Immunocytochemical study of the hypothalamo-neurohypophysial system. II. Distribution of neurophysin, vasopressin and oxytocin in the normal and osmotically stimulated rat.

Antisera, with cross reactive antibodies removed by affinity chromatography, were used in the immunoperoxidase-bridge technique to study the distribution of oxytocin and vasopressin together with neurophysin in the hypothalamo-neurohypophysial system of the rat. The hormones were demonstrated in different areas of the supraoptic nucleus (SON) and paraventricular nucleus (PVN), in neurosecretory fibres of the hypothalamo-neurohypophysial tract, median eminence, and in nerve terminals of the neurohypophysis. Intact normal and rats with hereditary hypothalamic diabetes insipidus (Brattleboro stra

Increased plasma arginine vasopressin in clinical adrenocortical insufficeincy and its inhibition by glucosteroids.

Trichloroacetic acid extracts of plasma were fractionated on a CG-50 resin column and the 50% acetic acid eluents chromatographed on silicic acid-impregnated glass paper in butanol-acetic acid-water. The specific arginine vasopressin (AVP) zone was eluted and assayed for antidiuretic activity in the diuretic rat. Thioglycolate inactivation was used to confirm AVP activity. Recovery of as little as 4 muU AVP per ml plasma ranged between 80 and 90%. In normal subjects after an overnight fast, plasma AVP ranged between 2.5 and 10.0 muU per ml. AVP secretion was inhibited by hemodilution and stimu

Lifetime adversities and patterns of maternal prenatal HPA function from hair glucocorticoids.

BACKGROUND: The associations between both early life and later adversities and prenatal stress physiology, particularly HPA axis function, are still largely unknown. Inconclusive previous studies highlight the necessity for adopting life-course approaches. This study examined the associations between adverse childhood experiences (ACEs), adverse prenatal life events (APEs), and patterns of maternal prenatal HPA function assessed via joint-trajectories of hair glucocorticoid concentrations (HGC). Particularly, ACEs were tested as a potential moderator. METHODS: We analyzed data from 269 women

Association of diurnal cortisol rhythm with chronic pain: Evidence from a prospective cohort study in community-dwelling adults.

Despite clinical evidence linking hypothalamic-pituitary-adrenal (HPA) axis dysfunction to chronic pain, epidemiological findings remained mixed. Data from 1246 respondents aged 34-84 at baseline, obtained from the Midlife in the United States (MIDUS) study and its subproject, the National Study of Daily Experiences (NSDE), were used to examine associations between salivary diurnal cortisol rhythms and chronic pain outcomes over a seven-year follow-up period, using mixed-effects logistic regression models adjusted for sociodemographics, lifestyle, and health-related factors. Furthermore, to ex

α-GalCer regulates acute stress-induced steroidogenesis by modulating lipid metabolism in female BALB/c mice.

The immune system orchestrates the hypothalamus-pituitary-adrenal (HPA) axis response to stress. However, the impact of invariant natural killer T (iNKT) cell activation on stress-induced glucocorticoid levels remains poorly understood. Alpha-galactosylceramide (α-GalCer), a specific agonist for iNKT cells, activates iNKT cells to produce inflammatory cytokines including interleukin (IL)-4 and interferon (IFN)-γ. Our findings indicate that treatment with α-GalCer 3 hours before acute restraint stress suppressed the elevation of adrenocorticotropic hormone (ACTH) but did not affect the increase

Prenatal phenol exposure and child behaviour: insights into the hypothalamic-pituitary-adrenal axis from two prospective mother-child cohorts.

BACKGROUND: Synthetic phenols are widely used chemicals with potential neurodevelopmental toxicity. Human studies are often limited by small sample sizes and exposure misclassification. Identifying the biological pathways affected by these substances is crucial for understanding key drivers of toxicity. We aimed to study associations between prenatal exposure to synthetic phenols and child behaviour, exploring the potential mediating role of maternal steroid hormones. METHODS: We pooled data from two European cohorts: the Barcelona Life Study Cohort (BiSC; Barcelona, Spain, 2018-21, N=1080) a

The Glucocorticoid Receptor Co-Chaperone FKBP51 in the Adrenal Cortex Is Not Involved in Regulating Hypothalamic-Pituitary-Adrenal Activity in the Mouse.

The FK506 binding protein 51 (FKBP51) is a stress-modulating protein implicated in stress-related psychiatric disorders. While FKBP51 has been extensively studied in the brain and other tissues, its role in the adrenal gland, a key part of the stress response, remains unexplored. To investigate Fkbp5 gene expression and FKBP51 function in the adrenal gland, we examined expression in C57Bl/6 male and female mice and generated an adrenal cortex-specific Fkbp5 knockout (ASFkbp5-/-). We found Fkbp5 mRNA expression throughout the adrenal gland in both male and female mice. However, deletion of Fkbp

Cumulative environmental stress and emerging cardiometabolic risk during childhood.

