Outer Membrane Proteins and Efflux Pumps Mediated Multi-Drug Resistance in Salmonella: Rising Threat to Antimicrobial Therapy.

Despite colossal achievements in antibiotic therapy in recent decades, drug-resistant pathogens have remained a leading cause of death and economic loss globally. One such WHO-critical group pathogen is Salmonella. The extensive and inappropriate treatments for Salmonella infections have led from multi-drug resistance (MDR) to extensive drug resistance (XDR). The synergy between efflux-mediated systems and outer membrane proteins (OMPs) may favor MDR in Salmonella. Differential expression of the efflux system and OMPs (influx) and positional mutations are the factors that can be correlated to

Hepatitis C: is a vaccine the solution?

Hepatitis C Virus (HCV) infection is a major cause of chronic hepatitis, cirrhosis, and hepatocellular carcinoma. Following acute infection, 20% of people eliminate the virus over weeks or months and are often asymptomatic. The remaining 80% of people will develop chronic disease, of which approximately 20% will eventually develop liver cirrhosis and 1-5% will develop liver cancer. About 150 million people are chronically infected with HCV, and more than 350,000 people die every year from hepatitis C related liver diseases. The economic cost of hepatitis C is significant both to the individual

The central role of macrophages in trypanosomiasis-associated anemia: rationale for therapeutical approaches.

Bovine African trypanosomiasis causes severe economical problems on the African continent and one of the most prominent immunopathological parameters associated with this parasitic infection is anemia. In this report we review the current knowledge of the mechanisms underlying trypanosomiasis-associated anemia. In first instance, the central role of macrophages and particularly their activation state in determining the outcome of the disease (i.e. trypanosusceptibility versus trypanotolerance) will be discussed. In essence, while persistence of classically activated macrophages (M1) contribute

Need for epidemiological evidence from the developing world to know the cancer-related risk factors.

The existing evidence on cancer etiology has mostly come from epidemiological studies conducted in the developed world. Now there is an urgent need to gather information on cancer risks in developing countries. Due to recent economic, demographic and health transitions, cancers are on the rise in many developing countries. Future epidemiological studies in these countries should address changing diet, level of physical activity, various environmental and occupational exposures, smoking habits and infections, relative to cancers. In many low resource settings western and conventional lifestyles

Streamlined CRISPR-based assays for detection and subtyping of H5 and H7 avian influenza.

Avian influenza viruses (AIVs) are zoonotic pathogens that pose an increasing global threat due to their potential for significant economic losses in agriculture, and the risk of a pandemic should human-to-human transmission occur. In light of this, we need rapid, sensitive, and specific tests to detect and differentiate circulating AIVs. Current AIV diagnostic methods require specialized laboratory equipment, limiting their use in the field and in low-resource settings. Here, we develop three CRISPR-based Streamlined Highlighting of Infections to Navigate Epidemics (SHINE) assays that address

Development of a compact cryogenic Penning trap with permanent magnets: An intermediate step toward the Shanghai Penning Trap.

Penning traps, renowned for their unparalleled precision in determining fundamental properties, such as mass and magnetic moments, are cornerstone instruments in modern physics. Their applications span from nuclear structure studies to stringent tests of quantum electrodynamics and CPT invariance. Although Penning traps have been demonstrated for fundamental studies, often employing superconducting magnets, their high cost and operational complexity remain challenges. In this work, we report the development of a compact cryogenic Penning trap that utilizes a permanent magnet to provide a confi

Age and ocular toxoplasmosis: a narrative review.

Toxoplasma gondii is an extremely "successful" opportunistic parasite that infects most warm-blooded animals, including humans. While the infection is generally largely asymptomatic, the infection of the eye presenting as ocular toxoplasmosis (OT) is a potentially blinding consequence. OT remains the most common cause of infectious retinochoroiditis and places a considerable socio-economic burden on societies, particularly in developing countries. Age is one of several factors influencing the clinical presentation and outcomes of OT. Older patients often exhibit more severe disease manifestati

Exploring the Potential of Microbial Coalbed Methane for Sustainable Energy Development.

By allowing coal to be converted by microorganisms into products like methane, hydrogen, methanol, ethanol, and other products, current coal deposits can be used effectively, cleanly, and sustainably. The intricacies of in situ microbial coal degradation must be understood in order to develop innovative energy production strategies and economically viable industrial microbial mining. This review covers various forms of conversion (such as the use of MECoM, which converts coal into hydrogen), stresses, and in situ use. There is ongoing discussion regarding the effectiveness of field-scale pilot

Rationale for combined therapies in severe-to-critical COVID-19 patients.

An unprecedented global social and economic impact as well as a significant number of fatalities have been brought on by the coronavirus disease 2019 (COVID-19), produced by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Acute SARS-CoV-2 infection can, in certain situations, cause immunological abnormalities, leading to an anomalous innate and adaptive immune response. While most patients only experience mild symptoms and recover without the need for mechanical ventilation, a substantial percentage of those who are affected develop severe respiratory illness, which can be fa

Plant-based production of a protective vaccine antigen against the bovine parasitic nematode Ostertagia ostertagi.

