Single-cell Sequencing of Circulating Human Plasmablasts during Staphylococcus aureus Bacteremia.

Staphylococcus aureus is the major cause of healthcare-associated infections, including life-threatening conditions as bacteremia, endocarditis, and implant-associated infections. Despite adequate antibiotic treatment, the mortality of S. aureus bacteremia remains high. This calls for different strategies to treat this infection. In past years, sequencing of Ab repertoires from individuals previously exposed to a pathogen emerged as a successful method to discover novel therapeutic monoclonal Abs and understand circulating B cell diversity during infection. In this paper, we collected peripher

Detection of Divergent Highly Pathogenic Avian Influenza A(H5N1) Clade 2.3.4.4b Virus, São Paulo, Brazil, 2025.

In 2025, we detected highly pathogenic avian influenza H5N1 virus in dead waterfowl at Ibirapuera Park, São Paulo, Brazil. Genomic characterization indicated a reassortant virus that emerged from locally circulating low pathogenicity avian influenza viruses and highly pathogenic North American lineages. Our results highlight cross-species transmission risk and underscore the need for enhanced surveillance.

Management of PFO in paradoxical embolic stroke with hemorrhagic conversion: a case report.

A paradoxical embolism is defined as a venous thrombus that crosses through a heart defect, into the systemic circulation, usually through a patent foramen ovale. Treatment varies between closure of patent foramen ovale vs. medical management based on a variety of individual risk factors and the cardiac defect's characteristics. We describe a case of paradoxical stroke complicated by hemorrhagic conversion, ultimately requiring an IVC filter.

Recombinant strains of HIV type 1 in the United Kingdom.

Twenty-five recombinant (mosaic) HIV-1 genomes were detected among 151 samples comprising 118 non-B subtype sequences and 33 samples containing subtype B sequences. Seven of the 25 mosaic patterns were similar to characterized circulating recombinant forms (two A/E, four A/G, and one D/F) and one was a MAL-like A/D recombinant. Eighteen of the recombinants had evidence of subtype A sequences in at least one region of their genome. One sample was found to contain a novel recombinant form (pol F, env K). Two samples could not be characterized unambiguously as recombinant forms and a further one

Immunogenicity against Far Eastern and Siberian subtypes of tick-borne encephalitis (TBE) virus elicited by the currently available vaccines based on the European subtype: systematic review and meta-analysis.

Tick-borne encephalitis (TBE) virus, which is usually divided into European, Far Eastern and Siberian subtypes, is a serious public health problem in several European and Asian countries. Vaccination is the most effective measure to prevent TBE; cross-subtype protection elicited by the TBE vaccines is biologically plausible since all TBE virus subtypes are closely related. This manuscript systematically explores available data on the cross-subtype immunogenicity elicited by the currently available Western vaccines based on the European subtype. Completed immunization course of 3 doses of both

Bullous pemphigoid and linear IgA dermatosis sera recognize a similar 120-kDa keratinocyte collagenous glycoprotein with antigenic cross-reactivity to BP180.

Circulating IgG from a large subset of bullous pemphigoid (BP) patients reacted on immunoblot with a 120-kDa protein in conditioned keratinocyte culture medium and in keratinocyte cell extracts. A protein with a similar molecular weight was recognized by circulating IgA from a subset of patients with linear IgA dermatosis (LAD). Both affinity-purified 120-kDa-specific BP IgG and 120-kDa-specific LAD IgA bound to the roof of salt-split skin. Both proteins recognized are collagenous glycoproteins. Deglycosylation with N-glycosidase F resulted in an identical reduction in molecular weight for bot

A cross-sectional examination of immune adaptations during pregnancy in the ECHO Cohort.

BACKGROUND: Pregnancy requires finely tuned immune changes that support implantation, placental development, maternal-fetal tolerance, and preparation for labor, yet the normative trajectories of circulating inflammatory proteins across gestation remain poorly defined. This cross-sectional study investigates how circulating inflammatory proteins vary with gestational age in pregnancy and examines the impacts of fundamental biological characteristics, such as gravidity and fetal sex. METHODS: Data were drawn from 1154 pregnant individuals from six study sites of the National Institutes of Heal

Lipoprotein subclasses compensate the classic atherogenic lipid profile of women with polycystic ovary syndrome, but common antiandrogenic treatments worsen their lipidome.

OBJECTIVES: Patterns of circulating inflammatory glycoproteins and lipoprotein subclasses are not well-defined in polycystic ovary syndrome (PCOS). We aimed to describe these profiles and effects of drug intervention on those parameters. DESIGN: Cross-sectional and longitudinal interventional prospective studies. METHODS: The cross-sectional study included 340 patients with PCOS and 92 non-hyperandrogenic women. Glycoprotein and lipoprotein profiles were measured by proton-nuclear magnetic resonance spectroscopy. We also analyzed their changes after treatment with metformin, an oral contrace

Susceptibility of Mammals to Highly Pathogenic Avian Influenza: A Qualitative Risk Assessment From the Belgian Perspective.

AIMS: The world experienced a huge number of outbreaks of highly pathogenic avian influenza (HPAI) in birds, which could represent one of the largest registered epidemics of infectious disease in food-producing animals. Therefore, mammals, including humans, are continuously exposed to HPAI viruses leading to sporadic and sometimes unusual mammal infections. The aim of this paper is to assess the risk of crossing the avian/mammalian species barrier by the currently circulating HPAI viruses, focusing on the epidemiological situation of Belgium, a representative country for Western Europe. Identifying the Shared Metabolite Biomarkers and Potential Intervention Targets for Multiple Sarcopenia-Related Phenotypes.

