BACKGROUND: Hyperkinetic movement disorders arise from dysfunction within cortico-basal ganglia-cerebellar loops. They frequently involve psychiatric and cognitive symptoms, reflecting impairment of both motor and non-motor domains within these loops. ADCY5 (MxMD-ADCY5) and SGCE (MYC/DYT-SGCE) related movement disorders are childhood-onset monogenic hyperkinetic conditions, both characterized by myoclonus, dystonia, and frequent psychiatric manifestations. Previous evidence suggests predominant basal ganglia involvement in MxMD-ADCY5 and cerebellar involvement in MYC/DYT-SGCE.

Aphasia is increasingly understood as a disorder of disrupted language networks rather than damage to isolated language regions. However, most resting-state functional connectivity studies treat people with aphasia as a single group or classify individuals by overall severity, potentially obscuring qualitative differences in network organization across aphasia types. The present study examined how resting-state functional connectivity varies across aphasia types and how aphasia severity relates to network organization following left-hemisphere stroke. Resting-state fMRI data were analyzed from

BACKGROUND: While analyzing distance-dependent functional connectivity strength (FCS) yields valuable information about brain activity, whether the FCS alterations found in obsessive-compulsive disorder (OCD) are linked to inter-regional anatomical distance is still unknown. This study investigated distance-associated functional reorganization in OCD and its associations with clinical assessment data and neurotransmitter receptor/transporter densities.
METHODS: To quantify FCS across global, long-range, and short-range scales, we analyzed resting-state functional MRI data from 99 OCD patients

BACKGROUND: Emotional disorders (EDs), including anxiety disorders, major depression (MDD), and post-traumatic stress disorder (PTSD), show overlapping symptoms and neurobiological heterogeneity, limiting current diagnostic frameworks. Transdiagnostic biomarkers are critical for precision psychiatry.
METHODS: Resting-state functional connectivity (FC) from 261 ED patients and 201 healthy controls (HCs) was decomposed via nonnegative matrix factorization (NMF). Differential FC features defined latent disease factors, classifying patients into subtypes with distinct network/structural profiles.

BACKGROUND/OBJECTIVES: Spastic paraplegia type 4 (SPG4), the most prevalent pure Hereditary Spastic Paraplegia (HSP), is mainly characterized by progressive lower limb spastic weakness, due to corticospinal system degeneration. However, more recent studies suggest a widespread pathophysiologic involvement of additional systems. We here investigated alterations in resting-state functional connectivity (FC) and large-scale brain network topology in SPG4, and their relationship with clinical features.
METHODS: Forty patients with SPG4 and 40 age- and sex-matched healthy controls underwent 3T MRI

INTRODUCTION: Monoaminergic neurotransmitters help regulate the interplay between the central nervous system and the immune system. Aberrant monoaminergic functional connectivity is presumably involved in MS-related fatigue, but mechanisms remain unclear.
AIM: To investigate how static and dynamic functional connectivity of monoaminergic circuits relate to fatigue in MS.
METHODS: 60 healthy controls (HC) and 217 people with MS (pwMS) completed the Fatigue Scale for Motor and Cognitive Functions (FSMC) and underwent functional brain MRI. Brain regions were assigned to resting-state networks.

Lesion-symptom mapping is widely used to identify causal relationships between brain structures and behaviour, and has played a central role in neuropsychologically informed network models of cognition. However, even recent approaches remain constrained by a one-to-one mapping framework, which oversimplifies the complex relationships between network-level damage and cognitive deficits. In addition, the non-orthogonality of cortical and white matter damage makes it difficult to disentangle their distinct contributions. Here, we used graph-based multilayer network analysis to address these limit

Clinical observation has identified cerebellar cognitive affective syndrome, which is characterized by various non-motor dysfunctions such as social disorders and anxiety. Increasing evidence has revealed reciprocal mono-/poly-synaptic connections of cerebello-cerebral circuits, forming the concept of the cerebellar connectome. In this study, we demonstrate that neurons in the cerebellar nuclei (CN) of male mice project to a subset of zona incerta (ZI) neurons through long-range glutamatergic and GABAergic transmissions, both capable of encoding acute stress. Furthermore, activating or inhibit

Functional networks characterized by coherent neural activity across distributed brain regions have been observed to emerge early in neurodevelopment. Synchronized maturation across regions that relate to functional connectivity (FC) could be partially reflected in the developmental changes in underlying microstructure. Nevertheless, covariation of regional microstructural properties, termed "microstructural connectivity" (MC), and its relationship to the emergence of functional specialization during the early neurodevelopmental period remain poorly understood. We investigated the evolution of

BACKGROUND: Severe worry is a core component of anxiety and depressive disorders and is independently associated with significant morbidity and mortality. However, the neural basis of worry is poorly understood. We investigated effective connectivity (EC) using functional magnetic resonance imaging of a naturalistic worry induction and reappraisal task in late life.
METHODS: A total of 112 participants age >50 years with varying worry severity completed a personalized, in-scanner worry induction and reappraisal task. We calculated voxelwise EC in neutral, worry, and reappraisal conditions wit