The development of effective recombinant vaccines against parasitic nematodes has been challenging and so far mostly unsuccessful. This has also been the case for Ostertagia ostertagi, an economically important abomasal nematode in cattle, applying recombinant versions of the protective native activation-associated secreted proteins (ASP). To gain insight in key elements required to trigger a protective immune response, the protein structure and N-glycosylation of the native ASP and a non-protective Pichia pastoris recombinant ASP were compared. Both antigens had a highly comparable protein st

Clade 2.3.4.4 H5 chimeric cold-adapted attenuated influenza vaccines induced cross-reactive protection in mice and ferrets.

Clade 2.3.4.4 H5Nx avian influenza viruses (AIVs) have circulated globally and caused substantial economic loss. Increasing numbers of humans have been infected with Clade 2.3.4.4 H5N6 AIVs in recent years. Only a few human influenza vaccines have been licensed to date. However, the licensed live attenuated influenza virus vaccine exhibited the potential of being recombinant with the wild-type influenza A virus (IAV). Therefore, we developed a chimeric cold-adapted attenuated influenza vaccine based on the Clade 2.3.4.4 H5 AIVs. These H5 vaccines demonstrate the advantage of being non-recombin

Role of transcription factors in porcine reproductive and respiratory syndrome virus infection: A review.

Porcine reproductive and respiratory syndrome (PRRS) is an infectious disease caused by the PRRS virus that leads to reproductive disorders and severe dyspnoea in pigs, which has serious economic impacts. One of the reasons PRRSV cannot be effectively controlled is that it has developed countermeasures against the host immune response, allowing it to survive and replicate for long periods. Transcription Factors acts as a bridge in the interactions between the host and PRRSV. PRRSV can create an environment conducive to PRRSV replication through transcription factors acting on miRNAs, inflammat

High-quality nuclear genome for Sarcoptes scabiei-A critical resource for a neglected parasite.

The parasitic mite Sarcoptes scabiei is an economically highly significant parasite of the skin of humans and animals worldwide. In humans, this mite causes a neglected tropical disease (NTD), called scabies. This disease results in major morbidity, disability, stigma and poverty globally and is often associated with secondary bacterial infections. Currently, anti-scabies treatments are not sufficiently effective, resistance to them is emerging and no vaccine is available. Here, we report the first high-quality genome and transcriptomic data for S. scabiei. The genome is 56.6 Mb in size, has a

A review of pretreatment and analysis of macrolides in food (Update Since 2010).

Macrolides are versatile broad-spectrum antibiotics whose activity stems from the presence of a macrolide ring. They are widely used in veterinary medicine to prevent and treat disease. However, because of their improper use and the absence of effective regulation, these compounds pose a threat to human health and the environment. Consequently, simple, quick, economical, and effective techniques are required to analyze macrolides in animal-derived foods, biological samples, and environmental samples. This paper presents a comprehensive overview of the pretreatment and analytical methods used f

Hit-to-Lead Optimization of Benzoxazepinoindazoles As Human African Trypanosomiasis Therapeutics.

Human African trypanosomiasis (HAT) is a neglected tropical disease caused by infection with either of two subspecies of the parasite Trypanosoma brucei. Due to a lack of economic incentive to develop new drugs, current treatments have severe limitations in terms of safety, efficacy, and ease of administration. In an effort to develop new HAT therapeutics, we report the structure-activity relationships around T. brucei for a series of benzoxazepinoindazoles previously identified through a high-throughput screen of human kinase inhibitors, and the subsequent in vivo experiments for HAT. We iden

Computer-Aided Multi-Target Management of Emergent Alzheimer's Disease.

Alzheimer's disease (AD) represents an enormous global health burden in terms of human suffering and economic cost. AD management requires a shift from the prevailing paradigm targeting pathogenesis to design and develop effective drugs with adequate success in clinical trials. Therefore, it is of interest to report a review on amyloid beta (Aβ) effects and other multi-targets including cholinesterase, NFTs, tau protein and TNF associated with brain cell death to be neuro-protective from AD. It should be noted that these molecules have been generated either by target-based or phenotypic method

Structural Basis for the Inhibition of a Phospholipase A2-Like Toxin by Caffeic and Aristolochic Acids.

One of the main challenges in toxicology today is to develop therapeutic alternatives for the treatment of snake venom injuries that are not efficiently neutralized by conventional serum therapy. Venom phospholipases A2 (PLA2s) and PLA2-like proteins play a fundamental role in skeletal muscle necrosis, which can result in permanent sequelae and disability. This leads to economic and social problems, especially in developing countries. In this work, we performed structural and functional studies with Piratoxin-I, a Lys49-PLA2 from Bothropspirajai venom, complexed with two compounds present in s

Duration of immunity to norovirus gastroenteritis.

The duration of immunity to norovirus (NoV) gastroenteritis has been believed to be from 6 months to 2 years. However, several observations are inconsistent with this short period. To gain better estimates of the duration of immunity to NoV, we developed a mathematical model of community NoV transmission. The model was parameterized from the literature and also fit to age-specific incidence data from England and Wales by using maximum likelihood. We developed several scenarios to determine the effect of unknowns regarding transmission and immunity on estimates of the duration of immunity. In t

Challenges in pig artificial insemination.

Semen extended in a liquid state, together with conventional artificial insemination (AI), is the sole sperm technology used by the pig industry. Sperm technologies known for many years, such as cryopreservation, and others developed during recent years, such as sperm sexing, have not yet been integrated into commercial use. Moreover, there has recently been an explosion of new technologies, such as sperm mediated gene transfer or encapsulated spermatozoa which need additional supportive techniques before they can be economically applied to pig breeding. The speed with which the aforementioned
