Effect of Benzo[a]pyrene on Cellular Senescence in MCF7 Breast Cancer Cells.

Studies on the effect of environmental chemicals on cancer has primarily focused on the early stages, while the impact on later stages, particularly cancer progression, has been less explored. Cellular senescence, a response to stress such as DNA damage and oxidative stress, has been reported to play a crucial role in cancer progression. Environmental chemicals are suspected to promote cancer progression, yet it is unclear whether this is mediated through cellular senescence. Benzo[a]pyrene (BP) is known to induce DNA damage, a key trigger of senescence, but whether it directly induces senesce

Insights into pubertal development: a narrative review on the role of epigenetics.

PURPOSE: Puberty is a key phase of growth and development, characterized by psychophysical transformations. It is driven by a combination of genetic, hormonal, and environmental variables. Epigenetic mechanisms, including histone post-translational modifications and chromatin remodeling, microRNAs, and DNA methylation, play important roles in orchestrating the developmental processes. We describe environmental factors that may interact with genetics, and factors influencing puberty onset, focusing in particular on epigenetic mechanisms that can help understand the timing and variations that le

The expression of DNMTs is dramatically decreased in peripheral blood mononuclear cells of male patients with juvenile idiopathic arthritis.

INTRODUCTION: Juvenile idiopathic arthritis (JIA) is an autoimmune rheumatic disease, which affects primarily the joints in children under 16 years old. The etiology of JIA is yet unknown but research has shown that JIA is a multifactorial disease implicating several genes and environmental factors. Environmental factors affect immune cells via epigenetic mechanisms. One of the most important epigenetic mechanisms is DNA methylation catalyzed by DNA methyltransferases (DNMTs) and usually associated with gene silencing. In this study, we analyzed the expression of three DNA methyltransferases n

Polymorphisms of DNA repair genes XRCC1 and XPD and their associations with risk of esophageal squamous cell carcinoma in a Chinese population.

Esophageal squamous cell carcinoma (ESCC), which is prevalent in China, is believed to be induced by environmental carcinogens. Accumulating evidence has shown that individual variation in DNA repair capacity resulting from genetic polymorphism influences risk of environmental carcinogenesis. We therefore investigated the associations between genetic polymorphisms in the DNA repair genes XRCC1 (Arg194Trp and Arg399Gln) and XPD (Asp312Asn and Lys751Gln) and risk of ESCC in an at-risk Chinese population. Genotypes were determined by a PCR-based approach in 433 patients with ESCC and 524 frequenc

A molecular mechanism of nickel (II): reduction of nucleotide excision repair activity by structural and functional disruption of p53.

Nickel is a major carcinogen that is implicated in tumor development through occupational and environmental exposure. Although the exact molecular mechanisms of carcinogenesis by low-level nickel remain unclear, inhibition of DNA repair is frequently considered to be a critical mechanism of carcinogenesis. Here, we investigated whether low concentrations of nickel would affect p53-mediated DNA repair, especially nucleotide excision repair. Our results showed that nickel inhibited the promoter binding activity of p53 on the downstream gene GADD45A, as a result of the disturbance of p53 oligomer

Trichloroethylene-Induced DNA Methylation Changes in Male F344 Rat Liver.

Trichloroethylene (TCE), a common environmental contaminant, causes hepatocellular carcinoma in mice but not in rats. To understand the mechanisms of the species-specific hepatocarcinogenecity of TCE, we examined the methylation status of DNA in the liver of rats exposed to TCE at 0 or 1000 mg/kg b.w. for 5 days using MeDIP-chip, bisulfite sequencing, COBRA, and LC-MS/MS. The related mRNA expression levels were measured by qPCR. Although no global DNA methylation change was detected, 806 genes were hypermethylated and 186 genes were hypomethylated. The genes with hypermethylated DNA were enric

Cementum as a source of DNA in challenging forensic cases.

Each forensic case is characterized by its own uniqueness. Deficient forensic cases require additional sources of human identifiers to assure the identity. We report on two different cases illustrating the role of teeth in answering challenging forensic questions. The first case involves identification of an adipocere male found in a car submersed in water for approximately 2 years. The second scenario, which involves paternity DNA testing of an exhumed body, was performed approximately 2.8 years post-mortem. The difficulty in anticipating the degradation of the DNA is one of the main obstacle

Pro and anti-inflammatory diets as strong epigenetic factors in inflammatory bowel disease.

Inflammatory bowel disease (IBD) is the consequence of a complex interplay between environmental factors, like dietary habits, that alter intestinal microbiota in response to luminal antigens in genetically susceptible individuals. Epigenetics represents an auspicious area for the discovery of how environmental factors influence the pathogenesis of inflammation, prognosis, and response to therapy. Consequently, it relates to gene expression control in response to environmental influences. The increasing number of patients with IBD globally is indicative of the negative effects of a food supply

The frequency distribution of Aspergillus section Nigri from clinical and environmental samples in Iran.

BACKGROUND: Aspergillus section Nigri is widely distributed in decaying plant materials in our environment. Although most of these species are known for food spoilage and some have industrial applications, some of these species, including Aspergillus tubingensis, A. welwitschiae, and A. niger, are etiologic agents of human and animal aspergillosis. Moreover, the frequency of each species in different environments is correlated with meteorological conditions. The present study aimed to investigate the frequency of Aspergillus section Nigri in clinical samples and environmental sources in severa

Isolation of a methanogenic bacterium, Methanosarcina sp. strain FR, for its ability to degrade high concentration of perchloroethylene.

