Posttranslational lysine acetylation and methylation are generally considered mutually exclusive modifications of the ε-amino group. The recent discovery of N-acetyl-N-methyl lysine (KAcMe) on histone H4 showed that both marks can coexist on the same lysine side chain, raising the question of whether this dual modification is enzymatically reversible. Here, we synthesized model peptides containing N-acyl lysine or N-acyl-N-methyl lysine residues and tested their processing by Zn2+-dependent HDACs and NAD+-dependent sirtuins. While the corresponding unmethylated substrates were deacylated by th

Acyl glucuronides (AG), reactive metabolites formed via the glucuronidation of carboxylic acid-containing drugs such as non-steroidal anti-inflammatory drugs (NSAIDs), are closely associated with drug-induced liver injury (DILI), particularly idiosyncratic types. AG can covalently bind to proteins, potentially disrupting their function and triggering immune responses. Their toxicity is influenced by chemical instability, stereoselectivity, and interactions with specific uridine 5'-diphospho-glucuronosyltransferase (UGT) isoforms. Transporters such as multidrug resistance-associated protein 2 (

AIM: Eltrombopag, a thrombopoietin receptor agonist for aplastic anaemia, can cause hepatotoxicity. It is metabolized by uridine diphosphate-glucuronosyltransferase (UGT) 1A1 to form an acyl glucuronide metabolite; however, the relationships among serum concentrations of the acyl glucuronide metabolite, UGT1A1 genotypes and hepatotoxicity remain unclear.
METHOD: We examined the serum concentrations of eltrombopag and its acyl glucuronide metabolite, UGT1A1 genotypes and occurrence of hepatotoxicity (abnormal liver function tests or drug-induced liver injury [DILI]) in 104 samples from 42 pati

Microbial production of 1,3-butanediol (1,3-BDO) offers a renewable route to this versatile C4 chemical. However, the low performance of CoA-acylating butyraldehyde dehydrogenase (Bld), which contains a catalytic cysteine, limits efficient production in recombinant Escherichia coli (E. coli). In this study, wild-type Clostridium saccharoperbutylacetonicum Bld and its variant Bld* were biochemically characterized and engineered to improve conversion of 3-hydroxybutyryl-CoA (3-HB-CoA) to 3-hydroxybutyraldehyde (3-HBA). Enzyme activity was strongly reduced by the product, 3-HBA, and this reductio

Atomic carbon is an attractive intermediate because it can form four new covalent bonds in a single transformation, but its synthetic use is limited by its short lifetime. Herein, we report an oxidation-triggered carbon-atom insertion into acyl chlorides using bench-stable N-isocyaniminophosphorane (PINC). Unlike our previous thermal/Rh-catalyzed protocol, this method proceeds at room temperature to afford cyclic α-chloro ketones. Mechanistic studies support oxidation-induced diazo formation from a phosphazine intermediate, followed by carbene generation.

Chemical modification of proteins is an important tool for the development of protein-based therapeutics, a rapidly growing field in disease treatment. Currently, no universal approach exists for site-specifically introducing desired functionalities into proteins with minimal disruption. Here, we present a reactive peptide tag/probe pair system for site-specific covalent protein labeling, leveraging the interaction between a lysine-containing histidine tag (KH6 or H6K) and a binuclear nickel (II)-nitrilotriacetic acid (BisNi²+-NTA) probe conjugated to a lysine-reactive N-acyl-N-alkyl sulfonami

To improve microbial nitrogen removal efficiency in aquaculture wastewater, we added different carbon-chain-length acyl-homoserine lactones (AHLs) to continuous-flow reactors, including N-butyryl-L-homoserine lactone (C4-HSL), N-hexanoyl-L-homoserine lactone (C6-HSL), N-octanoyl-L-homoserine lactone (C8-HSL), N-dodecanoyl-L-homoserine lactone (C12-HSL). We investigated their effects on nitrogen removal performance, sludge characteristics, and microbial community functions, and further analyzed the underlying mechanisms with metagenomics. The results showed that, compared with the nitrate conce

Hydroxyl-containing small-molecule drugs are widely used in oncology and inflammatory diseases but often suffer from poor aqueous solubility, off-target toxicity, and rapid systemic clearance, which collectively limit their therapeutic index. In parallel, hypochlorous acid (HOCl), a highly reactive myeloperoxidase-derived oxidant, is markedly upregulated in a variety of inflammatory disorders, including psoriasis, and thus represents an attractive endogenous trigger for site-selective prodrug activation. Here we report an HOCl-activated prodrug platform that enables controlled release of hydro

Riboflavin responsive multiple acyl-CoA dehydrogenase deficiency (RR-MADD) is an inherited metabolic disorder which is good responsive to riboflavin treatment. The phenotypic spectrum of adult-onset RR-MADD is highly heterogeneous. In this study, we described three patients with adult-onset RR-MADD presented with muscle weakness and spinal cord involvement. These three patients presented with adult-onset limb weakness, dyspnea, along with sensory levels changes (patient 1 below T2 level, patient 2 below T6 level, and patient 3 below T12 level, respectively). All patients displayed elevated acy

The bacterial Quorum Sensing system refers to the recognition of signaling molecules called autoinducers produced by bacteria when a certain cell density is reached in the environment. Those cell-density-dependent autoinducers regulate and coordinate diverse functional processes, such as bioluminescence, biofilm production, sporulation, and even the expression of some virulence factors, among others. There is a wide variety of autoinducers, and for Gram-negative bacteria, the canonical autoinducers are the N-acyl-homoserine lactones (AI-1). Presently, the production of autoinducers in Leptospi

