Short communication: effects of dietary inulin and animal density on stress in pigs.

Cortisol is a key glucocorticoid hormone involved in the physiological stress response of animals. Its secretion, largely regulated by the hypothalamic-pituitary-adrenal (HPA) axis, is essential for animal welfare and growth. Recent studies have shown that this stress-regulatory system operates in coordination with the gut microbiota, communicating with the brain through the gut-brain axis. Given that the diet can modulate microbial composition, dietary interventions thus offer potential in modulating HPA activity. Inulin is a prebiotic that promotes beneficial gut microbiota such as Lactobaci

Dietary Fructose Alters Duodenal Mucin Glycosylation and Mucus Production in High-Fat Diet-Fed Mice.

Fructose, a key component of modern diets, is closely linked to the growing prevalence of pediatric obesity and metabolic alterations. Although numerous studies highlight its systemic consequences, including altered carbohydrate and lipid metabolism and increased cardiovascular risk, the direct impact of fructose, particularly its role in modulating mucin composition, a key determinant of the mucosal barrier, remains poorly explored. This study investigated whether fructose supplementation modifies high-fat diet (HFD)-induced changes in duodenal mucin production and whether these effects vary

Mechanism of recombinant Trichinella spiralis antigen p53 alleviating experimental colitis via the IDO-AhR Axis.

BACKGROUND: Inflammatory Bowel Disease (IBD) is a chronic inflammatory disorder characterized by disrupted intestinal immune homeostasis. Emerging evidence indicates that parasitic infections may exert anti-inflammatory effects by modulating host immunity, although the underlying molecular mechanisms remain incompletely elucidated. This study aimed to investigate whether recombinant Trichinella spiralis (T. spiralis) antigen p53 (rTs p53) protects against experimental colitis by regulating the indoleamine 2, 3-dioxygenase (IDO)-aryl hydrocarbon receptor (AhR) axis. METHODS: In vitro assays we

Dahuang Huanglian Decoction alleviates dysbiosis by inhibiting GBP5/NLRP3 signaling pathway-mediated pyroptosis of colonic epithelial cells.

ETHNOPHARMACOLOGICAL RELEVANCE: Currently, the prevalence of dysbiosis is increasing, but its treatment options are limited. Dahuang Huanglian Xiexin Decoction (XXD) is a traditional herbal prescription recorded in the Treatise on Typhoid Fever, with a longstanding application in the treatment of digestive system diseases. It consists mainly of three classical chinese medicinal herbs: Dahuang (Rheum palmatum L.), Huangqin (Scutellaria baicalensis Georgi), and Huanglian (Coptis chinensis Franch.). Previous studies demonstrated the efficacy of XXD in treating dysbiosis. However, the exact underl

Improving insulin resistance by sulforaphane via activating the Bacteroides and Lactobacillus SCFAs-GPR-GLP1 signal axis.

Background: Insulin resistance (IR) is closely associated with non-alcoholic fatty liver disease (NAFLD), and the gut microbiome contributes to the development of NAFLD. Sulforaphane (SFN) is a phytochemical in cruciferous vegetables that could improve lipid metabolism disorder. However, whether SFN can alleviate IR in NAFLD by regulating the intestinal flora remains unclear. Methods: SFN was administered to high fat diet (HFD)-fed Wistar rats for 10 weeks. Gut microbiota was analysed by 16S rRNA sequencing and the short chain fatty acids (SCFAs) by gas chromatography. The expression of tight

Heart Failure Stimulates Tumor Growth by Circulating Factors.

BACKGROUND: Heart failure (HF) survival has improved, and nowadays, many patients with HF die of noncardiac causes, including cancer. Our aim was to investigate whether a causal relationship exists between HF and the development of cancer. METHODS: HF was induced by inflicting large anterior myocardial infarction in APCmin mice, which are prone to developing precancerous intestinal tumors, and tumor growth was measured. In addition, to rule out hemodynamic impairment, a heterotopic heart transplantation model was used in which an infarcted or sham-operated heart was transplanted into a recipi

Enteroendocrine-derived glucagon-like peptide-2 controls intestinal amino acid transport.

OBJECTIVE: Glucagon-like peptide-2 (GLP-2) is co-secreted with GLP-1 from gut endocrine cells, and both peptides act as growth factors to expand the surface area of the mucosal epithelium. Notably, GLP-2 also enhances glucose and lipid transport in enterocytes; however, its actions on control of amino acid (AA) transport remain unclear. Here we examined the mechanisms linking gain and loss of GLP-2 receptor (GLP-2R) signaling to control of intestinal amino acid absorption in mice. METHODS: Absorption, transport, and clearance of essential AAs, specifically lysine, were measured in vivo by Liq

Intestinal immunomodulation. Role of regulative peptides and promising pharmacological activities.

