Effects of alcohol concentration and temperature on the dynamics and stability of mutant Staphylococcal lipase.

The stability and activity of lipase in organic media are important parameters in determining how quickly biocatalysis proceeds. This study aimed to examine the effects of two commonly used alcohols in industrial applications, methanol (MtOH) and ethanol (EtOH) on the conformational stability and catalytic activity of G210C lipase, a laboratory-evolved mutant of Staphylococcus epidermidis AT2 lipase. Simulation studies were performed using an open-form predicted structure under 30, 40 and 50% of MtOH and EtOH at 25 °C and 45 °C. The overall enzyme structure becomes more flexible with increasin…

Molecular Determinants of Efficient Cobalt-Substituted Hemoprotein Production in E. coli.

Exchanging the native iron of heme for other metals yields artificial metalloproteins with new properties for spectroscopic studies and biocatalysis. Recently, we reported a method for the biosynthesis and incorporation of a non-natural metallocofactor, cobalt protoporphyrin IX (CoPPIX), into hemoproteins using the common laboratory strain Escherichia coli BL21(DE3). This discovery inspired us to explore the determinants of metal specificity for metallocofactor biosynthesis in E. coli. Herein, we report detailed kinetic analysis of the ferrochelatase responsible for metal insertion, EcHemH (E.…

A Novel Alkaline-Stable Chitin Deacetylase from Chitiniphilus eburneus for Eco-Friendly Preparation of Specific Deacetylated Products.

Chitin deacetylase is the key enzyme responsible for the biological deacetylation of chitin. However, its limited sources and low catalytic efficiency toward α-chitin present significant barriers to the bioproduction of chitosan. Herein, a chitin deacetylase Ce1425 sourced from Chitiniphilus eburneus was identified, which displayed optimal catalytic efficiency at pH 10.0 and 55 °C, maintaining 91.92% of its activity at pH 12.0. The presence of Co2+ enhanced its activity by 167.38%. Ce1425 exhibited the highest specific activity toward (GlcNAc)3 (8.45 U/mg) and was capable of catalyzing the dea

A Novel Hydrolase Capable of Efficiently Degrading the Highly Toxic ZEN Derivative.

Zearalenone (ZEN) and its toxic derivatives (α-ZAL and α-ZEL) pose significant challenges to food and feed safety, while efficient degrading enzymes for these toxins remain limited. This study identified a novel XyZHD exhibiting higher catalytic activity toward α-ZAL and α-ZEL than ZEN under the assay conditions. Recombinant XyZHD showed optimal activity at pH 9.0 and 45 °C, and its activity was significantly affected by Zn2+ and Fe3+. Structural analysis combined with mutagenesis studies provided insights into residues associated with catalytic activity. The S153E mutant showed a 3.1-fold inc

Engineered aldehyde dehydrogenases for amide bond formation.

Amide bond formation is widely used in pharmaceutical synthesis, typically involving stoichiometric coupling reagents to activate carboxylic acid substrates for a condensation reaction. As an alternative approach, we repurposed aldehyde dehydrogenases into oxidative amidases by creating a more hydrophobic and spacious catalytic pocket for amines to capture the thioester intermediate. This biocatalyst efficiently facilitates the formation of amide bonds between diverse aldehydes and amines. We also developed a two-step enzymatic cascade to synthesize amides from broadly available aliphatic alco

Enhanced Enzyme Cascade Reactions Through Coencapsulation in Biocompatible Silica Nanoconfinement.

Enzyme cascade reactions, where the product of one enzyme serves as the substrate for another, are fundamental to cellular metabolism and essential for constructing efficient multi-enzyme systems. Spatial confinement of sequential enzymes can enhance cascade efficiency through proximity effects and substrate channeling, mimicking the organization of natural metabolic pathways. Here, we coencapsulated glucose oxidase (GOX) and horseradish peroxidase (HRP) within silica nanocapsules (SiNCs) using an enzyme-friendly synthesis method. The encapsulated enzymes retained high catalytic activity, and

Heterogeneous dual-enzyme cascade platform based on COF and MOF for targeted valorization of rice protein.

