Pathways to age of onset of heroin use: a structural model approach exploring the relationship of the COMT gene, impulsivity and childhood trauma.

BACKGROUND: The interaction of the association of dopamine genes, impulsivity and childhood trauma with substance abuse remains unclear. OBJECTIVES: To clarify the impacts and the interactions of the Catechol -O-methyltransferase (COMT) gene, impulsivity and childhood trauma on the age of onset of heroin use among heroin dependent patients in China. METHODS: 202 male and 248 female inpatients who meet DSM-IV criteria of heroin dependence were enrolled. Impulsivity and childhood trauma were measured using BIS-11 (Barratt Impulsiveness Scale-11) and ETISR-SF (Early Trauma Inventory Self Report

Psychosocial pathways to inconsistent condom use among male sex workers: personality, drug misuse and criminality.

BACKGROUND: This research compared street male sex workers in Santo André, Brazil, that reported consistent condom use with those that revealed inconsistent condom use with their clients, concerning personality aspects, impulsiveness, alcohol and drug consumption, depressive symptoms, sociodemographic data and criminal involvement. METHODS: Eighty-six male sex workers were evaluated in face-to-face interviews at their place of work. A 'snowball' sampling procedure was used to access this hard-to-reach population. FINDINGS: Male sex workers with inconsistent condom use showed greater involvem

Impulsivity-related brain volume deficits in schizophrenia-addiction comorbidity.

Despite a high prevalence of schizophrenia patients with comorbid substance abuse, little is known about possible impacts on the brain. Hence, our goal was to determine whether addicted and non-addicted schizophrenic patients suffer from different brain deficits. We were especially interested to determine if grey matter volumes were affected by impulsivity. We hypothesized that (comorbid) substance abuse would be associated with enhanced impulsivity and that this enhanced impulsivity would be related to grey matter volume deficits in prefrontal areas. We employed a voxel-based morphometry appr

Interaction between serotonin transporter promoter and dopamine receptor D4 polymorphisms on decision making.

Appropriate decision making is an important brain function to maintain our lives. The Iowa gambling task (IGT) is a tool for decision making under ambiguity. The aims of this study were to evaluate the influence of serotonin transporter linked polymorphic region (5-HTTLPR) and dopamine receptor D4 (DRD4) polymorphisms and their interaction on IGT performance. One hundred fifty-nine normal subjects were involved in this study. All subjects performed the IGT and were genotyped for the triallelic 5-HTTLPR and DRD4 48 bp uVNTR polymorphisms. After controlling for gender, age, and impulsiveness, th

A pilot study of impulsivity and compulsivity in pathological gambling.

We examined the relationship between gambling severity, impulsivity and obsessionality/compulsivity in 38 pathological gamblers, representing the complete Minnesota sample of a randomized, placebo-controlled clinical trial of paroxetine for the treatment of pathological gambling (PG), using Pearson correlations and linear regression models at baseline and treatment endpoint. At baseline, Pathological Gambling Modification of the Yale-Brown Obsessive-Compulsive Scale (PG-YBOCS) scores correlated significantly with those of the Eysenck Impulsiveness Questionnaire (EIQ) Impulsiveness subscale and

A psycho-genetic study of associations between the symptoms of binge eating disorder and those of attention deficit (hyperactivity) disorder.

OBJECTIVE: Some recent studies have reported intriguingly strong correlations between ADHD and obesity. This study examined whether ADHD symptoms were more pronounced in adults with symptoms of binge eating disorder (BE) than in their non-binging obese counterparts, and whether the links were stronger with inattentive vs impulsive/hyperactive symptoms. We also assessed the role of the dopamine D3 receptor in ADHD symptoms since the DRD3 gene has been associated with impulsivity and drug addiction - both relevant features of ADHD. METHODS: A case (BE: n=60) double-control (normal weight: n=61

Childhood-onset pathologic skin picking: clinical characteristics and psychiatric comorbidity.

There has been little research examining clinical correlates of childhood-onset pathologic skin picking in a sample of individuals with a primary diagnosis of pathologic skin picking. Using a sample of 40 consecutive subjects with current pathologic skin picking, we compared subjects with childhood-onset (before 10 years of age) pathologic skin picking to those with later onset on a variety of clinical measures. Symptom severity was examined by assessing time spent picking per day, intensity and frequency of thoughts and urges to pick, and social and occupational functioning. Of the 40 subject

The relationship of behavioural undercontrol to alcoholism in higher-functioning adults.

