Critical concentration of 'unbound' cadmium in the rabbit renal cortex.

Critical concentration of 'unbound' cadmium in the rabbit renal cortex was around 13 micrograms Cd/g, which was far lower than the critical concentration of 'total' cadmium in the renal cortex, 200-300 micrograms Cd/g, and a little lower than the critical concentration of non-MT-Cd in the renal cortex, 35-60 micrograms Cd/g, respectively. The above result may suggest that it is necessary to identify the chemical form of 'active' cadmium in the renal cortex.

Sirenomelia: analysis in the cadmium- and lead-treated golden hamster.

Sirenomelia, a fusion of the lower extremities, is believed to result from a median, bilateral symmetric defect of the caudal portion of the embryo at a very early stage in development. Anomalies of the gastrointestinal and urogenital systems are commonly associated with this malformation. Sirenomelia is not an embryo-lethal condition but typically is incompatible with postnatal life when combined with the associated malformations. In this study, intravenous treatment of hamsters with a combination of cadmium and lead on the morning of the eighth day of gestation resulted in 1.4, 22.2, and 35.…

Assessment of renal function in workers previously exposed to cadmium.

Cadmium induced renal effects were examined in 60 workers (58 men, 2 women) previously exposed to cadmium. Tubular damage in the form of beta 2-microglobulinuria was found in 40%, and urinary albumin and orosomucoid increased significantly with increasing urinary cadmium and increasing relative clearance of beta 2-microglobulin. It is suggested that increased albumin excretion is secondary to the tubular damage. In no case was typical glomerular proteinuria found that could be related to cadmium. Histories of renal stones were more common among the workers with high urinary cadmium concentrati…

Effects of cadmium on the placental uptake and transport to the fetus of nutrients.

Cadmium (Cd) is known to produce malformations, growth inhibition of the fetus, and placental necrosis in rodents at higher doses (3-4 mg/kg body wt). We studied the influence of Cd in various doses (0.5-4 mg/kg body wt) and at different survival intervals on the placental uptake and transfer to the fetus of vitamin B12 (vit. B12), zinc (Zn), alpha-aminobutyric acid (AIB), and deoxyglucose (DOX). These were chosen to represent various mechanisms of membrane transport. We show that vit. B12, which is accumulated in the placenta by a presumed receptor-mediated mechanism, is most easily disturbed…

Normal and abnormal midfacial development in the cadmium-treated hamster.

Clefts of the midface ranging in severity from a notched lip to complete facial disorganization were observed in the offspring of golden hamsters injected with cadmium chloride on the morning of the eighth day of gestation. Other malformations frequently observed were microphthalmia, anophthalmia, and diencephalic encephaloceles. Histologic examination of embryos on the 10th, 11th, 12th, and 13th days of gestation revealed a marked deficiency of mesenchyme in the frontonasal process, which led to a foreshortened and deformed nasal septum. These data confirm that the teratogenicity of cadmium i…

Interaction of cadmium and zinc during prenatal development in the rat.

Cadmium chloride, zinc chloride, or a mixture of the two, labeled with 115m-Cd or 65-Zn was administered intraperitoneally to Wistar rats on day 9 of gestation. On day 20 fetuses of Cd-treated rats exhibited malformations, but those of rats given zinc or zinc plus cadmium did not. No radioactive cadmium was recovered in the fetuses or fetal membranes, although some was found in the placentas. Simultaneous administration of zinc did not alter the distribution of cadmium, but cadmium significantly increased the amount of zinc in the fetus and placenta. In a second experiment, cadmium or cadmium …

Cytogenetic investigations on leucocytes of workers from a cadmium plant.

Chromosome analysis has been performed on workers from a cadmium plant who were exposed to fumes and dust of cadmium and lead. The 35 workers were classified, according to the type and duration of exposure, into a group (cadmium service) exposed to high levels of lead and cadmium in the absence of zinc (23 people) and a group of rolling-mill workers subjected mostly to zinc but also to lower levels of lead and cadmium (12 people). A higher yield of severe chromosome anomalies (chromatid exchange, disturbance of spiralization, chromosome translocation, ring and dicentric chromosomes) together w…

Benchmark dose estimation for cadmium-induced renal tubular damage among environmental cadmium-exposed women aged 35-54 years in two counties of China.

BACKGROUND: A number of factors, including gender, age, smoking habits, and occupational exposure, affect the levels of urinary cadmium. Few studies have considered these influences when calculating the benchmark dose (BMD) of cadmium. In the present study, we aimed to calculate BMDs and their 95% lower confidence bounds (BMDLs) for cadmium-induced renal tubular effects in an age-specific population in south-central China. METHODS: In this study, urinary cadmium, β2-microglobulin, and N-acetyl-β-D-glucosaminidase levels were measured in morning urine samples from 490 randomly selected non-smo…

Increase in intracellular Cd(2+) concentration of rat cerebellar granule neurons incubated with cadmium chloride: cadmium cytotoxicity under external Ca(2+)-free condition.

