Formulation of Aloe vera based curcumin topical gel and its in-vitro evaluation.

Curcumin is a polyphenolic compound obtained from the rhizome of plant. Curcuma longa possesses antioxidant, anti-inflammatory and wound-healing properties. The current study was designed to formulate an Aloe vera-based curcumin topical gel. To enhance curcumin's solubility, it was first complexed with β-cyclodextrin, given its hydrophobic nature. While Carbopol, carboxy methyl cellulose and guar gum were used in various concentrations as gelling agents for preparation of the formulations. The effect of propylene glycol as a permeation enhancer was also observed. The prepared formulations were

Evaluation of novel nystatin nanoemulsion for skin candidosis infections.

One of the most common fungal skin infections is candidosis. Topical application of drugs at the pathological sites offers potential advantage of direct drug delivery to the site of action. The main aim of this work was to evaluate an optimal nystatin nanoemulsion for topical application avoiding undesirable side effects as systemic absorption and toxicity. Surface morphology and droplet size distribution of nystatin nanoemulsion was determined by transmission electronic microscopy and dynamic light scattering. Vertical diffusion Franz-type cells and high-performance liquid chromatography were

Increased skin barrier disruption by sodium lauryl sulfate in mice expressing a constitutively active STAT6 in T cells.

Atopic dermatitis (AD) is a pruritic, chronic inflammatory skin disease that affects 10-20% of children and 1-3% of adults worldwide. Recent studies have indicated that the ability of Th2 cytokines, such as interleukin-4 (IL-4) to regulate skin barrier function may be a predisposing factor for AD development. The present studies examined the ability of increased Th2 activity to affect cutaneous barrier function in vivo and epidermal thickening. Mice that express a constitutively active Signal Transducer and Activator of Transcription 6 (STAT6VT) have increased Th2 cells and a predisposition to

Mechanisms of esophageal protection, gastroprotection and ulcer healing by melatonin. implications for the therapeutic use of melatonin in gastroesophageal reflux disease (GERD) and peptic ulcer disease.

Melatonin is a potent reactive oxygen metabolite scavenger and antioxidant that has been shown to influence many physiological functions of the gastrointestinal (GI) tract including secretion, motility, digestion and absorption of nutrients. The role of melatonin in gastroduodenal defense and ulcer healing has been the subject of recent investigations. Melatonin produced in the GI mucosa plays an important role in protection against noxious agents thus contributing to the maintenance of GI integrity and to esophageal protection, gastroprotection and ulcer healing. This review was designed to s

cR10-modified liposomes for efficient topical delivery of lenvatinib to the posterior segment: Enhanced suppression of choroidal neovascularization.

Choroidal neovascularization (CNV) is a key pathological process underlying multiple retinal diseases and remains difficult to treat due to physiological barriers that limit drug delivery to the posterior segment of the eye. Current anti-vascular endothelial growth factor (VEGF) therapies rely on intravitreal injection, which is invasive and associated with potential complications. lenvatinib, a multi-targeted tyrosine kinase inhibitor, exhibits potent anti-angiogenic activity; however, its poor aqueous solubility and limited ocular bioavailability restrict its ophthalmic application. In this

Design of binder proteins specific to IL-13.

Interleukin-13 (IL-13) drives inflammatory responses in type 2 inflammatory disorders by binding to the IL-4Rα/IL-13Rα1 receptor complex. Anti-IL-13 monoclonal antibodies such as tralokinumab and lebrikizumab showed substantial clinical efficacy in autoimmunology diseases such as atopic dermatitis and asthma. However, their large size has limited the possible application in topical immunotherapy treatment. Here, we employed structure-based computational design to develop compact minibinder proteins targeting IL-13, based on the crystal structure of the IL-13-tralokinumab complex. Five designed

Development and evaluation of microemulsion foam formulation for topical delivery of betamethasone dipropionate.

Betamethasone dipropionate (BDP) is widely recognized as a therapeutic agent for psoriasis. However, the current topical formulations exhibit certain limitations, including significant systemic absorption and substantial residue on the skin surface post-application. Thus, this study aimed to develop a novel microemulsion (ME) foam system, which can enhance the targeting and bioavailability of BDP at the diseased skin. Through the pseudo-ternary diagrams, it was determined that when the ratio of surfactant to cosurfactant was 6:1, the ME region reached its maximum. The optimized formulation was

Pharmacological Management of Oral and Esophageal Candidiasis: A Clinical Pharmacotherapy Perspective.

Candida spp. are common components of normal microbiota in the oral cavity. However, Candida albicans can be a primary cause of superficial infections in the oral cavity and esophagus, especially in immunocompromised individuals. While these infections are rarely life-threatening, they can significantly impair quality of life and, in severe cases, progress to hematogenous dissemination. Oral candidiasis often exhibits as pseudomembranous, erythematous (atrophic), chronic hyperplastic, denture stomatitis, or angular cheilitis. Esophageal candidiasis is typically diagnosed by upper endoscopy, wh
