Predicting Bioconcentration Factors and Concentrations of Per- and Polyfluoroalkyl Substances in Plants with Machine Learning.

Bioconcentration factors (BCF) and concentrations of per- and polyfluoroalkyl substances (PFAS) in crops were predicted by four machine learning models using a newly compiled data set composed of 11 crops, 11 plant tissue compartments, and 21 PFAS, as well as diverse soil properties and cultivation conditions. The eXtreme Gradient Boosting (XGBoost) achieved the best performance, with a coefficient of determination (R2) of 0.951 for Log BCF and 0.943 for Log C. Feature importance analysis showed that soil pH and PFAS carbon chain length are key predictors of BCF. Soil PFAS concentration is the

Predicting the tabletability of binary mixtures from individual powder compaction behavior.

Successful development of direct-compression formulations can be hindered by poor compactibility of active pharmaceutical ingredients active pharmaceutical ingredients (APIs), necessitating rational formulation strategies. This work presents a neural network model trained on a large tableting dataset comprising over 200 formulations prepared from 33 powders, including 17 APIs, to predict tablet tensile strength across the full compaction pressure range directly from material properties and formulation composition for binary mixtures. The predictive accuracy of the neural network model was comp

Uncovering complex correlations between multidimensional factors and breastfeeding duration using XGBoost.

BACKGROUND: Breastfeeding duration is influenced by a complex interplay of obstetric, physiological, and sociodemographic factors. Traditional linear statistical models often struggle to capture the high-dimensional, non-linear synergistic effects among these variables, while complex deep learning models are prone to overfitting on small clinical datasets. METHODS: To address these challenges, this study proposes an interpretable machine learning framework based on eXtreme Gradient Boosting (XGBoost). We analyzed clinical follow-up data from 210 postpartum women. The framework incorporates Ex

Multivariate and Online Transfer Learning With Uncertainty Quantification.

Untreated periodontitis causes inflammation within the supporting tissue of the teeth and can ultimately lead to tooth loss. Modeling periodontal outcomes is beneficial as they are difficult and time-consuming to measure, but disparities in representation between demographic groups must be considered. There may not be enough participants to build group-specific models, and it can be ineffective, and even dangerous, to apply a model to participants in an underrepresented group if demographic differences were not considered during training. We propose an extension to the RECaST Bayesian transfer

Dual-Attention Multimodal Framework for Molecular Property Prediction.

Molecular property prediction plays a vital role in drug discovery and chemical research by facilitating the efficient selection and optimization of candidate compounds. Traditional Quantitative Structure-Activity Relationship models and early machine learning methods often rely on handcrafted molecular descriptors, which limit their generalizability and predictive performance. Recent advancements have shown that the powerful representation capabilities of Large Language Models on textual molecular data, combined with the relational learning strengths of Attention-based Graph Neural Networks,

Machine learning-based integration identifies a 10-gene predictive signature and its classification patterns in schizophrenia.

BACKGROUND: Schizophrenia (SCZ) is a highly heritability psychological disorder, however the exact etiology remains unclear, and lack of the reliable and effective biomarkers for diagnosis and treatments in the management of SCZ, thus exploring the novel biomarkers in SCZ may enhance the efficacy of its predictive, preventive, and personalized medicine (PPPM/3PM) approach. METHODS: Based on differentially expressed genes (DEGs) and weighted gene co-expression network (WGCNA) analyses from five brain datasets, we screened SCZ-key genes and then developed a novel machine learning (ML) framework

Challenges in Preprocessing Routine Laboratory Data for Machine Learning.

Post-Covid syndrome remains a major clinical challenge due to the lack of specific biomarkers and reliance on exclusion criteria. Routine laboratory data offers potential for identifying biological signatures, but their use in machine learning is hampered by data quality issues. This study investigates the impact of preprocessing on classification performance using 52 laboratory parameters from the German NAPKON cohort (n = 1,292: 1,130 Covid recovered, 162 post-Covid patients), measured at four time points. Data preprocessing included unit harmonization, handling of missing values and statist

EIGNN: An Explainable Imaging-Genetic Neural Network for Robust Alzheimer's Disease Risk Prediction.

Accurate risk prediction and early diagnosis are crucial for the early intervention of Alzheimer's disease (AD). Current prediction models usually have limited power in capturing the complex interplays between the heterogeneous inputs or lack biological explainability required for clinical adoption and new diagnosis biomarkers discovery. Inspired by pioneering works on biologically informed network and multi-modal learning, we presented an Explainable Imaging-Genetic Neural Network (EIGNN), integrating genetic and neuroimaging data to generate accurate, robust, and explainable AD risk predicti

Modeling Individual-Level Uncertainty From Missing Data in Multifactorial Breast Cancer Risk Prediction.