The relationship between circulating metabolites and sarcopenia-related phenotypes remains unclear. We explored the causality between circulating metabolites and sarcopenia-related phenotypes. Instrumental variables for the human metabolome were derived from the recently published GWAS, which included 690 plasma metabolites. Summary statistics for four sarcopenia phenotypes (whole-body lean mass (WBLM), usual walking pace, appendicular lean mass (ALM), and handgrip strength (HGS)) (both sexes, males and females) were obtained from relevant GWASs. We used MR to evaluate the association between

Small RNAs are differentially expressed in autoimmune and non-autoimmune diabetes and controls.

OBJECTIVE: Diabetes is a heterogeneous disease and a precise diagnosis of diabetes subgroups is necessary to initiate proper early treatment and clinical management of the disease. Circulating small RNAs (sRNAs) are potentially diagnostic biomarkers in diseases, including diabetes. Here we aimed to examine whether profiles of circulating sRNAs differed between patients with autoimmune and non-autoimmune diabetes and non-diabetic controls. DESIGN: This cross-sectional case-control study included participants from the third survey of the HUNT study. METHODS: We performed sRNA sequencing in ser

Circulating Carboxylated Osteocalcin Correlates With Skeletal Muscle Mass and Risk of Fall in Postmenopausal Osteoporotic Women.

BACKGROUND: Bone and skeletal muscle represent a single functional unit. We cross-sectionally investigated body composition, risk of fall and circulating osteocalcin (OC) isoforms in osteoporotic postmenopausal women to test the hypothesis of an involvement of OC in the bone-muscle crosstalk. MATERIALS AND METHODS: Twenty-nine non-diabetic, non-obese, postmenopausal osteoporotic women (age 72.4 ± 6.8 years; BMI 23.0 ± 3.3 kg/m2) underwent to: 1) fasting blood sampling for biochemical and hormone assays, including carboxylated (cOC) and uncarboxylated (uOC) osteocalcin; 2) whole-body dual ener

Failure of selective shunting to intercostal arteries to prevent spinal cord ischemia during experimental thoracoabdominal aortic occlusion.

Paraplegia from spinal cord ischemia during thoracoabdominal aneurysm repair remains an unpredictable and unpreventable complication. In an effort to prevent spinal cord ischemia during aortic cross-clamping, preoperative angiographic localization of the blood supply to the spinal cord was performed in dogs. Sixteen animals underwent 60 minutes of thoracoabdominal aortic cross-clamping either without (control, n = 8) or with (shunted, n = 8) a selective shunt. Shunting was performed from the aortic arch to that isolated aortic segment angiographically shown to supply the thoracolumbar anterior

Genetic and Epidemiological Evidence of Avian Influenza A(H9N2) Detection Among Poultry in Ghana, 2022.

Avian influenza viruses continue to pose significant zoonotic and pandemic threat globally, with low-pathogenic avian influenza A(H9N2) being of particular concern due to sustained circulation in poultry, adaptability, and repeated human spillover. This study investigated the detection and genetic characterization of influenza viruses at the animal-human interface in Ghana in 2022, using a nationwide cross-sectional One Health approach. Samples were collected from poultry, pigs, the environment, and animal handlers across backyard farms, commercial farms, and live bird markets. Laboratory test

Detection and structural characterization of bat-associated dengue virus serotype 2: A country-level first detection in Pakistan.

Bats (order Chiroptera) are recognized as reservoirs for diverse zoonotic viruses, including members of the Flavivirus genus. Despite global reports of bat-associated Dengue Virus (DENV), its presence in Pakistan has remained undocumented. This study investigates DENV circulation in bat populations across Islamabad/Rawalpindi, Lahore, Mardan, Swat, and Lower Dir. Fecal samples were screened using CprM-specific primers, yielding 14 amplified viral isolates (BatDENV01-BatDENV14). Phylogenetic analysis confirmed all sequences as Dengue Virus serotype 2 (DENV-2), clustering with bat-associated iso

Unrecognized Dengue Transmission in Socially Vulnerable Peri-Urban Neighborhoods of a Temperate Argentine City: Integrating Serology with Knowledge, Attitudes, and Practices.

Background/Objectives: Dengue is an emerging arboviral disease in temperate South America, where urban expansion, climate variability, and social vulnerability favor transmission. In Argentina, the endemic circulation of dengue was established in the late 1990s and outbreaks are reported every three or four years. Methods: This cross-sectional study assessed dengue virus (DENV) seropositivity and associated sociodemographic, environmental, and knowledge-attitudes-practices (KAPs) factors in three socioeconomically vulnerable peripheral neighborhoods of Santa Fe, Argentina, between December 201

Spatial interaction between retinal and choroidal circulation in retinal vein occlusion.

Retinal vein occlusion (RVO) is a leading cause of vision loss, yet the interaction between retinal and choroidal circulation in this condition remains incompletely understood. This retrospective, cross-sectional study aimed to quantitatively evaluate choroidal structural changes in treatment-naïve unilateral RVO using ultra-widefield imaging. Twenty-nine patients with unilateral branch or central RVO and their unaffected fellow eyes were analyzed. Mean choroidal thickness (MCT) and choroidal vascular index (CVI) were measured on widefield swept-source optical coherence tomography, while choro

Fetoplacental vascular effects of maternal adrenergic antihypertensive and cardioprotective medications in pregnancy.

Maternal cardiovascular diseases, including hypertension and cardiac conditions, are associated with poor fetal outcomes. A range of adrenergic antihypertensive and cardioprotective medications are often prescribed to pregnant women to reduce major maternal complications during pregnancy. Although these treatments are not considered teratogenic, they may have detrimental effects on fetal growth and development, as they cross the fetoplacental barrier, and may contribute to placental vascular dysregulation. Medication risk assessment sheets do not include specific advice to clinicians and women
