Fosfomycin enhances phagocyte-mediated killing of Staphylococcus aureus by extracellular traps and reactive oxygen species.

The successful treatment of bacterial infections is the achievement of a synergy between the host's immune defences and antibiotics. Here, we examined whether fosfomycin (FOM) could improve the bactericidal effect of phagocytes, and investigated the potential mechanisms. FOM enhanced the phagocytosis and extra- or intracellular killing of S. aureus by phagocytes. And FOM enhanced the extracellular killing of S. aureus in macrophage (MФ) and in neutrophils mediated by extracellular traps (ETs). ET production was related to NADPH oxidase-dependent reactive oxygen species (ROS). Additionally, FOM…

Autophagy mediates the delivery of thrombogenic tissue factor to neutrophil extracellular traps in human sepsis.

BACKGROUND: Sepsis is associated with systemic inflammatory responses and induction of coagulation system. Neutrophil extracellular traps (NETs) constitute an antimicrobial mechanism, recently implicated in thrombosis via platelet entrapment and aggregation. METHODOLOGY/PRINCIPAL FINDINGS: In this study, we demonstrate for the first time the localization of thrombogenic tissue factor (TF) in NETs released by neutrophils derived from patients with gram-negative sepsis and normal neutrophils treated with either serum from septic patients or inflammatory mediators involved in the pathogenesis of…

DNase expression allows the pathogen group A Streptococcus to escape killing in neutrophil extracellular traps.

The innate immune response plays a crucial role in satisfactory host resolution of bacterial infection. In response to chemotactic signals, neutrophils are early responding cells that migrate in large numbers to sites of infection. The recent discovery of secreted neutrophil extracellular traps (NETs) composed of DNA and histones opened a novel dimension in our understanding of the microbial killing capacity of these specialized leukocytes. M1 serotype strains of the pathogen Group A Streptococcus (GAS) are associated with invasive infections including necrotizing fasciitis (NF) and express a …

CXCL16-driven neutrophil extracellular traps promote osteoporosis in APP/PS1 mice by disrupting bone remodeling.

Patients with Alzheimer's disease (AD) often exhibit abnormal bone loss alongside cognitive impairment. However, the mechanisms linking these two conditions remain insufficiently understood. We first employed amyloid precursor protein/presenilin 1 (APP/PS1) mice as the experimental model and demonstrated that they exhibit significant bone loss concurrent with cognitive impairment. Subsequently, we analyzed public datasets for AD and osteoporosis (OP) and identified CXC chemokine ligand 16 (CXCL16) as a shared hub gene between the two diseases. Transcriptomic sequencing further showed increased

Macrophage extracellular traps amplify retinal endothelial anoikis via the S1P-S1PR axis in diabetic retinopathy.

OBJECTIVE: To investigate the pathogenic role of macrophage extracellular traps (METs) in proliferative diabetic retinopathy (PDR), focusing on endothelial dysfunction and inflammatory activation. METHODS: Transcriptomic data from PDR patients were first analyzed to identify METs-associated pathways. Anoikis-related subgroups were defined using the median gene set variation analysis (GSVA) enrichment score of the anoikis gene set, and enriched pathways were selected for subsequent validation. In vitro, diabetic retinopathy models were established using high-glucose and METs stimulation. Human …

Neutrophil extracellular traps in colorectal cancer: a paradoxical nexus of tumor defense and disease progression.

Neutrophil extracellular traps (NETs) are extracellular networks of decondensed chromatin and granular proteins generated by neutrophils in response to activating stimuli. Growing evidence indicates that NETs participate in colorectal cancer (CRC) development through their interactions with chronic inflammation, immune regulation, coagulation, and tumor progression. However, their biological effects vary across disease settings, and the regulatory mechanisms specific to CRC have not been fully clarified. This review examines the roles of NETs at different stages of CRC, including the transitio…

Neutrophils and neutrophil extracellular traps in diabetes mellitus and its complications: Mechanisms and therapeutic implications.

Diabetes mellitus is a chronic metabolic disease that has become a global health concern, leading to complications such as retinopathy, nephropathy, vasculopathy, and delayed wound healing. Neutrophils, as essential immune cells, protect the host by releasing neutrophil extracellular traps (NETs)-web-like structures composed of DNA and antimicrobial proteins that trap and kill pathogens. However, in diabetes, persistent hyperglycemia and inflammation induce excessive NET formation, resulting in endothelial damage, thrombosis, and tissue injury, which aggravate disease progression. Recent studi…

Neutrophils and Neutrophil Extracellular Traps in Hepatic Ischemia-Reperfusion Injury: Molecular Mechanisms and Therapeutic Strategies.

