The effect of an intrauterine thread on tissue changes in the endometrium of the pseudopregnant rat.

1. Changes in the fresh weight of uterine horns bearing unilateral intrauterine threads have been measured during early pseudopregnancy in the rat.2. Such a thread, which acts as an intrauterine contraceptive device, causes an increase in weight in the horn which carries it, compared to the untreated contralateral horn, on days 2, 4, 5 and 7 of pseudopregnancy. This increase is observed when the device has been present either for 25 or for 45 days.3. The fresh and dry weights of samples of endometrial tissue together with the concentration of chloride and potassium in these samples have also b

Ionic environment of neurones and glial cells in the brain of an amphibian.

1. Experiments were performed to determine the relative contribution of the blood plasma and of the cerebrospinal fluid (c.s.f.) to the ionic environment of neurones and glial cells within the brain of the amphibian Necturus maculosus.2. The concentrations in the blood plasma of untreated control animals were 99 +/- 2 mM for Na(+) and 2.0 +/- 0.1 mM for K(+). In the c.s.f. the corresponding values were 112 +/- 2 mM-Na(+) and 1.9 +/- 0.1 mM-K(+).3. By keeping animals in K(+)-rich water it was possible to raise chronically the concentrations of K(+) in the blood plasma up to almost 5 times the n

Extracellular pH Mapping as Therapeutic Readout of Drug Delivery in Glioblastoma.

An acidic extracellular space is a hallmark of the tumor microenvironment. Acidosis has been postulated to promote the aggressive and invasive characteristics of tumors and also inhibit the therapeutic response, particularly in the context of novel immunotherapies. Therefore, methods to quantitatively measure the extracellular pH (pHe) are needed. Here we describe a magnetic resonance spectroscopic imaging (MRSI) technique termed Biosensor Imaging of Redundant Deviation in Shifts (BIRDS), which uses the pH-dependent chemical shifts of nonexchangeable protons of lanthanide-based contrast agents…

Ca2+ influx via the Na+/Ca2+ exchanger is enhanced in malignant hyperthermia skeletal muscle.

Malignant hyperthermia (MH) is potentially fatal pharmacogenetic disorder of skeletal muscle caused by intracellular Ca(2+) dysregulation. NCX is a bidirectional transporter that effluxes (forward mode) or influxes (reverse mode) Ca(2+) depending on cellular activity. Resting intracellular calcium ([Ca(2+)]r) and sodium ([Na(+)]r) concentrations are elevated in MH susceptible (MHS) swine and murine muscles compared with their normal (MHN) counterparts, although the contribution of NCX is unclear. Lowering [Na(+)]e elevates [Ca(2+)]r in both MHN and MHS swine muscle fibers and it is prevented b

Growth-limiting intracellular metabolites in yeast growing under diverse nutrient limitations.

Microbes tailor their growth rate to nutrient availability. Here, we measured, using liquid chromatography-mass spectrometry, >100 intracellular metabolites in steady-state cultures of Saccharomyces cerevisiae growing at five different rates and in each of five different limiting nutrients. In contrast to gene transcripts, where approximately 25% correlated with growth rate irrespective of the nature of the limiting nutrient, metabolite concentrations were highly sensitive to the limiting nutrient's identity. Nitrogen (ammonium) and carbon (glucose) limitation were characterized by low intrace

Long-term activation of the glutamatergic system associated with N-methyl-D-aspartate receptors after postischemic hypothermia in gerbils.

OBJECTIVE: The objective of this study was to investigate whether hypothermia would suppress secondary damage in the chronic postischemic stage, in terms of glutamate excitotoxicity. METHODS: Gerbils underwent 5 minutes of ischemia via bilateral common carotid artery occlusion. Seven groups were studied, as follows: 1) ischemia without treatment group; 2) intraischemic hypothermia group; 3) postischemic hypothermia group (32 degrees C for 4 h); 4) MK-801 treatment group (2 mg/kg, every other day for 1 mo); 5) postischemic hypothermia with MK-801 treatment for 1 week group (2 mg/kg, every othe

Activated Platelet-Released Heat Shock Protein 90α Triggers Autophagy-Dependent Neutrophil Extracellular Trap Formation and Amplifies Sepsis.

Platelets are crucial to the development of thrombosis and coagulation abnormalities in sepsis, but the mechanisms by which they contribute … ytes. When activated, platelets translocate HSP90α to their plasma membrane and release it into the extracellular space in both free and exosome-associated forms. Extracellular HSP90α acts as a damage-associated molecular pattern that binds to toll-like receptor 4 (TLR4) on neutrophils. This binding activates a downstream MyD88-Beclin 1 signaling pathway, triggering …

Brain injury after hypoxia-ischemia in newborn rats: relationship to extracellular levels of excitatory amino acids and cysteine.

Changes of EAAs and cysteine were similar during HI in the infarcted, undamaged and border-zone regions. During reperfusion the concentrations of glutamate, aspartate and cyste …

Cancer-Derived Extracellular Vesicles: Their Role in Sarcoma.

Soft tissue sarcomas (STS) are rare malignancies with limited responses to anticancer therapy. Extracellular vesicles (EVs) are a heterogeneous group of bi-lipid layer sacs secreted by cells into extracellular space. Investigations of tumor-derived EVs have revealed their functional capabilities, including cell-to-cell communication and their impact on tumorigenesis, progression, and metastasis; however information on the roles of EVs in sarcoma is currently limited. In this review we investigate the role of various EV cargos in sarcoma and the mechanisms by which those cargos can affect the r…

Selective retention and degradation of molecules with a single mutant alpha1(I) chain in the Brtl IV mouse model of OI.

We investigated the secretion, matrix incorporation and interactions of molecules with one and two mutant alpha1(I) collagen chains in the Brtl IV murine model for Osteogenesis Imperfecta, carrying a Gly-349 to Cys substitution in one col1a1 allele. We detected a significant deviation from the expected 25 and 50% content of the molecules with no (37-46%) and one (26-40%) mutant chains in skin and bone as well as in fibroblast and osteoblast cell culture media. Steady-state labeling with (35)S-Cys demonstrated incomplete secretion of the mutant collagen in cell culture, particularly molecules c

N-Glycosylation is essential for the secretion of extracellular superoxide dismutase.

Extracellular superoxide dismutase (EC-SOD or SOD3) protects against various oxidative stress-related diseases by scavenging reactive superoxides in the extracellular space. It is the only SOD isozyme that is secreted and glycosylated (at asparagine 89). However, the physiological roles of its glycosylation are poorly understood. In this study, we found that the glycosylation site on EC-SOD is well conserved and that a glycosylation-deficient EC-SOD mutant retains its enzymatic activity, but is not secreted. This impairment in secretion may, in part, be due to the ability of the mutants to for…

Bilateral atrial appendage thrombosis in light chain cardiac amyloidosis: a case report.

Light chain cardiac amyloidosis (AL-CA) is a severe and progressively infiltrative disease caused by the deposition of misfolded monoclonal immunoglobulin light chains in the myocardial extracellular space. It is associated with poor prognosis and typically presents as diastolic heart failure, arrhythmias, and atrial appendage thrombosis. We report a case of light chain cardiac amyloidosis with bilateral atrial appendage thrombosis. The patient died from refractory heart failure four months after diagnosis.
