In November-December 2002, stool samples from a random sample of the human population (N = 190) in the Garu area of northern Ghana were checked for intestinal helminths, using a single Kato smear and duplicate coprocultures for each subject. All 190 subjects were subsequently treated with a single, 400-mg dose of albendazole and 146 of them were successfully re-examined 21-28 days post-treatment. Prior to treatment, 75.5% of the Kato smears were found to contain 'hookworm-like' eggs (with a geometric mean egg count among the positives of 578 eggs/g faeces), and the third-stage larvae of Oesoph…

The efficacy of intraruminal albendazole (ABZ) capsules (Proftril-Captec) and the effect of treatment on productivity parameters were studied in two experiments totally on 466 ewes naturally infected with gastrointestinal nematodes and trematodes D. dendriticum. Ovoscopical tests revealed that treated animals remained negative during 10-12 weeks after the administration of capsules and that pasture contamination with helminths was significantly reduced. Necropsy revealed 96.9-99.2% efficacy against nematodes Nematodirus spp., Oesophagostomum spp., Cooperia spp., Trichostrongylus spp. and Trich…

The anthelmintic effects of levamisole (100-50 mg kg-1 live bodyweight), ivermectin (2 mg kg-1 bodyweight), albendazole and fenbendazole (both 100 mg kg-1 bodyweight), were tested against second stage Toxocara canis larvae in mice. Anthelmintic treatment on days 2 to 7 after infection inclusive resulted in significant (P less than 0.05) retention of larvae in the liver, and fewer larvae migrated subsequently to the brain and the musculature of treated mice compared to untreated controls. Most of the larvae retained in the liver subsequently died there and were not recoverable by day 35 after i…

OBJECTIVES: We report two cases of cutaneous larva migrans in pregnant women.
METHODS: Due to pregnancy, systemic therapy was not considered. Limited efficacy of cryotherapy and unavailability of topical thiabendazole led to the in-office preparation of 10% albendazole ointment.
RESULTS: Application of this compound thrice daily under occlusion for 10 days led to complete resolution without recurrence. No safety signals were reported during pregnancy follow-up and until birth.
CONCLUSION: Topical albendazole may be an effective, safe, and painless treatment of cutaneous larva migrans. Large…

Alveolar echinococcosis (AE), caused by the larval stage of Echinococcus multilocularis, is a severe zoonosis requiring prolonged anthelmintic therapy. Albendazole (ABZ) remains the treatment of choice; however, its poor aqueous solubility and low systemic bioavailability limit therapeutic efficacy and necessitate prolonged high-dose regimens. Improving the delivery of existing antiparasitic drugs therefore represents a valuable therapeutic strategy. This study evaluated the in vivo chemoprophylactic and therapeutic efficacy of an innovative nanomedicine formulation comprising ABZ encapsulated…

BACKGROUND: Albendazole (ABZ) and atovaquone (ATO) achieve killing efficacy on Echinococcus granulosus (Egs) by inhibiting energy metabolism, but their utilization rate is low. This study aims to analyze the killing efficacy of ABZ-ATO loading nanoparticles (ABZ-ATO NPs) on Egs.
METHODS: Physicochemical properties of NPs were evaluated by ultraviolet spectroscopy and nanoparticle size potentiometer. In vitro experiments exmianed the efficacy of ATO, ABZ, or ATO-ABZ NPs on protoscolex activity, drug toxicity on liver cell LO2, ROS production, and energy metabolism indexes (lactic dehydrogenase…

BACKGROUND: Cystic echinococcosis (CE) is a common neglected parasitic disease. Nanoparticles containing drugs have been widely utilized in various formulations for several purposes, including improving the bioavailability of drugs by increasing the solubility and dissolution rate of the nanoparticles. The purpose of this study was to evaluate the effects of solid lipid nanoparticles containing albendazole and conjugated to albumin (B-SLN + ABZ) as a novel treatment approach for hydatid cysts in vivo.
METHODS: Albendazole-loaded solid lipid nanoparticles were prepared by emulsification and so…

Alveolar echinococcosis (AE) is a chronic and fatal infectious parasitic disease, which has not been well-researched. Current recommended therapies for AE by the World Health Organization include complete removal of the infected tissue followed by two years of albendazole (ABZ), administered orally, which is the only effective first-line anti-AE drug. Unfortunately, in most cases, complete resection of AE lesions is impossible, requiring ABZ administration for even longer periods. Only one-third of patients experienced complete remission or cure with such treatments, primarily due to ABZ's low…

BACKGROUND: Cystic echinococcosis (CE), a widespread helminthic disease caused by the larval stage of the dog tapeworm Echinococcus granulosus represents a public health concern in humans. Albendazole (ABZ) is the first-line treatment for CE; however therapeutic failure of ABZ against CE occurs because of size and location of formed cysts as well its low aqueous solubility and consequently its erratic bioavailability in plasma. Serious adverse effects have also been observed following the long-term use of ABZ in vivo.
METHODS: We evaluated the apoptotic effects of ABZ-loaded β-cyclodextrin (A…

Cystic echinococcosis (CE) is a zoonosis caused by Echinococcus spp., affecting both humans and animals' lives. Current treatment of CE by oral administration of albendazole (ABZ) is hampered by several limitations. The poor aqueous solubility and the rapid metabolism of ABZ in the liver are the main issues, leading to lack of efficacy of the treatment. In the present study, we developed a nanocrystalline (NC) formulation of ABZ to be delivered intradermally using dissolving microneedles (DMNs). The NC formulation was developed using milling in an ultrasmall-scale device. Following several scr…

BACKGROUND: Albendazole (ABZ), a benzimidazole (BZ) anthelmintic (AH), is commonly used for treatment of soil-transmitted helminths (STHs). Its regular use increases the possibility that BZ resistance may develop, which, in veterinary nematodes is caused by single nucleotide polymorphisms (SNPs) in the β-tubulin gene at positions 200, 167 or 198. The relative importance of these SNPs varies among the different parasitic nematodes of animals studied to date, and it is currently unknown whether any of these are influencing BZ efficacy against STHs in humans. We assessed ABZ efficacy and SNP freq…

Neurocysticercosis is the most common parasitic infection of the central nervous system caused by larvae of Taenia solium. Spinal cysticercosis is an uncommon site of cysticercal infection, and isolated intramedullary involvement is even rarer. We present a case of 10-year-old girl who presented with gradual onset paraparesis with sensory loss and bowel and bladder incontinence. Magnetic resonance imaging (MRI) of spine revealed a cystic lesion with mural nodule (scolex) which was diagnostic for cysticercosis. Patient was treated with antihelminthic, which led to marked clinico-radiological im

BACKGROUND: Annual mass treatment with albendazole and ivermectin is the mainstay of current strategies to interrupt transmission of Wuchereria bancrofti in Africa. More-effective microfilarial suppression could potentially reduce the time necessary to interrupt transmission, easing the economic burden of mass treatment programs in countries with limited resources.
METHODS: To determine the effect of increased dose and frequency of albendazole-ivermectin treatment on microfilarial clearance, 51 W. bancrofti microfilaremic residents of an area of W. bancrofti endemicity in Mali were randomized…
