Schizont transcriptome variation among clinical isolates and laboratory-adapted clones of the malaria parasite Plasmodium falciparum.

BACKGROUND: Malaria parasites are genetically polymorphic and phenotypically plastic. In studying transcriptome variation among parasites from different infections, it is challenging to overcome potentially confounding technical and biological variation between samples. We investigate variation in the major human parasite Plasmodium falciparum, generating RNA-seq data on multiple independent replicate sample preparations of merozoite-containing intra-erythrocytic schizonts from a panel of clinical isolates and from long-term laboratory-adapted clones, with a goal of robustly identifying differ

Dynamic control of carbon-ion radiation dose distribution amid respiratory motion challenges: A review of prediction research using geant4 and deep learning physics-constrained models.

BACKGROUND: Carbon ion radiotherapy (CIRT) offers superior dose conformity and biological effectiveness; however, its precision is highly sensitive to respiratory motion, leading to dose uncertainty and interplay effects that compromise treatment accuracy. METHODS: A comprehensive literature synthesis was conducted on studies employing motion-resolved imaging techniques (e.g. SMEIR), dynamic dose prediction, 4D robust planning, and hybrid adaptive methods for CIRT. The study emphasised the use of Geant4 Monte Carlo simulations combined with deep learning models, including physics-informed neu

Detecting local adaptation under weak genetic structure in an endemic damselfly: an integrative eco-evolutionary approach.

BACKGROUND: Insects comprise one of Earth's most diverse animal groups, but the adaptive capacity of most species, especially those with weak genetic structure, remains understudied. Psolodesmus mandarinus is an endemic damselfly in Taiwan, where its populations show latitudinal variation in wing traits despite limited genetic differentiation in mitochondrial and ribosomal sequences. We hypothesised that weak genome-wide structure may obscure the signals of local adaptation driven by environmental variation. To test this, we integrated genome-wide SNPs, phenotypic measurements, environmental a

Divergent Cardiovascular Adaptations and Gene Regulation in High-Elevation Natives and Recent Colonizers of the Qinghai-Tibetan Plateau.

High elevation imposes unrelenting and unavoidable hypoxia on species inhabiting these environments, providing an excellent natural setting for studying convergent or divergent evolution. By integrating measures of phenotypic variation, gene regulation, and functional performance, our study demonstrates that recent colonizers of high-elevation environments exhibit fundamentally different cardiovascular changes compared to long-term natives of these environments. Through the studying of heart morphological phenotypes, we showed that recent colonizers exhibit signs of cardiac hypertrophy, reflec

Evolutionary adaptation of anaerobic and aerobic metabolism to high sulfide and hypoxic hydrothermal vent crab, Xenograpsus testudinatus.

The vent crab, Xenograpsus testudinatus (xtcrab), is adapted to inhabit shallow-water, high sulfide and hypoxic hydrothermal vent. Our previous study revealed sulfide tolerance of vent xtcrabs which sulfide: quinone oxidoreductase (xtSQR) paralogs aid in sulfide detoxification. However, the mechanisms of how xtcrab adapts to high sulfide-hypoxic conditions in the vent area remain to be explored. In the present study, we tested the tolerance of xtcrab to sulfide-induced hypoxia, and investigated their aerobic and anaerobic responses in situ and in the laboratory. Comparisons were made to a non-

Functional Divergence of Scorpion Pedipalps: Musculoskeletal Specialization Toward Opposing Performance Optima.

When selective pressures for different functions act simultaneously on a structure, morphological diversification can be shaped by adaptation toward distinct functional optima. Systems may evolve along a performance gradient, optimizing different aspects of function in response to ecological demands. We investigated two scorpion species representing the morphological extremes of chela (pincer) shape. Scorpion chelae exhibit remarkable morphological diversity associated with ecological roles, and their performance varies along a force-velocity continuum. To explore how structural and muscular a

Cranial anatomy of Acynodon adriaticus and extreme durophagous adaptations in Eusuchia (Reptilia: Crocodylomorpha).

