Comprehensive in vitro and whole-genome characterization of probiotic properties in Pediococcus acidilactici P10 isolated from Iranian broiler chicken.

This study presents a comprehensive characterization of Pediococcus acidilactici strain P10, a novel probiotic isolated from native broiler chickens, integrating in vitro analyses with whole-genome sequencing. P10 demonstrates promising probiotic attributes, supported by both phenotypic and genomic evidence. The strain was non-hemolytic and exhibited high survival rates under simulated gastrointestinal conditions (95-99% in acidic pH, 55% in bile salts), with genomic analysis confirming the presence of associated stress resistance genes. Importantly, P10 displayed potent broad-spectrum antimic

Genomic and functional insights into commensal streptococci with anti-pneumococcal activity.

BACKGROUND: Streptococcus pneumoniae (pneumococcus), S. mitis, and S. oralis are closely related bacteria that colonize the human upper respiratory tract. While pneumococcus is a leading cause of global morbidity and mortality, S. mitis and S. oralis are generally considered commensals, rarely causing disease in immunocompetent hosts. Here, we characterized the genomes of seven commensal streptococcal strains (S. oralis A22 and S. mitis B22–G22) identified as potential biotherapeutics due to their bacteriocin-mediated antipneumococcal activity. RESULTS: Comparative genomic analyses revealed ke

Microcin MccI47 selectively inhibits enteric bacteria and reduces carbapenem-resistant Klebsiella pneumoniae colonization in vivo when administered via an engineered live biotherapeutic.

The gastrointestinal (GI) tract is the reservoir for multidrug resistant (MDR) pathogens, specifically carbapenem-resistant (CR) Klebsiella pneumoniae and other Enterobacteriaceae, which often lead to the spread of antimicrobial resistance genes, severe extraintestinal infections, and lethal outcomes. Selective GI decolonization has been proposed as a new strategy for preventing transmission to other body sites and minimizing spreading to susceptible individuals. Here, we purify the to-date uncharacterized class IIb microcin I47 (MccI47) and demonstrate potent inhibition of numerous Enterobact

Purification and characterization of two new cell-bound bioactive compounds produced by wild Lactococcus lactis strain.

Novel compounds and innovative methods are required considering that antibiotic resistance has reached a crisis point. In the study, two cell-bound antimicrobial compounds produced by Lactococcus lactis ID1.5 were isolated and partially characterized. Following purification by cationic exchange and a solid-phase C18 column, antimicrobial activity was recovered after three runs of RPC using 60% (v/v) and 100% (v/v) of 2-propanol for elution, suggesting that more than one antimicrobial compound were produced by L. lactis ID1.5, which were in this study called compounds AI and AII. The mass spect

[Efficacy of enterocin S760 in treatment of mice with anthrax infection due to Bacillus anthracis M-71].

The therapeutic efficacy of enterocin S760, a broad spectrum antimicrobial peptide produced by Enterococcus faecium LWP760 was tested on mice infected with Bacillus anthracis M-71 to induce anthrax (second Tsenkovsky's vaccine). Intraperitoneal four-, two- or one-fold administration of the peptide in a dose of 25 mg/kg for 10 days for prophylactic (1 hour after the contamination) and therapeutic (24 hours after the contamination) purposes prevented or cured the infection in 90-100% of the mice versus the 100-percent lethality in the control (untreated animals). The antimicrobial activity of en

The ecology, epidemiology and virulence of Enterococcus.

Enterococci are Gram-positive, catalase-negative, non-spore-forming, facultative anaerobic bacteria, which usually inhabit the alimentary tract of humans in addition to being isolated from environmental and animal sources. They are able to survive a range of stresses and hostile environments, including those of extreme temperature (5-65 degrees C), pH (4.5-10.0) and high NaCl concentration, enabling them to colonize a wide range of niches. Virulence factors of enterococci include the extracellular protein Esp and aggregation substances (Agg), both of which aid in colonization of the host. The

[The nature of typhimuricin--a substance produced by Salmonella typhimurium LT2 and relation of its synthesis to the cryptic plasmid].

The effect of typhimuricin on Escherichia coli K12 cells and the properties of this substance produced by Salmonella typhimurium LT2 have been studied. The obtained data permit one to conclude that typhimuricin is similar to the aminoacid L-valine or the chemical very similar to it in properties. The synthesis of typhimuricin is not controlled by the cryptic plasmid present in Salmonella typhimurium LT2 cells. The experiments aimed at isolation of enterobacterial strains producing low molecular mass antibiotics--microcines have been done. It was found that the capability to produce macrocines

[A comparison of antibiotic resistance patterns and pyocine types between strains of Pseudomonas aeruginosa from animal and human sources (author's transl)].

