Unraveling the Pathogenetic Potential and Immune Crosstalk of Yersinia spp. in Inflammatory Bowel Disease.

Yersinia enterocolitica and Yersinia pseudotuberculosis have been identified as microbiological factors in the multifactorial pathophysiology of inflammatory bowel disease (IBD). Although conventionally associated with self-limiting gastroenteritis, Yersinia has been increasingly investigated for its potential role in IBD, particularly in genetically predisposed individuals. Whether this reflects causality, opportunistic colonization, or secondary enrichment within inflamed tissue, however, remains unresolved. Polymerase chain reaction (PCR)-based studies have detected Yersinia DNA in the inte

Glucose sensing and homeostasis by adipocyte GPCR.

The adipose tissue regulates energy homeostasis, which is one of the vital processes for organismal survival, and its dysregulation causes metabolic diseases including obesity and type 2 diabetes. Glucose is utilized by the adipose tissue for energy production and storage to regulate systemic glucose homeostasis. The G-protein-coupled receptors (GPCRs) expressed in the adipose tissues play a crucial role in adipocyte function by responding to hormonal, neural, and metabolic signals; thereby, influencing insulin sensitivity, glucose uptake and lipid metabolism. The specific contribution of adip

Piezo ion channels in the digestive system: Mechanotransduction pathways and therapeutic targeting strategies.

Piezo ion channels, notably Piezo1 and Piezo2, are key mechanosensors that transduce mechanical forces into intracellular signals, playing indispensable roles in digestive physiology. These channels regulate essential functions such as intestinal motility, epithelial barrier integrity, bile secretion, and host-microbiota balance. Emerging evidence links aberrant Piezo signaling to a wide range of gastrointestinal disorders, including functional bowel diseases, inflammatory conditions, and digestive cancers. However, translating these insights into therapeutic applications remains challenging.

Activation of autophagy with PF-06409577 alleviates heatstroke-induced organ injury.

Heat waves are a significant environmental issue threatening global human health. Extreme temperatures can lead to various heat-related illnesses, with heatstroke being among the most severe. Currently, there are no effective treatments to mitigate the multi-organ damage caused by heatstroke. We found that heat stress activated autophagy. Knockdown of the autophagy-related gene 7 (ATG7) or knockout of the autophagy initiation regulatory genes UNC-51-like autophagy activating kinase 1/2 (ULK1/ULK2) increased cell death. Mindful Eating: A Deep Insight Into Fructose Metabolism and Its Effects on Appetite Regulation and Brain Function.

Fructose, a common sweetener in modern diets, has profound effects on both metabolism and brain function, primarily due to its distinct metabolic pathways. Unlike glucose, fructose bypasses critical regulatory steps in metabolism, particularly the phosphofructokinase-1 (PFK-1) feedback inhibition, leading to uncontrolled metabolism and increased fat storage. This review delves into the metabolic consequences of fructose consumption, including its limited role in directly stimulating insulin secretion, which affects satiety signaling and contributes to increased food intake. The small intestine

Enteropathogenic Escherichia coli revisited - New insights into old EPEC isolates using whole genome sequencing.

Enteropathogenic E. coli (EPEC) cause severe diarrhoeal disease in children under the age of three in low- and middle-income countries. Upon ingestion of contaminated food or water, the bacteria colonise the small intestine. EPEC are grouped into typical and atypical isolates based on the presence or absence of the genes encoding the bundle-forming pilus (Bfp), with tEPEC (Bfp+) adhering to cells in microcolonies, whereas aEPEC show a more diffuse adherence phenotype. All EPEC express a type III secretion system (T3SS) encoded on the locus of enterocyte effacement (LEE) pathogenicity island, w

Mannose-modified Prunus persica kernel protein nanoparticles loading baicalin coated with Lycium barbarum polysaccharide for ulcerative colitis treatment.

Baicalin (Ba) is a promising anti-inflammatory agent for treating ulcerative colitis (UC), but its efficacy is constrained by poor solubility and colon-targeting efficiency. Lycium barbarum polysaccharide (LBP), an anionic polysaccharide, not only restores gut microbiota dysbiosis but also exhibits excellent upper gastrointestinal stability and biocompatibility, making it a candidate for developing colon-targeted carriers. In this paper, the polymeric nanoparticles (Ba@MP/LBP NPs) were constructed using LBP, mannose, and Prunus persica kernel protein (PKP) for targeted delivery of baicalin to

Chain Length-Dependent Inhibition of Human and Rat Placental Aromatase by Benzalkonium Disinfectants: Experimental, Quantitative Structure-Activity Relationship, and In Silico Docking Insights.

Benzalkonium chloride (BAC) compounds are a subclass of cationic surfactants widely used as disinfectants. The current research explored the inhibition of BACs on human and rat aromatase activity, revealing a structure-dependent mechanism. In human placental microsomes, BACs (C10, C12, C14, C16, and C18) significantly suppressed aromatase activity with IC50 and Ki values following the order C10 > C12 > C14 > C16 > C18, indicating that inhibitory potency increases with alkyl chain length. Enzyme kinetics and Lineweaver-Burk analyses suggested mixed/noncompetitive inhibition, where BACs bind bot

Rapid cyclic stretching of cultured human visceral smooth muscle cells promotes a synthetic, proinflammatory phenotype.

