Anaphylaxis is an acute, potentially life-threatening reaction, often triggered by foods and largely mediated by immunoglobulin (Ig)E. The human microbiota is known to influence oral tolerance, but the microbial mechanisms directly involved in IgE-mediated anaphylaxis remain unknown. Here, we demonstrate that human saliva and jejunum harbor peanut (PN)-degrading bacteria that metabolize immunodominant allergens (Ara h 1 and 2), reducing IgE binding and anaphylaxis. Isolated Rothia and Staphylococcus species degraded PN allergens in vitro, generating proteins with reduced IgE binding and limite

Antibiotic drugs have the potential to induce persistent impairment to the ecosystem within soil and natural water bodies. To address this issue, this study utilized ofloxacin (OFL) as the research subject; ZnSA@PEG was synthesized employing agricultural waste-derived peanut shell biochar as the primary material, with sodium alginate (SA) serving as the substrate. The biochar was activated using ZnCl2, followed by the incorporation of polyethylene glycol (PEG) into the SA, and subsequently cross-linked with CaCl2. The effect of ZnSA@PEG on OFL removal efficiency was investigated under differen

Whether epigenetic factor UTX, a histone H3 lysine 27 (H3K27) demethylase, is critical for type 2 immunity, including allergic sensitization and antigen-driven anaphylaxis, is unclear. We used UTXfl/fl x Lck-Cre mice with UTX-deficient T cells (UTX-TCD) to determine whether T cell-specific UTX expression regulates antigen-specific IgE production after airway sensitization to peanut and anaphylaxis following intraperitoneal (i.p.) peanut challenge. UTX-TCD mice sensitized via the airway with peanut and lipopolysaccharide (LPS), a bacterial component and environmental adjuvant found in house dus

INTRODUCTION AND OBJECTIVES: Eosinophilic esophagitis (EoE) is frequently miss-diagnosed or overlooked for several years because of the invasiveness of investigations and the non-specificity of symptoms in childhood. Due to the lack of specific recommendations in children, its management remains very heterogeneous, especially concerning allergy testing. The aim of this study is to analyze our population and practices, in comparison with the literature, with a focus on allergic management, to harmonize and optimize our practice.
MATERIAL AND METHODS: We included all children with a diagnosis o

BACKGROUND: Peanut allergy frequently causes severe allergic reactions. Diagnosis includes detection of IgE to peanuts in serum or by skin prick tests. While children may have allergic sensitization without having clinical peanut allergy, oral peanut challenge is often required for accurate diagnosis. The conjunctival provocation test is used for diagnosis and evaluation of treatment effect in inhalant allergies, but it has not been evaluated as a tool for diagnosing peanut allergy.
OBJECTIVE: To investigate whether the conjunctival provocation tests may be feasible, accurate and safe in diag

BACKGROUND: Oral food challenge is the main tool for diagnosing food allergy, but there is little data on the reaction profiles of young children undergoing challenges, nor how these reactions compare to reactions on accidental ingestion in the community.
OBJECTIVES: To compare reaction profiles from food challenges and parent-reported reactions on accidental ingestion, and assess predictors of severe reactions.
METHODS: HealthNuts is a longitudinal population-based cohort study of 5276 1-year-old infants. Infants underwent skin prick tests and those with identifiable wheals were offered foo

BACKGROUND: Although several studies have assessed the effects of nut consumption (tree nuts, peanuts, and soy nuts) on blood pressure (BP), the results are conflicting.
OBJECTIVE: The aim was to conduct a systematic review and meta-analysis of published randomized controlled trials (RCTs) to estimate the effect of nut consumption on BP.
DESIGN: The databases MEDLINE, SCOPUS, ISI Web of Science, and Google Scholar were searched for RCTs carried out between 1958 and October 2013 that reported the effect of consuming single or mixed nuts (including walnuts, almonds, pistachios, cashews, hazeln

