Evaluation of the impact of postnatal maternal separation stress on enamel formation in an experimental murine model.

BACKGROUND: The adverse effects of postnatal maternal separation (MS) on offspring development are well documented; however, its specific impact on dental enamel formation remains inadequately explored. This study examines the effects of maternal-separation-induced toxic stress on dental enamel formation in a murine model. METHODS: A total of 24 Wistar rat offspring (Rattus norvegicus) were utilized, divided into a control group (n = 12) and an MS group (n = 12). The MS group underwent 4 h of daily separation from their mothers from postnatal day 2 to day 21, while the control group remained

Fam83h mutation causes mandible underdevelopment via CK1α-mediated Wnt/β-catenin signaling in male C57/BL6J mice.

Truncation mutations in FAM83H are the major cause of autosomal dominant hypocalcified amelogenesis imperfecta. Some studies also indicated that FAM83H could be involved in osteogenic differentiation; however, the function of FAM83H in bone formation was rarely explored. This study aimed to explore the effect of Fam83h mutation on skeletal development. We generated Fam83h c.1186C>T (p.Q396*) knockin C57/BL6J mice by CRISPR/Cas9 technology and found that the Fam83hQ396⁎/Q396⁎ male mice presented skeletal development retardation that was inconspicuous at birth but progressively worsened as they …

Evidence for Bicarbonate Secretion by Ameloblasts in a Novel Cellular Model.

Formation and growth of hydroxyapatite crystals during amelogenesis generate a large number of protons that must be neutralized, presumably by HCO3 (-)ions transported from ameloblasts into the developing enamel matrix. Ameloblasts express a number of transporters and channels known to be involved in HCO3 (-)transport in other epithelia. However, to date, there is no functional evidence for HCO3 (-)transport in these cells. To address questions related to HCO3 (-)export from ameloblasts, we have developed a polarized 2-dimensional culture system for HAT-7 cells, a rat cell line of ameloblast o…

NBCe1 in mouse and human ameloblasts may be indirectly regulated by fluoride.

Enamel biomineralization results in a release of protons into the enamel matrix, causing an acidification of the local microenvironment. This acidification, which may be enhanced by more rapid mineral deposition in the presence of fluoride, must be neutralized by the overlying ameloblasts. The electrogenic sodium bicarbonate co-transporter NBCe1 has been localized in mouse ameloblasts, and has been proposed to have a role in matrix pH regulation. In this study, transcript analysis by PCR showed NBCe1-A present in human ameloblasts, whereas mouse ameloblasts expressed NBCe1-B. In situ hybridiza

Changes in ultrastructural distribution of dental basement membrane heparan sulfate during early mouse tooth development.

With the invagination of the deep surface of the tooth buds of mouse embryos (15 days of gestation), heparan sulfate in the region of the lamina lucida of the dental basement membrane began to disappear. Subsequently, as the outline of the future dentino-enamel junction, which elaborates the molar cusp pattern, was formed (16-17 days of gestation), heparan sulfate disappeared not only in the lamina lucida but also at the external side of the lamina densa. However, prior to or during predentin matrix deposition (18 days of gestation), heparan sulfate reappeared in the dental basement membrane.

Solute carrier family 9 member A5 regulated by TGF-β is necessary for dental enamel formation.

OBJECTIVES: In this study, we aimed to elucidate the functional significance of transforming growth factor-β (TGF-β)-regulated SLC9A5 (NHE5) in enamel formation. METHODS: Mouse ameloblast-derived mHAT9d cells were treated with TGF-β1, TGF-β2, or TGF-β3, and the expression of Slc9a family members was analyzed via RNA sequencing (RNA-seq) and quantitative PCR (qPCR). Slc9a5-deficient mice were generated to examine enamel morphology. Enamel volume, mineral density, and structure were assessed using micro-computed tomography (μCT), scanning electron microscopy (SEM), and electron probe microanaly

Nephrocalcinosis associated with continuous enamel hypoplasia and severe alveolar bone loss: a case report and literature review.

Enamel-renal syndrome (ERS) is a rare manifestation of nephrocalcinosis that has been associated with generalized enamel hypoplasia. The purpose of this paper was to describe, for the first time, the association of enamel-renal syndrome with severe localized periodontal bone loss. A 13-year-old boy presented with: generalized hypoplastic enamel; intrapulpal calcifications; retention of primary teeth; delayed eruption of permanent teeth; enlarged dental-follicles; misshaped roots of permanent teeth; gingival overgrowth; severe localized alveolar bone loss; and severe malocclusion. His parents w

Mechanism and timing of fluoride effects on developing enamel.

