Loss of Function of Phosphodiesterase 11A4 Shows that Recent and Remote Long-Term Memories Can Be Uncoupled.

Systems consolidation is a process by which memories initially require the hippocampus for recent long-term memory (LTM) but then become increasingly independent of the hippocampus and more dependent on the cortex for remote LTM. Here, we study the role of phosphodiesterase 11A4 (PDE11A4) in systems consolidation. PDE11A4, which degrades cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP), is preferentially expressed in neurons of CA1, the subiculum, and the adjacently connected amygdalohippocampal region. In male and female mice, deletion of PDE11A enhances remote

MicroRNA epigenetic signatures in human disease.

MicroRNAs (miRNAs) are short non-coding RNAs that act as important regulators of gene expression as part of the epigenetic machinery. In addition to posttranscriptional gene silencing by miRNAs, the epigenetic mechanisms also include DNA methylation, histone modifications and their crosstalk. Epigenetic modifications were reported to play an important role in many disease onsets and progressions and can be used to explain several features of complex diseases, such as late onset and fluctuation of symptoms. However, miRNAs not only function as a part of epigenetic machinery, but are also epigen

Parkinsonian syndromes presenting with circadian rhythm sleep disorder- advanced sleep-phase type.

BACKGROUND: Circadian rhythm sleep disorder-advanced sleep-phase type is a relatively uncommon disorder, mostly seen among the elderly population. Impaired circadian rhythms have been reported in neurodegenerative conditions; however, there are no reports of any circadian rhythm sleep disorder among patients with Parkinsonian syndromes. We report two patients who presented with this circadian rhythm disorder, and were then diagnosed with a Parkinsonian syndrome. The cases. A 65-year-old retired man presented with history of abrupt change in sleep schedules, sleeping around 6.30-7 p.m. and waki

Extracellular matrix protein 1 gene (ECM1) mutations in lipoid proteinosis and genotype-phenotype correlation.

The autosomal recessive disorder lipoid proteinosis results from mutations in extracellular matrix protein 1 (ECM1), a glycoprotein expressed in several tissues (including skin) and composed of two alternatively spliced isoforms, ECM1a and ECM1b, the latter lacking exon 7 of this 10-exon gene (ECM1). To date, mutations that either affect ECM1a alone or perturb both ECM1 transcripts have been demonstrated in six cases. However, lipoid proteinosis is clinically heterogeneous with affected individuals displaying differing degrees of skin scarring and infiltration, variable signs of hoarseness and

Cortical perivascular satellitosis in intractable epilepsy; a form of cortical dysplasia?

This report describes two cases of cortical dysplasia in patients with intractable epilepsy: the first case presents a 12-year-old male with a simple partial seizure of his left arm (case 1), while the second case presents an 8-year-old female with a complex partial seizure (case 2). Magnetic resonance images showed high signal intensity on T2-weighted and fluid attenuated inversion-recovery images in the cerebral cortex of right frontal convexity in case 1 and in the medial temporal lobe and anterior amygdala in case 2. The latter lesion showed focal contrast enhancement. Light microscopy rev

TRPV4 is involved in NLRP3 and NLRP1 inflammasomes activation after pilocarpine-induced status epilepticus in mice : Concise title: TRPV4-mediated inflammasome activation in epilepsy.

The nucleotide-binding oligomerization domain (NOD)-like receptor family pyrin domain-containing 1 (NLRP1) and NLRP3 inflammasomes are activated in temporal lobe epilepsy (TLE), leading to neuroinflammation, pyroptosis, and neuronal injury. Protein phosphatase 2 A (PP2A) and the toll-like receptor 4 (TLR4)–p38 MAPK signaling pathway participate in regulating these inflammasomes. Although blocking transient receptor potential vanilloid 4 (TRPV4) alleviates inflammation and neuronal damage after pilocarpine-induced status epilepticus (PISE), the underlying mechanisms remain unclear. Here, we fou

Pyruvate dehydrogenase autoantibodies in autoantibody-negative patients with seizures are associated with reduced pyruvate dehydrogenase activity.