Rice protein is a high-quality plant-based resource, but its poor solubility limits applications. Deep enzymatic hydrolysis is required to improve functionality and produce bioactive peptides. However, industrial implementation is hindered by the instability and non-reusability of free proteases. Herein, a substrate-matched heterogeneous dual-enzyme cascade platform was constructed by placing two proteases in distinct framework microenvironments according to their catalytic roles and substrate-accessibility requirements. α-Chymotrypsin was covalently immobilized onto magnetic Fe3O4@DQTp nanoca

A Biomimetic Study of the Behavior of N-Cyclopropyl-Based Single Electron Transfer Probes in the Context of Monoamine Oxidase-Catalyzed Oxidations.

Monoamine oxidase-A and -B are important flavoenzymes involved in the oxidative metabolism of various biogenic amines. Mechanisms including polar/nucleophilic, hydride transfer, and single electron transfer (SET) have been proposed for the initial steps of the catalytic mechanism. The most compelling evidence for the latter comes from the observed inhibitory behavior of N-cyclopropyl compounds. Enzyme inactivation presumably occurs when the primary radical portion of the distonic radical cation, resulting from cyclopropyl ring opening, couples to the enzyme. Previously, we hypothesized that th

Selective Enrichment of α-Glucosylated Trilobatin via Cascade Enzymatic Glycosylation-Hydrolysis in Polysaccharide-Based Aqueous Reaction Systems.

Cyclodextrin glucanotransferase (CGTase)-catalyzed glycosylation is an effective approach to improve flavonoid solubility and bioavailability, while poor substrate solubility and low selectivity often limit its practical application. Herein, a polysaccharide-based sugar-aqueous system coupled with cascade glycosylation-hydrolysis was developed for selective glycosylation of trilobatin. Compared with conventional organic solvents and deep eutectic solvents, the sugar-aqueous system significantly enhanced catalytic performance, achieving 83.62% conversion with α-cyclodextrin. Subsequent amyloglu

Intracellular assembly of artificial enzymes for cytoplasmic enantioselective Mannich reactions.

Artificial enzymes constructed by introducing abiological catalytic cofactors into protein scaffolds are emerging as promising biocatalysts for reactions that have no natural equivalents. However, the design of artificial enzymes for in cellulo stereoselective biosynthesis remains largely unexplored because of the numerous challenges posed by the complex cellular environment. Here, we present an efficient strategy for the intracellular assembly of artificial enzymes via covalent anchoring by incubating cells that are expressing target protein with a synthetic catalytic cofactor, in the manner

Atomically Modulated WSe2 Clusters with Enhanced Biocatalytic Activity for Acute Liver Injury Management.

Two-dimensional WSe2 has attracted considerable attention owing to its tunable electronic structure and intrinsic catalytic activity. However, the catalytic redox performance of pure WSe2 remains limited by insufficient charge-transfer efficiency and suboptimal catalytic kinetics in complex oxidative environments. Herein, we constructed a series of ultrasmall WSe2 clusters via atomic-level regulation. This strategy could modulate their electronic configuration and enhance their catalytic performance. The resulting approximately 2 nm M-WSe2 (M = Ce, Pt, Fe) clusters exhibited total antioxidant

Mechanism of Hyaluronic Acid Hydrolysis Catalyzed by Snake Venom Hyaluronidase.

Hyaluronidases are widely distributed in nature being ubiquitous in snake species (svHyal). They catalyze the hydrolysis of β-1,4-glycosidic bonds in hyaluronic acid, a critical constituent of the extracellular matrix. This facilitates the spread of venom toxins into the bloodstream, exacerbating tissue damage and systemic toxicity─a rationale for their common designation as "spreading factors". While svHyals are not directly toxic, they substantially contribute to the morbidity and mortality associated with snakebite envenomation, the world's most lethal neglected tropical disease. Despite th

Liquid-to-gel transitions of phase-separated coacervate microdroplets enabled by endogenous enzymatic catalysis.

