Fine-Tuning Treatments with PN HPT Devices to Improve Asian Skin Quality: The ACES Consensus.

INTRODUCTION AND PURPOSE: Biological differences between Asians and Caucasians contribute to variations in the prevalence of skin disorders such as melanocytoses and skin aging. The current regenerative medicine indications were primarily developed for Caucasians; adapting them to the specific needs of Asian individuals may be beneficial. Five distinguished Asian experts had this purpose in mind when they convened to develop evidence-based and experience-based recommendations on maximizing the benefits of natural-origin ingredient Polynucleotides High Purification Technology (PN HPT™) in indiv

An independent component analysis confounding factor correction framework for identifying broad impact expression quantitative trait loci.

Genome-wide expression Quantitative Trait Loci (eQTL) studies in humans have provided numerous insights into the genetics of both gene expression and complex diseases. While the majority of eQTL identified in genome-wide analyses impact a single gene, eQTL that impact many genes are particularly valuable for network modeling and disease analysis. To enable the identification of such broad impact eQTL, we introduce CONFETI: Confounding Factor Estimation Through Independent component analysis. CONFETI is designed to address two conflicting issues when searching for broad impact eQTL: the need to

Whole-blood CD3/CD28-stimulated cytokine responses in children with preschool wheeze, asthma, and healthy controls.

BACKGROUND: Childhood asthma and wheezing disorders are heterogeneous conditions that may be reflected by distinct immune response patterns. In this study, we investigated whether CD3/CD28-stimulated cytokine responses differ between children with asthma, preschool wheeze, and healthy controls. METHODS: Baseline samples from 511 children in the All Age Asthma cohort (ALLIANCE; 196 children with asthma, 181 with preschool wheeze, and 134 healthy controls) were analyzed. Whole blood was stimulated with anti-CD3/CD28 antibodies for 48 h, and 37 cytokines were quantified using multiplex immunoass

Associations between affective distress, alcohol use, and cognitive flexibility: moderating effects of COMT Val158Met genotype.

BACKGROUND: Alcohol use is common during emerging adulthood and has been associated with impairments in executive functioning, including cognitive flexibility. Individual differences in affective distress, early life adversity, and genetic variation in dopaminergic signaling pathways may contribute to variability in cognitive flexibility among young adults. The present study examined the relationships among proximal affective distress, distal early life stress, alcohol use, and the functional COMT Val158Met polymorphism. METHODS: Seventy-two college-aged adults completed measures of adverse c

Association of age, hormonal, and lifestyle factors with the Leydig cell biomarker INSL3 in aging men from the European Male Aging Study cohort.

BACKGROUND: Aging in men is accompanied by a broad range of symptoms, including sexual dysfunction, cognitive and musculoskeletal decline, obesity, type 2 diabetes, cardiovascular disease and hypertension, organ degeneration/failure, and increasing neoplasia, some of which are associated with declining levels of Leydig cell-produced testosterone. High natural biological variance, together with multiple factors that can modulate circulating testosterone concentration, may influence its interpretation and clinical implications. Insulin-like peptide 3 is a biomarker of Leydig cell function that m

Exploring the possibility of predicting human head hair greying from DNA using whole-exome and targeted NGS data.

BACKGROUND: Greying of the hair is an obvious sign of human aging. In addition to age, sex- and ancestry-specific patterns of hair greying are also observed and the progression of greying may be affected by environmental factors. However, little is known about the genetic control of this process. This study aimed to assess the potential of genetic data to predict hair greying in a population of nearly 1000 individuals from Poland. RESULTS: The study involved whole-exome sequencing followed by targeted analysis of 378 exome-wide and literature-based selected SNPs. For the selection of predicto

Mechanisms of genetic susceptibility to type I diabetes: beyond HLA.