In order to examine the cadmium cytotoxicity unrelated to external Ca(2+), the effects of micromolar CdCl(2) on intracellular Cd(2+) concentration, cellular content of glutathione, and cell viability of rat cerebellar granule neurons were examined under normal Ca(2+) and external Ca(2+)-free conditions, using a laser confocal microscope with fluorescent probes, fluo-3-AM, 5-chloromethylfluorescein (CMF) diacetate, and propidium iodide. CdCl(2) (10-300 microM) dose-dependently increased the intensity of fluo-3 fluorescence. Exposure to CdCl(2) equally enhanced the fluo-3 fluorescence under both…

[Current status of cadmium exposure among Japanese, especially regarding the safety standard for cadmium concentration in rice and adverse effects on proximal renal tubular function observed in farmers exposed to cadmium through consumption of self-grown rice].

Because the staple food in Japan is rice, which absorbs cadmium (Cd) from the soil efficiently, rice is the main source of exposure to Cd in the Japanese population. In addition, there have been many Cd-contaminated farming areas in Japan. Therefore, a safety standard for the Cd concentration in rice was set as 0.4 ppm by the Japanese government. This safety standard has been followed for decades without any appropriate scientific or legal basis. However, recent epidemiological studies of female Japanese farmers exposed to Cd through self-grown rice, that is, a series of Japanese Multi-centere…

Assessment of cadmium and lead as biomarkers of prostate cancer severity.

This study examines the relationship between cadmium, lead exposure and prostate cancer by quantifying elemental concentrations in prostate biopsy tissue. Tissue samples were obtained from a total of 142 patients (64.8% black versus 35.2% white, mean age of 60.9 years, and mean BMI of 29.4) undergoing prostate biopsies. Only 11 (7.7%) of these patients had Gleason score of less than or equal to 6. Majority (59.2%) of them had a score of 3 + 4, 4 + 3 (19%), or higher (14.1%). Associations between tissue cadmium and age, race, body mass index (BMI), smoking status, prostate-specific antigen (PSA…

Disentangling the role of selenium in antagonizing the toxicity of arsenic and cadmium.

Cadmium (Cd) and inorganic arsenic (As) compounds are considered to be among the major public health hazards. This is due to both the high intrinsic toxicity of these substances and the often difficult to avoid exposure of the general population through contaminated water and food. One proposed method to reduce the toxic effects of As and Cd on animals and humans is the use of selenium (Se). As discussed in our previous article, laboratory studies show that this micronutrient can have a beneficial effect on the detoxification of As and Cd in the body through the formation of non-toxic complexe…

Competitive and synergistic effects of phosphate and cadmium coadsorbed on goethite.

Soil acidification in southern China increases the environmental risk posed by heavy metals, especially cadmium (Cd), and has been accelerated by anthropic activities. Long-term fertilizer use increases the quantity of phosphate (P) in soils, which affects the biogeochemistry of heavy metals in soil environments. In this study, various analytical methods were used to investigate the critical mechanisms controlling P(V)-Cd(II) interactions with goethite (α-FeOOH, a typical iron oxide widely distributed in red soil areas) with added P(V)/Cd(II) in different molar ratios (to simulate the annual P…

Is Environmental Cadmium Exposure Causally Related to Diabetes and Obesity?

Cadmium (Cd) is a pervasive toxic metal, present in most food types, cigarette smoke, and air. Most cells in the body will assimilate Cd, as its charge and ionic radius are similar to the essential metals, iron, zinc, and calcium (Fe, Zn, and Ca). Cd preferentially accumulates in the proximal tubular epithelium of the kidney, and is excreted in urine when these cells die. Thus, excretion of Cd reflects renal accumulation (body burden) and the current toxicity of Cd. The kidney is the only organ other than liver that produces and releases glucose into the circulation. Also, the kidney is respon…

The mechanism of the cadmium-induced toxicity and cellular response in the liver.

Cadmium is a toxic element to which man can be exposed at work or in the environment. Cd's most salient toxicological property is its exceptionally long half-life in the human body. Once absorbed, Cd accumulates in the human body, particularly in the liver. The cellular actions of Cd are extensively documented, but the molecular mechanisms underlying these actions are still not resolved. The liver manages the cadmium to eliminate it by a diverse mechanism of action. Still, many cellular and physiological responses are executed in the task, leading to worse liver damage, ranging from steatosis,…