The human brain operates as a complex network, and understanding its functional connectivity is a core challenge in neuroscience. Functional near-infrared spectroscopy (fNIRS) offers a non-invasive, portable method for studying brain activity and connectivity, providing valuable insights into the brain's network dynamics. In this study, we used fNIRS to examine the functional connectivity of the human brain during the Dual n-back task, a cognitive challenge that varies in memory load (0-back, 1-back, and 2-back). Data were collected from 24 channels in the frontal cortex and pre-processed with

Cardiorespiratory fitness may protect against or alleviate depression through neuroplastic mechanisms within disrupted brain networks underlying cognitive and affective symptoms. There is a scarcity of research examining the effects of cardiorespiratory fitness on functional brain networks in representative samples spanning adulthood. Further, little is known about the interaction of cardiorespiratory fitness with depression in these networks. To address this problem, this study made use of the NKI-Rockland sample dataset and magnetic resonance imaging to examine whether functional brain netwo

OBJECTIVE: Subjects with left temporal lobe epilepsy may either show altered hemispheric language lateralization or retain typical, left lateralization. Examining the integrity of white matter pathways involved in the adaptation or maintenance of language lateralization in these patients could have important clinical implications for preserving or potentiating compensatory language mechanisms.
METHODS: We combined task functional magnetic resonance imaging and structural diffusion metrics to determine the dependency of lobe-based language laterality on white matter integrity in healthy partic

BACKGROUND: Major depressive disorder (MDD) is a main risk factor of suicide, emphasizing the urgent need for understanding the neurobiological mechanisms underlying suicide attempts (SAs) in depressive patients. We hypothesized that aberrant frequency-specific functional connectivity patterns underlying an executive and inhibition task might be associated with SA in depression.
METHODS: The current study enrolled 143 subjects including 43 healthy controls and 87 patients with MDD (43 patients with SA and 44 without SA), who attended a GO/NOGO task during the magnetoencephalography recording.

Cognitive control refers to a set of mental processes that enable individuals to flexibly and adaptively engage in goal-directed behavior. Adolescence is characterized by the emergence and subsequent rapid development of adult-like cognitive control abilities, and as such, there is great interest in understanding the neural basis of this maturational process. The goal of the present study was to investigate how the resting-state and task-based functional connectivity (FC) patterns of two brain networks implicated in control processes, the fronto-parietal network (FPN) and the cingulo-opercular

Obsessive-Compulsive Disorder (OCD) may involve an imbalance between goal-directed and habitual learning systems, and this study investigates the structural and functional brain networks underpinning these systems in OCD. Using predefined brain regions, structural and functional connectivity networks were constructed, and methods such as network-based statistics, average connectivity strength, structural-functional coupling, and partial least squares path modeling were employed to compare OCD patients and healthy controls. The results revealed that OCD patients showed increased structural conn

Schizophrenia (SZ) is a complex psychiatric disorder that is currently defined by symptomatic and behavioral, rather than biological, criteria. Neuroimaging is an appealing avenue for SZ biomarker development, as several neuroimaging-based studies have shown measurable group differences in brain structure, as well as functional brain alterations in both static and dynamic functional network connectivity (sFNC and dFNC, respectively), between SZ and controls. The recently proposed filter-banked connectivity (FBC) method extends the standard dFNC sliding-window approach to estimate FNC within an

Behavioral and imaging studies suggests that emotional biases in the perception of faces associated with major depression disorder (MD) may be embedded within a broader sensory processing deficit. Increased cortical acetylcholine in MD suggest that this deficit may be related to abnormal attention modulation of sensory areas. It is not clear, however, whether these problems are a manifestation of the disease or whether they precede symptom onset. To investigate this, we applied functional magnetic resonance imaging (fMRI) to look for brain activity changes that participants with a family risk

The central amygdaloid nucleus (CeA) and the lateral habenular nucleus (LHb) are essential nuclei playing modulatory roles in encoding noxious stimuli. Their interaction has recently been demonstrated in chronic pain-induced depression. However, little is known about the CeA-LHb pathway in a formalin-induced pain model. In the present study, we aimed to clarify whether the CeA-LHb pathway modulates the formalin-induced pain model using a neuroanatomical tracing method combined with a designer receptor exclusively activated by a designer drugs strategy (DREADD). The results revealed that the Ce

BACKGROUND: The resting-state functional connectivity (FC) patterns of the fronto-parietal network (FPN) and language network (LN) underlying bipolar disorder (BD) are obscure. This study aimed to uncover abnormal FC patterns of FPN and LN underlying BD and their evolution following treatment.
METHODS: Imaging data at rest state and clinical variables were acquired from 82 patients with BD (with 43 finishing the follow-up) and 88 healthy controls (HCs). Seed-based FC analysis was performed, and correlations between FCs and clinical variables were investigated with whole-brain multiple regress
