Optimisation and validation of cost-effective water-based solutions of acid fuchsin for detecting fingermarks in blood.

Fingermark impressions in blood are commonly encountered at violent crime scenes and represent a critical trace that can link an individual to the scene. A range of techniques are available for detecting and enhancing bloody impressions; however, many chemical methods involve using hazardous solvents or require alternative light sources to visualise fluorescence. This is particularly challenging for bloody impressions on dark substrates. An alternative treatment is the protein dye known as acid fuchsin (commonly known as 'Hungarian Red'), which can be visualised under both white light and fluo

Nanoremediation and Antioxidant Potential of Biogenic Silver Nanoparticles Synthesized Using Leucena's Leaves, Stem, and Fruits.

Synthetic dyes are persistent organic environmental pollutants that can cause extensive damage to living beings and to the ecosystem as a whole. Cost-effective, sustainable, and efficient strategies to deal with this type of pollution are necessary as it commonly resists conventional water treatment methods. Silver nanoparticles (AgNPs) synthesized using the aqueous extract from the leaves, stem, and fruits of Leucaena leucocephala (Leucena) were produced and characterized through UV-vis, TEM, EDS, SDL, XPS, XRD, and zeta potential, and they proved to be able to promote adsorption to remediate

What's in My Ink: An Analysis of Commercial Tattoo Ink on the US Market.

As tattoos continue to rise in popularity, the demand for tattoo ink has surged. Historically, tattoo inks have been underregulated in the US market. This study analyzes inks from nine different brands that are common in the United States, ranging from major to small manufacturers. Out of 54 inks, 45 contained unlisted additives and/or pigments. Major, unlisted adulterants include poly(ethylene glycol), propylene glycol, and higher alkanes. Many of the adulterants pose possible allergic or other health risks. Taken together, the results from this study highlight the potential for a significant

Stolen Horses and Scented Garments: Vegetal and Mineral Yellow in Arabic Technical Literature.

Mediaeval Arabic technical literature shows a keen interest in yellow dyes, paints, varnishes, inks, and even perfumes. Recipes reveal that yellow was viewed as just one step away from gold, with preparations for these two colours often sharing ingredients and techniques. In the unfolding of procedures and applications to different materials, from skin to textiles, Arabic sources also offer a glimpse into daily life and shared tastes, presenting luxury objects along with their imitations. This paper traces the role played by yellow and gold in inks, cosmetic dyes, and coloured, scented fabrics

Tattooing: immediate and long-term adverse reactions and complications.

Tattooing has become a popular global trend in industrialised countries, with the highest prevalence rates of up to 30-40 % in the adult population younger than 40 years. Common tattoo inks may contain heavy metals, polycyclic aromatic hydrocarbons, and primary aromatic amines, toxic if exceeding permissible limits. It is estimated that about 14.36 mg of ink is injected per cm2 of skin, at a depth of 1-3 mm. The injected pigment is internalised by neutrophils, fibroblasts, and macrophages or dendritic cells. About 60-90 % of the pigment is then transported to the lymph nodes via the lymphatic

Round Bodies Detected by Treponema pallidum Immunohistochemical Stain in Two Cases of Cutaneous Syphilitic Gummata.

Tertiary syphilis may present a diagnostic challenge due to negative nontreponemal serologies in up to 30% of cases and frequent lack of identifiable spirochetes on histopathology or other direct detection tests. We report 2 cases of round bodies staining with Treponema pallidum immunohistochemistry by light microscopy in biopsies from cutaneous syphilitic gummata. In 1 case, the finding was validated 3 times by 2 independent laboratories; in the other case, T. pallidum was detected by polymerase chain reaction in the biopsy sample. Spirochete round bodies have previously been reported in the

A versatile vessel casting method for fine mapping of vascular networks using a hydrogel-based lipophilic dye solution.

Efficient labeling of the vasculature is important for understanding the organization of vascular networks. Here, we propose VALID, a vessel-labeling method that enables visualization of vascular networks with tissue clearing and light-sheet microscopy. VALID transforms traditional lipophilic dye solution into hydrogel by introducing gelatin and restrains the dye aggregation, resulting in complete and uniform vessel-labeling patterns with high signal-to-background ratios. VALID also enhances the compatibility of lipophilic dyes with solvent-based tissue-clearing protocols, which was hard to ac

Some experiments and remarks regarding the possible formation of blood stains on the Turin Shroud: stains attributed to the crown of thorns, the lance wound and the belt of blood.

The Turin Shroud (TS) is a Christian relic interpreted to be the burial cloth of Jesus of Nazareth. It exhibits red discolorations that have been interpreted as blood stains and that are the subjects of a highly controversial discussion. We conducted experiments to identify theoretically possible explanations for the stains attributed to the crown of thorns, the lance wound and the belt of blood. In the experiments with a focus on the stains attributed to the crown of thorns, a very similar stain pattern as on the TS could be provoked by simulating the following sequence of events: blood from

Rhodamine-based fluorescent sensors for the rapid and selective off-on detection of salicylic acid and their use in plant cell imaging.

Salicylic acid (SA) is a key hormone that regulates plant growth and immunity, and understanding the physiologic processes induced by SA enables the development of highly pathogen-resistant crops. Here, we report the synthesis of three new SA-sensors (R1-R3) from hydroxyphenol derivatives of a rhodamine-acylhydrazone scaffold and their characterization by NMR and HRMS. Spectroscopic analyses revealed that structural variations in R1-R3 resulted in sensors with different sensitivities for SA. Sensor R2 (with the 3-hydroxyphenyl modification) outperformed R1 (2-hydroxyphenyl) and R3 (4-hydroxyph

Factors affecting insulation failure in reusable surgical devices.

