Predictive mapping of human risk for West Nile virus (WNV) based on environmental and socioeconomic factors.

A West Nile virus (WNV) human risk map was developed for Suffolk County, New York utilizing a case-control approach to explore the association between the risk of vector-borne WNV and habitat, landscape, virus activity, and socioeconomic variables derived from publically available datasets. Results of logistic regression modeling for the time period between 2000 and 2004 revealed that higher proportion of population with college education, increased habitat fragmentation, and proximity to WNV positive mosquito pools were strongly associated with WNV human risk. Similar to previous investigatio

Increase in the size of the amino acid pool is sufficient to activate translation of early trypsin mRNA in Aedes aegypti midgut.

Early trypsin is a female-specific protease present in the midgut of the yellow fever mosquito Aedes aegypti during the first 4-6 h after ingestion of a blood meal. Transcription of the early trypsin gene occurs after adult emergence under control of juvenile hormone, but the transcript remains untranslated before feeding. Early trypsin was in vitro translated using mRNA extracted from midguts of unfed and fed females, indicating that there are not structural features in the early trypsin mRNA that impede translation in vitro. Eight single protein meals exhibiting different molecular weights a

Herd immunity to filarial infection is a function of vector biting rate.

Despite the existence of an impressive body of work on human immune responses against filarial infections, the occurrence of a protective response to infection remains unclear. Here, we use a combined modelling and comparative data analysis framework to address this issue for human infections with the filarial parasite, Wuchereria bancrofti. By analogy with previous work, the analysis involves the comparison of observed field patterns of infection with epidemiological patterns predicted by a mathematical model of parasite immunity. Unlike most other human helminths, which are transmitted by in

Detection of Dengue Virus in Aedes aegypti Eggs Supports Vertical Transmission in Natural Settings.

Dengue remains one of the most important mosquito-borne viral diseases worldwide, and vector control continues to be the cornerstone of disease prevention. Vertical transmission has been proposed as a mechanism contributing to the maintenance of dengue virus (DENV) during interepidemic periods. In this study, field-collected egg pools and second-generation eggs derived from field-collected females from the state of Rio de Janeiro were analyzed by RT-qPCR, Sanger sequencing, plaque assay, and transmission electron microscopy. The second-generation egg samples (E1-E4), corresponding to F2 eggs o

Chronic active Epstein-Barr virus disease in children: Clinical outcomes and prognostic risk factors from a 149-patient cohort.

Chronic active Epstein-Barr virus disease (CAEBV), an Epstein-Barr virus (EBV)+ T/Natural Killer (NK)-cell lymphoproliferative disorder, remains critically undefined in treatment and prognostic stratification due to its rarity. We analysed 149 paediatric systemic CAEBV patients, of whom 134 received LDEP (liposomal doxorubicin, etoposide, pegylated asparaginase and methylprednisolone) chemotherapy as a bridge to haematopoietic stem cell transplantation (HSCT). Diagnostic delay was common (median 6.0 months), with 41.6% presenting atypically with localized symptoms at onset. Haemophagocytic lym

Robust Bioconjugated Antigens Induce Immune Responses Preventing Malaria Infection and its Transmission.

Malaria remains a major global health challenge, with current vaccines providing only limited reductions in case numbers. This study introduces an innovative approach that utilizes engineered Escherichia coli to bioconjugate and assemble the pre-erythrocytic circumsporozoite protein-derived RTS,S antigen and the sexual-stage Pfs47 subdomain into biopolymer particles (BPs), generating the dual-antigen Pfs47-RTS,S-BP vaccine. Compared to single-antigen BPs, the fused Pfs47-RTS,S-BP formulation shows synergistic enhancement in immunogenicity and protective efficacy. Pfs47‑RTS,S‑BP retains its phy

Re-emergence of DENV-3 in Paraguay after two decades: a genomic and epidemiological investigation.

Dengue is one of the most rapidly expanding mosquito-borne viral diseases worldwide, posing a persistent public health challenge across tropical and subtropical regions. Here, we report the re-emergence of dengue virus serotype 3 (DENV-3) in Paraguay following approximately 15 years without confirmed detection. In early 2025, twelve laboratory-confirmed cases were identified in the metropolitan region of Asunción through routine epidemiological surveillance and molecular diagnostics. To investigate the origin and evolutionary context of these infections, whole-genome sequencing and phylogeneti

Burden of malaria in young adults in Niger: A prospective study in a military setting.

In Niger, malaria remains a major public health issue and cause of economic loss, but its impact on young adults remains poorly documented. Although this population is central to the country's development, it is rarely studied. Our study aims to fill this gap by assessing the burden of malaria in three cohorts of military recruits monitored in different training centers in Niger, a population representative of urban youth. A prospective cohort study was conducted from September to December 2023 in three military health centers. Participants (aged 17-55) had free access to care. All consultatio

Early circulating biomarkers to predict plasma leakage in dengue fever.

BACKGROUND: Dengue, a mosquito-borne viral infection, poses a rapidly growing burden, particularly in low- and middle-income countries. Without early identification of patients at risk of severe outcomes (dengue haemorrhagic fever, severe dengue, and plasma leakage- the latter typically occurring on days 5-7 of illness), untriaged admissions lead to hospital overcrowding and suboptimal care. METHODS: This nested case-control study compared early-stage plasma samples (within the first 96 hours of fever) from dengue patients with and without plasma leakage. Thirty-four potential biomarkers, sel

Toward ecological forecasting of West Nile virus in Florida: Insights from two decades of sentinel chicken surveillance.

West Nile Virus (WNV) is the leading cause of mosquito-borne disease in the United States, yet transmission activity remains difficult to predict. The present study used 20 years of digitized WNV seroconversion data from 526 sentinel chicken coops across Florida to develop spatiotemporal models with landscape and climate variables to predict WNV seroconversion at monthly and seasonal timescales. We found several environmental predictors hypothesized to impact WNV transmission were important at both timescales. Seroconversion of WNV was predicted to decrease with higher maximum temperatures dur

Construction and Evaluation of a Chimeric Japanese Encephalitis Virus Vaccine Candidate Strain with Chaoyang Virus as the Backbone.

BACKGROUND: Pathogenic flaviviruses pose a serious threat to human health, and vaccines are an effective means of prevention and control. Although related vaccines have achieved significant progress, safety and efficacy limitations still exist, urgently requiring the development of novel vaccine platforms. The insect-specific flavivirus Chaoyang virus (CYV), with a structure similar to pathogenic flaviviruses and limited to insect cell replication, has potential as a safe vaccine vector. METHODS: To systematically evaluate CYV's potential as a universal flavivirus vaccine backbone and provide

Resurgence of Dengue Virus Serotype 3 in Minas Gerais, Brazil: A Case Report.

This report provides a detailed overview of the resurgence of DENV-3 in the state of Minas Gerais, Brazil, which is a concerning scenario in the context of dengue, a mosquito-borne viral disease. Historically, Brazil has grappled with dengue epidemics caused primarily by the DENV-1 and DENV-2 serotypes. However, in 2023, a significant shift in this pattern was observed as DENV-3 made a notable resurgence. This resurgence was characterized by the increase in DENV-3 cases within the country and the region of the Americas. Given the absence of sustained DENV-3 circulation in Brazil in previous ye