About 50 peptides, and a similar number of peptide receptors, are known to be present in the gut and this amount is likely to rise significantly over the next few years. While there has been a massive research effort to define their functions and their anatomical distribution in the central nervous system (CNS), the understanding of their roles in the gut is far more limited. Classically, the physiological functions include the control of motility, fluids, electrolytes, and digestive enzymes secretion, or vascular and visceral pain function, and more recently, the role-played in cell prolifera

Gastric emptying. Risk versus benefit in the treatment of acute poisoning.

This review examines the various clinical options used to elicit gastric emptying, viz. drug-induced emesis, mechanical pharyngeal stimulation, gastric lavage, and catharsis. Apomorphine and syrup of ipecac are the 2 drugs most frequently used for induction of emesis. Both agents act centrally and, in addition, syrup of ipecac has a peripheral action. Toxins ingested or foods previously eaten may inhibit or enhance emetic action by interfering with mediating and conducting mechanisms. Studies indicate that both syrup of ipecac and apomorphine are similarly effective in inducing emesis; however

Neuro-modulation and bariatric surgery for type 2 diabetes mellitus.

Obesity has reached epidemic proportions and is continuing to grow into one of the leading healthcare issues worldwide. With this development, bariatric surgery has emerged as an acceptable treatment for morbid obesity, generally achieving meaningful and sustained weight loss. In a surprising turn of events, bariatric surgery was also found to be the most effective therapy for type 2 diabetes mellitus (T2DM). This observation has sparked a great deal of research that has improved our understanding of T2DM pathophysiology; it has facilitated the development of medical treatment and is expanding

Oligopeptides from Gynura divaricata improve glycemic control via inhibition of gluconeogenesis and gut-brain axis regulation.

BACKGROUND: Oligopeptides derived from dietary sources are regarded as ideal functional ingredients for nutritional interventions in diabetes due to their favorable bioavailability, target specificity, and safety profiles. Gynura divaricata (GD), a medicinal food plant, has shown hypoglycemic properties; however, the potential of GD-derived oligopeptides in glycemic control and their mechanistic underpinnings remain largely unexplored. PURPOSE: This study aimed to investigate the anti-diabetic efficacy of GD oligopeptides and elucidate their mechanisms of action, particularly via the gut-micr

Evaluation of F18 enterotoxigenic Escherichia coli intestinal attachment and early disease onset in nursery pigs.

INTRODUCTION: Enterotoxigenic Escherichia coli (ETEC) is a major cause of post-weaning diarrhea and reduced performance in nursery pigs. While ETEC pathogenesis is well established, the early epithelial and functional responses to F18 ETEC infection remain poorly defined. This study investigated the effects of F18 ETEC on bacterial attachment, intestinal function, and early epithelial cell responses. METHODS: Ten days post-weaning, 24 individually housed pigs (n = 6/treatment) were orally inoculated with 5 ml of F18 ETEC at 107, 108, or 109 colony-forming units (cfu)/ml, or with a negative co

Whole-body Bacteriophage Distribution Characterized by a Physiologically based Pharmacokinetic Model.

In 2019 there were over 2.8 million cases of antibiotic-resistant bacterial infection in the US with gram negative organisms having up to a 6% rate of mortality. Bacteriophage (phage) therapy holds great promise to treat such infections. However, the biologic features which influence the pharmacokinetics (PK) of phage have been difficult to characterize due to a lack of standardized protocols of phage purification, tissue assay, and labeling. Here we present robust methods for ultrapure phage preparation as well as non-destructive highly stable attachment of radio-iodide to phage using the wel

Exploring optimal folic acid supplementation levels for lactating-pregnant rabbit does with different litter size.

The lactation-pregnancy overlap in the industrialized 49-d breeding model increases nutritional demands for lactating-pregnant rabbit does. This study examined the effects of folic acid (FA) on the production performance and intestinal microflora of does with different litter sizes (LS, or number of kits). A total of 144 third-parity Hyplus does, aged 11 mo and weighing approximately 5.00 ± 0.07 kg, were divided into 4 treatment groups: control group (basal diet) and FA groups (basal diet + 15, 30, and 45 mg/kg FA). Does with LS of 5, 7, and 9 were selected after weaning. Our findings revealed