291 strains of Pseudomonas aeruginosa from human bacteriological samples were compared with 102 strains which had been isolated from animals for frequency of pyocine-types and patterns of antibiotic resistance. 146 strains had been isolated from hospitalized patients and 145 from out-patients. More strains of animal origin were sensitive to carbenicillin and tetracyclines compared to strains of human origin. Since the frequency of different pyocine-types is the same in all three groups, it is concluded, that antibiotic resistance may be transferred from man to animal and vice versa via transfe

Gut microbiome modulation by cricket, pea, and whey protein using the SHIME in vitro simulator.

Entomophagy is increasingly popular, and Acheta domesticus offers an ecologically sustainable protein alternative, but the effects on the human gut microbiome need further investigation. In this study, we investigated the impact of the intake of three isolated proteins: pea (plant), whey (animal), and cricket (insect) on gut microbiome of a single-donor using the Simulator of the Human Intestinal Microbial Ecosystem (SHIME®). Cricket protein intake was associated with potential beneficial taxa such as Bifidobacterium and Lactobacillus, genes related to vitamin biosynthesis and bacteriocin tran

Vaginal Weissella confusa MV0113 suppresses pathogens and restores microbiota to alleviate bacterial vaginosis: integrated genomic and in vivo evidence.

Bacterial vaginosis (BV), the most common vaginal infection in women of reproductive age, shows high recurrence after antibiotic therapy, highlighting the need for novel probiotic approaches. This study aimed to evaluate the probiotic properties of the vaginal isolate Weissella confusa MV0113 and its potential role in preventing and managing BV. MV0113 remained sensitive to clinically relevant antibiotics and produced high levels of lactic acid and hydrogen peroxide. Its cell-free supernatant inhibited Candida albicans and Gardnerella vaginalis biofilms, showing greater activity than the refer

Valorisation of acid whey permeate for high-purity nisin Z production using artisanal Lactococcus lactis isolates.

The growing demand for sustainable bioprocessing has positioned whey, a side stream of the dairy industry, as a valuable substrate for producing high-value compounds. This study investigated the production and purification of nisin, a potent lantibiotic, using permeate (10 kDa cut-off) of ultrafiltered acid whey (PUFAW) devoid of lactoferrin as growth medium. Three strains of Lactococcus lactis subsp. lactis, including two artisanal isolates (IM 143 and IM 145) and a reference strain (IM 407), were evaluated for nisin production in PUFAW supplemented with yeast extract (YE) and mineral salts.

Phenotypic comparison and DNA sequencing analysis of a wild-type and a pediocin-resistant mutant of Listeria ivanovii.

Listeria ivanovii is one of the two pathogenic species within the genus Listeria, the other being Listeria monocytogenes. In this study, we generated a stable pediocin resistant mutant Liv-r1 of a L. ivanovii strain, compared phenotypic differences between the wild-type and the mutant, localised the pediocin-induced mutations in the chromosome, and analysed the mechanisms behind the bacteriocin resistance. In addition to pediocin resistance, Liv-r1 was also less sensitive to nisin. The growth of Liv-r1 was significantly reduced with glucose and mannose, but less with cellobiose. The cells of L

Characterization of functional, safety, and probiotic properties of Enterococcus faecalis UGRA10, a new AS-48-producer strain.

Enterococcus faecalis UGRA10, a new AS-48-producer strain, has been isolated from a Spanish sheep's cheese. The inhibitory substance produced by E. faecalis UGRA10 was purified and characterized using matrix-assisted laser desorption ionization-time of flight mass spectrometry, confirming its identity with AS-48 enterocin (7.150 Da). Subsequent genetic analysis showed the existence of the as-48 gene cluster on a plasmid of approximately 70-kb. The UGRA10 strain was examined for safety properties such as enterococci virulence genes, biogenic amine production, and antibiotic resistance. As for m

Antibiotic resistance in Providencia stuartii isolated in hospitals.

A total of 238 isolates of Providencia stuartii obtained from infected patients in six Dublin hospitals were grouped by using serological and bacteriocin typing methods and tested for sensitivity to a number of antimicrobial agents. Most isolates were resistant to several of these agents. Resistance to tetracycline, resistance to penicillin, resistance to polymyxin, and probably resistance to nitrofurantoin was intrinsic. Plasmid screening coupled with resistance transfer studies showed that both chromosome-encoded and plasmid-coded resistance mechanisms were clinically important. Ampicillin r

Bioprotective cultures as a clean label strategy for food preservation: From concept to market.

Bioprotective cultures and their derived antimicrobial agents represent a promising clean label strategy to enhance food safety and extend shelf-life while aligning with consumer demand for minimally processed products. This review critically analyses the scientific foundations and mechanisms of action of bioprotective agents, with a particular focus on lactic acid bacteria. It elucidates how they inhibit spoilage and pathogenic microorganisms through different strategies (competitive exclusion, acidification, and the production of bacteriocins and other compounds). This work also evaluates in…
