Pinning Down the Mechanism of Transport: Probing the Structure and Function of Transporters Using Cysteine Cross-Linking and Site-Specific Labeling.

Transporters are crucial in a number of cellular functions, including nutrient uptake, cell signaling, and toxin removal. As such, transporters are important drug targets and their malfunction is related to several disease states. Treating transporter-related diseases and developing pharmaceuticals targeting transporters require an understanding of their mechanism. Achieving a detailed understanding of transporter mechanism depends on an integrative approach involving structural and computational approaches as well as biochemical and biophysical methodologies. Many of the elements of this tool

Community resilience assessment and literature analysis.

Earlier and current disaster-related research emphasised the sociological/behavioural perspective. This led to a significant amount of literature devoted to descriptive context of natural, man-made and technological disasters and sequelae. This paper considers a next step involving a more expanded approach in research methodology. The phases include: (1) the development of a comprehensive database of ideas provided by authors of scholarly and scientific papers; (2) the development of computer-supported algorithms to prepare an array of scenarios representing relationships, gaps and inconsisten

miRNA target enrichment analysis reveals directly active miRNAs in health and disease.

microRNAs (miRNAs) are short non-coding regulatory RNA molecules. The activity of a miRNA in a biological process can often be reflected in the expression program that characterizes the outcome of the activity. We introduce a computational approach that infers such activity from high-throughput data using a novel statistical methodology, called minimum-mHG (mmHG), that examines mutual enrichment in two ranked lists. Based on this methodology, we provide a user-friendly web application that supports the statistical assessment of miRNA target enrichment analysis (miTEA) in the top of a ranked li

FDG-PET in Infection Imaging of Immuno-compromised Patients.

Infectious syndromes represent a monumental challenge in the management of immunocompromised patients, directly driving high rates of morbidity and critical mortality. This clinical cohort comprised oncological populations undergoing aggressive cytoreductive chemotherapy, end-stage renal disease patients dependent on specialized long-term dialysis access networks, and individuals undergoing treatment with advanced antiretroviral regimens. Because these individuals suffer from blunted or suppressed cellular immune mechanisms, standard structural imaging methodologies (such as standard digital r

Mild and fast access to indenopyrans via tandem Michael addition/cyclization of enynones and 1,3-indandiones.

A novel and efficient strategy for constructing fused heterocyclic indenopyrans is established through a tandem Michael addition/cyclization reaction between enynones and 1,3-indandiones. A slight alteration in the reaction duration facilitated the formation of another indenopyran isomer. The methodology is mild, rapid, operationally simple, and atom-economical, providing good to excellent yields. Furthermore, DFT calculations were performed to evaluate the stability of the isomers, which in turn confirmed the kinetically/thermodynamically controlled product. UV-visible studies revealed that t

A structure-based approach towards the identification of novel antichagasic compounds: Trypanosoma cruzi carbonic anhydrase inhibitors.

Trypanosoma cruzi carbonic anhydrase (TcCA) has recently emerged as an interesting target for the design of new compounds to treat Chagas disease. In this study we report the results of a structure-based virtual screening campaign to identify novel and selective TcCA inhibitors. The combination of properly validated computational methodologies such as comparative modelling, molecular dynamics and docking simulations allowed us to find high potency hits, with KI values in the nanomolar range. The compounds also showed trypanocidal effects against T. cruzi epimastigotes and trypomastigotes. All

[Radiological procedures in the traumatised wrist].

Injuries of the wrist are difficult to diagnose because of the complex and narrow anatomic structures. On the basis of precise clinical examination, X-rays, CT and MRI are valuable additional tools that can be used. In the case of bone injury, spiral computer tomography with multiplanar reformatting is currently the method of choice. MRI is indicated for the identification of soft tissue or ligamentous injury and avital fragments or necrosis. Other diagnostic tools for the wrist are currently of minor importance. Technical and methodological innovations allow ever better visualisation and clas

Quantitation of renal function based on two-compartmental modeling of renal pelvis.

The primary functions of the kidney are: (i) to get rid of the body waste materials that are either ingested or produced by metabolism, and (ii) to control the volume and composition of the body fluids. Herein, we provide a noninvasive methodology to assess physiological function of the kidneys. For this purpose, we analyze the renograms with 2-compartmental modelling of the kidney-renal outflow system, and therefrom compute the amount of flow of renal radionuclide into and out of the renal pelvis compartment. The derived information of uptake (k/A) and washout (e(beta/2V2)t sinhAt) rates can

Decoding adaptive immunity: advanced strategies in T and B cell repertoire analysis.

BACKGROUND: The immune repertoire (IR) reflects the adaptive immune system of an organism by encompassing the diversity and composition of receptor sequences expressed by T and/or B lymphocytes. Despite significant advances in the efficiency and accuracy of antigen discrimination facilitated by high-throughput sequencing technologies, IR analysis remains challenging due to its extraordinary heterogeneity and the high dimensional nature of sequencing data. MAIN BODY: This review provides an overview of the key steps involved in IR profiling, highlighting factors that impact data accuracy, inclu

Assessment of air kerma-area product in computed tomography using small solid-state dosemeters.

Technological advances in computed tomography (CT) have highlighted limitations of traditional dose descriptors such as computed tomography dose indexvol and dose-length product, particularly for wide-beam and modulated acquisitions. This study investigates a CT-specific, operational air kerma-area product derived from time-resolved measurements using small solid-state dosemeters mounted at the gantry aperture. A mathematical framework is presented that accounts for tube rotation, bow-tie filtration, beam geometry, and the longitudinal dose profile. Model validation was performed using measure

Beyond the Transcriptome: Leveraging Cellular Indexing of Transcriptomes and Epitopes by Sequencing (CITE-seq) for Deeper Cellular Insights.

The advent of single-cell genomics has revealed profound cellular heterogeneity, yet transcriptomic profiling alone often fails to show the functional proteomic state of a cell. Cellular indexing of transcriptomes and epitopes by sequencing (CITE-seq) emerged to bridge this critical gap, enabling the simultaneous quantification of RNA and surface protein expression within individual cells. This transformative technology leverages oligonucleotide-conjugated antibodies to assess protein abundance using a panel of marker-specific tags, which are cocaptured with cellular messenger RNA in single-ce

Fast simulation of soft x-ray near-edge spectra using a relativistic state-interaction approach: Application to closed-shell transition metal complexes.

Spectroscopic techniques based on core-level excitations offer powerful tools for probing molecular and electronic structures with high spatial resolution. However, accurately calculating spectral features at the L or M edges is challenging due to the significant influence of spin-orbit and multiplet effects. While scalar-relativistic effects can be incorporated with minimal computational cost, accounting for spin-orbit interactions requires complex frameworks that can be computationally expensive. In this work, we develop a reduced-cost state-interaction approach for simulating near-edge soft

Quantitative ultrasound methods to assess bone mineral status in children: technical characteristics, performance, and clinical application.

Measurement of bone mineral status may be a useful tool in identifying the children who could be exposed to an increased risk of osteoporosis in adulthood. Dual energy x-ray absorptiometry and peripheral quantitative computed tomography may be used to this purpose, but the exposure to ionizing radiation is a limiting factor for preventive studies in large populations of children. In the last years, quantitative ultrasound (QUS) methods have been developed to assess bone mineral status in some peripheral skeletal sites such as calcaneus, phalanges of the hand, and tibia. QUS techniques are safe