PURPOSE: Multifactorial breast cancer (BC) risk prediction models use a range of predictors to estimate an individual's chance of developing BC. Data on risk factors are often incomplete, and point estimates calculated when data are missing can mask considerable uncertainty. Quantifying this uncertainty is critical for effective risk communication. METHODS: We used Monte Carlo simulation methods to estimate the distribution of 10-year BC risk for individuals with missing data, using the BOADICEA multifactorial model as an example. Multivariate imputation by chained equations with large repres

Cyclist behavioral persistence across consecutive intersections: Re-ID based analysis and intention prediction.

The safety risks associated with cyclists at signalized intersections are often analyzed within isolated units, an approach that fails to account for the inherent behavioral dependencies across consecutive intersections. Ignoring such cross-intersection persistence results in a fragmented understanding of cyclist decision-making and hinders the early identification of risky behaviors. This study investigates cyclists' behavioral persistence across consecutive signalized intersections using high-resolution trajectory data collected from unmanned aerial vehicles and roadside cameras. A re-identi

Methodologic Insights on Building and Evaluating Models for Early Warning of Hypotension during Surgery.

BACKGROUND: Hypotension prediction has attracted considerable attention in the medical community, leading to numerous publications on the topic. Several data-driven models have been proposed, but framing, data selection, and evaluation metrics differ widely in the literature. METHODS: Using datasets from noncardiac and cardiac surgery and a forward framing, the authors assess how data selection affects model performance. The study compares models trained and tested with or without segments containing ongoing hypotension at prediction time or interventions that could affect the classification

A simple personalised prediction model for hip fracture risk based on mechanistic parameters.

Fragility hip fractures in older adults are a major burden for the individual and the society, as they result in increased morbidity, mortality and substantial health care expenditure. Most fragility hip fractures are caused by falls, resulting in an impact on the postero-lateral aspect of the hip, inducing a force that exceeds the femoral bone strength. The risk of fracture depends on the rate of falling, the fall-induced impact force, and the femoral bone strength. This article presents the framework of a novel fragility hip fracture risk calculator based on a combined mechanistic and stocha

Leveraging regularity in COVID-19 growth rate dynamics for epidemic wave forecasting.

Real-time forecasting of infectious diseases is essential for public health decision-making. Traditional forecasting methods for epidemics exhibiting repeated waves, such as influenza and RSV, rely on strongly regular patterns in the incidence data, with stable duration (i.e., one year), peak timing, magnitude and initial incidence. In 2022-2023, COVID-19 epidemic waves became more regular compared to early in the pandemic, but they were nonetheless characterised by an absence of seasonality that shaped their overall trajectories, rendering existing methods unfit for use. Furthermore, the tota

Development and external validation of a risk prediction model for hypertension among individuals with established hypothyroidism.

BACKGROUND: Hypothyroidism represents a multisystem endocrine disorder involving cardiovascular, renal, and metabolic alterations beyond thyroid hormone deficiency. However, validated tools for hypertension (HTN) risk stratification specifically designed for patients with hypothyroidism remain lacking. This study aimed to develop and externally validate a hypothyroidism-specific HTN prediction model incorporating routinely available clinical variables. METHODS: This retrospective multicohort study included patients with hypothyroidism from a hospital-based cohort and an independent external p

LMF-CP: An Interpretable Multimodal Late-Fusion Framework for Compound Carcinogenicity Prediction.

Accurately predicting the carcinogenicity of compounds is of great significance for drug discovery, clinical drug safety, and chemical risk assessment. Traditional methods for assessing carcinogenicity rely on animal testing, which suffers from limitations such as time-consuming processes, high costs, significant interspecies differences, and low predictive throughput. In recent years, computational modeling-based prediction methods (such as Quantitative Structure-Activity Relationships, QSAR) have made some progress, but they still face challenges such as insufficient molecular feature inform

Management of Atypical Haemolytic Uraemic Syndrome With Triggers: Diagnostic and Treatment Algorithms From an Asia-Pacific Perspective.

Complement-amplifying events/conditions associated with thrombotic microangiopathy (TMA) include pregnancy/postpartum period, severe hypertension, autoimmune diseases, drug exposures, infections and organ transplantation. Some of these 'triggers' may exist comorbidly with atypical haemolytic uraemic syndrome (aHUS; a complement-mediated form of TMA), unmask previously undiagnosed aHUS, or occur secondary to aHUS, thus creating a considerable diagnostic challenge. A major goal in patients presenting with TMA is to differentiate complement-mediated aHUS from other causes of TMA such that appropr