OBJECTIVE: We investigated the presence and potential functional relevance of antimitochondrial autoantibodies in patients suspicious for autoimmune encephalitis (AIE) associated with psychiatric symptoms and/or seizures, who were negative for known antineuronal autoantibodies. METHODS: We screened serum samples from 387 patients autoantibody-negative for known antineuronal autoantibodies with psychiatric disturbances and/or epileptic seizures, including patients with temporal lobe epilepsy of unknown etiology. Various techniques, including immunoblotting, immunoprecipitation, mass spectromet

Prolonged postictal hypercapnia is associated with delayed recovery of consciousness after generalized convulsive seizure.

BACKGROUND: Postictal impairment of breathing and arousal is a significant contributor in many SUDEP cases. Central CO2 chemoreception, which mediates ventilatory response, is often blunted after generalized convulsive seizures (GCS). It is plausible that the central chemoreceptors that mediate arousal in response to hypercapnia are similarly impaired after GCS, thereby contributing to delayed recovery of consciousness (ROC). Therefore, we hypothesized that greater severity of postictal hypercapnia is associated with longer latency to early signs of ROC following GCS. METHODS: Epilepsy patien

Targeted optical uncaging of an adenosine A1 receptor agonist regulates excitability of hippocampal slices derived from epileptic mice.

Photocaged adenosine A1 receptor (A1R) agonists combined with spatiotemporally selective illumination represents a promising therapeutic strategy for mesial temporal lobe epilepsy (mTLE). This ex vivo study investigated whether the A1R agonist N6-cyclopentyladenosine (CPA) suppresses hippocampal excitability in the intrahippocampal kainic acid (IHKA) mouse model and whether photocaged CPA (cCPA) enables subregion-selective and closed-loop reduction of hippocampal excitability. Hippocampal slices from male IHKA and saline-injected control mice were exposed to CPA (10-900 nM) while electrically

Regulation of MAP2, GFAP, and calcium in the CA3 Region Following Kainic Acid Exposure to  organotypic hippocampal slice culture.

BACKGROUND: Neurodegeneration due to neurotoxicity is one of the phenomena in temporal lobe epilepsy. Experimentally, hippocampal excitotoxicity process can occur due to kainic acid exposure, especially in the CA3 area. Neuronal death, astrocyte reactivity and increased calcium also occur in hippocampal excitotoxicity, but few studies have investigated immediate effect after kainic acid exposure. The organotypic hippocampal slice culture (OHSC) is a useful model for studying the neurodegeneration process, but there are still many protocol differences. In this study, minor modifications were ma

Glial changes in the dentate gyrus of neuronal-specific PTEN knockout mice correlate with changes in cell proliferation.

Dysregulated hippocampal neurogenesis is a feature of temporal lobe epilepsy (TLE), marked by increased neuronal proliferation. The tumor suppressor gene phosphatase and tensin homolog (PTEN) regulates neuronal proliferation, and its deletion is implicated in TLE. We have previously shown that deletion of neuronal subset-specific (NS)-PTEN in mice increases the number of proliferating cells throughout the dentate gyrus, including subregions that are typically devoid of neurons but rich in glial cells, most notably the Hilus and Molecular Layer. In this study, we hypothesized that NS-PTEN knock

Brain morphology normative modelling platform for abnormality and centile estimation: Brain MoNoCle.

Normative models of brain structure estimate the effects of covariates such as age and sex using large samples of healthy controls. These models can then be applied to, for example, smaller clinical cohorts to distinguish disease effects from other covariates. However, these advanced statistical modelling approaches can be difficult to access, and processing large healthy cohorts is computationally demanding. Thus, accessible platforms with pre-trained normative models are needed. We present such a platform for brain morphology analysis as an open-source web applicationhttps://cnnplab.shinyapp

Transient epileptic amnesia: a retrospective cohort study of 127 cases, including CSF amyloid and tau features.

