Enhanced presynaptic beta 2-adrenoceptor-mediated facilitation of the pressor responses in the prehypertensive SHR.

The effects of beta-adrenoceptor agonists and antagonists on pressor responses of the isolated perfused mesenteric arteries to periarterial nerve stimulation (PNS) in the prehypertensive 4-week-old spontaneously hypertensive rat (SHR) and the age-matched Wistar Kyoto rats (WKY) were examined. The systolic arterial blood pressure (SBP) of SHR and WKY were not significantly different at this young age. The pressor responses of the mesenteric arteries to PNS at various stimulating frequencies, however, were significantly greater in SHR than WKY. Cocaine, isoproterenol (a nonselective beta-adrenoc

Role of spinal glutamatergic transmission in the ascending limb of the micturition reflex pathway in the rat.

This study was undertaken to evaluate the role of glutamate receptors at spinal synapses on the ascending limb of the micturition reflex. In urethane-anesthetized female rats, a tungsten electrode was inserted stereotaxically into the dorsal part of the rostral pons to record field potentials which were evoked by electrical stimulation of the pelvic nerve (PLN) (1-15 V, 0.05 ms pulse duration at 100-300 Hz, 5-30 ms train duration). The effects of glutamate receptor antagonists administered intrathecally (i.t.) on the PLN-evoked field potentials in the dorsal part of the rostral brainstem were

Reorganization of sympathetic preganglionic connections in cat bladder ganglia following parasympathetic denervation.

1. Experiments were undertaken to examine the mechanisms involved in the reorganization of sympathetic efferent pathways to the urinary bladder of the cat following chronic unilateral, parasympathetic preganglionic denervation of the bladder. 2. Electrical stimulation (10-30 Hz) of the hypogastric nerve in cats with an intact bladder innervation or on the normally innervated side of the bladder in unilaterally denervated preparations elicited low-amplitude (10-25 cmH2O) transient (10-30 s) bladder contractions and non-synaptic axonal volleys on bladder postganglionic nerves. However, after chr

Differences in Hebbian stimulation effects between biceps and triceps brachii in humans.

Animal and human studies indicate that monosynaptic corticospinal connections are more prevalent in biceps than triceps brachii motoneurons. Based on this evidence, we hypothesized that Hebbian stimulation, which targets corticospinal-motoneuronal connections, would enhance corticospinal excitability more in the biceps than the triceps brachii. To test this hypothesis, we assessed motor-evoked potential (MEP) size using transcranial magnetic stimulation (TMS) at resting motor threshold (MEP-RMT) and maximum stimulator output (MEP-100%) immediately and up to 30 min poststimulation. During Hebbi

Biomineralized Shewanella Exploits Tumor Lactate Metabolism via Biological Nanowires for Radiotherapy-Synergized Bioelectric Ablation and Systemic Immunity.

Bioelectronic therapy is an emerging strategy that ablates tumor cells via electrical signals. However, achieving precisely controllable bioelectronic ablation remains challenging. Here, we report a living bioelectronic platform, termed PSDT, for radiotherapy (RT)-activated tumor intervention and immune activation. PSDT consists of a Pd/Pt-modified Shewanella core (SDT) encapsulated within a ROS-responsive polymeric shell (PEI-Se-Se-PEG). The ROS-responsive shell keeps the platform inert systemically and confines its activity to irradiated tumors, achieving spatiotemporal control. Upon RT acti

A high-density microelectrode array integrated microfluidic platform for quantitative investigation of functional connectivity in interconnected human dopaminergic neurospheroids.

The quantitative investigation of functional connectivity across spatially organized neuronal networks remains a major challenge in human three-dimensional (3D)in vitromodels. Although neurospheroids, brain organoids, and microfluidic systems enable controlled structural organization and guided neurite growth, their integration with high-resolution electrophysiological interfaces remains limited, often requiring post hoc adaptations that compromise recording stability and long-term reproducibility. Here, we present a polydimethylsiloxane (PDMS) microfluidic platform natively engineered for sea

Exercise-driven neuromuscular junction plasticity: integrating motor-neuron adaptation, synaptic remodeling, and glial support.