The purpose of this study was to investigate the associated factors of insulation failure (IF) in reusable endoscopic instruments. The insulation coating of reusable endoscopic instruments underwent routine visual checks, hand washing to remove visible stains, and mechanized sterilization. We recorded the cleaning number and usage period of all instruments. The instruments were tested for IF using a detector. IF was found in eight of 69 devices (11.6%). Examining by clinical specialty, we found IF in 4 of 28 gastrointestinal (14.3%), 3 of 20 gynecological (15.0%), 1 of 12 urological (8.3%), an

The Encapsulation of Natural Organic Dyes on TiO2 for Photochromism Control.

Titanium dioxide (TiO2) plays a pivotal role in photocatalytic reactions and holds great promise for the cosmetic and paint industries due to its white color and high refractive index. However, the original color of TiO2 changes gradually to blue or yellow with UV irradiation, which affects its color realization. We encapsulated TiO2 with several natural organic dye compounds, including purpurin, curcumin, and safflower, to control its photochromism and realize a range of different colors. The chemical reaction between TiO2 and dyes based on their functional group was investigated, and the lig

CuCeTA nanoflowers as an efficient peroxidase candidate for direct colorimetric detection of glyphosate.

Conventional nanozyme-based pesticide detection often requires the assistance of acetylcholinesterase. In this work, a CuCeTA nanozyme was successfully designed for the direct colorimetric detection of glyphosate. Direct detection can effectively avoid the problems caused by cascading with natural enzymes such as acetylcholinesterase. By assembling tannic acid, copper sulfate pentahydrate and cerium(III) nitrate hexahydrate, CuCeTA nanoflowers were prepared. The obtained CuCeTA possessed excellent peroxidase-like activity that could catalyze the oxidation of 3,3',5,5'-tetramethylbenzidine (TMB

Large-scale interrogation of retinal cell functions by 1-photon light-sheet microscopy.

Visual processing in the retina depends on the collective activity of large ensembles of neurons organized in different layers. Current techniques for measuring activity of layer-specific neural ensembles rely on expensive pulsed infrared lasers to drive 2-photon activation of calcium-dependent fluorescent reporters. We present a 1-photon light-sheet imaging system that can measure the activity in hundreds of neurons in the ex vivo retina over a large field of view while presenting visual stimuli. This allows for a reliable functional classification of different retinal cell types. We also dem

A Perspective on the Use of Fluorescent Imaging to Reveal Mechanisms of Breakup.

Purpose: Tear film instability, which can lead to rapid tear film breakup (TBU), is considered to be a major etiological factor in dry eye. However, experimental support for many of the proposed theories for TBU mechanisms is relatively scarce. The major aim of this perspective is to show that fluorescence studies of TBU can be used to provide experimental evidence for two proposed underlying mechanisms of TBU, evaporation and divergent flow. Methods: To understand the effects of TBU on tear film fluorescence, we show that local fluorescence is the product of three main factors: tear film thick

Phosphate based new organic polymer networks for efficient dye sorption and catalyst loading for chemo-selective reactivity.

Phosphate based organic polymer networks (OPNs) have been synthesized for the first time for dye sorption and heterogeneous catalysis. The OPNs were sythesized by the polycondensation of POCl3 with di- and tri-hydroxy organic linkers e.g., quinol, 4,4'-biphenol, phloroglucinol and 1,3,5-(4-hydroxyphenyl)benzene. These show remarkably selective adsorption of cationic dyes methylene blue and propidium iodide via the electrostatic interaction of the polymers with the dyes. These OPNs also allow the in situ synthesis and stabilization of gold nanoparticles within the polymer networks which demonst

Ratiometric fluorescence sensing of formaldehyde in food samples based on bifunctional MOF.

A new fluorescence strategy was described for ratiometric sensing of formaldehyde (FA) with bifunctional MOF, which acted as a fluorescence reporter as well as biomimetic peroxidase. With the assistance of H2O2, NH2-MIL-101 (Fe) catalyzes the oxidation of non-luminescent substrate o-phenylenediamine (OPD) to produce fluorescent product (oxOPD) with the maximum emission at 570 nm. Besides, intrinsic fluorescence of MOF (λem = 445 nm) was quenched by oxOPD through inner filter effect (IFE). However, FA and OPD reacted to generate Schiff bases, which competitively consumed OPD inhibiting the gene

Characterizing paint technologies and recipes in Levantine and Schematic rock art: El Carche site as a case study (Jalance, Spain).

This paper contributes to current debates on the technologies and practices of prehistoric artists using the rock art site of el Carche (Jalance, Spain) as a case study. The site preserves both Levantine and Schematic paintings, yet poorly understood from an analytical point of view. In the past, it has even been argued how little differentiation there is between these two post-Paleolithic traditions in terms of paint composition. Our aim with this paper was to identify pigments, paint recipes and technologies and decipher the order of the superimpositions, both between Levantine motifs of dif

An Approach to Developing Cyanines with Upconverted Photosensitive Efficiency Enhancement for Highly Efficient NIR Tumor Phototheranostics.

Upconverted reactive oxygen species (ROS) photosensitization with one-photon excitation mode is a promising tactic to elongate the excitation wavelengths of photosensitive dyes to near-infrared (NIR) light region without the requirement of coherent high-intensity light sources. However, the photosensitization efficiencies are still finite by the unilateral improvement of excited-state intersystem crossing (ISC) via heavy-atom-effect, since the upconverted efficiency also plays a decisive role in upconverted photosensitization. Herein, a NIR light initiated one-photon upconversion heavy-atom-fr