OBJECTIVE: To prospectively evaluate the relationship between cumulative environmental stress and cardiometabolic risk in middle childhood, and to examine whether hair cortisol, a measure of hypothalamic pituitary adrenal-axis activity, mediates this relationship. METHODS: In a cohort of children from low-income households (n = 320; 59% Hispanic, 23% Black, body mass index (BMI) percentile >50th at enrollment), environmental stressors including family and neighbourhood factors representing disadvantage/deprivation, and cortisol concentrations from hair samples, were measured over five timepoi

Genotypic variation in the promoter region of the CRH-248 gene interacts with early rearing experiences to disrupt the development of the HPA axis in infant rhesus macaques (Macaca mulatta).

Aberrant functioning of the hypothalamic-pituitary-adrenal (HPA) axis is a hallmark of conditions such as depression, anxiety disorders, and post-traumatic stress disorder. Early-life adversity and genetic variation can interaction to disrupt HPA axis regulation, potentially contributing to certain forms of psychopathology. This study employs a rhesus macaque model to investigate how early parental neglect interacts with a single nucleotide polymorphism within the promoter region of the corticotropin-releasing hormone (CRH-248) gene, impacting the development of the HPA axis. For the initial s

Electroacupuncture Ameliorates Hypothalamic‒Pituitary‒Adrenal Axis Dysfunction Induced by Surgical Trauma in Mice Through the Hypothalamic Oxytocin System.

Electroacupuncture (EA) can effectively reduce surgical stress reactions and promote postoperative recovery, but the mechanisms remain unclear. The present study aims to examine the effects of EA on the hyperactivity of the hypothalamic‒pituitary‒adrenal (HPA) axis and investigate its potential mechanisms. Male C57BL/6 mice were subjected to partial hepatectomy (HT). The results showed that HT increased the concentrations of corticotrophin-releasing hormone (CRH), corticosterone (CORT), and adrenocorticotropic hormone (ACTH) in the peripheral blood and upregulated the expression of CRH and glu

Ginsenoside Rg3 modulates spatial memory and fear memory extinction by the HPA axis and BDNF-TrkB pathway in a rat post-traumatic stress disorder.

Post-traumatic stress disorder (PTSD) is a serious mental disorder that can develop after exposure to extreme stress. Korean red ginseng, whose major active component is ginsenoside Rg3 (Rg3), is a widely used traditional antioxidant that has anti-inflammatory, anti-apoptotic and anxiolytics effects. This study investigated whether the administration of Rg3 ameliorated the memory deficit induced by a single prolonged stress (SPS) in rats. Male rats were dosed with Rg3 (25 or 50 mg/kg) once daily for 14 days after exposure to SPS. Rg3 administration improved fear memory and spatial memory might

Chronic Systemic Dexamethasone Regulates the Mineralocorticoid/Glucocorticoid Pathways Balance in Rat Ocular Tissues.

Central serous chorioretinopathy (CSCR) is a retinal disease affecting the retinal pigment epithelium (RPE) and the choroid. This is a recognized side-effect of glucocorticoids (GCs), administered through nasal, articular, oral and dermal routes. However, CSCR does not occur after intraocular GCs administration, suggesting that a hypothalamic-pituitary-adrenal axis (HPA) brake could play a role in the mechanistic link between CSCR and GS. The aim of this study was to explore this hypothesis. To induce HPA brake, Lewis rats received a systemic injection of dexamethasone daily for five days. Con

Intermittent fasting ameliorates di-(2-ethylhexyl) phthalate-induced precocious puberty in female rats: A study of the hypothalamic-pituitary-gonadal axis.

Di-(2-ethylhexyl) phthalate has been reported to interfere with the development and function of animal reproductive systems. However, hardly any studies provide methods to minimize or prevent the adverse effects of DEHP on reproduction. The energy balance state of mammals is closely related to reproductive activities, and the reproductive axis can regulate reproductive activities according to changes in the body's energy balance state. In this study, the effects of every other day fasting (EODF), as a way of intermittent fasting, on preventing the precocious puberty induced by DEHP in female r

Gut-Brain-Microbiota Axis: Antibiotics and Functional Gastrointestinal Disorders.

Gut microbiota composition and function are major areas of research for functional gastrointestinal disorders. There is a connection between gastrointestinal tract and central nervous system and this is mediated by neurotransmitters, inflammatory cytokines, the vagus nerve and the hypothalamic-pituitary-adrenal axis. Functional gastrointestinal disorders are prevalent diseases affecting more than one third of the population. The etiology of these disorders is not clarified. Visceral hyperalgesia is the main hypothesis for explaining clinical symptoms, however gut-brain axis disorder is a new t

The influence of hypertensive environment on adipose tissue remodeling measured by fluorescence lifetime imaging in spontaneously hypertensive rats.

There is a lack of information correlating low adiposity with hypertension experienced by Spontaneous Hypertensive Rats (SHR) or overweight and normotension in Wistar-Kyoto (WKY). We aimed to investigate this lipodystrophy phenomenon by measuring fluorescence lifetime (FLIM), optical redox ratio (ORR), serum levels of hypothalamic-pituitary-adrenal (HPA) and/or hypothalamic-pituitary-thyroid (HPT) hormones axes between Wistar, WKY and SHR before and after establishment of hypertension. Under high blood pressure, we evaluated serum adipokines. Brown adipose tissue was characterized as lower ORR
