Review: fetal programming of polycystic ovary syndrome by androgen excess: evidence from experimental, clinical, and genetic association studies.

CONTEXT: Polycystic ovary syndrome (PCOS) is a common endocrine disorder of premenopausal women, characterized by hyperandrogenism, polycystic ovaries, and chronic anovulation along with insulin resistance and abdominal obesity as frequent metabolic traits. Although PCOS manifests clinically during adolescence, emerging data suggest that the natural history of PCOS may originate in intrauterine life. EVIDENCE ACQUISITION: Evidence from experimental, clinical, and genetic research supporting the hypothesis for the fetal origins of PCOS has been analyzed. EVIDENCE SYNTHESIS: Female primates, e

Endotoxemia severely affects circulation during normoxia and asphyxia in immature fetal sheep.

OBJECTIVE: The purpose of the present study was to determine whether endotoxins (lipopolysaccharides, LPS) affect the fetal cardiovascular system in a way likely to cause brain damage. METHODS: Thirteen fetal sheep were chronically instrumented at a mean gestational age of 107 +/- 1 days. After control measurements of organ blood flow (microsphere method), blood gases, and acid base balance were obtained, seven of 13 fetuses received LPS (53 +/- 3 microg/kg fetal weight) intravenously. Sixty minutes later, asphyxia was induced by occlusion of the maternal aorta for 2 minutes. Measurements of

Fetal microchimerism and the two-stage model of preeclampsia.

Fetal cells cross the placenta during pregnancy and some have the ability to persist in maternal organs and circulation long-term, a phenomenon termed fetal microchimerism. These cells often belong to stem cell or immune cell lineages. The long-term effects of fetal microchimerism are likely mixed, potentially depending on the amount of fetal cells transferred, fetal-maternal histocompatibility and fetal cell-specific properties. Both human and animal data indicate that fetal-origin cells partake in tissue repair and may benefit maternal health overall. On the other hand, these cells have been

Swedish intrauterine growth reference ranges for estimated fetal weight.

Fetal growth restriction is a strong risk factor for perinatal morbidity and mortality. Reliable standards are indispensable, both to assess fetal growth and to evaluate birthweight and early postnatal growth in infants born preterm. The aim of this study was to create updated Swedish reference ranges for estimated fetal weight (EFW) from gestational week 12-42. This prospective longitudinal multicentre study included 583 women without known conditions causing aberrant fetal growth. Each woman was assigned a randomly selected protocol of five ultrasound scans from gestational week 12 + 3 to 41

Gestational dexamethasone alters fetal neuroendocrine axis.

This study tested whether the maternal transport of dexamethasone (DEXA) may affect the development of the neuroendocrine system. DEXA (0.2mg/kg b.w., subcutaneous injection) was administered to pregnant rats from gestation day (GD) 1-20. In the DEXA-treated group, a decrease in maternal serum thyroxine (T4), triiodothyronine (T3), and increase in thyrotropin (TSH) levels (hypothyroid status) were observed at GDs 15 & 20 with respect to control group. The reverse pattern (hyperthyroid status) was observed in their fetuses at embryonic days (EDs) 15 & 20. Although the maternal body weight was d

Slow rising serial chorionic gonadotropins predict poor pregnancy outcome despite sonographic viability.

PURPOSE: To determine the prognosis of women with slow rising beta-hCG levels when viability is detected by ultrasound. METHODS: Serum beta-hCG levels were obtained every two to three days in the early first trimester. Doubling-time (DT) of beta-hCG levels was defined as DT exceeding 3.2 days. Sonography was performed at eight weeks and then after 12 weeks. RESULTS: There were 158 consecutive pregnancies evaluated and 111 (70%) had normal rising beta-hCG levels, viable ultrasound at eight weeks, and viable pregnancies after 12 weeks. There were 22 pregnancies with slow rising beta-hCG levels

Abnormal growth patterns in diabetes in pregnancy: a longitudinal study.

In this longitudinal study we sought to investigate the presence of different abnormal fetal growth patterns in the pregnant diabetic. At least three serial ultrasound examinations were performed during the third trimester on 522 diabetic and 93 control subjects. Growth curves were established for femur length, abdominal circumference and head circumference. In addition, daily growth rate was calculated for fetal weight and all morphometric measurements. The study revealed: a) in the gestational diabetes group large-for-gestational age infants, two accelerated growth patterns (early mean blood

Calcium metabolism in pregnancy and the perinatal period: a review.

Calcium homeostasis is a complex process involving calcium, other involved ions, and three calcitropic hormones, parathyroid hormone, calcitonin, and 1,25-dihydroxyvitamin D3. The principal maternal adjustment during pregnancy is an increasing parathyroid hormone secretion which maintains the serum calcium concentration in the face of a falling albumin level, an expanding extracellular fluid volume, an increasing renal excretion, and placental calcium transfer. The placenta transports calcium ions actively, making the fetus hypercalcemic relative to its mother, which in turn stimulates calcito

Multi-resolution tone mapping for high dynamic range medical ultrasound images.