Acynodon adriaticus, a small eusuchian from the Late Cretaceous of Italy, is known for its well-preserved cranial and postcranial material. Despite its excellent preservation, many details remain hidden due to the physical overlap between the elements and matrix obliteration. We used Micro-CT scans to reveal previously overlooked anatomical features and describe in detail the cranial and dental anatomy of this taxon, shedding new light on its palaeoecology. The holotypic specimen, SC 57248, represents a mature individual exhibiting signs of hyperossification, developed ornamentation, and vario

Identification and molecular characterization of H9N2 viruses carrying multiple mammalian adaptation markers in resident birds in central-western wetlands in India.

We report here a targeted risk-based study to investigate the presence of influenza A viruses at the migratory-wild-domestic bird interface across the major wetlands of central India's Maharashtra state during the winter migration season. The H9N2 viruses have been isolated and confirmed in 3.86% (33/854) of the fecal samples of resident birds. To investigate the genetic pools of H9N2 circulating in resident birds, we sequenced two isolates of H9N2 from distant wetlands. Sequence and phylogenetic analyses have shown that these viruses are triple reassortants, with HA, NA, NP, and M genes belon

Mutation rate dynamics reflect ecological change in an emerging zoonotic pathogen.

Mutation rates vary both within and between bacterial species, and understanding what drives this variation is essential for understanding the evolutionary dynamics of bacterial populations. In this study, we investigate two factors that are predicted to influence the mutation rate: ecology and genome size. We conducted mutation accumulation experiments on eight strains of the emerging zoonotic pathogen Streptococcus suis. Natural variation within this species allows us to compare tonsil carriage and invasive disease isolates, from both more and less pathogenic populations, with a wide range o

Rapid adaptation of Chironomus yoshimatsui to acetylcholinesterase inhibitors (pyraclofos and pirimicarb) in a multi-generation study.

We evaluated the real effects of pollutants through a multi-generation study. We tested whether short-term exposure (48 h) of successive (first and second) generations of Chironomus yoshimatsui neonates (<24-h-old) to two acetylcholinesterase inhibitor insecticides, pyraclofos, and pirimicarb, would change insecticide sensitivity and life-cycle parameters over four generations. Additionally, we tested whether acetylcholinesterase (AChE) activity levels would be associated with this sensitivity change. Sensitivities (48 h EC50 value, using immobility as the endpoint) in chironomids (<24-h-old)

Convergent evolution in bacteria from multiple origins under antibiotic and heavy metal stress, and endophytic conditions of host plant.

The focus of this work is to study the convergent evolution in bacteria from multiple origins under antibiotic and heavy metal stress, and endophytic conditions of host plant cultivated on the Yamuna river bank. Forty-one endophytic bacteria (EB) were isolated from green leafy vegetables (GLV's) and were found to be resistant to a wide range of antibiotics (AB) and heavy metals (HM) tested. Further, they showed susceptibility to Quinolones group of antibiotics, and the HM, Cadmium, Chromium, and Mercury. Twenty-seven percent of these bacteria endowed with Class I integron. Phasome analysis of pathogenic and commensal Neisseria species expands the known repertoire of phase variable genes, and highlights common adaptive strategies.

Pathogenic Neisseria are responsible for significantly higher levels of morbidity and mortality than their commensal relatives despite having similar genetic contents. Neisseria possess a disparate arsenal of surface determinants that facilitate host colonisation and evasion of the immune response during persistent carriage. Adaptation to rapid changes in these hostile host environments is enabled by phase variation (PV) involving high frequency, stochastic switches in expression of surface determinants. In this study, we analysed 89 complete and 79 partial genomes, from the NCBI and Neisseria

Reciprocal cross infection of sticklebacks with the diphyllobothriidean cestode Schistocephalus solidus reveals consistent population differences in parasite growth and host resistance.

BACKGROUND: In host-parasite evolutionary arms races, parasites are generally expected to adapt more rapidly, due to their large population sizes and short generation times. There exist systems, though, where parasites cannot outpace their hosts because of similar generation times in both antagonists. In those cases concomitant adaptation is expected. METHODS: We tested this hypothesis in the three-spined stickleback-Schistocephalus solidus tapeworm system, where generation times are comparable in both organisms. We chose two populations of sticklebacks which differ prominently in the prevale

A morphological adaptation? The prevalence of enlarged external occipital protuberance in young adults.