Biomolecular condensates formed by liquid-liquid phase separation (LLPS) play a crucial role in organizing biochemical processes within living cells. The phase transition of these condensates from a functional liquid-like state to a pathological gel-like or solid-like state is believed to be linked to cellular dysfunction and various diseases. Here, we present a biomimetic model to demonstrate that endogenous enzyme-catalyzed crosslinking within condensate-mimicked coacervate microdroplets can promote a liquid-to-gel phase transition. We identify the transformation in physical characteristics

Exploring combi-lipase catalysis of Eversa® Transform and Candida rugosa lipases for enzymatic hydrolysis of crude palm oil: Effects on fatty acid yield and retention of carotene and tocols.

Several lipases have been used to catalyze the hydrolysis of crude palm oil (CPO) to concentrate palm phytonutrients. Previously, the retention of phytonutrients during hydrolysis has been improved with a small addition of ethylenediaminetetraacetic acid. However, the production of free fatty acids (FFA) via enzymatic hydrolysis has yet to be optimised. Thus, for the first time, this study explored the combined use of AY 30SD (from Candida rugosa) and Eversa® Transform 2.0, ET 2.0 (from Aspergillus oryzae) to investigate the effect of reaction time and temperature on the lipases' performance t

Chemoenzymatic Dynamic Kinetic Resolution of a Tertiary Alcohol Based on Compartmentalization of Oxovanadium and Lipase Catalysts by Means of a Polydimethylsiloxane Thimble.

Addressing the challenging field of chemoenzymatic dynamic kinetic resolution (DKR) of tertiary alcohols, for which so far only one example exists in the literature, we combined biocatalytic esterification and oxovanadium-catalyzed racemization, operating both steps in two different compartments of one reactor. The compartmentalization of the two heterogeneous catalysts, namely, immobilized lipase A from Candida antarctica (CAL-A) or its mutant and oxovanadium species on mesoporous silica, was achieved using a polydimethylsiloxane thimble, avoiding contact of the oxovanadium with water, thus m

Improved biodiesel production through the chaotic motion of magnetic whole-cell biocatalysts in an electrical-induced magnetic microchannel reactor.

This study focuses on the application of magnetic whole cells biocatalysts (MWCBs) ofCandida catenulataas the intracellular lipase source for methanolysis of soybean oil in a microchannel reactor under a direct current-induced magnetic field (DC-IMF) and an alternating current-induced magnetic field (AC-IMF). The results showed the active mixing of MWCBs under both IMF systems improved the performance of the microreactor to achieve higher fatty acid methyl ester yield (YEFME) and transesterification rate (R)however, the DC-IMF was more efficient than the AC-IMF. Enzymatic Synthesis of Saturated Bioisosteres of Ortho-Substituted Benzenes by Artificial Photoenzyme.

Saturated bioisosteres of ortho-substituted benzenes are of significant interest due to their enhanced pharmacokinetic properties, such as improved metabolic stability and reduced toxicity, making them valuable in drug design and development. However, efficient synthesis of them remains a challenge in organic chemistry. Herein, we report the biocatalytic synthesis of saturated bioisosteres of ortho-substituted benzenes using engineered artificial photoenzymes. Mechanistic Investigation on the C-C Bond Cleavage of α-Hydroxy Ketone Catalyzed by Engineering P450 Monooxygenase Enzyme F182L-CYP199A4.

In addition to the typical oxygen insertion reaction, some cytochrome P450 also catalyze the C-C cleavage, and most P450 employs Cpd I to trigger lyase reactions. There is also evidence to support an alternative mechanism, in which a species earlier than the formation of Cpd I was suggested to perform the lyase reaction. To understand the detailed reaction mechanism, we performed quantum mechanics/molecular mechanics (QM/MM) calculations to explore the cleavage mechanism of mutated CYP199A4 (F182L-CYP199A4). Our calculation results reveal that the peroxoanion (Fe(III)-O-O2-) rather than oxo-fe