Hepatic ischemia-reperfusion injury (HIRI) is an unavoidable clinical challenge in liver transplantation, major hepatectomy and trauma resuscitation, with no approved specific therapeutics to date. As core innate immune effector cells, neutrophils are the central driver of the sterile inflammatory cascade in HIRI. Centered on the Mac-1-Syk core regulatory axis, this review systematically elaborates the neutrophil recruitment cascade and intracellular signaling network in HIRI, focuses on the sequential formation pathways of neutrophil extracellular traps (NETs) and their multi-dimensional inju…

Exploring Neutrophil Extracellular Traps in Canine Soft Tissue Sarcomas: A Preliminary Study With Translational Insights.

BACKGROUND: Neutrophil extracellular traps (NETs) have gained attention in oncology due to their role in inflammation-driven tumour progression. While their involvement has been documented in various human malignancies, including select sarcoma subtypes, data on soft tissue sarcomas (STS) remain sparse, particularly in veterinary species. OBJECTIVES: This study aimed to investigate the presence and potential significance of NET formation in spontaneously occurring canine STS, with an additional focus on possible translational relevance to human oncology. METHODS: This study examined 30 canin…

Neutrophil extracellular traps promote neuronal ferroptosis through STING-mediated AMPK dysregulation after traumatic brain injury.

Neuronal ferroptosis exacerbates neurological dysfunction after traumatic brain injury (TBI), yet the upstream signals driving this process remain poorly defined. Here, we identify neutrophil extracellular traps (NETs) as critical mediators of post-TBI ferroptotic injury. In patients, NET accumulation closely accompanied markers of neuronal ferroptosis and correlated with neurological severity. In a mouse TBI model, genetic ablation of peptidylarginine deiminase 4 (PAD4) or enzymatic degradation of NET-derived DNA markedly attenuated neuronal lipid peroxidation, suppressed ferroptosis-associat…

Neutrophil extracellular traps drive cholangiocyte injury via ZNF709-TP53-MDM2 pathway in primary biliary cholangitis.

BACKGROUND: Primary biliary cholangitis (PBC) is a chronic autoimmune liver disease in which many patients respond inadequately to ursodeoxycholic acid (UDCA) and may progress to liver failure. Although research on PBC has primarily centered on lymphocyte-mediated mechanisms, increasing evidence implicates neutrophils in autoimmunity, yet their role in PBC remains unclear. METHODS: Cytokine screening of patient sera collected from our medical center was performed to identify NETs-inducing factors. Transcriptomic profiling was conducted to determine the molecular effects of NETs on cholangiocy

Comprehensive analysis of neutrophil extracellular traps-associated inflammatory genes for patients with idiopathic pulmonary fibrosis.

Neutrophil extracellular traps (NETs) facilitate inflammation and epithelial-mesenchymal transition (EMT), promoting the progression of pulmonary fibrosis. Various machine learning methods were used to screen for prognostic genes. Based on prognostic genes, a risk model was constructed to assess their ability for prognosis prediction of idiopathic pulmonary fibrosis (IPF). Mendelian Randomization (MR) analysis evaluated causal associations between IPF and prognostic genes, while GSE122960 examined cell-type-specific expression. A bleomycin-induced pulmonary fibrosis mouse was established, coll…

Neutrophil extracellular traps induced by a monoclonal anti-phosphatidylserine/prothrombin antibody activate platelets in vitro.

Antiphospholipid syndrome (APS) is an autoimmune thrombotic disorder characterized by the presence of antiphospholipid antibodies, including anti-phosphatidylserine/prothrombin antibodies (aPS/PT). While neutrophil extracellular traps (NETs) are implicated in the pathogenesis of APS, the role of aPS/PT in NET induction and its contribution to thrombosis remain unclear. This study aimed to clarify the effects of NETs induced by a monoclonal aPS/PT antibody on platelet activation and their potential contribution to thrombo-inflammatory responses. NETs were induced by stimulating peripheral blood…

NADPH oxidase 2 (NOX2)-independent inducers of neutrophil extracellular traps promote resolution of chronic inflammation.

Reactive oxygen species (ROS) generated by NADPH oxidase 2 (NOX2) are essential for antimicrobial defense but also for the resolution of inflammation. NOX2 deficiency, as observed in chronic granulomatous disease (CGD), predisposes to persistent sterile inflammation. Current therapeutic strategies largely rely on nonspecific immunosuppression or require residual NOX2 activity, while systemic ROS-inducing therapies are limited by toxicity. Here, we present a NOX2-independent approach to restore inflammation-resolving ROS signaling using N-alkylaminoferrocene-based prodrugs (pro-NAAFs), which am

Neutrophil extracellular traps capture the human pathogen, Candida albicans, in blood and delay hyphal transformation.

In tissues, neutrophils neutralize Candida albicans through phagocytosis and delay C. albicans hyphae growth by deploying neutrophil extracellular traps (NETs). However, in the bloodstream, the dynamic interactions between NETs and C. albicans are far less understood. Here, we employ a microfluidic assay and measure a significant increase in intact NETs in blood within 3 h after adding C. albicans yeast or hyphae. We show that C. albicans yeast can be captured efficiently on NETs, thereby delaying the transition to hyphal growth. We measure higher amounts of intact NETs in blood samples from i…
