How close are we to a cAMP- and cGMP-theory-based pharmacological therapy for fragile X syndrome?

Recent advances in targeting cAMP and cGMP pathways offer hope for treating fragile X syndrome, a leading cause of inherited intellectual disability. PDE4 and PDE2 inhibitors have shown promise in animal models, improving memory, social behavior, and cognitive function. Clinical trials are underway, raising optimism for future therapies.

Once upon a Testis: The Tale of Cyclic Nucleotide Phosphodiesterase in Testicular Cancers.

Phosphodiesterases are key regulators that fine tune the intracellular levels of cyclic nucleotides, given their ability to hydrolyze cAMP and cGMP. They are critical regulators of cAMP/cGMP-mediated signaling pathways, modulating their downstream biological effects such as gene expression, cell proliferation, cell-cycle regulation but also inflammation and metabolic function. Recently, mutations in PDE genes have been identified and linked to human genetic diseases and PDEs have been demonstrated to play a potential role in predisposition to several tumors, especially in cAMP-sensitive tissue

Variations in cyclic adenosine 3',5'-monophosphate and cyclic guanosine 3',5'-monophosphate content and efflux from the photosensitive pineal organ of the pike in culture.

The photoreceptor cells of the pike pineal organ transduce 24-h light/dark (LD) information to synchronize the clocks driving the melatonin (MEL) rhythm. In fish, the nocturnal rise in MEL synthesis is associated with an increase in cyclic adenosine 3', 5'-monophosphate (cAMP) production and with Ca2+ entry, through voltage-gated channels. Light induces inhibition of MEL synthesis and a depression of cAMP content, as well as closure of Ca2+ channels. Cyclic guanosine 3',5'-monophosphate (GMP) levels also are reduced upon acute illumination but this second messenger of phototransduction does no

[Cyclic nucleotides during radiotherapy of breast cancer].

Levels of cAMP and cGMP were higher in breast cancer tissue as compared with benign tumors. Large-fraction irradiation was followed by a decrease in cGMP concentration matched by a rise in cAMP/cGMP ratio in cases of radiosensitive tumors. Small-fraction treatment did not affect cyclic nucleotide levels. Radiotherapy was followed by a decrease in initially raised blood plasma cyclic nucleotide concentration.

Distinct Long- and Short-Term Adaptive Mechanisms in Pseudomonas aeruginosa.

Heterogeneous environments such as the chronically infected cystic fibrosis lung drive the diversification of Pseudomonas aeruginosa populations into, e.g., mucoid, alginate-overproducing isolates or small-colony variants (SCVs). In this study, we performed extensive genome and transcriptome profiling on a clinical SCV isolate that exhibited high cyclic diguanylate (c-di-GMP) levels and a mucoid phenotype. We observed a delayed, stepwise decrease of the high levels of c-di-GMP as well as alginate gene expression upon passaging the SCV under noninducing, rich medium growth conditions over 7 day

Analysis of Drosophila STING Reveals an Evolutionarily Conserved Antimicrobial Function.

The vertebrate protein STING, an intracellular sensor of cyclic dinucleotides, is critical to the innate immune response and the induction of type I interferon during pathogenic infection. Here, we show that a STING ortholog (dmSTING) exists in Drosophila, which, similar to vertebrate STING, associates with cyclic dinucleotides to initiate an innate immune response. Following infection with Listeria monocytogenes, dmSTING activates an innate immune response via activation of the NF-κB transcription factor Relish, part of the immune deficiency (IMD) pathway. DmSTING-mediated activation of the i

Cyclic AMP resets the circadian clock in cultured Xenopus retinal photoreceptor layers.

The Xenopus retinal photoreceptor layer contains a circadian oscillator that regulates melatonin synthesis in vitro. The phase of this oscil … lin and 3-isobutyl-1-methylxanthine increased the accumulation of intracellular cyclic AMP, but not cyclic GMP, in photoreceptor layers. The results indicate that cyclic AMP-dependent pathways regulate the photoreceptor circadian oscillator and suggest that a decrease in cyclic AMP may be involved in circadian entrainment by light and/or dopamine.

Immunologic and neuropharmacologic stimulation of mucous glycoprotein release from human airways in vitro.

Human bronchial airways obtained after surgical resection were maintained in tissue culture for 24-48 h. Incorporation of [(3)H]- or [(14)C] … d cholinergic stimulation of human tissues induce the formation of guanosine 3',5'-phosphoric acid (cyclic GMP), and 8-bromo cyclic GMP added to the airways led to increased mucous secretion. Thus, it seems likely that neurohormones capable of stimulating cyclic GMP formation in human airways may lead to increased mucous glycoprotein release.

Flagellar motility, extracellular proteases and Vibrio cholerae detachment from abiotic and biotic surfaces.

Vibrio cholerae of serogroups O1 and O139, the causative agent of Asiatic cholera, continues to be a major global health threat. This pathogen utilizes substratum-specific pili to attach to distinct surfaces in the aquatic environment and the human small intestine and detaches when conditions become unfavorable. Both attachment and detachment are critical to bacterial environmental survival, pathogenesis and disease transmission. However, the factors that promote detachment are less understood. In this study, we examine the role of flagellar motility and hemagglutinin/protease (HapA) in vibrio

A photoaffinity probe covalently modifies the catalytic site of the cGMP-binding cGMP-specific phosphodiesterase (PDE-5).

