Prediction of efficacy of antimicrobial agents in treatment of infections due to Chlamydia trachomatis.

Although Chlamydia trachomatis is readily eradicated by systemic therapy in patients with acute urethritis, systemic therapy is less satisfactory in treatment of chronic trachoma. The activities of antimicrobial agents against C. trachomatis in cell cultures when the antimicrobial agents are added 1 hr after the C. trachomatis (minimal inhibitory concentration [MIC]) predicts efficacy of the drugs in the treatment of urethritis but does not necessarily predict efficacy in the treatment of chronic ocular trachoma. Concentrations of antimicrobial agents required to eradicate C. trachomatis when

In vivo susceptibility of the Legionnaires disease bacterium to ten antimicrobial agents.

The susceptibility of the Legionnaires disease bacterium to various antimicrobial agents was determined by inoculation of embryonated eggs via the yolk sac. When administered prophylactically, the minimal dose of drug preventing all deaths due to the infection was as follows: rifampin, 0.02 mg; gentamicin, 0.25 mg; streptomycin, 0.39 mg; erythromycin, 0.62 mg; sulfadiazine, 1.56 mg; chloramphenicol, 2.50 mg; and cephalothin, 20.0 mg. Smaller amounts delayed deaths, and larger or equal amounts rendered the embryos free of infection. Oxytetracycline in the largest tested amount, 5.0 mg, protecte

Synthesis and antibacterial activity of some derivatives of tolypomycinone. Relationship between structure and activity in ansamycins.

3-Aminotolypomycinoes and 3,16-diamino-16,17-dihydrotolypomycinones are formed by the addition of primary and secondary amines to tolypomycinone, obtained by mild hydrolysis of the antibiotic tolypomycin Y.3-Amino-16,17-dihydrotolypomycinones are formed by the addition of primary and secondary amines to 16,17-dihydrotolypomycinone. In vitro microbiological tests showed high antibacterial activity in compounds obtained by the addition of primary amines, which must be unbranched in the alpha position to the nitrogen atom to position 3 of the naphthoquinone ring. The relationship between structur

Activity of cefamandole and other cephalosporins against aerobic and anaerobic bacteria.

The activity of cefamandole was comparable to that of cephalothin, cefazolin, and cephaloridine against Staphylococcus aureus, Streptococcus pyogenes, and Diplococcus pneumoniae. In contrast, cefamandole was considerably more active than cephalothin, cefazolin, or cephaloridine against gram-negative facultative bacilli, including Haemophilus influenzae, the most striking disparities being noted with indole-positive Proteus and Enterobacter. Bacteroides fragilis was more susceptible to cefoxitin than to cefamandole or cefazolin (median minimal inhibitory concentration, approximately 8, 32, and

Nosocomial enterococcal blood stream infections in the SCOPE Program: antimicrobial resistance, species occurrence, molecular testing results, and laboratory testing accuracy. SCOPE Hospital Study Group.

Characteristics of nosocomial enterococcal blood stream infection (NEBSI) isolates obtained from patients at 41 U.S. hospitals participating in the SCOPE Program were studied. Isolates from 480 episodes of NEBSI were characterized according to species and antimicrobial susceptibility profile. Selected isolates were also identified to species and vancomycin resistance genotype using polymerase chain reaction based methods. Polymerase chain reaction genotyping and ribotyping were used as genetic markers for molecular epidemiologic typing. Enterococci were the third most common cause of nosocomia

Comparative Analysis of Chlorhexidine Derivatives and Alternative Agents on Streptococcus mutans Viability, Biofilm Formation, and Gene Expression.

This study evaluates the antimicrobial and antibiofilm effects of chlorhexidine and related compounds against S. mutans, focusing on bacterial viability, biofilm formation, and expression of key biofilm genes. Eight compounds were tested for their chemical properties, MICs, and interactions via FIC analyses. Time-kill assays assessed bactericidal activity over 24 h, while cytotoxicity was evaluated in human gingival fibroblasts. Gene expression of biofilm-related genes was analyzed to determine transcriptional modulation. Chlorhexidine gluconate and alexidine showed the strongest antimicrobial

Assessment of Enterococcus faecalis biofilm response to amoxicillin and ciprofloxacin using a dynamic in vitro pharmacokinetic-pharmacodynamic model of catheter-associated urinary tract infection.