The neuromuscular junction (NMJ) is a dynamic synapse whose structure and function change with activity. This review synthesizes evidence from molecular studies, in vivo imaging, computational models, and human exercise research to explain how exercise remodels the presynaptic motor neuron terminal, postsynaptic muscle endplate, perisynaptic Schwann cells (PSCs), and extracellular matrix (ECM). Three conclusions emerge. First, endurance, resistance, high-intensity interval training (HIIT), and neuromuscular electrical stimulation (NMES) produce partly distinct NMJ signatures: endurance trainin

Multisession epidural direct current stimulation of the auditory cortex mitigates age-related transcriptomic dysregulation in Wistar rats.

Age-related hearing loss (ARHL) disrupts ascending auditory inputs, impairing auditory signal transmission, triggering cortical hyperexcitability, and increasing the risk of age-related cognitive decline. In early aging, multisession epidural direct current stimulation (DCS) of the auditory cortex (AC) preserves auditory thresholds and prevents cortical hyperexcitability in Wistar rats. Here, we hypothesized that multisession DCS could halt transcriptional dysregulation in the AC at the earliest stages of aging. We have characterized age-related transcriptional changes in the AC to assess DCS-

The Neural Bases of Tinnitus: Lessons from Deafness and Cochlear Implants.

Subjective tinnitus is the conscious perception of sound in the absence of any acoustic source. The literature suggests various tinnitus mechanisms, most of which invoke changes in spontaneous firing rates of central auditory neurons resulting from modification of neural gain. Here, we present an alternative model based on evidence that tinnitus is: (1) rare in people who are congenitally deaf, (2) common in people with acquired deafness, and (3) potentially suppressed by active cochlear implants used for hearing restoration. We propose that tinnitus can only develop after fast auditory fiber

Posttetanic enhancement of striato-pallidal synaptic transmission.

The striato (Str)-globus pallidus external segment (GPe) projection plays major roles in the control of neuronal activity in the basal ganglia under both normal and pathological conditions. The present study used rat brain slice preparations to characterize the enhancement of Str-GPe synapses observed after repetitive conditioning stimuli (CS) of Str with the whole cell patch-clamp recording technique. The results show that 1) the Str-GPe synapses have a posttetanic enhancement (PTE) mechanism, which is considered to be a combination of an augmentation and a posttetanic potentiation; 2) the de

Disynaptic and polysynaptic statocyst pathways to an identified set of premotor nonspiking interneurons in the crayfish brain.

Nonspiking giant interneurons (NGIs) in the crayfish brain occupy a key position in the neuronal circuit for eyestalk motor control and, hence, play a crucial role in the central compensation process following unilateral deprivation of the statocyst that functions as an equilibrium sensory system. The synaptic organization of their input pathways, however, remains unknown. In the present study we identified 11 local interneurons that were polysynaptically connected to NGIs by making simultaneous intracellular recordings. We also identified three other local interneurons that connected to NGIs

Prefrontal control of the amygdala.

Accumulating evidence indicates that phobic and posttraumatic anxiety disorders likely result from a failure to extinguish fear memories. Extinction normally depends on a new learning that competes with the original fear memory and is driven by medial prefrontal cortex (mPFC) projections to the amygdala. Although mPFC stimulation was reported to inhibit the central medial (CEm) amygdala neurons that mediate fear responses via their brainstem and hypothalamic projections, it is unclear how this inhibition is generated. Because the mPFC has very sparse projections to CEm output neurons, the mPFC

Dendritic calcium accumulation regulates wind sensitivity via short-term depression at cercal sensory-to-giant interneuron synapses in the cricket.

An in vivo Ca2+ imaging technique was applied to examine the cellular mechanisms for attenuation of wind sensitivity in the identified primary sensory interneurons in the cricket cercal system. Simultaneous measurement of the cytosolic Ca2+ concentration ([Ca2+]i) and membrane potential of a wind-sensitive giant interneuron (GI) revealed that successive air puffs caused the Ca2+ accumulation in dendrites and diminished the wind-evoked bursting response in the GI. After tetanic stimulation of the presynaptic cercal sensory nerves induced a larger Ca2+ accumulation in the GI, the wind-evoked bur

Tetanus-induced asynchronous GABA release in cultured hippocampal neurons.