An individual's predisposition to Type I diabetes (T1D) is largely determined by complex interactions between several genetic loci and other, nonheritable factors. In T1D, the HLA locus has been known for decades to contribute 50% of the inherited risk. Outside the HLA are many proposed candidate loci with smaller effects, but only two confirmed candidate genes, the INS-VNTR and the CTLA-4 genes, which together do not contribute more than 15% of the risk. Because of the high frequency of the disease-associated DNA variants of these genes, understanding the biological mechanisms of such DNA var

Repeated treatments of ciprofloxacin and kresoxim-methyl alter their dissipation rates, biological function and increase antibiotic resistance in manured soil.

The dissipation of ciprofloxacin (CIP, 1.0 and 10.0mg/kg) and kresoxim-methyl (KM, 1.0 and 2.0mg/kg) in manure-amended soil, the variations in soil enzyme activities and microbial functional diversities, and CIP-induced bacterial community tolerances were studied using a chromatographic analysis, enzyme colorimetric and titration analyses, and the BIOLOG EcoPlate method. Three successive treatments of individual and combined samples of CIP and KM at low and high concentrations were performed at 60d intervals. The dissipation half-life of CIP increased, but that of KM decreased in manured soil

Population genetics of Aedes aegypti (Diptera: Culicidae) in India reveals high connectivity and weak geographic and climatic structuring.

Understanding the genetic structure and diversity of Aedes aegypti (Linnaeus, 1762) populations is critical for predicting transmission dynamics and implementing effective vector control strategies. This study assessed the genetic diversity, population structure, and influence of geography and climate on Ae. aegypti populations across India. Of 308 genotyped individuals, 257 high-quality genotypes from 13 collection sites across 6 climatic regions were retained for analysis using 10 microsatellite loci. High genetic diversity was observed across all populations, consistent with substantial eff

Lower Limb Entheseal Variation in Modern Athletes and an Early Medieval Population: Implications for Activity Reconstruction.

OBJECTIVES: Entheses are commonly used to infer past activity patterns, yet their interpretation is limited by the combined effects of mechanical loading and intrinsic biological factors. This study addresses this issue by integrating an Early Medieval population from Pohansko (Czechia; 9th-10th centuries CE) with a Spanish modern professional athlete reference sample characterised by well-defined activity regimes. MATERIALS AND METHODS: Lower limb entheseal areas of the gluteus medius, gluteus maximus, vasti, gastrocnemius, and soleus muscles were quantified in the archaeological individuals

Using point-of-care devices to examine covariation among blood nutritional-physiological parameters and their relationships with poxvirus infection, habitat urbanization, and male plumage coloration in house finches (Haemorhous mexicanus).

The development of inexpensive and portable point-of-care devices for measuring nutritional physiological parameters from blood (e.g., glucose, ketones) has accelerated our understanding and assessment of real-time variation in human health, but these have infrequently been tested or implemented in wild animals, especially in relation to other key biological or fitness-related traits. Here we used point-of-care devices to measure blood levels of glucose, ketones, uric acid, and triglycerides in free-ranging house finches (Haemorhous mexicanus)-a common songbird in North America that has been w

Short-term biological variation study of plasma hemophilia and thrombophilia parameters in a population of apparently healthy Caucasian adults.

OBJECTIVES: Biological variation (BV) data obtained in a standardized way is valuable to assess the analytical requirements and the utility of a reference interval. Our study aimed to determine the short-term BV of thrombophilia (protein S, protein C, activated protein C resistance (APCR) and factor VIII) and hemophilia (factors VIII, IX and XI) parameters in plasma. Coagulation factors V and XII were also evaluated. Based on the obtained data, we assessed analytical performance specifications for the parameters. Finally, we intended to provide a robust tool for comparison of serial measuremen

Peripheral biomarkers of cognitive response to dopamine receptor agonist treatment.