CONTEXT: Certain individuals are sensitive enough to react to peanuts and peanut oil, sometimes with deadly effect. It is thus crucial to have an accurate testing methodology for the assessment of allergies and immune reactivities to peanuts and their components, such as agglutinins and oleosins. Currently, skin-prick testing is performed only with the water-soluble components of peanut proteins and can produce false negatives. Testing with all possible food antigens and with both immunoglobulin G (IgG) and immunoglobulin E (IgE) antibodies may offer a more accurate alternative.
OBJECTIVE: Th

BACKGROUND: Recent findings show food allergy is rarely the cause of chronic urticaria. However; reports showed up to 5% of chronic idiopathic urticaria (CIU) was food induced urticaria (FIU) and the remission rate with food avoidance in CIU was varied. According to recent studies, skin prick test (SPT) is not a gold standard for investigating the culprit food allergen in CIU. The clinical response for food avoidance is still unclear.
OBJECTIVE: The purpose of the present study is to investigate the association of food allergen and SP7 the clinical response after positive food avoidance in ad

BACKGROUND: Food Allergy Herbal Formula-2 (FAHF-2) prevents anaphylaxis in a murine model of peanut allergy. Multiple food allergies (MFA) are common and associated with a higher risk of anaphylaxis. No well-characterized murine model of sensitization to multiple food allergens exists, and no satisfactory therapy for MFA is currently available.
OBJECTIVE: To determine the effect of FAHF-2 in a murine model of MFA.
METHODS: C3H/HeJ mice were orally sensitized to peanut, codfish, and egg concurrently. Oral FAHF-2 treatment commenced 1 day after completing sensitization and continued daily for

BACKGROUND: Initial food-allergic reactions are often poorly recognized and under-treated.
METHODS: Parents of food-allergic children were invited to complete an online questionnaire, designed with Kids with Food Allergies Foundation, about their children's first food-allergic reactions resulting in urgent medical evaluation.
RESULTS: Among 1361 reactions, 76% (95% CI 74-79%) were highly likely to represent anaphylaxis based on NIAID/FAAN criteria. Only 34% (95% CI 31-37%) of these were administered epinephrine. In 56% of these, epinephrine was administered by emergency departments; 20% by p

To date, there are sparse data on epinephrine treatment for food-related anaphylaxis in adults. We sought to establish the frequency of more than one epinephrine treatment for adult patients who present with food-related anaphylaxis to the emergency department (ED). We performed a chart review, at two academic centers, of all adults presenting to the ED for food allergy (ICD9CM codes 693.1, 995.0, 995.1, 995.3, 995.7, 995.60-995.69, 558.3, 692.5, and 708.X) between January 1, 2001 and December 31, 2006. We focused on causative foods; treatments, including the number of epinephrine treatments g

The search for natural alternatives to synthetic antioxidants and antimicrobials is growing tremendously as consumers become concerned with currently used compounds. Peanut phytoalexins may become a viable source of these compounds. Recent developments have shown compounds derived from peanuts possess both high antioxidant activity and antimicrobial capabilities. Some of these compounds have been identified as stilbenes and other low molecular weight phenolic compounds. Patents have been awarded for increasing the amounts of these compounds in peanut plants and in the peanut seeds. Increasing

BACKGROUND: Maternal allergy is believed to be a risk factor for peanut allergy (PNA) in children. However, there is no direct evidence of maternal transmission of PNA susceptibility, and it is unknown whether maternal peanut exposure affects the development of PNA in offspring.
OBJECTIVE: To investigate the influence of maternal PNA on offspring reactions to the first peanut exposure, and whether maternal low-dose peanut exposure during pregnancy and lactation influences these reactions and peanut sensitization in a murine model.
METHODS: Five-week-old offspring of PNA C3H/HeJ mothers (PNA-

Peanut and tree nut allergies present multiple challenges in their presentation and management. These challenges have become increasingly relevant in recent years, as these allergies appear to have become more common. An estimated 1-2% of the population in the USA is allergic to peanut or tree nuts. Peanut allergy typically presents with symptoms in one of the first few exposures to peanut. Diagnosis is based on clinical history along with skin prick test, or quantitation of allergen-specific immunoglobulin E (IgE), and oral food challenges when indicated. Once the diagnosis is confirmed, the
