Protease-Mediated Growth of Staphylococcus aureus on Host Proteins Is opp3 Dependent.

Staphylococcus aureus has the ability to cause infections in multiple organ systems, suggesting an ability to rapidly adapt to changing carbon and nitrogen sources. Although there is little information about the nutrients available at specific sites of infection, a mature skin abscess has been characterized as glucose depleted, indicating that peptides and free amino acids are an important source of nutrients for the bacteria. Our studies have found that mutations in enzymes necessary for growth on amino acids, including pyruvate carboxykinase (ΔpckA) and glutamate dehydrogenase (ΔgudB), reduc

Aging influences protein digestion, absorption and amino acid metabolism.

Proteins are essential biomolecules that play indispensable roles in maintaining cellular homeostasis and regulating systemic metabolic processes. Protein deficiency is closely associated with age-related chronic diseases. Our purpose is to reveal the reason of protein deficiency in the elderly and potential treatments. A systematic literature search mainly on human studies addressing gastrointestinal aging and amino acid homeostasis was conducted. This review investigated the effects of aging on protein metabolism and summarized age-related physiological changes that collectively disrupt prot

LAT1 acts as a crucial transporter of amino acids in human thymic carcinoma cells.

L-type amino acid transporter 1 (LAT1, SLC7A5) incorporates essential amino acids into cells. Recent studies have shown that LAT1 is a predominant transporter in various human cancers. However, the function of LAT1 in thymic carcinoma remains unknown. Here we demonstrate that LAT1 is a critical transporter for human thymic carcinoma cells. LAT1 was strongly expressed in human thymic carcinoma tissues. LAT1-specific inhibitor significantly suppressed leucine uptake and growth of Ty82 human thymic carcinoma cell lines, suggesting that thymic carcinoma takes advantage of LAT1 as a quality transpo

Fetal malnutrition: a possible cause of the fetal alcohol syndrome.

The effects of ethanol ingestion during pregnancy on total folate levels in fetal tissues and on the concentrations of free amino acids in fetal and maternal plasma were examined in the rat. No differences were observed between the ethanol-fed and the control groups in total folates in fetal brain and liver. However, the concentration of fetal plasma histidine was reduced by 50% as a result of maternal ethanol consumption; the maternal plasma histidine level was not affected. It is suggested that fetal malnutrition in an essential amino acid, histidine, could impair fetal protein synthesis pro

Regulation of placental amino acid transport in health and disease.

Abnormal fetal growth, i.e., intrauterine growth restriction (IUGR) or fetal growth restriction (FGR) and fetal overgrowth, is associated with increased perinatal morbidity and mortality and is strongly linked to the development of metabolic and cardiovascular disease in childhood and later in life. Emerging evidence suggests that changes in placental amino acid transport may contribute to abnormal fetal growth. This review is focused on amino acid transport in the human placenta, however, relevant animal models will be discussed to add mechanistic insights. At least 25 distinct amino acid tra

Mutations disrupting selenocysteine formation cause progressive cerebello-cerebral atrophy.

The essential micronutrient selenium is found in proteins as selenocysteine (Sec), the only genetically encoded amino acid whose biosynthesis occurs on its cognate tRNA in humans. In the final step of selenocysteine formation, the essential enzyme SepSecS catalyzes the conversion of Sep-tRNA to Sec-tRNA. We demonstrate that SepSecS mutations cause autosomal-recessive progressive cerebellocerebral atrophy (PCCA) in Jews of Iraqi and Moroccan ancestry. Both founder mutations, common in these two populations, disrupt the sole route to the biosynthesis of the 21st amino acid, Sec, and thus to the

Hepatic dysfunction during hyperalimentation.

Liver biopsy specimens were studied in 26 patients in whom liver function abnormalities developed during intravenous hyperalimentation (IVH). The clinical manifestations and duration of IVH were evaluated in relation to the morphological changes seen in the liver. Early hepatic changes consisted of fatty metamorphosis, and progressive intrahepatic cholestasis developed as IVH was continued. Essential fatty acid deficiency, amino acid imbalance, caloric excess, and toxic manifestations of certain amino acids are postulated as causative factors. The hepatic steatosis secondary to IVH may be trea

Impaired BCAA Catabolism Contributes To Acute Lung Injury By Triggering Oxidative Stress and Inflammatory Response Via the MAPK Pathway.

Acute lung injury (ALI) is a severe respiratory failure characterized by rapid alveolar injury, severe hypoxemia, and an uncontrolled inflammatory response. Branched-chain amino acids (BCAAs) are a group of essential amino acids, including valine, leucine, and isoleucine. Accumulating evidence shows that BCAA metabolism has long been implicated in the etiology of type 2 diabetes, myocardial ischemia/reperfusion, heart failure, cancer, and other conditions. However, the contribution of BCAA metabolism regulation in ALI remains largely elusive. Here, we found that branched-chain α-keto acids (BC

Plasma metabolomics of early parenteral nutrition followed with enteral nutrition in pancreatic surgery patients.