RATIONALE: Using biological markers to objectively measure addiction severity or to identify individuals who might benefit most from pro-cognitive treatment could potentially revolutionize neuropsychopharmacology. We investigated the use of dopamine receptor mRNA levels in circulating blood cells as predictors of cognitive response following dopamine agonist treatment, and as biomarkers of the severity of stimulant drug dependence. METHODOLOGY: We employed a double-blind, placebo-controlled cross-over design, administering a single dose of the selective dopamine D(2/3) receptor agonist pramip

Detection, imputation, and association analysis of small deletions and null alleles on oligonucleotide arrays.

Copy-number variation (CNV) is a major contributor to human genetic variation. Recently, CNV associations with human disease have been reported. Many genome-wide association (GWA) studies in complex diseases have been performed with sets of biallelic single-nucleotide polymorphisms (SNPs), but the available CNV methods are still limited. We present a new method (TriTyper) that can infer genotypes in case-control data sets for deletion CNVs, or SNPs with an extra, untyped allele at a high-resolution single SNP level. By accounting for linkage disequilibrium (LD), as well as intensity data, call

Association of megalin genetic polymorphisms with prostate cancer risk and prognosis.

PURPOSE: Megalin, an endocytic receptor expressed by prostate epithelial cells, can internalize biologically active androgens bound to sex hormone binding globulin. Genetic variation within megalin could potentially influence levels of steroid hormone uptake. EXPERIMENTAL DESIGN: Forty haplotype-tagging single-nucleotide polymorphisms (htSNP) were analyzed in a population-based, case-control study of 553 Caucasian men who were diagnosed with prostate cancer between the ages of 40 and 64 years from the Seattle-Puget Sound region and 534 control men. Prostate cancer risk was estimated using adj

Adult skin acute stress responses to short-term environmental and internal aggression from exposome factors.

Exposome factors that lead to stressed skin can be defined as any disturbance to homeostasis from environmental (meteorological factors, solar radiation, pollution or tobacco smoke) and/or internal exposure (unhealthy diet, hormonal variations, lack of sleep, psychosocial stress). The clinical and biological impact of chronic exposome effects on skin functions has been extensively reviewed, whereas there is a paucity of information on the impact of short-term acute exposure. Acute stress, which would typically last minutes to hours (and generally no more than a week), provokes a transient but

Morphological and genetic variation of the pine shoot tunnel beetle Placusa pinearum (Staphylinidae) in China.

Placusa pinearum, a newly described beetle species found living in pine shoot tunnels bored by the pine tip moth, Dioryctria rubella (Pyralidae), has potential as a vector to transport biological agents for controlling this moth pest of pine trees because of the high degree of niche overlap (co-occurrence) between them. In order to help determine the suitability of potential biological control vectors, it is useful to obtain knowledge concerning the intraspecific variation of the vector. We examined P. pinearum collected from 14 different geographical populations in China using morphological a

Estimating the Irreducible Uncertainty in Visual Diagnosis: Statistical Modeling of Skill Using Response Models.

BACKGROUND: For the representative problem of prostate cancer grading, we sought to simultaneously model both the continuous nature of the case spectrum and the decision thresholds of individual pathologists, allowing quantitative comparison of how they handle cases at the borderline between diagnostic categories. METHODS: Experts and pathology residents each rated a standardized set of prostate cancer histopathological images on the International Society of Urological Pathologists (ISUP) scale used in clinical practice. They diagnosed 50 histologic cases with a range of malignancy, including

Sex-specific interaction effects of Syntaxin 1A coexpression network and childhood trauma on adult depressive symptoms.

BACKGROUND: Disruptions in synaptic function and exposure to early life trauma are both implicated in the development of depression, though the underlying mechanisms remain poorly understood. METHODS: To investigate this relationship, we developed an expression-based polygenic score (ePRS) that captures individual variations in the expression of genes co-expressed with the synaptic protein Syntaxin 1A (STX1A) in the prefrontal cortex (PFC). This polygenic score allows us to explore the impact of variations on synaptic function and childhood trauma on the susceptibility to depressive symptoms