Tetrachloroethylene (PCE) is a toxic compound essentially used as a degreasing and dry-cleaning solvent. A methanogenic and sulfate-reducing consortium that dechlorinates and mineralizes high concentrations of PCE was derived from anaerobically digested sludge obtained from a waste water treatment plant (Bourg-en-Bresse, France). A methanogenic bacterium, strain FR, was isolated from this acclimated consortium. On the basis of morphological and physiological characteristics, strain FR was classified in the genus of Methanosarcina. Phylogeny analysis with the 16S rRNA gene sequence revealed tha

Possible role of oxidative damage in metal-induced carcinogenesis.

This review presents and evaluates evidence relevant to the mechanisms of metal carcinogenicity with special emphasis on the emerging hypothesis of the oxidative nature of metals' effect on DNA. The carcinogenic transition metals are capable of in vivo binding with the cell nucleus and causing promutagenic damage that includes DNA base modifications, inter- and intramolecular crosslinking of DNA and proteins, DNA strand breaks, rearrangements, and depurination. The chemistry of that damage and the resulting mutations observed in vitro and in metal-induced tumors are both characteristic for oxi

New perspectives on DNA methylation modifications in ocular diseases.

The methylation of DNA is a prevalent epigenetic modification that plays a crucial role in the pathological progression of ocular diseases. DNA methylation can regulate gene expression, thereby affecting cell function and signal transduction. Ophthalmic diseases are a kind of complex diseases, and their pathogenesis involves many factors such as genetic, environmental and individual differences. In addition, inflammation, oxidative stress and lipid metabolism, which abnormal DNA methylation is closely related to, are also considered to be major factors in eye diseases. The current understandin

Visible light-induced plasmid DNA damage catalyzed by a CdSe/ZnS-photosensitized nano-TiO2 film.

Visible light-induced photocatalytic degradation of environmental pollutants with improved TiO2 has attracted much attention in pollution control and management. The degradation of nucleic acids is of great significance for biological contaminants such as viruses. In the presentwork, visible light-induced plasmid DNA damage catalyzed by a CdSe/ZnS-photosensitized nano-TiO2 film (QDs-TiO2 film) was investigated by atomic force microscopy (AFM) and agarose gel electrophoresis. Illuminated by visible light, the supercoiled pUC18 DNA could be damaged into nicked-circle and linear conformations by

Molecular mechanisms of cadmium induced mutagenicity.

Cadmium is a human carcinogen of worldwide concern because it accumulates in the environment due to its extremely long half-life. Its compounds are classified as human carcinogens by several regulatory agencies. Cadmium affects cell proliferation, differentiation, apoptosis and other cellular activities and can cause numerous molecular lesions that would be relevant to carcinogenesis. For a long time cadmium has been considered as a non-genotoxic carcinogen, as it is only weakly mutagenic in bacterial and mammalian cell test systems. Recently, we presented evidence that when assayed in a test

[Application of circular dichroism to the study of interactions between small molecular compounds and DNA].

Circular dichroism (CD) is an useful technique for monitoring DNA conformation changes resulting from changes in environmental conditions, such as temperature, ionic strength, and pH, and also for the study of the interaction between DNA and ligands (including small molecules and proteins). CD spectroscopy of DNA arises from the asymmetric backbone sugars and by the helical structures often adopted by nucleic acids. By the interpretation of induced circular dichroism (ICD) of ligand signals resulting from the coupling of electric transition moments of the ligand, DNA bases within the asymmetri

Analysis of DNA damage in sea lamprey (Petromyzon marinus) spermatozoa by UV, hydrogen peroxide, and the toxicant bisazir.

In this study we sought to demonstrate that Comet assay can be applied to sea lamprey sperm DNA fragmentation and used to describe the relationship between sperm DNA damage and sperm fertilizing ability. We show that the assay can be used reliably and accurately, and unlike in the case of mammals, there is no need for additional steps related to improvement of efficacy of lysis and DNA decondensation. This agrees with the presence of histone proteins in lamprey sperm. An increase in DNA fragmentation was noted during short-term storage of milt on ice (0-4 days). We demonstrated genotoxic effec

The genotoxic burden of toxic metals: bridging molecular mechanisms, biomonitoring, and multidisciplinary interventions.

We review the genotoxic effects of trace metal exposure and evaluate an efficient treatment strategy. Toxic metal(loid)s cause significant risks to animal health and ecosystems because of their toxicity and bioaccumulation potential. We then examine the mechanisms of genotoxicity, which include direct DNA damage, oxidative stress, epigenetic alterations, and interference with DNA repair mechanisms. This review highlights the health implications of toxic metal exposure: symptoms like increased cancer risks, reproductive and developmental toxicity, and chronic health disorders. Various assessmen

Persistent interferon signaling and clonal expansion mark early events in DNA methylation damage-induced liver cancer.

N-Nitrosodimethylamine (NDMA), a probable human carcinogen, induces toxic and mutagenic DNA O 6-methylguanine (O 6MeG) adducts that are repaired by O 6-methylguanine-DNA methyltransferase (MGMT). To elucidate how early-life environmental mutagenic exposures promote latent liver tumorigenesis, we performed longitudinal studies in wild-type and MGMT-deficient (Mgmt -/-) mice. Neonatal mice received NDMA intraperitoneally on postnatal days 8 and 15 and were followed for up to 10 months post-exposure. Phenomics, transcriptomics, phosphoproteomics, spatial transcriptomics, and histopathology reveal