BACKGROUND: Catheter-associated urinary tract infections (CAUTIs) are among the most common types of healthcare-associated infections. Among uropathogens, Enterococcus faecalis is particularly notable for its strong capacity to form biofilms on urinary catheters, thereby decreasing its susceptibility to antibiotics commonly used to treat CAUTIs. However, current antibiotic dosing strategies still rely on planktonic phenotypes, limiting their applicability to biofilm-associated infections. To address this limitation, a dynamic in vitro pharmacokinetic-pharmacodynamic model was developed to asse

A pharmacometric model assessing the in vitro synergistic effect of a bacteriophage-polymyxin B combination in a clinical multidrug-resistant Klebsiella pneumoniae isolate.

BACKGROUND: Klebsiella pneumoniae represents one of the most critical pathogens globally and bacteriophage (phage) therapy offers a promising alternative for treatment. Phage therapy poses challenges due to its high specificity and fast resistance development in bacteria. One promising option to overcome these challenges is combining phages with conventional antibiotics. OBJECTIVE: This study investigates phage kinetics and pharmacodynamic interactions between novel phage pK8 and polymyxin B against a clinical multidrug-resistant K. pneumoniae. METHODS: Time-kill assays with time-dissolved s

In-vitro analysis of biofilm formation and synergistic antibiotic-phage therapy for amoxicillin-resistant Escherichia coli.

Biofilm formation by Escherichia coli (E. coli) significantly enhances bacterial resistance to antibiotics, complicating treatment, particularly in amoxicillin-resistant strains. Bacteriophage therapy demonstrates potential in treating biofilm-related infections, and the combination of phages and antibiotics (phage-antibiotic synergy, PAS) further enhances efficacy. This is the first study to evaluate PAS using penicillin-class antibiotics against multidrug-resistant (MDR) Gram-negative bacteria, E. coli. E. coli-specific lytic bacteriophages were isolated and characterized. PAS was evaluated

Benzyl isothiocyanate exhibits antibacterial and antibiofilm activity against Fusobacterium nucleatum while preserving viability of human bone marrow mesenchymal stem cells: an in vitro study.

BACKGROUND: Regenerative endodontic procedures require effective disinfection and preservation of stem cell viability, which are essential for successful tissue regeneration. Conventional intracanal medicaments possess antibacterial properties but may negatively impact stem cell survival. Benzyl isothiocyanate (BITC) is a bioactive compound with known antibacterial properties and favorable biocompatibility. METHODS: This study evaluated the antibacterial efficacy of BITC against Fusobacterium nucleatum in both planktonic and biofilm forms and assessed its biocompatibility on human bone marrow

Evaluation of Cecure as a postharvest sanitizer for controlling Listeria monocytogenes on fresh apples.

Recent listeriosis outbreaks linked to fresh apples highlight the need to control Listeria monocytogenes during postharvest handling. Commercially used sanitizers have shown limited efficacy against L. monocytogenes on apple surfaces, indicating a need for alternative interventions. This study evaluated the efficacy of Cecure, a generally recognized as safe (GRAS) antimicrobial solution, in controlling Listeria monocytogenes in solutions, on inoculated apples, and preventing cross-contamination. Cecure demonstrated minimum inhibitory concentrations (MIC) and minimum bactericidal concentrations

High incidence of antibiotic resistance amongst isolates of Helicobacter pylori collected in Nottingham, UK, between 2001 and 2018.

Introduction. Helicobacter pylori is the leading cause of peptic ulcers and gastric cancer. The most common treatment regimens use combinations of two or three antibiotics and a proton pump inhibitor (PPI) to suppress stomach acid. The World Health Organization designated clarithromycin-resistant H. pylori as a high priority pathogen for drug development, due to increasing antibiotic resistance globally.Hypothesis/Gap Statement. There is no routine surveillance of H. pylori primary antimicrobial sensitivities in the UK, and published data are lacking.Aim. This study aimed to characterize antim