Externalising behaviours, including the personality characteristics of behavioural undercontrol (BU), represent one of several genetically influenced domains that impact on the alcoholism risk. Because genes explain only about 60% of the vulnerability toward alcohol use disorders (AUDs), an optimal understanding of how such behaviours affect the risk requires evaluation of their impact in the context of additional influences. Few studies have addressed this question regarding BU among relatively well-functioning adults. This paper presents results from testing a BU-based mediational model of r

Cognition and motor control as a function of Delta9-THC concentration in serum and oral fluid: limits of impairment.

Cannabis use has been associated with increased risk of becoming involved in traffic accidents; however, the relation between THC concentration and driver impairment is relatively obscure. The present study was designed to define performance impairment as a function of THC in serum and oral fluid in order to provide a scientific framework to the development of per se limits for driving under the influence of cannabis. Twenty recreational users of cannabis participated in a double-blind, placebo-controlled, three-way cross-over study. Subjects were administered single doses of 0, 250 and 500 mi

Alex is a 9-year-old boy brought to you, his primary care provider, for a "fifth opinion." You have cared for Alex since he was adopted from a Romanian orphanage at 3 years of age. He has been physically healthy with normal growth parameters and no evidence of fetal alcohol syndrome. Alex has long-standing history of social difficulties, impulsivity, lying, controlling, manipulative behaviors, violent outbursts at home with subsequent lack of remorse, and excessive chatter. You referred Alex to an interdisciplinary child development clinic 2 years ago, where he was diagnosed with reactive atta

Differences in Mental Health, Risk Behaviors, and Social Determinants Among Anabolic-Androgenic Steroid Consumers With and Without Co-occurring Methamphetamine Use in Australia.

Objective: People who use anabolic-androgenic steroids (AAS) frequently engage in polypharmacy, with evidence highlighting recurrent co-use with amphetamine-like stimulants. While the independent risks of each substance are established, little is known about their combined impact in treatment-engaged populations. This study compared people who co-use AAS with methamphetamine (AAS-M) and without (AAS-non-meth [AAS-NM]) across mental health, risk behaviors, and social determinants. Methods: Intake data were drawn from Lives Lived Well (LLW), a large Australian non-government alcohol and other dr

Frequency of Suicidal Ideation and Associations With Autonomic, Motor, and Cognitive Dysfunction in People With REM Sleep Behavior Disorders.

BACKGROUND AND OBJECTIVES: REM sleep behavior disorder (RBD) is characterized by loss of REM sleep atonia leading to dream enactment behavior. Isolated RBD (iRBD) is a known prodrome of alpha-synuclein pathology including Parkinson disease (PD) and dementia with Lewy bodies (DLB). Depression is also a common prodromal symptom of PD/DLB, and death by suicide is greater among individuals with PD. Patients with RBD are at greater risk of comorbid depression, but the prevalence of suicidal ideation in this population is unknown. We aim to assess the frequency of suicidal ideation and whether it is

Shared genetic and environmental influences between the broad avoidant/restrictive food intake disorder phenotype and neurodevelopmental traits: a twin study.

IMPORTANCE: Avoidant/restrictive food intake disorder (ARFID) is a feeding and eating disorder characterized by extremely restricted dietary variety and/or quantity resulting in significant physical health impairment and psychosocial dysfunction. ARFID frequently co-occurs with neurodevelopmental conditions, yet the extent to which this co-occurrence reflects shared genetic or environmental influences remains largely unknown, as few twin or genetic studies of ARFID have been conducted. OBJECTIVE: To examine the extent to which genetic and environmental influences contribute to the association

Sustained-release bupropion versus naltrexone in the treatment of pathological gambling: a preliminary blind-rater study.

BACKGROUND: Pathological gambling (PG) is a relatively common and highly disabling impulse control disorder. A range of psychotherapeutic agents, including selective serotonin reuptake inhibitors, mood stabilizers, and opioid antagonists, has been shown to be effective in the treatment of PG. The use of selective serotonin reuptake inhibitors and opioid antagonists for PG is consistent with the observation that PG shares features of both the obsessive-compulsive spectrum disorders and addictive disorders. The aim of the study is to compare the effectiveness of sustained-release bupropion versu

Nerve growth factor gene polymorphisms may be associated with heroin dependence in women but do not mediate specific personality traits.

Heroin dependence (HD) is a complex disease with a substantial genetic contribution and is associated with traits of impulsivity and specific personality traits. The neurotrophic factor nerve growth factor (NGF) may mediate the reward processes in HD. This study aims to investigate whether NGF gene polymorphisms are associated with the co-occurrence of HD and impulsivity/specific personality traits in HD patients. To minimize the potential confounding effects of population stratification, we selected a homogeneous Han Chinese population and recruited 1364 participants (831 HD patients and 533