A novel transition-metal-free radical coupling of 2-azaallyl anions for the synthesis of α-amino ketones has been developed. Easily accessible thioesters and 2-azaallyl anions undergo single electron transfer (SET) to generate acyl radicals, which participate in intermolecular radical coupling to generate α-amino ketones with good functional group tolerance and yields (31 examples, up to 98% yield). A telescoped gram-scale synthesis and derivatization of the product illustrate the potential synthetic utility of this method. Radical trapping and radical clock experiments support the proposed ra

BACKGROUND: Factors leading to severe COVID-19 remain partially known. New biomarkers predicting COVID-19 severity that are also causally involved in disease pathogenesis could improve patient management and contribute to the development of innovative therapies. Autophagy, a cytosolic structure degradation pathway is involved in the maintenance of cellular homeostasis, degradation of intracellular pathogens and generation of energy for immune responses. Acyl-CoA binding protein (ACBP) is a key regulator of autophagy in the context of diabetes, obesity and anorexia. The objective of our work wa

Over the past decade, significant progress has been made in the direct C-H acylation of naphthalenes, occurring at the α or β-positions to yield valuable ketones through Friedel-Crafts acylation or transition-metal-catalysed carbonylative coupling reactions. Nevertheless, highly regioselective acylation of naphthalenes remains a formidable challenge. Herein, we developed a nickel-catalysed reductive ring-opening reaction of 7-oxabenzonorbornadienes with acyl chlorides as the electrophilic coupling partner, providing a new method for the exclusive preparation of β-acyl naphthalenes.

Chiral sulfur pharmacophores are crucial for drug discovery in bioscience and medicinal chemistry. While the catalytic asymmetric synthesis of sulfoxides and sulfinate esters with stereogenic-at-sulfur(IV) centres is well developed, the synthesis of chiral sulfinamides remains challenging, which has primarily been attributed to the high nucleophilicity and competing reactions of amines. In this study, we have developed an efficient methodology for the catalytic asymmetric synthesis of chiral sulfinamides and sulfinate esters by the sulfinylation of diverse nucleophiles, including aromatic amin

The leinamycin family of polyketides are promising antitumor antibiotics, yet several aspects of their biosynthesis remain elusive. All lein … has revealed strict functional selectivity for a single ACP. Furthermore, we propose a cryptic transacylation step must occur prior to polyketide off-loading and cyclization. Altogether, these mechanistic investigations suggest that an atypical in-series mechanism underpins sulfur incorporation in the leinamycin family, and provides significant progress tow …

Aerobic organisms continuously generate internal superoxide and hydrogen peroxide, which can damage enzymes and impair growth. To avoid this problem cells maintain high levels of superoxide dismutases, catalases, and peroxidases. Surprisingly, we do not know the primary sources of these reactive oxygen species (ROS) in living cells. However, in vitro studies have shown that flavoenzymes can inadvertently transfer electrons to oxygen. Therefore, it seems plausible that substantial ROS may be generated when large metabolic fluxes flow through flavoproteins. Such a situation may arise during the

Herein, we report visible-light-driven hydroacylation of unactivated alkenes. We employed benzimidazolines as new acyl donors and achieved perfect regioselectivity, high functional-group tolerance, and excellent substrate generality. We also performed mechanistic experiments to elucidate the detailed reaction mechanism. This is the first example of (1) hydroacylation of unactivated alkenes using (2) easily prepared acyl donors under (3) visible-light irradiation. Our findings offer a new strategy to synthesize a wide variety of ketones under mild conditions.

Cancer cells use acetate to support the higher demand for energy and lipid biosynthesis during uncontrolled cell proliferation, as well as for acetylation of regulatory proteins. Acyl-CoA thioesterase 12 (Acot12) is the enzyme that hydrolyzes acetyl-CoA to acetate in liver cytosol and is downregulated in hepatocellular carcinoma (HCC). A mechanistic role for Acot12 in hepatocarcinogenesis was assessed in mice in response to treatment with diethylnitrosamine(DEN)/carbon tetrachloride (CCl4) administration or prolonged feeding of a diet that promotes non-alcoholic steatohepatitis (NASH). Loss-of-function mutations in patatin-like phospholipase domain-containing protein 1 (PNPLA1) cause autosomal recessive congenital ichthyosis, and altered PNPLA1 activity is implicated in the pathogenesis of atopic dermatitis and other common skin diseases. To examine the hypothesis that PNPLA1 catalyzes the synthesis of acylceramides and acyl acids, we expressed and partially purified a soluble, truncated form of PNPLA1 in Escherichia coli, (PNPLA1trun) along with the related protein PNPLA2 (ATGL, adipose triglyceride lipase) and coactivator CGI-58. Liposomal substrates were incubated with re

BACKGROUND: Multiple acyl-coenzyme A dehydrogenase deficiency (MADD) is a rare metabolic disorder affecting fatty acid oxidation. Incidence at birth is estimated at 1:250 000, but type III presents in adults. It is characterized by nonspecific symptoms but if undiagnosed may cause ketoacidosis and rhabdomyolysis. A review of 350 patients found less than one third presented with metabolic crises. Our objective is to describe an adult with weakness after carbohydrate restriction that developed a pulmonary embolism and ketoacidosis, and was diagnosed with MADD type III.