Ultrasound diagnostics is a key tool in obstetrics for detecting fetal anomalies and monitoring pregnancy, but image quality often declines in obese patients due to reduced contrast resolution. This pilot study develops and preliminarily validates a novel tone-mapping algorithm for enhancing contrast resolution in high dynamic range (HDR) ultrasound images. The method employs multi-resolution fusion with depth-adaptive weighting and depth compensation to improve contrast, enhance tissue differentiation, reduce noise, and preserve fine details. The algorithm was tested on 20 fetal ultrasound im

Effect of inhibiting prostaglandin synthesis in pregnant sheep with 4-aminoantipyrine under normothermic and hyperthermic conditions.

Infusion of the prostaglandin synthetase inhibitor 4-aminoantipyrine at a rate of 20 mg/min via the fetal tarsal vein during normothermic conditions (ambient temperatures = 24 degrees C) significantly decreased the concentrations of prostaglandins E2 and F2 alpha in maternal and fetal arterial plasma and in uterine and umbilical vein plasma. The infusion was associated with a small but significant rise in fetal temperature and a fall in fetal arterial pH. Respiratory alkalosis developed in fetuses during hyperthermia by raising the ambient temperature to 43 degrees C for 8 hours. In contrast,

Prenatal exposure to gas stoves, mold, and water damage: Associations with gestational duration and fetal growth in the ECHO cohort.

Epidemiological evidence on prenatal exposure to household environmental factors (gas stoves, mold, and dampness) and adverse birth outcomes remains limited and inconsistent. We examined associations of these exposures with gestational duration and fetal growth in 11,483 mother-infant dyads from 31 sites in the United States Environmental influences on Child Health Outcomes (ECHO) Cohort. Participants reported presence of a gas stove, mold/mildew (outside the shower/bathtub), and water damage in the home where they lived for the longest duration during their pregnancy (2000-2023). Gestational

[The programming effects of the placenta on offspring neurobehavioral development].

Neurobehavioral development in offspring is shaped by multiple interacting determinants. As a critical organ during gestation and fetal development, the placenta exerts programming effects on offspring neurobehavioral outcomes through diverse mechanisms. This article systematically examines the programming role of placental factors from the following perspectives: placental barrier function, placental pathological alterations, neuroendocrine pathways, epigenetic regulation, and immunomodulatory mechanisms. In addition, it explores potential mechanisms underlying sex-specific influences on offs…

Early maternal obesity shapes offspring development through immune mechanisms: a systematized review.

In recent years, rates of maternal obesity before conception have continued to rise globally. Maternal obesity in early pregnancy may alter intrauterine immune mechanisms, impact fetal metabolic programming, and increase the risk of later-life obesity. This study aimed to review the literature on changes in immune parameters and weight-related outcomes in offspring reported in human and animal studies of maternal obesity. A systematized literature search was conducted using PubMed and Web of Science for relevant human and animal studies published between 2010 and 2025. Study screening was faci

Protective Effects of Maternal Administration of a Standardized Extract of Cultured Lentinula edodes Mycelia against Radiation-Induced Fetal Morphological Abnormalities.

Exposure to ionizing radiation during early pregnancy poses serious risks to fetal development, particularly during organogenesis. In this study, we evaluated the protective effects of AHCC®, a standardized extract of Lentinula edodes mycelia with known immunomodulatory and metabolic properties, using an ICR mouse model. Intraperitoneal administration of AHCC prior to 1.4-Gy γ-irradiation on gestational day 9 significantly reduced the incidence of tail malformations and diaphragmatic hernias, without affecting implantation or fetal survival. Additionally, lifelong AHCC ingestion across three g…

Maternal Adiponectin Decreases Placenta Nutrient Transport in Mice.

Women with obesity who develop gestational diabetes have lower serum adiponectin throughout pregnancy, suggesting that low levels impair the ability to handle metabolic challenges during pregnancy. The placenta expresses adiponectin receptors, and adiponectin could therefore indirectly affect the developing fetus. Here, we aimed to investigate how elevated maternal and fetal adiponectin affect placental function, fetal growth, and metabolism during pregnancy in normal-weight and obese mice. Wild-type (wt) and adiponectin-overexpressing (APNtg) mice were fed normal chow or a high fat/high sucro

A review of the implications of maternal monosodium glutamate consumption on offspring health.

BACKGROUND AND AIMS: Monosodium glutamate (MSG), a globally used food additive for over a century, is considered generally safe. Although inconsistent, many animal studies have shown that MSG is associated with multiple health conditions, necessitating a closer examination of its potential health impact, especially on fetal development. Human data on MSG's effects on offspring during pregnancy are lacking. This review examines existing literature to assess the impact of maternal MSG consumption on offspring health. METHODS: A systematic literature search of human and animal studies was conduc…
