory and fundamental therapy is not available to us to help patients. Effective blood glucose control and vigilant screening programs for foot problems are all we have to offer. Clinical observation of neuropathic syndromes and measures of nerve function have not led to significant understanding of pathogenesis. The primary source of understanding of pathways to nerve damage come from animal studies, despite the fears that the model in diabetes in no way re