Bowel smooth muscle experiences mechanical stress constantly during normal function and pathologic mechanical stressors in disease states. We tested the hypothesis that pathologic mechanical stress could alter transcription to induce smooth muscle phenotypic class switching. To test this hypothesis, primary human intestinal smooth muscle cells (HISMCs), seeded on electrospun aligned poly-ε-caprolactone nano-fibrous scaffolds, were subjected to pathologic, high-frequency (1 Hz) uniaxial 3% cyclic stretch (loaded) or kept unloaded in culture for 6 hours. Gut microbiota in hypothyroidism: pathogenic mechanisms and opportunities for precision microbiome interventions.

Hypothyroidism is a common endocrine disorder characterized by insufficient thyroid hormone synthesis or secretion, most frequently caused by Hashimoto's thyroiditis, an autoimmune condition that leads to chronic thyroid gland damage. Despite the widespread use of levothyroxine replacement therapy, a substantial proportion of patients continue to experience persistent symptoms and metabolic dysregulation even after achieving biochemical euthyroidism. These observations have prompted growing interest in non-hormonal contributors to hypothyroidism, particularly the role of the gut microbiota. Re

Saccharomyces cerevisiae TAD1 Mutant Strain As Potential New Antimicrobial Agent: Studies on Its Antibacterial Activity and Mechanism of Action.

Human infections caused by pathogenic bacteria remain a major global health concern. Among them, Staphylococcus aureus, Escherichia coli, Klebsiella pneumoniae, and Salmonella typhi are particularly prevalent and associated with significant morbidity and mortality. While antibiotics have long been the cornerstone of bacterial infection treatment, the widespread and often inappropriate use of these drugs has led to the emergence of multidrug-resistant (MDR) strains. This escalating resistance crisis underscores the urgent need for alternative therapeutic strategies. Amid the escalating global a

Emergence of porcine-derived hybrid Escherichia coli with dual-disease manifestations in intestinal and extraintestinal niches.

Escherichia coli, a significant zoonotic pathogen, continuously evolves to produce hybrid strains with diverse pathogenic characteristics, threatening public health. These hybrid strains are capable of causing multiple diseases across various tissues within a single host or among genetically diverse hosts. They have been reported to exhibit heightened virulence, enhanced infectiousness, and increased resistance to environmental clearance. Here, we identified a hybrid aEPEC/ExPEC isolate, PE164, from the bloody stool of a piglet with severe diarrhoea, which exhibited both intestinal and extrain

Magnitude of Fruit Juice-Drug Interactions Due to Osmolality-Dependent Fluid Secretion: Differences among Apple, Orange, and Grapefruit Juices.

We recently reported that the gastrointestinal (GI) fluid volume is influenced by the solution osmolality, and proposed that this effect may play a role in beverage-drug interactions. Here, we investigated whether osmolality-dependent fluid secretion can explain the difference in the magnitudes of fruit juice-drug interactions depending on the type of fruit juice (grapefruit juice (GFJ), orange juice (OJ), and apple juice (AJ)). The osmolality of GFJ, OJ, and AJ used in this study was found to be 552, 686, and 749 mOsm/kg, respectively. Measurements of intestinal fluid movement following bever

Exploring the metabolic patterns and response mechanisms of bile acids during fasting: A study with poultry as an example.

Fasting is beneficial to alleviate fatty liver, lose weight and improve reproductive function. However, previous studies have shown that, during fasting, disorders of bile acid metabolism were strongly associated with intestinal inflammation. The physiological and biochemical parameters and gene expression of multiple tissues of chickens at every critical time node were measured by ELISA and qPCR. In addition, association analysis was performed based on liver transcriptome sequencing and cecum metabolome data. At the cellular level, the regulatory effects of cecal metabolites on host bile acid

Gut microbiota are involved in leptin-induced thermoregulation in the Mongolian gerbil (Meriones unguiculatus).

Leptin is a hormone that is secreted by adipocytes and may promote energy expenditure by increasing thermogenesis. Our previous studies have shown that thermo-transient receptor potentials (thermo-TRPs) and gut microbiota are associated with thermoregulation in Mongolian gerbils, which are characterized by relative high serum leptin concentrations. Here, we tested whether leptin can stimulate non-shivering thermogenesis (NST) in Mongolian gerbils, and whether thermo-TRPs and gut microbiota are involved in leptin-induced thermogenesis. First, gerbils were given acute leptin treatment (ALT) with

Highly efficient expression of Rasamsonia emersonii lipase in Pichia pastoris: characterization and gastrointestinal simulated digestion in vitro.

BACKGROUND: Acidic lipases with high catalytic activities under acidic conditions have important application values in the food, feed and pharmaceutical industries. However, the availability of acidic lipases is still the main obstacle to their industrial applications. Although a novel acidic lipase Rasamsonia emersonii (LIPR) was heterologously expressed in Escherichia coli, the expression level was unsatisfactory. RESULTS: To achieve the high-efficiency expression and secretion of LIPR in Pichia pastoris GS115, the combinatorial optimization strategy was adopted including gene codon prefere

The preventative effects of Lactococcus Lactis metabolites against LPS-induced sepsis.

INTRODUCTION: Sepsis is a syndrome of organ dysfunction caused by a dysregulated host response to infection and septic shock. Currently, antibiotic therapy is the standard treatment for sepsis, but it can lead to drug resistance. The disturbance of the gut microbiota which is affected by sepsis could lead to the development of organ failure. It is reported that probiotics could shape the gut microbiota, potentially controlling a variety of intestinal diseases and promoting whole-body health. METHODS: In this study, we evaluated the preventive effects of intra- and extracellular products of pr