Asynchronous GABA release was studied in cultured hippocampal neurons using paired whole-cell recordings. Tetanization of the presynaptic GABAergic neuron was accompanied by a train of IPSCs which showed tetanic depression. Asynchronous IPSCs (asIPSCs) also developed during the train and continued for 1.85+/-0.3 s after the stimulation. The threshold frequency for evoking asIPSCs was 10 Hz, while maximal asynchronous activity was achieved at 40 Hz. Perfusion with EGTA-AM blocked asIPSCs. The elevation of [Ca(2+)](i) that accompanies presynaptic action potential firing triggers asynchronous rel

Facilitated NMDA receptor-mediated synaptic plasticity in the hippocampal CA1 area of dystrophin-deficient mice.

The contribution of the cytoskeletal membrane-associated protein dystrophin in glutamatergic transmission and related plasticity was investigated in the hippocampal CA1 area of wild-type and dystrophin-deficient (mdx) mice, using extracellular recordings in the ex vivo slice preparation. Presynaptic fiber volleys and field excitatory postsynaptic potentials (fEPSPs) mediated through N-methyl-D-Aspartate receptors (NMDAr) or non-NMDAr were compared in both strains. Comparable synaptic responses were observed in wild-type and mdx mice, suggesting that basal glutamatergic transmission is not alte

Incidence of Malignancy in Appendicectomy Specimens.

Understanding how different neuronal types connect and communicate is critical to interpreting brain function and behavior. However, it has remained a formidable challenge to decipher the genetic underpinnings that dictate the specific connections formed between neuronal types. To address this, we propose a novel bilinear modeling approach that leverages the architecture similar to that of recommendation systems. Our model transforms the gene expressions of presynaptic and postsynaptic neuronal types, obtained from single-cell transcriptomics, into a covariance matrix. The objective is to cons

Immunology of stiff person syndrome and other GAD-associated neurological disorders.

Antibodies against glutamic acid decarboxylase (GAD), the rate-limiting enzyme for the synthesis of GABA, are associated with an array of distinct, mostly autoimmune, neurological conditions. In all associated syndromes, namely stiff person syndrome, cerebellar ataxia, epilepsy, limbic encephalitis or abnormal eye movements, anti-GAD antibodies are detected at high titers and play a fundamental role in diagnosis, but do not correlate with disease severity, diversity of symptomatology or response to therapies. Despite considerable efforts, including in vitro (enzymatic assays) and in vivo (anim

Endocannabinoids in nervous system health and disease: the big picture in a nutshell.

The psychoactive component of the cannabis resin and flowers, delta9-tetrahydrocannabinol (THC), was first isolated in 1964, and at least 70 other structurally related 'phytocannabinoid' compounds have since been identified. The serendipitous identification of a G-protein-coupled cannabinoid receptor at which THC is active in the brain heralded an explosion in cannabinoid research. Elements of the endocannabinoid system (ECS) comprise the cannabinoid receptors, a family of nascent lipid ligands, the 'endocannabinoids' and the machinery for their biosynthesis and metabolism. The function of the

Galanin modulates neuronal and synaptic properties in the rat supraoptic nucleus in a use and state dependent manner.

The magnocellular neurons of the hypothalamic supraoptic nucleus (SON) synthesize and secrete oxytocin (OXT) and vasopressin (AVP) from their dendrites. These peptides, and several other neurotransmitters, have been shown to modulate afferent glutamatergic neurotransmission in the SON. The neuropeptide, galanin (GAL) is also localized in SON magnocellular neurons and in afferent fibers in the nucleus. We show that GAL dose-dependently reduces evoked excitatory postsynaptic currents (eEPSCs), alters paired pulse ratio and decreases mEPSC frequency, but not amplitude or decay kinetics in both OX

Capacitance measurements at the calyx of Held in the medial nucleus of the trapezoid body.

We have recently applied Lindau-Neher's capacitance measurement technique to study vesicle trafficking at the calyx-type synapse in the rat medial nucleus of the trapezoid body (MNTB) in slice conditions. This application made the MNTB synapse an excellent model for the study of exocytosis and endocytosis at conventional active zones. However, the application was only made at calyces that are presumably equivalent to a single-compartment circuit because their passive current transients decayed mono-exponentially. Here, we determined whether the application could be extended to majority of caly