The cGMP-binding cGMP-specific phosphodiesterase (PDE-5) contains distinct catalytic and allosteric binding sites, and each is cGMP-specific. Cyclic nucleotide phosphodiesterase inhibitors, such as 3-isobutyl-1-methylxanthine (IBMX), are believed to compete with cyclic nucleotides at the catalytic sites of these enzymes, but the portion of PDE-5 that accounts for interaction of either of these inhibitors of the substrates themselves with the catalytic domain of the enzymes has not been identified. IBMX was derivatized to yield the photoaffinity probe 8([3-125I,-4-azido]-benzyl)-IBMX, which is

Effects of phosphodiesterase inhibitors on human lung mast cell and basophil function.

1. The non-hydrolysable cyclic AMP analogue, dibutyryl (Bu2)-cyclic AMP, inhibited the stimulated release of histamine from both basophils and human lung mast cells (HLMC) in a dose-dependent manner. The concentrations required to inhibit histamine release by 50% (IC50) were 0.8 and 0.7 mM in basophils and HLMC, respectively. The cyclic GMP analogue, Bu2-cyclic GMP, was ineffective as an inhibitor of histamine release in basophils and HLMC. 2. The non-selective phosphodiesterase (PDE) inhibitors, theophylline and isobutyl-methylxanthine (IBMX) inhibited the IgE-mediated release of histamine fr…

The effect of cyclic nucleotides on secretin secretion in canine duodenal mucosa in vitro.

We examined the effects of cyclic nucleotides and calcium on secretin release from canine duodenal mucosal explants incubated in organ culture media. Time course studies revealed that at pH 7.4, 5 and 10 mM dibutyryl cyclic adenosine monophosphate (DBcAMP) increased secretin release progressively, reaching a peak at 2 hours. Two mM of DBcAMP at pH 7.4 did not increase secretin release but at pH 4.5, all 3 doses potentiated secretin release. DBcAMP-stimulated secretin release was not dependent on the influx of extracellular calcium. Graded doses of 3-isobutyl-1-methylxanthine (IBMX) did not sti

Therapeutic potentials of natural compounds acting on cyclic nucleotide phosphodiesterase families.

Intracellular cyclic AMP and/or cyclic GMP are characterized in the 1960th. These second messengers, hydrolysed specifically by cyclic nucleotide phosphodiesterase (PDE), play a major role in intracellular signalling. Natural products have been a rich source of drug discovery, Theophylline and Methylxanthine originated from tea leaves used for asthma treatment, whereas, Papaverine, a natural isoquinolein originated from Papaver somniferum traditionally used in impotency, altogether as caffeine where firstly described as PDE-inhibiting compounds. Since that time, the knowledge in PDE field has …

Pinacidil showed no effect on cyclic AMP or cyclic GMP contents in rabbit aortic strips. These results indicate that pinacidil, like nifedipine and hydralazine, do not seem to directly act on the process from the formation of Ca2+-calmodulin to the actinmyosin interaction in the contracting mechanism of …

Phosphodiesterase 8 (PDE8), a novel target for neurodegenerative diseases.

Phosphodiesterases (PDEs) constitute a superfamily of enzymes comprising 11 distinct families that hydrolyze cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP), thereby precisely regulating the spatiotemporal dynamics of intracellular cyclic nucleotides. Among these PDEs, PDE8 is distinguished by its high affinity and specificity for cAMP and exerts diverse biological effects. The PDE8 family comprises two genes, Pde8a and Pde8b, which are widely distributed throughout the brain and are expressed in both neuronal and glial cells. The widespread distribution of PDE8

HmsC Controls Yersinia pestis Biofilm Formation in Response to Redox Environment.

Yersinia pestis biofilm formation, controlled by intracellular levels of the second messenger molecule cyclic diguanylate (c-di-GMP), is important for blockage-dependent plague transmission from fleas to mammals. HmsCDE is a tripartite signaling system that modulates intracellular c-di-GMP levels to regulate biofilm formation in Y. pestis. Previously, we found that Y. pestis biofilm formation is stimulated in reducing environments in an hmsCDE-dependent manner. However, the mechanism by which HmsCDE senses the redox state remains elusive. Using a dsbA mutant and the addition of Cu2+ to simulat

Influence of thymosin on the level of cyclic nucleotides in rats with experimental varicocele.

Experimental varicocele was induced by partial ligation of the left renal vein in rats, and the effect of thymosin (fraction 5), a hormone secreted by the thymus, on the level of cyclic nucleotides was investigated. The levels of cAMP and cGMP in plasma and testicular tissue were estimated using radioimmunoassay techniques. The results showed that the level of cAMP in bilateral testicular tissues in rats with experimental varicocele decreased considerably, especially in the left testicular tissue as compared with that in the control group (P < 0.05). 4 weeks after thymosin administration (0.5

Identification of small molecules inhibiting diguanylate cyclases to control bacterial biofilm development.

Biofilm formation by pathogenic bacteria is an important virulence factor in the development of numerous chronic infections, thereby causing a severe health burden. Many of these infections cannot be resolved, as bacteria in biofilms are resistant to the host's immune defenses and antibiotic therapy. An urgent need for new strategies to treat biofilm-based infections is critically needed. Cyclic di-GMP (c-di-GMP) is a widely conserved second-messenger signal essential for biofilm formation. The absence of this signalling system in higher eukaryotes makes it an attractive target for the develop