Nutrition support is essential for surgical patients. Patients undergoing pancreaticoduodenectomy (PD) require tremendous nutrient support but also faced with risks of infection and gastrointestinal complications. Early parenteral nutrition has recently shown benefits while limited information provided about the influence on metabolism. This prospective single-center cohort study used plasma metabolomics to clarify metabolic alteration after early parenteral nutrition followed with enteral nutrition. Patients undergoing pancreaticoduodenectomy (n = 52) were enrolled. 36 patients received paren

Disorders of branched chain amino acid metabolism.

The three essential branched-chain amino acids (BCAAs), leucine, isoleucine and valine, share the first enzymatic steps in their metabolic pathways, including a reversible transamination followed by an irreversible oxidative decarboxylation to coenzyme-A derivatives. The respective oxidative pathways subsequently diverge and at the final steps yield acetyl- and/or propionyl-CoA that enter the Krebs cycle. Many disorders in these pathways are diagnosed through expanded newborn screening by tandem mass spectrometry. Maple syrup urine disease (MSUD) is the only disorder of the group that is assoc

The effects of maternal protein restriction on the growth of the rat fetus and its amino acid supply.

Maternal protein deficiency causes fetal growth retardation which has been associated with the programming of adult disease. The growth of the rat fetus was examined when the mothers were fed on diets containing 180, 90 and 60 g protein/kg. The numbers of fetuses were similar in animals fed on the 180 and 90 g protein/kg diets but the number was significantly reduced in the animals fed on the 60 g protein/kg diet. The fetuses carried by the mothers fed on the 90 g protein/kg diet were 7.5% heavier than those of mothers fed on 180 g protein/kg diet on day 19 of gestation, but by day 21 the situ

Rice Cultivation Reshapes Mercury Accumulation in Soil-Plant Systems.

Predicting the bioaccumulation of potent neurotoxic methylmercury (MeHg) in rice is essential for assessing and mitigating human exposure risks, particularly for Asians and infants. However, significant uncertainties lie in understanding and predicting MeHg accumulation in rice, partially due to the insufficient understanding of how rice plants regulate Hg biogeochemistry in soil-rice systems. Here, we conducted a pot experiment involving 32 commonly cultivated rice varieties under the same soil and ambient conditions. By analyzing MeHg contents in rice-paddy systems and amino acid profiles in

Novel amino acid residues in listerolysin O drive phagosome escape and pathogenicity in Listeria monocytogenes.

Listeria monocytogenes, a significant zoonotic foodborne intracellular pathogen, relies on its principal virulence factor, listeriolysin O (LLO), to escape from host cell phagosomes-a critical step for establishing intracellular infection and replication. LLO, composed of four structural domains with amino acids essential for its pore-forming abilities, still harbors many sites with undefined roles. We identified a critical domain within LLO (amino acids 251-255) crucial for bacterial pathogenesis and systematically investigated the individual roles of five amino acid sites within this domain.

Untargeted metabolomic profiling of serum from client-owned cats with early and late-stage chronic kidney disease.

Evaluation of the metabolome could discover novel biomarkers of disease. To date, characterization of the serum metabolome of client-owned cats with chronic kidney disease (CKD), which shares numerous pathophysiological similarities to human CKD, has not been reported. CKD is a leading cause of feline morbidity and mortality, which can be lessened with early detection and appropriate treatment. Consequently, there is an urgent need for early-CKD biomarkers. The goal of this cross-sectional, prospective study was to characterize the global, non-targeted serum metabolome of cats with early versu

Dietary Arecoline Supplementation Enhances Flesh Quality, Muscle Growth, and Protein Deposition in Grass Carp (Ctenopharyngodon idella): Insights Into TOR and FOXO Signaling Pathways.

Arecoline is an alkaloid isolated from the plant betel nut, which exerts a wide range of physiological effects by stimulating muscarinic receptors. To explore the effect of arecoline on the flesh quality of fish, grass carp were fed different levels of arecoline (0, 0.5, 1.0, 1.5, 2.0, and 2.5 mg/kg diet) for 63 days. Key findings demonstrated that arecoline significantly alleviated postmortem pH decline, reduced cooking losses, and decreased lactic acid content. It also enhanced the nutritional value of muscle by increasing the content of essential amino acids (EAAs) and polyunsaturated fatty

Structure and Activity Relationships of the Two-Component Lantibiotic Bicereucin.

Identified from the pathogen Bacillus cereus SJ1, the two-component lantibiotic bicereucin is featured by the presence of a series of nonproteogenic amino acids and exhibits potent synergistic activity against a broad spectrum of Gram-positive bacteria, including methicillin-resistant Staphylococcus aureus and vancomycin-resistant Enterococci, as well as hemolytic activity against mammalian cells. In this study, we performed site-directed mutagenesis on the nonproteogenic amino acids as well as truncation of dehydrobutyrine-rich N-terminal residues and evaluated the effects on both biological