BACKGROUND: Transient epileptic amnesia (TEA) is a late-onset epilepsy syndrome encompassing transient iterative amnesias and interictal cognitive impairment, two features that overlap with incipient neurodegenerative dementias. We, therefore, examined the yield of CSF amyloid and tau biomarkers in TEA. METHODS: In this retrospective study, 127 TEA patients with unremarkable imaging findings were divided into 2 groups, namely, CSF (n = 71) and no-CSF (n = 56). Both were compared for demographics; medical history; baseline neurological, cognitive, and behavioral features; baseline mesial tempo

Differential regulation of gene expression pathways with dexamethasone and ACTH after early life seizures.

Early-life seizures (ELS) are associated with persistent cognitive deficits such as ADHD and memory impairment. These co-morbidities have a dramatic negative impact on the quality of life of patients. Therapies that improve cognitive outcomes have enormous potential to improve patients' quality of life. Our previous work in a rat flurothyl-induction model showed that administration of adrenocorticotropic hormone (ACTH) at time of seizure induction led to improved learning and memory in the animals despite no effect on seizure latency or duration. Administration of dexamethasone (Dex), a cortic

Neonatal Subpial Hemorrhage: Clinical Factors, Neuroimaging, and Outcomes in a Quaternary Care Children's Center.

BACKGROUND: Subpial hemorrhages are underrecognized, underreported, and poorly understood. The spectrum of their clinical manifestations and consequences in neonates has not been fully described. Here, we describe the demographic, clinical, and radiographic characteristics of neonates with subpial hemorrhages. METHODS: We reviewed the medical records and neuroimaging studies of neonates with subpial hemorrhage who were admitted to our neonatal intensive care unit between September 2009 and December 2020. RESULTS: Of 114 neonates with intracranial hemorrhage, 31 (27%) had subpial hemorrhage.

Cerebral Waste Accumulation and Glymphatic Clearance as Mechanisms of Human Neurological Diseases.

The brain is a complex system that requires continual regulation of parenchymal pressure, osmolarity, and waste removal for optimal function; despite this, human brain lacks any obvious extension of lymphatic circulation for moderating fluid and waste regulation. We recapitulate herein a recent analysis of proteinaceous waste deposition in the human brain, its observed route of clearance, and the implications of abnormal accumulation along this clearance pathway as a potential mechanism of neurological diseases. This study uncovered an analogous staining pattern of cerebral phosphorylated tau

Subventricular zone-derived neural stem cell grafts protect against hippocampal degeneration and restore cognitive function in the mouse following intrahippocampal kainic acid administration.

Temporal lobe epilepsy (TLE) is a major neurological disease, often associated with cognitive decline. Since approximately 30% of patients are resistant to antiepileptic drugs, TLE is being considered as a possible clinical target for alternative stem cell-based therapies. Given that insulin-like growth factor I (IGF-I) is neuroprotective following a number of experimental insults to the nervous system, we investigated the therapeutic potential of neural stem/precursor cells (NSCs) transduced, or not, with a lentiviral vector for overexpression of IGF-I after transplantation in a mouse model o

Uneven interhemispheric connections between left and right primary sensori-motor areas.

To clarify the characteristics of interhemispheric connections, we investigated cortico-cortical evoked potentials (CCEP) in human. Fourteen patients with temporal lobe epilepsy who underwent invasive EEG monitoring with bilaterally implanted subdural electrodes were studied. Electric pulse stimuli were given in a bipolar fashion at two adjacent electrodes on and around the motor area (MA) or sensory area (SA), and CCEP responses were recorded by averaging electrocorticograms from the contralateral hemisphere. Seventy-two pairs of electrodes were stimulated, and 468 recordings were analyzed. F

In the adult hippocampus, chronic nerve growth factor deprivation shifts GABAergic signaling from the hyperpolarizing to the depolarizing direction.

GABA, the main inhibitory transmitter in adulthood, early in postnatal development exerts a depolarizing and excitatory action. This effect, which results from a high intracellular chloride concentration ([Cl(-)](i)), promotes neuronal growth and synaptogenesis. During the second postnatal week, the developmental regulated expression of the cation-chloride cotransporter KCC2 accounts for the shift of GABA from the depolarizing to the hyperpolarizing direction. Changes in chloride homeostasis associated with high [Cl(-)](i) have been found in several neurological disorders, including temporal l