Enthesophytes are bony projections that arise from the sites of ligament, tendon or joint capsule attachment to a bone. They are seen rarely in radiographic findings in young adults, as these bony adaptations are assumed to develop slowly over time. However, in recent years, the presence of an enlarged external occipital protuberance (EEOP) has been observed frequently in radiographs of relatively young patients at the clinic of the lead author. Accordingly, the aim of this project was to assess the prevalence of an EEOP in a young adult population. Analysis involved a retrospective analysis o

Dental eruption in East African wild chimpanzees.

Knowledge of chimpanzee development has played an essential role in our understanding of the evolution of human ontogeny. However, recent studies of wild ape dentitions have cast doubt on the use of developmental standards derived from captive individuals. Others have called into question the use of deceased wild individuals to infer normative development. We conducted a high resolution photographic study of living known-age subadults in the Kanyawara community (Kibale National Park, Uganda) to generate a comprehensive three year record of dental eruption (including tooth emergence ages). Thes

Male adaptations to minimize sexual cannibalism during reproduction in the funnel-web spider Hololena curta.

Males of many spider species risk being attacked and cannibalized while searching for, courting, and mating with conspecific females. However, there are exceptions. We show that the funnel-web spider, Hololena curta, has 3 adaptations that minimize risk to males during courtship and mating, and enhance reproductive success. First, males detected chemical or tactile signals associated with webs of virgin females, and differentiated them from webs of mated females, enabling males to increase encounter rates with virgin females and avoid aggressive mated females. Second, males produced stereotype

Secular increase in placental weight in Saudi Arabia.

BACKGROUND: Ramadan is an annual period of day-time fasting during which people in Saudi Arabia, including pregnant women, change their diets and physical activity. We recently reported that among babies who were in the second or third trimester of gestation during Ramadan placental growth slowed. We also found that, over the four years of the study, placental weight increased by 29 g per year. We have now extended our data collection in order to examine this trend in more detail. METHODS: We studied the birth records of 17 660 singletons born in King Saud Hospital, Unizah, Saudi Arabia, over

A poisonous surprise under the coat of the African crested rat.

Plant toxins are sequestered by many animals and the toxicity is frequently advertised by aposematic displays to deter potential predators. Such 'unpalatability by appropriation' is common in many invertebrate groups and also found in a few vertebrate groups. However, potentially lethal toxicity by acquisition has so far never been reported for a placental mammal. Here, we describe complex morphological structures and behaviours whereby the African crested rat, Lophiomys imhausi, acquires, dispenses and advertises deterrent toxin. Roots and bark of Acokanthera schimperi (Apocynaceae) trees are

Retinal illumination phase shifts the circadian rhythm of serotonin N-acetyltransferase activity in the chicken pineal gland.

The pineal gland of birds, in contrast to its mammalian counterpart, is a directly photosensitive organ. It has recently been demonstrated that light also acting via the retina acutely suppresses melatonin synthesis in the chicken pineal gland. The present study was aimed to investigate whether retinal illumination alone was capable of resetting the biological oscillator generating the circadian rhythm of pineal serotonin N-acetyltransferase (AA-NAT) activity in the chicken. Ocular exposure of chickens to 6 h low intensity white light (4 lux) potently suppressed AA-NAT activity (the penultimat

In vivo muscle activity in the hindlimb of the arboreal lizard, Chamaeleo calyptratus: general patterns and the effects of incline.

Arboreal animals often move on surfaces with variable and steep inclines, but the changes in hindlimb muscle activity in response to incline are poorly understood. Thus, we studied the hindlimb muscle activity in the arboreal specialist, Chamaeleo calyptratus, moving up and down 45 degrees inclines and on a horizontal surface. We quantified electromyograms (EMGs) from nine hindlimb muscles, and correlated EMGs with three-dimensional hindlimb kinematics. Kinematics changed little with incline, but the EMGs changed substantially. Most of the changes in EMGs were for amplitude rather than timing,
