Combination Disease-Modifying Therapy for Neurodegenerative Diseases Using Repurposed Drugs.

We review the positive effects of several existing drugs from different classes, such as chemical chaperones, glucagon-like peptide-1 receptor agonists (GLP-1 RAs), iron chelators, and cluster-Abelson tyrosine kinase inhibitors (c-Abl TKIs), in preclinical disease models and in available published human data following use of these drugs in individuals with common neurodegenerative diseases (NDs), including Parkinson's disease (PD), Alzheimer's disease (AD), amyotrophic lateral sclerosis (ALS), and Huntington's disease (HD). A concept of combinatory neuroprotective therapy using a drug-repurpos

A scalable multimodal graph neural network for drug combination response prediction.

Background Resistance to targeted cancer therapies, such as osimertinib in EGFR-mutant lung cancer, remains a major obstacle to effective treatment. Predicting synergistic drug combinations offers a promising strategy to overcome such resistance; however, the nonlinear and heterogeneous nature of molecular interactions makes this prediction highly challenging. This study introduces the Multimodal Molecular Drug Graph Neural Network (MMDGNN), a unified framework designed to enhance drug synergy prediction through advanced molecular representation learning. Methods MMDGNN integrates molecular fi…

Multiomics combined drugs to explore the potential mechanism and treatment of ankylosing spondylitis.

Ankylosing spondylitis (AS) is an immune-mediated disease with an unknown etiology, posing challenges in effective treatment. This study aims to investigate the underlying mechanisms and explore the potential of traditional Chinese medicine (TCM) as a treatment avenue. Employing a multiomics analysis and leveraging public databases, we scrutinized AS immune cell subpopulations and associated genes. Gene regulatory mechanisms were dissected, and molecular docking was performed to assess the therapeutic efficacy of TCM. Our findings revealed a significant elevation in effector CD8+ memory T (Tem

DeepSynBa: actionable drug combination prediction with complete dose-response profiles.

Many cancer monotherapies demonstrate limited clinical efficacy, making combination therapies a relevant treatment strategy. The extensive number of potential drug combinations and context-specific response profiles complicates the prediction of drug combination responses. Existing computational models are typically trained to predict a single aggregated synergy score, which summarises drug responses across different dosage combinations, such as Bliss or Loewe scores. This oversimplification of the drug-response surface leads to high prediction uncertainty and limited actionability, as these m…

The activity of combinations of currently used antigiardial drugs against Giardia duodenalis.

BACKGROUND: Drug-combination therapies are needed to combat treatment refractory giardiasis. However, data describing the activity of combinations of currently used drugs against Giardia duodenalis are unavailable. METHODS: The in vitro activity of combinations of currently used antigiardial drugs were investigated against G. duodenalis. RESULTS: Combinations of metronidazole with albendazole or quinacrine, and quinacrine with nitazoxanide, behaved additively, whereas combinations of metronidazole with nitazoxanide, albendazole or quinacrine behaved synergistically. Combinations of albendazo

Timely administration of drug combination improves chemoimmunotherapy of an immune-cold tumor.

An immunoactive complex consisting of a polyethyleneimine derivative (2E'), paclitaxel (PTX), and cyclic dinucleotide (CDN) was developed for chemoimmunotherapy of solid tumors. Each component uniquely contributes to stimulating innate immune response to tumors: 2E' carries PTX and CDN while stimulating antigen-presenting cells, PTX induces immunogenic cell death, and CDN activates the STING pathway. A single intratumoral injection of 2E'/PTX/CDN inhibited the growth of MOC1 oral squamous cell carcinoma and KPCY (2838c3) pancreatic tumors, achieving complete tumor regression in 80-100 % of mic

Melatonin combined with antineoplastic drugs or natural products for cancer treatment: An update.

Drug combinations have shown promise in suppressing drug resistance, improving drug efficacy, and reducing side effects in anticancer therapy. Considering that the anticancer activity of melatonin may be due to its antiproliferative, antioxidant, and immunomodulatory activities, the combined administration of this endogenous indoleamine with anticancer drugs has been extensively explored. This review provides an overview of the advances in the last five years in the anticancer activity of melatonin in combination with synthetic drugs and natural products. Papers on this topic were searched in …

Sacubitril Does Not Exert Proarrhythmic Effects in Combination with Different Antiarrhythmic Drugs.

Background: Previous studies suggest a direct effect of sacubitril on cardiac electrophysiology and indicate potential arrhythmic interactions between sacubitril and antiarrhythmic drugs. Therefore, the aim of this study was to explore the electrophysiologic effects of combining sacubitril with the antiarrhythmic drugs d,l-sotalol and mexiletine in isolated hearts. Methods and results: A total of 25 rabbit hearts were perfused using a Langendorff setup. Following baseline data collection, hearts were treated with mexiletine (25 µM, 13 hearts) or d,l-sotalol (100 µM, 12 hearts). Monophasic acti

Efficacy and safety of intranasal corticosteroids combined with mucoactive drugs for chronic rhinosinusitis.

BACKGROUND: Chronic rhinosinusitis (CRS) leads to a burden in life and economy. Better therapies need to be explored. OBJECTIVE: This stage I study aims to explore the efficacy and safety of intranasal corticosteroids combined with mucoactive drugs for CRS. METHODS: This randomized-controlled, double-blind study was conducted at 13 hospitals from 2021.01.31 to 2023.01.17. CRS patients were randomly assigned into the eucalyptol-limonene-pinene enteric capsules (ELP) group or placebo group, both using budesonide nasal spray (two doses of 64 μg into each nostril twice a day), and followed up at

Identifying Drug Combination Strategies for ZMYM2: FGFR1 Fusion Positive Leukemia.

Myeloid/lymphoid neoplasms with eosinophilia and tyrosine kinase gene fusions (MLN-TK) are a class of fusion protein-driven, poor prognosis leukemias. Leukemias harboring FGFR1 fusions have previously been referred to as 8p11.2 myeloproliferative syndrome (EMS) or stem cell leukemia/lymphoma (SCLL) and are currently referred to as Myeloid/lymphoid neoplasms with FGFR1 rearrangement based on the most recent WHO classification system. To identify new therapeutic options for MLN-TK patients, we evaluated clinical and ex vivo drug response data from a ZMYM2:FGFR1-positive patient who was successfu

Combination of ganciclovir and trifluridine prevents drug-resistance emergence in HSV-1.

Ocular herpes simplex virus 1 (HSV-1) infections can lead to visual impairment. Long-term acyclovir (ACV) prophylaxis reduces the frequency of recurrences but is associated with drug resistance. Novel therapies are needed to treat drug-resistant HSV-1 infections. Here, we describe the effects of trifluridine (TFT) in combination with ACV or ganciclovir (GCV) on HSV-1 replication and drug-resistance emergence. Wild-type HSV-1 was grown under increasing doses of one antiviral (ACV, GCV, or TFT) or combinations thereof (ACV + TFT or GCV + TFT). Virus cultures were analyzed by Sanger sequencing an