Fluoride appears to specifically interact with mineralizing tissues, causing an alteration of the mineralization process. In enamel, fluorosis results in a subsurface hypomineralization. This hypomineralized enamel appears to be directly related to a delay in the removal of amelogenins at the early-maturation stage of enamel formation. The specific cause for this delay is not known, although existing evidence points to reduced proteolytic activity of proteinases that hydrolyze amelogenin. This delay in hydrolysis of amelogenins could be due to a direct effect of fluoride on proteinase secretio

Prenatal exposure to environmental toxins and comprehensive dental findings in a population cohort of children.

Environmental toxins are known to have many impacts on growth and development in humans, starting in utero. Alterations in amelogenesis, caused by chemical and physical trauma that occur during the antenatal, perinatal and postnatal time periods, may result in developmental defects in deciduous and permanent tooth enamel, as demonstrated in animal studies. These defects can be clinically visible and result in a variety of morphological and functional problems in the dentition. Since enamel does not remodel after formation, it may serve as a permanent record of insults during organ development.…

Tributyltin impairs dentin mineralization and enamel formation in cultured mouse embryonic molar teeth.

Tributyltin (TBT), earlier used as an antifouling agent in marine paints, causes damage to the aquatic ecosystem, for example, impaired shell calcification in oysters. TBT affects hard tissue mineralization even in mammals: delayed bone mineralization has been observed in rodents exposed to TBT in utero. To see if TBT interferes with tooth development, especially dental hard tissue formation, we exposed mouse E18 mandibular first and second molars to 0.1, 0.5, 1.0, and 2.0 microM TBT chloride in organ culture for 7-12 days. The amount of enamel was assessed and the sizes of the first molars we

The chemical composition of tooth enamel in junctional epidermolysis bullosa.

The junctionalis form of epidermolysis bullosa (EBJ) is associated with a number of clinical problems involving tooth enamel, including increased susceptibility to caries. The aim here was to carry out a chemical characterization of the enamel of teeth from EBJ patients compared with that of unaffected controls. The results showed that while protein concentration, amino acid composition and carbonate content were similar in both groups, EBJ enamel contained a significantly reduced mineral per volume content, resulting in enamel hypoplasia. In addition, Western blotting revealed the presence of

Root canal treatment as a treatment modality for temporary tooth retention in adolescent patients.

In certain cases root canal treatment may be an adequate intermediate therapy, mainly for severely traumatized teeth in adolescent patients that probably cannot be retained permanently. The treatment aims at temporary retention of otherwise hopeless teeth until definite treatment, for example insertion of bridges or dental implants, may be performed. This kind of treatment can help to avoid the application of removable prostheses in young patients. This treatment modality requires intensive information to be presented to the patients and their parents on the benefits and risks of the treatment

Distribution of protein kinase C alpha and accumulation of extracellular Ca2+ during early dentin and enamel formation.

Activation of the protein kinase C (PKC)-related signal transduction system has been associated with phenotypic expression in a wide variety of cell types. In in vitro studies, it has often been activated by relatively small increases in the Ca2+ concentration ([Ca2+]) in the medium. The studies reported here explored the hypothesis that localized increases in the extracellular [Ca2+] and activation of the PKC-related pathway may be involved in early dentin and enamel formation. Whole-head, freeze-dried sections through the developing molars of 5-day-old rats were evaluated by methods that loc

Cells from Hertwig's epithelial root sheath do not transcribe amelogenin.

Recent experimental evidence has led to the interpretation that "enamel-like" material is deposited along the forming mouse molar root surface by cells of Hertwig's epithelial root sheath (HERS cells) and that this material is integral to the developmental program for cementogenesis. The experimental strategy described in this study was to examine selected developmental stages of root formation for mouse first and second mandibular molars in order to localize the cellular sites of amelogenin gene transcripts using high resolution in situ hybridization. Amelogenin is the major structural protei

Orthodontic loading modulates epithelial progenitor dynamics and enamel formation in the mouse mandibular incisor.

BACKGROUND: Adult stem cells (SCs) maintain tissue homeostasis through a balance of self-renewal and differentiation regulated by biochemical and mechanical cues. While biochemical pathways are well studied, the in vivo mechanisms by which external forces influence SCs and their progeny remain poorly understood. The continuously growing mouse incisor offers a tractable model for investigating these mechanisms because dental epithelial stem cells (DESCs) reside in a spatially restricted apical niche, and their progeny undergo stereotyped proliferation and differentiation along the apical-incisa
